Calcite deposits, composed of a mixture of calcium carbonate and silicon dioxide, were found in fractures and small cavities within the welded tuff of Yucca Mountain. This research investigation involves determining the presence of thermophilic, calciumprecipitating bacteria within these deposits. The possible existence of thermophilic bacteria may help to resolve the issue of whether these calcite deposits formed from precipitation of dissolved calcium carbonate in rain water transported from the overlying soil environment or as a result of upwelling of geothermally-heated waters transported from below the mountain. This report provides a rationale for the advances in instrumentation and understanding needed to assess claims of ancient and extraterrestrial life made on the basis of morphological biosignatures. Morphological biosignatures consist of bona fide microbial fossils as well as microbially influenced sedimentary structures. To be recognized as evidence of life, microbial fossils must contain chemical and structural attributes uniquely indicative of microbial cells or cellular or extracellular processes. When combined with various research strategies, high-resolution instruments can reveal such attributes and elucidate how morphological fossils form and become altered, thereby improving the ability to recognize them in the geological record …

Military aviation fuel systems can be an ideal environment for microorganisms. Microbial growth in hydrocarbon fuel systems arises because of the impracticality of keeping fuel tanks sterile and the inevitable presence of water from condensation. Microbial contaminants in aviation fuel systems are a concern because of their potential to degrade the fuel, accelerate tank corrosion, and threaten flight safety. This research addresses the concern of using more environmentally friendly Fuel System Icing Inhibitors (FSII), which are also biocidal. Members of the uncultivated bacterial division TM7 have been detected in the human mouth, but little information is available regarding their prevalence and diversity at this site. Human subgingival plaque samples from healthy sites and sites exhibiting various stages of periodontal disease were analyzed for the presence of TM7 bacteria. TM7 ribosomal DNA (rDNA) was found in 96% of the samples, and it accounted for approximately 0.3%, on average, of all bacterial rDNA in the samples as determined by real-time quantitative PCR. In this study, food samples were intentionally contaminated with Escherichia coli O157:H7, and then DNA was isolated by using four commercial kits. The isolated DNA samples were compared by using real-time PCR detection of the Shiga toxin genes. The four kits tested worked similarly.

Molecular investigations of microbial community structure and dynamics involve costly and time-consuming methods. This approach is limiting when rapid assessment and detection of microbial organisms are needed. In aquatic environments, especially freshwater environments which may be used as a water source, rapid detection of pathogenic microbes is essential. Likewise, monitoring for the presence or absence of various functional genes can be used to indicate the type of microbial community present in the environment of interest. In liquid culture, eight typical Gram-negative bacteria were ca. 10,000-fold more sensitive to cationic detergents than to the anionic detergent sodium dodecyl sulfate. Cetyltrimethylammonium bromide (CTAB) was inhibitory at concentrations ranging from 0.0006% to 0.01%. Four pseudomonads able to form biofilms were ca. 1000-fold more resistant to CTAB on Luria–Bertani agar plates than they were in liquid culture. A lasI mutant of Pseudomonas aeruginosa was only able to tolerate 0.1% CTAB on Luria–Bertani agar plates but could tolerate 5% CTAB when supplemented with homoserine lactone containing culture supernatants.

The surface morphology of Bacillus spores was resolved by atomic force microscopy in order to determine if characteristic surface features could be used to distinguish between closely related species. Four strains were studied: Bacillus anthracis Sterne strain, Bacillus thuringiensis var. kurstaki, Bacillus cereus strain 569, and Bacillus globigii var. niger. The spores were separated from a nutrient agar culture by filtering and centrifugation, suspended in deionized water, and immobilized on a graphite substrate by spin-coating. Atomic force microscopy imaging was done using intermittent contact mode, in air, under reduced humidity. Seven to ten spores of each species were fully characterized …

The potential for aerobic methyl tert-butyl ether (MTBE) degradation was investigated with microcosms containing aquifer sediment and groundwater from four MTBE-contaminated sites characterized by oxygen-limited in situ conditions. MTBE depletion was observed for sediments from two sites (e.g., 4.5 mg/liter degraded in 15 days after a 4-day lag period), whereas no consumption of MTBE was observed for sediments from the other sites after 75 days. For sediments in which MTBE was consumed, 43 to 54% of added [U-14C]MTBE was mineralized to14CO2. Molecular phylogenetic analyses of these sediments indicated the enrichment of species closely …

