After over 100 years of Zn and Pb mining in the Tri-State mining district (TSMD), former mines continue to be sources of metals. Metal contaminated soils can be remediated using plant-microbe interactions. Plants manipulate their microbiome and recruit microbes to increase metal tolerance. Remediating bacteria are site-specific and identifying native microbes can accelerate remediation efforts. Studies relating microbes and metal concentrations in remediated areas in the TSMD are scarce. I collected root and bulk zone soils associated with Andropogon virginicus from remediated and non-remediated sites in Webb City, MO. Bacterial communities play a central role in the biological nutrient removal and overall performance of wastewater treatment plants (WWTPs). However, very few is known about the spatial and seasonal dynamics of these communities. This study explores the spatial and seasonal variations in bacterial community composition across multiple treatment stages in two municipal WWTPs—Southwest (SWTP) and Northwest (NWTP)—in Springfield, Missouri. Using 16S rRNA gene amplicon sequencing, I analyzed 87 samples collected during summer and winter of 2024 from influent, sludge, aeration basins, clarifiers, and final effluent stages. Soybean plants rely on nitrogen for growth, however atmospheric nitrogen (N2) is not a usable form and needs to be converted into different forms that are available to the plant. Nitrogen fixing bacteria such as rhizobia form symbiotic relationships with Soybeans in exchange for shelter and excess plant sugars. As a result, the plant can get the nitrogen it needs. Soybeans form symbiotic relationships with nitrogen-fixing bacteria in their root nodules to acquire most of their nitrogen requirement; nitrogen is often one of the limiting nutrients in the soil. Members of the Bradyrhizobium, Sinorhizobium, Mesorhizobium, and Rhizobium genera can nodulate soybeans. Bradyrhizobium is preferentially selected over other types of rhizobia in varying environmental conditions and is often found in acidic soils. Sinorhizobium is often identified in the root nodules of soybeans in neutral and alkaline soil. Waterborne pathogens from human fecal matter pose a significant health risk in karst environments, where domestic wastewater can easily be transferred to the shallow groundwater system via old, leaky septic tanks and damaged sewer lines. This study monitored fecal indicator bacteria (FIB) and potential pathogens in Sequiota Spring located in the karst region of southwest Missouri, using Microbial Source Tracking (MST) and Illumina DNA sequencing of bacterial 16S rRNA genes. MST identified high levels of human fecal indicator bacteria (HFIB) (up to 110,000 cells/L) in July 2020. By June 2022, HFIB levels had dropped significantly (55-fold reduction) after the City …

Perfluorooctanesulfonic acid (PFOS) has been phased out of production in the United States, but because of its resistance to degradation and its persistence in the environment, PFOS can be found in drinking water across America. Oral exposure to PFOS has been linked to adverse developmental, immunological, hepatic, and endocrine health. Oral PFOS exposure may alter the gut microbiota and cause dysbiosis, which is implicated in the pathology of inflammatory diseases like Crohn’s disease, diabetes, and ulcerative colitis. Flowers host diverse communities of epiphytic bacteria, which can impact floral functions, pollinator interactions, and overall plant fitness. However, little is known about how the abundance of these bacteria change as flowers age, and how these shifts may correlate with floral longevity. In this study I investigate the change in bacterial abundance across the lifespan of a flower from anthesis (opening of bud to flower) to senescence (wilting of flower) and to explore the potential implications for floral longevity. This master's thesis examines the influence of environmental variables on cognitive processes in Bombus impatiens, focusing on the effects of ingested microbial communities on gut microbiota and their subsequent impact on associative color learning. The study implemented controlled feeding trials with isolated microbe species (Candida bombi, Metschnikowia reukaufii, Lactobacillus micheneri and a control with no microbes) to analyze shifts in gut microbial composition in foraging bees. Results indicated minor alterations in the microbial profiles of bees, yet no statistically significant correlation was found between these modifications and variations in learning or memory performance. Soybean plants fulfill most of their nitrogen requirement by developing symbiotic associations with four different rhizobial genera, including Bradyrhizobium, Sinorhizobium, Mesorhizobium, and genus Rhizobium. In general, members of Bradyrhizobium and Sinorhizobium have been reported as dominant root nodule endophytes under acidic soils and alkaline soil conditions, respectively. Bacterial impairment of water systems is a major issue facing mankind. Bacteria that are introduced into a system have the potential to cause harmful diseases to wildlife and humans. In Greene and Polk counties, many water systems have become bacterially impaired over the years. Despite this, little is known about the contamination of known harmful bacteria in this region. Biological nitrogen fixation by rhizobia in the root nodules of legumes is a significant source of agricultural nitrogen in global crop production systems. The influence of and interaction of factors involved in nodule endophyte selection remain poorly understood. In the present study, the influences of crop rotation (soybean-legume vs. cotton-legume) and recalcitrant soil organic matter (compost amendment) on the relative distribution of endophytic bacteria in the root nodules of greenhouse-grown soybean and lablab were investigated by extracting, amplifying, and sequencing 16S ribosomal RNA (rRNA) and nifH genes. Early life stress is considered a risk factor for development of migraine, which is a prevalent painful neurological disorder involving sensitization and activation of trigeminal neurons, and irritable bowel syndrome, a condition often comorbid with chronic migraine. The focus of my study was to determine the effects of early life stress and dietary inclusion of enriched chicken bone broth on trigeminal nociceptor sensitization and the gut microbiota.
