Plant-soil-microbe interactions are integral components of ecological networks and drive ecosystem function from the bottom-up. Whereas herbivores regulate ecosystem function from the top-down, having opposing and potentially complementary effects on ecosystem function. My thesis sought to understand how below-ground microbial communities contribute to herbivore preference and plant biomass across varied soil substrates. My thesis asked: how do substrate and soil microbial communities influence herbivore preference and plant biomass accumulation on swamp milkweed (Asclepias incarnata)? Lakes are important aquatic ecosystems that are extensively influenced by seasonal variations. In this study we investigate the seasonal dynamics and trends of three environmental variables, water temperature, conductivity and dissolved oxygen (DO), in Lake Louise, a sinkhole lake located in southern Georgia. These variables were measured in-situ along a depth profile from January 27, 2024 to January 17, 2025. We also accessed archived data from field campaigns in 2003, 2004, and 2017 in order to test whether a recent increase in the number of hurricanes impacting the region, five since August, 2017, may have significantly altered the seasonal dynamics …

This project analyzes the rise of cyanobacteria blooms in Rhode Island freshwater systems and identifies a policy gap in the state’s largely reactive management approach. Drawing on state reports and interviews with environmental experts, it highlights nutrient runoff, climate change, and limited public education as key drivers of bloom severity. To address this gap, I developed a prevention-focused public education poster designed for integration into existing state alert and outreach systems.

Taal Volcano Island’s hydrothermal environment supports extremophilic microbial communities. This study isolated, biochemically characterized, and genetically profiled Staphylococcus sp. from shoreline soils. Soil samples were collected near active vents and cultured on acidified gelatin agar (pH 5.0). Biochemical tests—Methyl Red, Spirit Blue Agar, Motility, and Acrolein—were performed. DNA extraction and 16S rRNA sequencing identified Staphylococcus candidatus, with 99 % sequence similarity to GenBank entries JN794602.1 and KX250303.1. Phylogenetic analysis clustered the isolate with S. ureilyticus, S. cohnii, and S. equorum. Caliciviridae is a family of non-enveloped, positive-sense, single stranded RNA viruses, including eleven defined genera that contain important pathogens of humans and animals. Within the Caliciviridae family, the genus Vesiviridae includes feline calicivirus, a leading cause of upper respiratory disease in cats, and vesicular exanthema of swine virus, a historical disease of domestic swine that has now been implicated in several marine mammal vesicular diseases, one of which has been confirmed zoonotic. Temperature limits plant growth and survival more than many other environmental variables, and increases in the frequency of extreme hot or cold weather events pose significant challenges for plant growth and crop yields. Previous studies have shown that bacteria, fungi, and other microorganisms associated with plants ( i.e., the plant microbiome) can influence host response to temperature stress. These same microbes have their own temperature tolerances. In this dissertation, water samples from the Chesapeake and Delaware Bay estuaries were analyzed to see how estuarine environmental gradients drive distributions, functional potential, growth estimates and gene expressions of bacteria. To answer this, the abundant and versatile taxa– Rhodobacterales and Actinobacteriota, along with broader particle-attached (PA) vs. free-living (FL) bacterial assemblages of these Bays were analyzed using a multifaceted ‘omics approach.

