Life After Death: Investigating The Resistance And Recovery Of Fungal Communities To Foundation Tree Mortality, Jessie F. Marlenee

Tree mortality can have cascading effects on the composition and resilience of interacting fungal communities. Yet, little is known about the impact tree mortality, distinct from abiotic drivers (i.e., drought), has on these communities. This dissertation focuses on the resilience of fungal communities to experimental tree mortality in a piñon-juniper woodland. In chapter one, I found that piñon mortality reduced piñon fine root biomass and piñon mycorrhizal fungal diversity, whereas juniper mycorrhizal fungi were unaffected by juniper mortality. The research presented in this dissertation focuses on microbial roles in biogeochemical cycling in Antarctic aquatic environments. The major objective of my research was to examine the impact of biotic and abiotic pressures on nutrient cycling in microbially dominated systems. I used three perennially ice covered lakes in the McMurdo Dry Valleys (MDVs) along with subglacial lakes Whillans and Mercer as natural case studies. The MDVs are located in Victoria Land, East Antarctica and have been studied since 1993 as part of the McMurdo Long Term Ecological Research Project. Quantifying the flow of energy and nutrients through food webs is foundational to understanding the structure and function of ecosystems. Here, I utilize the stable carbon and nitrogen isotope analysis of individual amino acids to trace the movement of essential amino acids through terrestrial and freshwater food webs in New Mexico, USA. I first explore isotopic patterns among co-occurring terrestrial plants and aquatic algae. I then combine this molecular isotopic approach with 16S and 18S rRNA sequencing to demonstrate the importance of gut microbiota as sources of essential amino acids to wild mammalian hosts. Quantifying microbial biomass is important for understanding microbial contribution to nutrient cycling and population flux. Widely accepted biomass estimation methods are sufficient for well-studied and high biomass systems, though are often not sensitive enough for systems with extremely low biomass. The McMurdo Dry Valleys of Antarctica are characterized as an extremely harsh environment with severe restrictions on life, with perhaps some of the lowest biomass in the world. We measured prokaryote biomass across Taylor and Wright Valleys using four estimation metrics to determine the most appropriate method for this ecosystem. Fungi perform several critical functions in the environment. Spatiotemporal distributions of fungal communities will mediate when and where these functions happen and how they vary across the landscape. I first explored tropical tree canopy fungal community variation at small spatial scales and documented near total turnover of fungi across sub-meter distances and among adjacent substrates. The second chapter analyzed fungal turnover over the course of three years, where community stability was driven primarily by abundant fungi. The McMurdo Dry Valleys (MDV) of Antarctica are on the threshold of widespread landscape scale change due to increasing temperature and solar radiation and altered hydrology: buried ice is melting, the soil active layer is thickening, thermokarst features are developing along streams, water tracks are expanding, and lake levels are rising. These changes will impact the microbial communities found in each of the affected habitats. My doctoral work focused on understanding the reciprocal relationship between fungi and their environment, namely how fungi respond to environmental flux, as well as how fungi can modify and structure their habitats, especially in the context of climate change. As such, I aimed my research on fungi with distinct adaptations to their environmental niches: endophytic fungi that inhabit plant tissue and thermophilic fungi that are capable of growing at the upper temperature limit for eukaryotic life. This dissertation examines how climate change affects mycorrhizal fungal communities in boreal and arctic ecosystems. In chapter one, I revealed that increases in fire severity and related increases in deciduous tree dominance result in greater Ascomycota relative abundance (RA) and subsequent declines in Basidiomycota RA. In chapter two I analyzed the effects of post-fire mycorrhizal fungal communites on host growth. There were trends at the fungal genus level that were largely reflected at the guild level across all hosts; however, there were some fungal genera that had the opposite effect on different host species. Advances in DNA sequencing and data analysis make it possible to address questions in population genetics and evolution at the genomic level. Fungi are excellent subjects for such studies, because they are found in diverse environments, have short generation times, can be maintained in culture and have relatively small genomes. My research employed genetic approaches using a variety of sequencing technologies and methods of analysis to explore questions in fungal evolution.

Plant-microbial interactions influence biogeochemical cycles. Plants and biological soil crusts are primary producers in drylands. Biocrusts include cyanobacteria, lichens, mosses, algae, fungi, bacteria, and archaea on the soil surface, some of which fix atmospheric nitrogen. I investigated controls on biocrust carbon fluxes and their contribution to ecosystem fluxes, the incorporation of plant-derived carbon into biocrusts, and the role of soil fungi in promoting performance of plants and biocrusts. Biocrusts responded to temperature and moisture differently by biome. Biocrusts in grasslands/shrublands contributed >25% of total summertime ecosystem respiration, but biocrusts in savannas/woodlands contributed < 1%. Biocrusts contributed < 2% to GPP in any biome. To augment their native photosynthesis, biocrusts may include 16% plant-derived carbon. Fungal connections improved plant and biocrust performance and reduced differences in the CN ratio between organisms compared to when connections were impeded. Investigation of interactions among biocrusts, plants, and fungi has improved understanding of resource cycling in drylands.

Grinnell Lake, Lake Josephine, and Swiftcurrent Lake, at an elevation of approximately 5,000 feet in northwestern Montana, are comparable with lakes in the montane zone defined by Pennak for lakes at elevations between 8,200 feet and 10,500 feet in the Rocky Mountains of northern Colorado. Five species of Copepoda, six Cladocera, and four Ostracoda were collected in the lakes studied. The population of microcrustacea in these lakes was most dense in or slightly above the masses of rooted aquatic vegetation growing to a height of six to 10 inches above the bottom wherever the water was between three feet and …