Extended tailing of low bacterial concentrations following breakthrough at the Narrow Channel focus area was observed for 4 months. Bacterial attachment and detachment kinetics associated with breakthrough and extended tailing were determined by fitting a one-dimensional transport model to the field breakthrough-tailing data. Spatial variations in attachment rate coefficient (k(f)) were observed under forced gradient conditions (i.e., k(f) decreased as travel, distance increased), possibly because of decreased bacterial adhesion with increased transport distance. When pore water velocity decreased by an order of magnitude at 9 days following injection, apparent bacterial attachment rate coefficients did not decrease with velocity as expected …

Streptococcus iniae is a gram-positive organism responsible for causing disease in both freshwater as well as saltwater fish. Every year fisheries lose fish by the tons due to diseases caused by this organism. In 1991, the first reported human case of disease associated with this organism was described. Since this initial case, there have been numerous other reported cases of S. iniae infections. This organism is a catalase negative, facultatively anaerobic organism that produces a capsule and when plated onto blood agar demonstrates β-hemolytic activity.

During amphibian somitogenesis, presumptive myotome cells change shape from round or polygonal to elongated and aligned parallel to the notochord (for reviews see Radice, et al., 1989; Keller, 2000). Although the final orientation of myotomal cells is always axial, the movements that achieve this final arrangement can differ greatly between species. The simplest movement is that seen in Bombina variegata, Pelobates fuscus, and Bufo bufo (Brustis et al., 1976; Brustis, 1979; Kielbowna, 1981 ). In order to determine the prevalence and distribution of the human pathogen, Escherichia coli O157:H7, in free-ranging deer, hunters were asked to collect and submit fecal samples from deer harvested during a regular firearm season (14–22 November 1998). Prior to the season, 47% of the hunters with permits in the southeastern Nebraska (USA) study area indicated a willingness to participate in the study. Approximately 25% of successful hunters in the area submitted deer fecal samples. Escherichia coli O157:H7 was cultured from four (0.25%) of 1,608 total samples submitted. To investigate temporal (ca 1 mo) and spatial dynamics of bacterioplankton in the Caribbean Sea, time-series measurements of chlorophyll a concentration, bacterial abundance, and thymidine incorporation rate were performed at 6 stations near Barbados, In addition, a series of incubation experiments was carried out with seawater samples collected from various depths and stations to determine bacterial growth rates and removal rates by bacterivory. Vertically integrated phytoplankton and bacterioplankton biomasses averaged 1290 and 1200 mg C m(-2), respectively, and heterotrophic bacterial production was 106 mg C m(-2), with in situ bacterial growth of 0.1 d(-1), Approximately halfway through the sampling period, …

Wild and domesticated animals can harbor a pathogenic Escherichia coli strain designated as O157:H7. Potential health problems could occur if strain O157:H7 is a more robust survivor in defecated waste than commonly used indicator bacteria. A laboratory study was conducted to assess E. coli O157:H7 survival relative to a nonpathogenie E. coli strain in two soils with different physical and chemical characteristics. Bacteria in the inoculated soils were enumerated on a weekly basis for 8 wk using a most probable number (MPN) technique. Archaea are traditionally thought of as “extremophiles,” but recent studies have shown that marine planktonic Archaea make up a surprisingly large percentage of ocean midwater microbial communities, up to 60% of the total prokaryotes. However, the basic physiology and contribution of Archaea to community microbial activity remain unknown. We have studied Archaea from 200-m depths of the northwest Mediterranean Sea and the Pacific Ocean near California, measuring the archaeal activity under simulated natural conditions (8 to 17°C, dark and anaerobic) by means of a method called substrate tracking autoradiography fluorescence in situ hybridization (STARFISH) that simultaneously detects specific cell types …

This study compares two different analytical methods for the determination of coliform bacteria within natural waters. The two methods are: 1) membrane Filtration, and 2) multiple tube fermentation. Water samples from ten different locations within the Appomattox River watershed were collected by technicians from the Piedmont Soil and Water Conservation District and the Piedmont Region TMDL Initiative in Farmville, Virginia.

Resistance to antimicrobial agents used to treat severe Campylobacter spp. gastroenteritis is increasing worldwide. We assessed the antimicrobial resistance patterns of Campylobacter spp. isolates of human and animal origin. More than half (n = 32) were resistant to sulphonamide, a feature known to be associated with the presence of integrons. Analysis of these integrons will further our understanding of Campylobacter spp. epidemiology.