Stream ecosystems promote variable microhabitats along a longitudinal gradient. These microhabitats support dynamic biotic assemblages. One such microhabitat in freshwater streams involves the external surface of crayfish. We investigated the microbiome of crayfish exoskeletons and the surrounding environment (water and substrate) in association with the presence and abundance of crayfish cleaning ectosymbionts (branchiobdellidans). These ectosymbionts actively consume the biofilm on crayfish as they live and reproduce in association with crayfish microhabitats. Sequenced microbes of the environment and crayfish were compared for dominant taxa. Rock pools are ephemeral aquatic environments that offer a unique opportunity to study complex communities and their responses to environmental changes. The urban James River rock pool system consists of over 700 individual rock pools varying in size and depth. This unique urban freshwater ecosystem has been studied for its invertebrate, plant, and zooplankton populations but not for its microbial community composition. Lake Sentani is a tropical lake in Indonesia, consisting of four interconnected sub-basins of different water depths. Harmful Algal Blooms (HABs) threaten aquatic ecosystems, local economies, and water quality. While extensively studied in lakes (lentic systems), their impact on flowing waters (lotic systems) remains underexplored. This study examined algal drivers in both ecosystems within the same watershed, focusing on Beech Fork Lake (lentic) and Twelve Pole Creek (lotic). Key environmental factors, including pH, turbidity, dissolved oxygen, and nutrient levels, were measured using in situ sensors and laboratory analysis. The Nebraska Water Quality Index, under development by the Nebraska Department of Environment and Energy, summarizes in a single value eight environmental parameters that have been monitored in Nebraska for nearly 20 years. Water quality parameters including those used in the Nebraska Water Quality Index have been shown in previous studies to impact bacterial growth. As such, this index has the potential to correlate with the freshwater microbial community. Microcystis spp. produce prolific annual blooms in freshwater systems worldwide. The success of these blooms depends heavily on the Microcystis spp. overcoming environmental factors such as oxidative stress caused by reactive oxygen species (ROS) like hydrogen peroxide. Most Microcystis genomes do not contain many genes for responding to oxidative stress, including those that encode the enzyme catalase. Other cyanobacteria such as Prochlorococcus that also lack hydrogen peroxide degrading enzymes have been shown to benefit from partnerships with heterotrophic bacteria. Non-native species are increasing in prevalence around the world, resulting in negative economic and ecological impacts. However, the broad distributions of non-native species also offer a system for investigating the response of host-associated microbial communities to environmental factors across a range of ecological scales. At the broadest scale, I investigated the geography of microbial communities in the non-native estuarine anemone Diadumene lineata on the west coast of the United States of America. Ice-covered lakes are vulnerable to environmental change, especially those in the Northern Hemisphere where ice cover is rapidly declining due to global warming. These changes can alter metabolic processes and disrupt carbon cycling driven by primary producers who form the base of the food chain and are key to sustaining ecosystem function. Quantifying the flow of energy and nutrients through food webs is foundational to understanding the structure and function of ecosystems. Here, I utilize the stable carbon and nitrogen isotope analysis of individual amino acids to trace the movement of essential amino acids through terrestrial and freshwater food webs in New Mexico, USA. I first explore isotopic patterns among co-occurring terrestrial plants and aquatic algae. Microphototrophs (algae and cyanobacteria) in karst environments have been intensively studied in aquatic epigean habitats. In recent decades knowledge about the communities inhabiting cave entrances and lampenflora has grown substantially, but the data about the communities in aphotic cave zone are scarce. This study aimed to investigate spatio-temporal presence of microphototrophs in the aphotic zone of Veternica Cave (Mt. Global climate change is a phenomenon resulting in more extreme weather patterns and species diversity loss. In this study, I explore the impacts of climate change on regional patterns of microbial productivity. Variation in microbial productivity was explained using temperature, salinity, and dissolved oxygen as predictors in regression models with data from the LTER and iLTER network of sites that ranged from the poles to the equator. I found a positive relationship between temperature and productivity. Antarctica and Arctic sites exhibited the strongest positive relationships supporting prior research demonstrating temperature as one of the driving forces of productivity change in …

The characterization of the most common photosynthetic biofilms in the Nerja Cave by the continuous monitoring of the in vivo chlorophyll a (Chl a) fluorescence and the incorporation of the irradiance as a new environmental variable related to previous studies in the cave, have allowed us to improve our knowledge about the photosynthetic pattern of the biofilms of the cave. In Nebraska and other regions of the Great Plains, the conifer Juniperus virginiana (eastern redcedar) is converting grasslands to dense woodlands. This is driven by the interacting drivers of fire suppression, altered grazing regimes, climate change and other anthropogenic factors, impacting the provisioning of ecosystem services. This vegetation state transition modifies water resource regulation and biogeochemical cycles leading to altered edaphic properties including soil microbial community composition. To restore these grasslands and control J. virginiana spread, prescribed extreme burns are implemented as a management tool through local prescribed burn associations. Background/Questions/Methods

The introduction of the Chinese chestnut blight in 1904 decimated native American chestnut (Castanea dentata (Marshall) Borkh.) populations. In this study we aim to: 1) document the location of individual chestnuts in our vicinity; 2) document healthy versus blight infected individuals; 3) characterize putative differences in the soil microbiome between infected and non-infected individuals, as well as between chestnuts and hardwood neighbors. With this information we hope to provide new insights into mechanisms that may enhance blight and fungal resistance in American chestnuts through the understanding of the interactions between the trees and their surrounding soil microbiome.

Microplastics and their threat to marine environments are well known, but studies in freshwater environments are few and far between. Microplastics can be created by the breaking down of plastics or in the process of making plastics. They affect biological lifeforms in a multitude of ways, water temperature, permeability of the sediment. Pollution in Texas waterways have motivated groups like the Environmental Protection Agency, Trash Free Texas, and Environment Texas to get involved.
