Microbial extracellular enzymes are critical for the breakdown of organic matter and nutrient cycling in aquatic ecosystems. Extracellular enzyme activity serves as an indicator of nutrient turnover rates, metabolic states, and nutrient limitations within an ecosystem. Coastal ecosystems are especially dynamic as the influx of freshwater from inland runoff or seawater intrusion from storm surges can lead to fluctuations in salinity which can restructure microbial communities and alter enzymatic activity. Leptoglossus phyllopus is a polyphagous pest that primarily feasts on citrus, tomatoes, and legumes. They must form a mutualistic symbiosis with Caballeronia every generation by ingesting the bacterium from their environment. However, not all species of Caballeronia confer the same fitness on the L. phyllopus host, raising the question of whether L. phyllopus possesses a partner choice mechanism that would allow it to distinguish between a good (GAOX1) and a suboptimal (Lep1P3) symbiosis partner. The common green iguana (Iguana iguana) was introduced to South Florida in 1965 and has since been an especially pernicious threat both environmentally and economically. This highly invasive species is also expanding in range and populations at an astonishing rate. Understanding how this reptile is able to adapt and thrive in novel environments is of imperative need in order to begin managing their disruptive populations. Notwithstanding their negative impacts and significance, the microbiome of wild populations has not yet been characterized. This project investigated the use of engineered bacterial reporter systems to detect cellular stress responses associated with antibiotic activity. While constructs were successfully introduced and tested against known antibiotics, the reporter consistently produced a blue signal across conditions, limiting the ability to distinguish specific stress responses. This suggests issues such as leaky expression, insufficient regulatory control, or lack of specificity in the reporter design. Although results were inconclusive, this work highlights key challenges in developing reliable biosensors and provides a foundation for future optimization. Amphibians are experiencing population declines due to habitat loss, climate change, and diseases, particularly chytridiomycosis caused by the fungus Batrachochytrium dendrobatidis. Protection from this fungal infection stems from the cutaneous microbiome. Pesticides have been shown to affect amphibians in a variety of ways, including reducing survival, locomotor performance, and accumulation in body tissues. Pesticides may also lead to changes in the cutaneous microbiome affecting amphibians who rely on these organisms as an anti-Bd defense mechanism. Temperature limits plant growth and survival more than many other environmental variables, and increases in the frequency of extreme hot or cold weather events pose significant challenges for plant growth and crop yields. Previous studies have shown that bacteria, fungi, and other microorganisms associated with plants ( i.e., the plant microbiome) can influence host response to temperature stress. These same microbes have their own temperature tolerances. Insect gut bacteria are vital contributors to host physiology, yet the broader prevalence and fitness implications of symbiont-mediated detoxification remain poorly understood. This thesis investigates these dynamics across three research chapters. Chapter 1 uses 16S rRNA sequencing to characterize how grasshopper gut microbial communities vary across species, dietary breadths, and gastrointestinal segments. Chapter 2 evaluates the detoxification capacity of over 150 bacterial isolates from diverse herbivores against the plant allelochemicals phenethyl isothiocyanate, rutin, and chlorogenic acid to determine if host dietary specialization predicts microbial metabolic ability. Lead contamination remains a significant global public health concern, particularly in Kabwe, Zambia, the site of a former lead mining operation. Bioremediation, which utilizes biological materials and processes to remove environmental contaminants, represents a promising approach for addressing lead pollution. However, the development of effective bioremediation systems requires a detailed understanding of the mechanisms underlying metal removal and the feasibility of implementation in low-resource settings. Most multicellular organisms depend on their microbial symbionts for various functions, such as digestion, nutrient supplementation, pesticide degradation, and defense. While many insects acquire their symbionts vertically from their parents, thousands of species of true bugs (Hemiptera) across different families acquire their microbial symbiont, Caballeronia, from the environment each generation. Acquiring symbionts from the environment (horizontal transmission) has various ecological consequences for the host. After over 100 years of Zn and Pb mining in the Tri-State mining district (TSMD), former mines continue to be sources of metals. Metal contaminated soils can be remediated using plant-microbe interactions. Plants manipulate their microbiome and recruit microbes to increase