Listeria monocytogenes continues to be a major foodborne pathogen that causes food poisoning and sometimes death in immunosuppressed people and abortion in pregnant women. Nanoparticles have recently drawn attentions for use in immunomagnetic separation techniques due to their greater surface area/volume ratio and better stability against sedimentation in the absence of a magnetic field. Interdigitated microelectrodes and microfluidics make material transfer more efficient and biological/chemical interaction between the surface and solution phase much quicker. Magnetic nanoparticles (Fe3O4) with a 30 nm diameter were functionalized with rabbit anti-L. monocytogenes antibodies via biotin-streptavidin bonds and then amalgamated with target bacterial cells …

Myxomycetes are a group of amoeboid organisms with the capacity of forming fruiting bodies that resemble some macrofungi. The ecology and global distribution of species within the group have been studied only during the last half century. For this reason, a number of questions regarding the nature of the interactions that exist between myxomycetes and their environment still lack the empirical evidence required to obtain complete answers. In the Neotropical region, species assemblages have been moderately well studied, but their biogeography and macroecology have received little attention. Nitric oxide (NO) plays an important role in physiological functions like vasodilation, neurotransmission, and inhibition of platelet aggregation. The endothelium-derived NO diffuses into the vascular lumen where it interacts with flowing blood as well as the smooth muscles where it modulates vascular tone. However, uncertainty exists on how NO escapes the rapid scavenging by hemoglobin (Hb) and reaches smooth muscles. Several proposed hypotheses include 1) a reduced reaction rate of NO with Hb contained inside red blood cells (RBCs) and 2) NO preservation in the bound form of s-nitrosohemoglobin or nitrite. As a serious expense for both the hospital and the patient, nosocomial infections create a burden on the health care industry that is not easily overcome. Among the infections commonly contracted in the hospital environment, those associated with the Clostridium difficile bacterium account for millions of cases each year. Largely due to the nature of C. difficile infection as a response to the disruption of the normal flora of the colon caused by antibiotic activity, no completely effective treatment for this condition has been identified. Use Of Irradiated And Formalin-Fixed Trichomonas Vaginalis To Examine Protective Immune Responses In The Mouse Intraperitoneal Model, Terryl L. Hostetler, James J. Daly, Timothy J. O'Brien, Max L. Baker

Gamma irradiated Trichomonas vaginalis was used to initiate a protective immune response in mice by intraperitoneal inoculation using host mortality as the measure of virulence. Irradiated trichomonads of a virulent strain gave some immune protection but no more so than the use of formalin-fixed, virulent trichomonads. A formalin-fixed virulent strain combined with complete Freund's adjuvant (CFA) gave complete protection. Metabolically produced antigens do not appear to be important in conferring protective immunity in these experiments. A common laboratory strain of T. vaginalis (ATCC 30001) was used as an avirulent control. It gave no protection against a virulent strain. Combining CFA …

In Vitro Growth Characteristics Of Two Cryptococcus Neoformans Isolates, Kristy L. Jones, Juneann W. Murphy

Cryptococcus neoformans is an opportunistic pathogen which attacks individuals with weakened immune systems. Two C. neoformans isolates, 184A and NU-2, were examined to determine characteristics that contribute to their difference in virulence. Both isolates were grown under tissue culture conditions, and different characteristics were tested at timed intervals. Isolate 184A was found to grow at a faster rate than isolate NU-2, with doubling times of 4 and 12 hours respectively. The polysaccharide capsule of isolate NU-2 doubled in size in four hours, while that of isolate 184A stayed approximately the same. The NU-2 yeast cell diameter without the polysaccharide capsule …

Adsorption of Rhesus monkey erythrocytes to the plasma membranes of measles virus infected cells is frequently carried out to detect the presence of plasma membrane-associated measles virus hemagglutinin. The hemagglutinin is a viral genome-coded structural glycoprotein of the measles virion that is associated with the plasma membrane of the host cell during measles virus replication. BGM/MV, anon-virogenic line of African green monkey kidney cells persistently-infected with measles virus, adsorbed Rhesus monkey erythrocytes in an inverse fashion relative to the number of cells present in the culture and the time post-seeding. Distribution Of Naegleria Fowleri In Selected Northeast Arkansas Lakes, Robert Dean Evans, Lawrence W. Hinck

Seven northeast Arkansas recreational lakes were examined for the presence of pathogenic and nonpathogenic Naegleria fowleri. Cultural differentiation and microscopic morphology were used as species determining tests, while mouse pathogenicity tests were conducted to determine virulence. Only one isolate met all criteria utilized for definite identification of Naegleria fowleri, although Naegleria type organisms were found in all of the lakes. None of the isolates were pathogenic in mice.

The influence of cold (4°C) used to prepare cells for metabolic investigations was examined with Trichomonas gallinae in this study. Cells washed with cold diluent in a refrigerated centrifuge were found to be less stimulated in their gas production at 37°C when exposed to glucose or maltose than cells washed at room temperature conditions. Such cold-prepared cells had higher initial glycogen contents after washing, faster endogenous glycogen degradation rates when incubated at 37 °C, but lower glycogen synthesis in the presence of glucose or maltose when compared to cells not prepared in the cold. Selected recreational waters of Arkansas were sampled for pathogenic free-living limax amoebae. Water quality parameters were determined for correlation with amoebic population densities and species diversity. Cultural criteria and animal inoculation revealed no pathogenic strains. The possibility of introduction and/or induction of pathogenic amoebic strains by environmental factors requires further ecological investigations.
