Antibiotic resistance is a rapidly emerging issue within public health. As resistance grows, the effectiveness of traditional antibiotic therapies diminishes while risk of treatment grows. Alongside novel therapies, public knowledge is a strong combatant of the resistance-related risks of antibiotic usage. Through a comprehensive review of current literature, this work serves as an accessible guide to the mechanisms of antibiotics, bacterial resistance, and the challenges related to current antibiotic usage. Additionally, the ability of current policy and emerging novel treatments, such as nanoparticles and bacteriophage therapy, to fight antibiotic resistance is evaluated. Typical antibiotic susceptibility testing (AST) of microbial samples is performed in homogeneous cultures in batch environments, which does not account for the highly heterogeneous and dynamic nature of antibiotic responses. The most common mutation found in P. aeruginosa lineages evolved during chronic cystic fibrosis infections in the human lung, a loss of function of repressor MexZ, increases basal levels of multidrug efflux MexXY, but does not increase resistance by traditional minimal inhibitory concentration (MIC) assays. Here, we use single cell microfluidics to show that P. aeruginosa’s response to aminoglycosides is highly heterogeneous, with only a subpopulation of cells surviving exposure. …

Macrophages are key cells of the innate immune system, playing essential roles in host defense, tissue homeostasis, and inflammation. Depending on stimuli, macrophages can polarize into distinct functional phenotypes, characterized as classically activated (M1) or alternatively activated (M2) macrophages. M2 macrophages are associated specifically with anti-inflammatory responses, tissue repair, and immune regulation. They contribute to wound healing and can regulate host responses during infection. Community-acquired respiratory distress syndrome (CARDS) toxin, a key virulence factor of Mycoplasma pneumoniae, plays a role in altering host immune responses, but its impact on M2 macrophage function and polarization remains unclear.

Listeria monocytogenes is a facultative intracellular foodborne pathogen capable of crossing the placental barrier, leading to miscarriage, stillbirth, and neonatal sepsis, and has a fetal/neonatal fatality rate of ~20–30%. Although the importance of canonical virulence factors such as Internalin A (InlA) and Internalin B (InlB) in L. monocytogenes crossing of the placental barrier has been demonstrated, clinical strains lacking functional canonical internalins have been reported to cause severe neonatal disease, suggesting additional mechanisms of placental translocation.

Antimicrobial resistance is a growing concern in medicine1 leading to a need for the development of novel compounds. Two compounds were tested in this study, 2-hydroxyethylhydrazine (HEH) and para-nitrophenyl-isocyanide (NPIC). HEH targets phosphatidylcholine (PC) production2, and NPIC targets mitochondrial function3. Choline (Cho) is a compound that bypasses the inhibition of HEH through the Kennedy pathway.2 Two-fold serial dilutions were conducted in 96-well plates to test the sensitivity of different bacteria to HEH and fungi to HEH and NPIC in different media types. Antibiotic resistance is a critical global challenge that requires the discovery of alternative therapeutic agents. Plants -derived compounds, such as those found in Ananas comosus (pineapple)peel, offer promising antibacterial potential. This study aimed to evaluate the antibacterial activity of pineapple peel extract using both in vitro and in silico approaches. Ethanolic extracts of pineapple peel were fractionated using silica gel chromatography, and the resulting fractions were analyzed by GC-MS. Antibacterial activity was tested against Gram-positive and Gram-negative pathogens using the agar well diffusion method, and the inhibition zones were recorded. Tidal flooding is common in the City of Norfolk, VA due to high rates of sea rise in the region, as well as the region’s subsidence. This has led to increases in flooding at high tide, especially during the spring tide portion of the tidal cycle. At high tide water from the Elizabeth and Lafayette Rivers can flood the landscape directly or can rise out of storm drains. This study measures whether microbial communities in the shallow coastal waters of Virginia Beach are capable of consuming methane in the water column. Using methane enrichment incubations and gas chromatography, we found no measurable methane oxidation of our samples over multi-day periods. These results suggest that these coastal waters may act as net sources of atmospheric methane.