The effect of humic acid (HA) on uptake and transfer of Cu by selected marine organisms from the microbial loop was determined. Bacteria grown with and without 15 µg Cu l-1 and with and without 10 mg Suwannee River Humic Acid (SRHA) l-1 were fed to Uronema sp. The Uronema were subsequently fed to Acartia tonsa to determine the effect of humic acid on the uptake and transfer of Cu from bacteria to copepods. The purpose of this study was to develop a more efficient method to test for phenol in both wastewater and non-wastewater. Phenol is a caustic and toxic contaminant that may be present in our water supplies. Phenol is used in various resins, pharmaceuticals, plastics and disinfectants. Environmental Protection Agency methods are available to test for phenol using gas chromatography along with mass spectrometry or flame ionization detection. Siderophores are low molecular mass compounds used by many microorganisms to scavenge dissolved iron, which is typically rare in environments. Pseudomonas aeruginosa (PAO1) was exposed to metals and methyl viologen in low-iron medium in order to monitor the abundance of a specific siderophore (pyoverdine) and monitor growth over time. Sequence analysis of the 330-kb genome of chlorella virus Paramecium bursaria chlorella virus 1 (PBCV-1) revealed an open reading frame, A237R, that encodes a protein with 34% amino acid identity to homospermidine synthase from Rhodopseudomonas viridis. Expression of the a237r gene product in Escherichia coli established that the recombinant enzyme catalyzes the NAD+-dependent formation of homospermidine from two molecules of putrescine. The a237r gene is expressed late in PBCV-1 infection. Both uninfected and PBCV-1-infected chlorella, as well as PBCV-1 virions, contain homospermidine, along with the more common polyamines putrescine, spermidine, and cadaverine. Human and livestock exposure to fecal pathogens via contaminated surface or groundwater is an important water quality concern for soils receiving animal wastes. The effects of manure application timing (spring or fall application) and soil management (no-tillage or conservation tillage) on fecal bacteria infiltration through shallow karst soils in central Kentucky (the Bluegrass region) have not been evaluated. We performed a field experiment to measure fecal coliforms and fecal streptococci in leachate from dairy manure-amended no-tillage and conservation tillage soils. Vertical profiles of virus-like particles (VLPs) and bacteria were determined by near-synoptic sampling through the water column and 15 to 25 cm into the sediment at 5 stations across the mouth of Chesapeake Bay, USA. VLPs were about 10 times more abundant in the pore water (grand mean = 3.6 x 10(8) VLPs ml(-1)) than in the water column (grand mean = 3.8 x 10(7) VLPs ml(-1)). Similarly, bacteria counts were about 3 times higher in the pore water (grand mean = 6.5 x 10(6) bacteria ml(-1)) than in the water column (grand mean = 2.4 x 10(6) bacteria ml(-1)). …

The transcriptional response of freshwater Synechococcus sp. PCC 7942 and oceanic Synechococcus sp. WH 7803 to ammonium, nitrate or iron was studied in single or multiple factor limited cultures. Both strains showed maximum production of NiR mRNA when grown in nitrate-containing media. When grown in ammonium-containing medium, they did not show any signal for NiR mRNA synthesis. The influence on the transcription of NiR mRNA by iron as sole limiting nutrient was also evaluated. Iron increased the NiR mRNA whether or not the positive effect of nitrate was already present. The focus of this study is to assess the efficacy of fecal coliform bacteria as a microbiological water quality indicator. The scientific and educational context of fecal coliform bacteria is explored through analysis of large sets of water quality data, focused field monitoring projects, and review of commonly used resources for school-based and volunteer water-related studies. Analyzed data sets include long term sampling (10 years), multiple sites, daily samples, storm event samples, and other parameters monitored in parallel with bacteria. Poultry production in Kentucky increased almost 200% between 1991 and 1995. Their waste is typically land applied, and fecal pathogen runoff and infiltration may cause nonpoint source groundwater pollution. We looked at the preferential flow of fecal coliforms through undisturbed soil blocks since fecal bacteria typically infiltrate the soil profile to contaminate groundwater. Poultry manure was uniformly distributed on top of sod-covered or tilled (upper 12.5 cm) soil blocks and the blocks were irrigated.
