A pumped storage system consists of an upper reservoir and lower reservoir separated by an elevation difference. During low demand energy periods such as nights and weekends water is pumped from the lower to the upper reservoir using available energy from conventional steam electric power plants. During high energy demand periods, such as mornings and afternoons of weekdays, upper reservoir water is allowed to drop back down through the same system of water conduits and turbines, generating electricity to conveniently meet abrupt electrical energy requirements. In February, 1982 I began a year-long study to determine if growth and reproduction in Daphnia Pulex were limited by the amount of food available in Boulder Basin, Lake Mead. To determine this, I made monthly collections of Daphnia Pulex and natural lake seston from an already established station in Boulder Basin. I cultured the Daphnia Pulex under simulated field conditions in a flow-through feeding apparatus using four different food regimes; 1-natural lake seston filtered through 80 um mesh to remove other zooplankton, 2-lake seston (as above) with an enrichment of 103 cells-ml-1 of Chlamydomonas reinhardti, 3-lake …

Analysis Of The Charleston, Illinois Water Supply For The Presence Of Organic Mutagens Utilizing The Ames Salmonella/Microsome Assay, Steven Lee Washburn

Water samples from the Charleston, Illinois water supply and selected agricultural run-off areas were tested for the presence of organic mutagenic chemicals. The Ames Salmonella/microsome mutagenicity assay utilizing strains TA98 and TA100 was used to analyze concentrated water samples. A special set of samples was exposed to the S9 activation system coupled with the TA98 strain to simulate the mammalian metabolic system.

No mutagens were detected in any of the samples tested. Several positive and negative control systems were included in the testing system. Negative control systems were used to determine the average spontaneous reversion rates for each strain for …

The response to periodic desiccation of periphyton communities developed in eddy environments or in areas exposed to variable, direct current was examined. Algal communities were incubated on clay tiles for between 2 and 12 weeks in the Colorado River, directly below Hoover Dam from October, 1983 - January, 1984. Mean daily discharge from the dam decreased over this period. To assess the influence of these discharge changes on community dynamics, samples collected over the first 12 weeks of the investigation were compared to samples of the same age collected over the final 8 weeks.

Monthly water samples collected from four sites in the Lost River Groundwater Basin, a shallow karst aquifer in the Bowling Green-Warren County area of Kentucky, represented samples from sites receiving conduit and diffuse flow. All sites were severely contaminated with bacteria, and on some occasions the surface water criteria for some heavy metals were exceeded.

Of the total 334 bacterial colonies identified 92.1% were verified as Escherichia coli by the API20E system. Zooplankton samples collected from throughout Lake Mead, in 1981-1982, demonstrate the presence of a statistically significant seasonal and spatial heterogeneity in zooplankton densities. Seasonally, the major zooplankton groups were most abundant in the spring and fall, coincident with maxima in chlorophyll-a concentrations. Successions among the various rotifers, cladocerans and copepods present in the reservoir were influenced by food availability, diapause, predation by planktivorous fish and, possibly, water temperatures. Spatial heterogeneity in zooplankton densities was unrelated to water temperature, pH, conductivity and dissolved oxygen concentrations, but was related to the abundance of phytoplankton (chlorophyll-a concentrations) and fish. At the request of the U.S. Bureau of Reclamation, Boulder City, Nevada, the U.S. Environmental Protection Agency's Environmental Monitoring Systems Laboratory at Las Vegas collected multispectral scanner imagery of Las Vegas Wash on October 1, 1982. A combined maximum likelihood classification and editing procedure was used to classify the multispectral scanner imagery into 12 categories of land cover. The classification identified four categories of marsh vegetation, one category of riparian, two categories of mixed scrub, and two desert categories. Turbid water and cultivated land formed an "other" category. Area tabulations were formed by georeferencing the classification to the Universal Transverse …

Charleston, Illinois, a city located in east-central Illinois, recently implemented a new concept in a raw water system. Due to storage capacity-sedimentation problems in the old Lake Charleston-Embarras River raw water supply, the Charleston Pump Storage Reservoir was constructed. The new system involved building a dike separating the Lake Charleston impoundment from the Embarras River. Water to increase the storage capacity of the reservoir can now be selectively pumped into the impoundment from the river.

Previous studies have shown the planktonic organisms of various natural water bodies to be quite sensitive to the levels of solar ultraviolet radiation incident at the water surface in summer. It has been assumed that the majority of the phytoplankton are simply carried about in the mass of the water which they inhabit with little control of their position or sunlight exposure. Photosynthetic microorganisms must strike a delicate balance with regard to solar radiation; they must obtain enough visible light for adequate photosynthesis while avoiding an excessive exposure to the injurious solar UV-B radiation.

A silica gel-petroleum (SGP) medium was developed which is reliable, approximately the same cost as that of the corresponding agar-based petroleum medium, and can be made sufficiently firm to use in streaking or the spread-plate technique. Advantages of the medium include: the incorporation of all of the major ions of seawater in accordance with Dittmar's Law (the salinity of which can easily be varied from 0 to 35 parts per thousand); a wide workable pH range (5.5 to 11.6); negligable pH drift(~ 0.2 pH units after 166 days); the elimination of all but negligable amounts of syneresis, the ability to …

The presence of large numbers of enteric viruses in wastewaters has resulted in concern regarding their significance and consequences for humans. Assessments of relative significance or consequences may most accurately be made following precise measurements of the range of virus types and amounts in wastewaters and sludges and after epidemiological considerations. In this study, results are presented pertaining to enteric virus isolation, concentration, and enumeration from primary wastewater sludges.