metal tolerance. Remediating bacteria are site-specific and identifying native microbes can accelerate remediation efforts. Studies relating microbes and metal concentrations in remediated areas in the TSMD are scarce. Influenza viruses (IV) affect populations globally, with the World Health Organization (WHO) estimating that IV are responsible for an estimated 3-5 million infections and 290,000 – 650,000 deaths annually1–3. More specifically, the 2019-2020 IV season is associated with an estimated 38 million illnesses and 22,000 deaths in the United States alone4. Increasing incidence of Harmful Algal Blooms (HABs) has led to fish kills, air quality hazards, and economic impacts. One economic impact is on tourism, one of the main sources of Florida’s income, where billions of dollars are lost to such events. The Florida Fish and Wildlife Conservation Commission Fish and Wildlife Research Institute (FWC-FWRI), with the help of collaborators, collects water samples from across the state for HAB monitoring. These samples are stored at ambient temperature for up to 24 hours which could directly affect data analyses. Abstract
Diabetes mellitus is a metabolic disorder characterized by elevated blood glucose levels due to impaired insulin secretion or diminished cellular response to insulin (resistance). Pancreatic β-cells depend on a well-organized cytoskeleton, particularly microtubules composed of α- and β-tubulin heterodimers, to transport insulin granules to the plasma membrane during glucose-stimulated insulin secretion (GSIS). Disruption of microtubule organization can impair granule trafficking and reduce insulin release. However, emerging environmental toxicants such as microplastics that interfere with β-cell cytoskeletal function remains largely unexplored.
Microplastics (MPs) are small plastic particles (< 5 mm) that have become widespread environmental contaminants and can enter biological systems. Previous studies have shown that MPs can induce cellular stress and protein misfolding, but their effects on microtubules in pancreatic β-cells are not well characterized. This study investigates whether exposure to fluorescent polystyrene microplastics alters microtubule organization and insulin secretion in βTC-6 cells.

Chronic wound infections pose significant healthcare challenges due to persistent biofilms, antibiotic resistance, and impaired healing pathways. These wounds are characterized by prolonged inflammation, microbial colonization, and disrupted tissue regeneration, leading to substantial morbidity and healthcare costs. This article reviews current knowledge on chronic wound types, their pathophysiology, and the mechanisms underlying biofilm formation to examine the latest advancements in antimicrobial-based strategies aimed at addressing these challenges. As an emerging crop, cultivation techniques for industrial hemp (Cannabis sativa L.) are still being developed and optimized. For example, irrigation and plastic mulching are some management practices that have been touted for their benefits in improving crop quality. Both techniques ensure ample water availability, which has often been attributed to improving hemp cannabinoid and terpene composition. Non-invasive fecal sampling is essential for wildlife gut microbiome (GMB) research, yet field conditions often limit preservation options. I compared gut bacterial communities in paired fecal samples preserved in stabilization tubes and air-dried in paper bags collected from anesthetized African lions (Panthera leo) and spotted hyenas (Crocuta crocuta) in Etosha National Park, Namibia. Additional opportunistic samples from carnivore kill sites were also analyzed. No significant differences in alpha or beta diversity were detected between preserved and dried samples, although bacterial beta diversity differed between preserved and opportunistic samples (PERMANOVA). Inland Pacific Northwest prairie is an endangered ecosystem that has been reduced by conversion to agriculture. There is growing interest in the role of soil microorganisms in prairie ecosystems, which may be crucial for restoration of the plant community through interspecies interactions within the soil near plant roots. One method for improving soil microorganism community composition is compost tea: enriched liquid compost extract. Compost tea is used in regenerative agriculture to improve crop yields and protect against pathogens, ostensibly through the activity of the added compost microbes. Worldwide, amphibian populations have experienced dramatic declines due to a variety of threats. Among them, disease has been particularly deleterious and continues to impact populations. In the United States and Canada, the three leading causes of amphibian mass mortality events are ranavirosis, chytridiomycosis, and severe perkinsea infection, all caused by infectious pathogens. While research and policy have helped populations recover from these diseases, more work needs to be done to develop practical and efficient treatments for these infections. The impact of land development on soil microbial diversity and antibiotic production is critical due to industrial expansion and the scarcity of effective