American Foulbrood is the most destructive bacterial infection of the honeybee (Apis mellifera) and is caused by the Gram-positive, spore forming bacterium Paenibacillus larvae. Current treatment methods rely on antibiotics, but antibiotics treatments are experiencing a reduction in efficacy due to the recent rise in antibiotic resistant strains of P. larvae. This has been a major catalyst for exploration of alternative treatment methods. Phage therapy is an alternative treatment method that uses viruses that exclusively infect bacteria, known as bacteriophages (phages), to combat bacterial infections. Novel methods are needed to traverse the blood-brain barrier (BBB) and deliver drugs to specific targets in the brain. To this end, MS2 bacteriophage was explored as a multifunctional transport and targeting vector. The MS2 capsid exterior was modified with two different targeting moieties for delivery across the BBB and targeting specific regions of interest in the brain. Successful modification of MS2 capsids with a brain targeting peptide and NMDAR2D-targeting antibody was confirmed by immunoblotting and fluorescence detection. To measure transport efficiency of MS2 particles across an in vitro BBB model, a highly sensitive RT-qPCR protocol was developed and implemented. …

This study identified evolutionary selective pressures within subcluster A3 actinobacteriophages. These phages are able to infect the clinically important genus Mycobacterium. Understanding the selective pressures on genes in these phage genomes is a step toward understanding the adaptations that result from short-term and long-term associations of phages and bacteria that have been co-evolving for perhaps billions of years. In this study 149 phamilies (phage protein families) of homologous gene sequences were analyzed using Datamonkey. Complete data were obtained for 57 phamilies. Industrial production of protein therapeutics demand rigorous testing and clearance of viruses. The U.S. Food and Drug Administration dictate the purity of pharmaceuticals with regards to viral contamination. As this testing is time consuming and expensive using mammalian cells and viruses, bacteriophages may provide a faster and cheaper alternative for membrane filtration processes. We used ultrafiltration membranes to filter protein solutions with viruses. Two bacteriophages were tested against membranes with two different pore sizes.