The pleomorphism of Coprinus radians was studied. The three distinct growth forms--sporophore, oidial stage, and ozonium stage--were examined in relation to ultrastructure, morphology and development. Scanning and transmission electron microscopy are correlated with photomicrographs of developmental stages. Development of the oidia and of the sporophore are of particular interest. The multinucleate oidia develop quite differently from oidia as generally described for Basidiomycete imperfect stages. Development of the oidia of this Coprinus is holoblastic and sympodial. The mushroom primordia are differentiated internally into gill, cap, and stipe hyphae while only l-2mm in diameter. At this stage they look superficially like sclerotial …

1. The waters of Las Vegas Bay, a heavily utilized recreational resource, receive discharges from a variety of municipal and industrial waste sources. The U.S. Environmental Protection Agency has determined, on the basis of numerous studies, that the present water quality violates state and federal standards and constitutes a public nuisance. Consultants have advised the Sewage and Wastewater Advisory Committee that rapid abatement of the alleged pollution conditions can be achieved by an advanced wastewater treatment (AWT) plant.

This report describes investigations into the physiology of microorganisms that are commonly involved in the oxidation of inorganic sulfur compounds. The metabolic activities of these bacteria play a potent role in several fields of economic importance such as strip mining operations, water pollution, corrosion, metallurgy, petroleum technology and soil fertility processes.

The oxidation of inorganic sulfur compounds was studied in the chemolithotrophs Thiobacillus denitrificans, Thiobacillus A2, Thiobacillus neapolitanus, and a photolithotroph Rhodopseudomonas palustris. Cell suspensions from all of the thiobacilli catalyzed completely the oxidation of sulfide, thiosulfate, and sulfite to sulfate. The oxidation of thiosulfate in R. palustris …

Phytoplankton samples were collected in Lake Mend 6 times from September 1910 to June 1971 for 8 stations at depths of 0. 3, 5, 10, 20, and 30 m. These samples were processed through a Millipore filter apparatus and 79 planktonic algae were identified. Algal divisions represented were Bacillariophyta, 42 species; Chlorophyta, 18 ; Cyanophyta, 9; Chrysophyta, 3; Cryptophyta, 3; Pyrrophyta, 2; and Euglenophyta, 2. Blue-green algae were dominant in late summer and fall; green algae, diatoms, and, cryptomonads in winter; and green algae in spring. The early summer flora was best represented by the Chlorophyta, Cryptophyta, and Chrysophyta. Palmer's …

Phytoplankton successions, applications of the general growth equation, and physical measurements have been employed to investigate events occurring at the interface between industrial and sewage effluent contained in Las Vegas Wash and the waters of Lake Mead, Nevada. The data indicate that the entering waters tend to form a density current interrupted at intervals by dynamic effects generated in the lake. The dynamic relationships described here for the spring months suggest that a much more thorough understanding of physical, chemical and biological interactions is necessary to permit solution of the numerous problems of Las Vegas Bay discussed or alluded to …

The temporal and spatial changes in chemical and biological properties of Lake Mead have been investigated, thereby indicating the sources of water pollution and the time of highest pollution potential. Planktonic organisms have been shown to indicate the presence of water problems. Macro- and micro-nutrient analyses have shown that primary productivity is not inhibited by limiting concentrations. A mathematical model has been developed, tested with one set of independent data, and shown worthy of management utility. Progressive increases in concentration of dissolved solids in the Colorado River water from Lake Powell to Imperial Dam seem to alter plankton dynamics and biological productivity of the river. Also, changes in biological productivity and micronutrients concentrations occur within the same reservoir. Development of a digital simulation model to predict micronutrients concentrations and biological productivity is necessary for diagnosing changes in plankton population and effluent-carrying capacity of the system. The microflora of Cane Branch of Beaver Creek in McCreary County, Kentucky, which drains an area that was strip-mined between 1955 and 1959, was studied and compared with that of Helton Branch which drains a comparable area where there has been no mining. Differences include: the establishment of Ferrcbacillus ferrooxidans, for which procedures were developed for direct colony isolation from the stream; fewer saprophytic bacteria; more numerous and more diversified filamentous and unicellular fungi; and characteristic differences in algal flora. Representatives of 42 genera of filamentous fungi were identified. Of these, 21 were isolated only from Cane Branch. Representatives of …

During the fall of 1959, monolayer behavior studies were made on two lakes in the southwest; Boulder Basin of Lake Mead, Nevada, and Lake Sahuaro near Phoenix, Arizona. These studies were made to evaluate the effect of geographical and climatological conditions on movement and behavior of monolayers. The Lake Sahuaro tests also were used to provide data for design and development of improved methods of application and maintenance of a film on this lake in anticipation of full scale, evaporation-reduction tests to be performed there during the summer of I960.