antibiotics. Low Impact Development (LID) aims to preserve ecosystems, but its effectiveness in maintaining microbial diversity is uncertain. This study compares soil samples from nine sites: undisturbed (UD), residential (RD), and Madla Greenway (GW) within the VIDA development area and Texas A&M University-San Antonio campus. Isolated microbes were screened for antibiotic production and genetically characterized. Cyanobacterial harmful algal blooms (cHABs) occur globally, excluding Antarctica, and often involve toxin-producing species such as Microcystis aeruginosa. These blooms threaten both ecosystems and public health, making it essential to understand their underlying drivers. Cyanobacteria generate reactive oxygen species (ROS) during photosynthesis, which can cause cellular damage unless neutralized. Some, like M. aeruginosa, lack the genetic capacity to detoxify ROS and rely on associated bacterioplankton for protection. Tree mortality can have cascading effects on the composition and resilience of interacting fungal communities. Yet, little is known about the impact tree mortality, distinct from abiotic drivers (i.e., drought), has on these communities. This dissertation focuses on the resilience of fungal communities to experimental tree mortality in a piñon-juniper woodland. In chapter one, I found that piñon mortality reduced piñon fine root biomass and piñon mycorrhizal fungal diversity, whereas juniper mycorrhizal fungi were unaffected by juniper mortality. Healthcare-acquired infections (HAIs) are a significant global challenge driven by biofilm-forming pathogens. Polymicrobial biofilms, involving interactions between multiple microbial species, exacerbate treatment difficulties due to their enhanced resistance to antimicrobial therapies. Borate bioactive glasses (BBGs) are an emerging class of biomaterials that have attracted significant interest in infection control. The incorporation of copper and zinc into the BBG matrix can effectively disrupt biofilm formation and bacterial colonization. Soybean plants rely on nitrogen for growth, however atmospheric nitrogen (N2) is not a usable form and needs to be converted into different forms that are available to the plant. Nitrogen fixing bacteria such as rhizobia form symbiotic relationships with Soybeans in exchange for shelter and excess plant sugars. As a result, the plant can get the nitrogen it needs. Arthropods, or phylum Arthropoda, are the largest group of animals, accounting for over three quarters of all animal records on NCBI Taxonomy. And yet, only 40% of all deposited sequences on NCBI Nucleotide come from arthropods. Clearly, arthropods are an understudied group of animals: they are small, often drab, and lead lives very alien to our own. While previous studies have examined plant intraspecific competition and soil microbial effects separately, fewer studies have investigated how these factors interact, particularly in the context of genotype-specific soil conditioning effects. Plant-soil feedbacks can significantly alter interspecific competitive outcomes, yet few studies have examined how local variation in soil conditioning influences plant responses to density-dependent competition. This study focuses on the bioaccumulation and biomagnification of mercury in freshwater food webs, with emphasis on bioindicators, such as macroinvertebrates, crayfish, and river otters. Despite regulatory efforts and advancements in monitoring, significant gaps remain in understanding mercury dynamics in freshwater systems—especially regarding its transport, methylation, and bioavailability across different ecosystems. To address these knowledge gaps, this study adopts a biocentric approach, emphasizing the role of living organisms in their environment. By utilizing macroinvertebrates, crayfish, and river otters as bioindicators, we aim to assess mercury bioavailability and distribution. Innovative design enhancements to microbial fuel cells (MFCs) are pivotal for improving their viability as renewable energy sources. This study introduces a dual approach to electrode modification, integrating polyaniline (PANI) and gold nanoparticles (AuNPs) on carbon felt (CF) anodes coupled with a unique cathode composed of DNA origami and chitosan. The synergistic effect of PANI and AuNPs significantly increases the hydrophilicity and conductivity of the anode, leading to a considerable surge in power density. Concurrently, the biocompatible DNA origami and chitosan-functionalized cathode facilitates efficient electron transfer without relying on conventional catalysts. Antibiotics are some of the most essential lifesaving drugs currently available to us against infectious diseases. Several pathogens are developing resistance to these drugs and many have become resistant to multiple antibiotics. Such multi-drug resistance is becoming a global issue and has been found in many environments including wastewater treatment plants (WWTPs). In this study, we sampled a local WWTP for antibiotic-resistant bacteria. This WWTP is in North Georgia and runs directly into Lake Lanier, which is the main source of drinking water for the Atlanta area as well as a popular spot for water recreation.