BACKGROUND: Lapsi (Choerospondias axillaris), a plant native to Nepal, has been traditionally used in Asian countries to treat cardiovascular conditions. However, its effects on immune regulatory function remain largely unexplored. This study aimed to in vivo evaluate the immunoregulatory properties of Lapsi fruit extract in mice on immunotoxic responses with analysis on in vitro mechanism for immune suppression, oxidative stress, and inflammatory response. Male Balb/c mice were intragastrically administered various doses of the extract for 21 days. Background: Filifactor alocis is a Gram-positive, obligate anaerobe that has emerged as a diagnostic marker of periodontitis. Neutrophils and macrophages are key players in oral immune homeostasis. Neutrophils are short-lived cells and constitutively undergo apoptosis. This apoptotic program can be accelerated or delayed by bacterial interactions. Macrophages clear apoptotic neutrophils via “efferocytosis,” a crucial mechanism for resolving inflammation and preventing tissue damage. The efferocytic process downregulates the expression and release of proinflammatory cytokines such as interleukin (IL)-6 and IL-23 while promoting anti-inflammatory responses. Recessive dystrophic epidermolysis bullosa (RDEB) is a devastating inherited blistering disorder caused by biallelic mutations in COL7A1, resulting in deficiency of type VII collagen (C7) and loss of dermal–epidermal adhesion. Beyond mechanical fragility, RDEB is characterized by chronic inflammation, progressive fibrosis, impaired wound healing, severe pain, and an exceptionally high risk of aggressive cutaneous squamous cell carcinoma. Emerging transcriptomic and cytokine profiling studies have identified interleukin-1 alpha (IL-1α) as a dominant and temporally upstream inflammatory mediator in RDEB skin. Lakes are important aquatic ecosystems that are extensively influenced by seasonal variations. In this study we investigate the seasonal dynamics and trends of three environmental variables, water temperature, conductivity and dissolved oxygen (DO), in Lake Louise, a sinkhole lake located in southern Georgia. These variables were measured in-situ along a depth profile from January 27, 2024 to January 17, 2025. Human cytomegalovirus (HCMV) causes the most common congenital viral infection in the United States, with well-known acute and late-onset neurological pathologies. Moreover, HCMV, like multiple herpesviruses, has been associated with neuroinflammation and neurodegeneration. Using a well-established neonatal murine (M)CMV infection model, we found that early-life infection drove adult-onset neuron loss and neuropathology in the retina and brain, without evident viral reactivation. Pathology was associated with the persistence of highly activated and inflammatory damage-associated microglia. Human immunodeficiency virus type 1 (HIV-1) is a retrovirus that integrates into the host cell's DNA as a provirus. Transcription from the provirus is regulated in large part by cellular proteins and epigenetic factors. These may be repressive or permissive to productive infection. The host factors that regulate this balance are therefore attractive targets for HIV-1 therapeutics. Indeed, proviral chromatin is the focus of two of the current HIV-1 cure strategies. The co-evolution of virus and host may partly be understood in terms of “cross-recognition”; that is, as a virus evolves to recognize and infiltrate a suitable cellular environment, the host reciprocally has evolved to recognize and thwart such an invasion. Mycobacterium tuberculosis (Mtb) causes the disease tuberculosis (TB) and is one of the leading causes of death worldwide by an infectious agent. Furthermore, the emergence and evolution of drug resistance has significantly hindered global health efforts to combat this deadly epidemic. Thus, there is a critical need for development of new antibiotics with unique mechanisms of action and design of novel strategies for optimizing the efficacy of existing therapeutic regimens. Biofilm Architecture Determines Proliferation Of Phages And Plasmids, James B. Winans

This thesis explores how biofilm architecture determines the spread and ecological consequences for the bacteria-dependent genetic elements phages and plasmids. Using Escherichia coli and Vibrio cholerae as model microbes, and the genetic elements T7 phages, λ phages, and conjugative plasmid pED208, we investigate how spatial structure influences phage infection dynamics, plasmid transfer, and interbacterial interactions within biofilms. We show that densely packed biofilms can protect sensitive bacteria from phage exposure and modulate competitive outcomes between obligately lytic and temperate phages. Although autophagy, which is the process by which cells reallocate nutrients and eliminate unwanted materials, has been studied for decades, the connections between autophagy, inflammation, and viral immunity are still under investigation. Our studies focus on two genes: murine Epg5, which is involved in fusion of the autophagosome and lysosome, and C. elegans atg-3, which is important for the elongation of the phagophore. Here, we find that Epg5 mutation protects against enteric viruses including norovirus and rotavirus. We identify IFN- responsive genes as a key alteration and observe that Epg5 mutant mice exhibit substantial alterations in the intestinal microbiota. Surprisingly, …

Streptococcus gallolyticus subsp. gallolyticus (Sgg) is a gram-positive, non-beta-hemolytic bacterium that belongs to the Lancefield group D of streptococcal pathogens. For the past several decades, associations between Sgg bacteremia and colorectal cancer (CRC) have been apparent, with gastroenterologists routinely recommending colonoscopies for positive cases of Sgg in either blood or stool samples. However, current research has not wholly established whether Sgg acts as a causative agent of CRC or if pre-existing cancers make the lumen of the colon more hospitable to its colonization. Copyback viral genomes (cbVGs) are truncated non-protein coding viral genomes produced when an RNA virus polymerase detaches from the replication template and resumes elongation downstream, copying back a sequence complementary to the nascent strand. cbVGs can stimulate antiviral immunity and compete for viral proteins to suppress virus replication, but technical limitations of PCR-based approaches have painted an incomplete picture of the diverse cbVG populations generated during infection. Here, we sought to improve our understanding of cbVG molecular structure by harnessing long-read RNA sequencing tools. The emergence of multidrug-resistant bacterial pathogens, particularly Mycobacterium tuberculosis, highlights the urgent need for new antimicrobial agents. Tuberculosis remains one of the world’s deadliest infectious diseases, claiming over a million lives annually and posing increasing challenges due to the thick, lipid-rich, acid-fast cell wall of Mycobacterium species that renders them naturally resistant to many antibiotics. In this study, a soil sample collected from the Campus of Fort Hays State University yielded a bacterial isolate exhibiting strong antimicrobial activity. Initial screening against Escherichia coli MM294 showed a 10 mm inhibition zone, and subsequent testing identified the isolate as Bacillus pumilus …
