Microsatellites Reveal The Population Genetic Structure Of Yellow Perch In Minnesota, Megan Boche

When analyzing a population’s genetic makeup, looking at variation of individuals and populations is a crucial factor. The management of yellow perch will be aided by this information particularly in regard to stocking or relocation. Microsatellite loci were used to analyze the genetic population structure of yellow perch Perca flavescens within and among 29 sampling areas throughout Minnesota’s basins. Seven microsatellite DNA loci were evaluated, with heterozygosities ranging from 0.41 to 0.78. The Lower Mississippi basin samples showed higher numbers of alleles per locus and greater expected heterozygosities compared to other basins, with Lake Pepin (PEP) exhibiting the highest average …

The continual habitat destruction and draining of wetland systems makes the need to understand the ecology of roadside wetlands all the more necessary. Roadside wetlands provide valuable habitat to countless aquatic invertebrates, which can provide insight into the health of the wetland system due to their diversity. The objective of this study was to analyze the populations of aquatic invertebrates in roadside wetlands in relation to water characteristics such as pH, salinity, conductivity, water depth and temperature, and dissolved oxygen. Training in specialized techniques and instrumentation is an important component of undergraduate and graduate education. While such training can occur in formal lab courses, it also occurs as students engage in research. Although training in a research lab setting is typically informal, implementing evidence-based practices from the classroom would be beneficial, particularly for learning complex techniques and instruments, for example, light microscopy. Using advanced microscopes for research is more complicated than using teaching microscopes in a course. Fish size structure is an important factor in a fishery population. Panfish such as black crappie Pomoxis nigromaculatus and bluegill Lepomis macrochirus play a major role in the health of fisheries. Five lakes from southern, northern, eastern, and western Minnesota were selected for analysis. Data were collected using standard sampling methods, including gill nets, electrofishing, and fyke nets. Length-frequency distributions were created to assess size structure in each region. Relative abundance was measured using catch per unit effort. Data analysis included a one-way ANOVA to compare Proportional Size Distribution (PSD) values and the distribution of fish sizes among regions. Chi-square …

Panfish including bluegill Lepomis macrochirus and black crappie Pomoxis nigromaculatus are gamefish species of significance found in Minnesota. Hoop nets and trap nets are two types of gear that can be used to sample these fish. Fisheries managers and anglers alike are interested in the best tactics to capture panfish. The behavior of fish, including bluegill and black crappie, change throughout the sampling season and make it difficult to assume accurate representations. Microplastics, plastic particles smaller than 5 mm, are an increasing environmental threat, particularly due to their widespread distribution and toxicity. While much research has focused on larger freshwater and marine ecosystems, the presence and impacts of microplastics in smaller inland lakes remain less understood. This study investigates the relationship between plant density and microplastic concentrations in Lake Bemidji, a smaller freshwater lake. A total of 30 water samples were collected across varying vegetation conditions, ranging from open water (0%) to 90% plant coverage. The North American beaver Castor canadensis is an ecosystem engineer and a keystone species to the environment. Beaver inhabit ponds and construct dams to build ponds that provide habitat for certain fish. Little information is provided on the species of fish that live in beaver ponds. The objective of this study is to sample the fish communities in beaver ponds while comparing the effectiveness of two styles of minnow traps. How white-tailed deer use the landscape in an urban environment can help us understand the effects of urbanization on deer populations. White-tailed deer form observable matriarchal hierarchies in studied populations. This study examines the matriarchal structure, home range size, and home range composition of 25 collared white-tailed deer within the city limits of Bemidji, MN. The results found no significant difference between age class and home range size or composition. However, movement metrics from yearling and mature age classes were more similar than the juvenile age class females. I propose this is because social acceptance between mature females was higher …

This study was aimed at setting the stage for future research which might determine if U. macrorhiza could be a biological method of control for invasive species. Acute diabetes characterized by high spikes in blood glucose levels in association with severe hyperglycemia can lead to an increased risk of ischemic stroke in the brain. To better understand high blood glucose levels, hyperglycemia, in the brain we explored its effects on endothelial cells. Endothelial cells line the inner walls of blood vessels and are directly impacted in hyperglycemia. Brain endothelial cells are a major component of the blood brain barrier (BBB) which tightly regulates the brain’s microenvironment. The depletion of the stratospheric ozone layer has led to increased levels of harmful UV-B radiation reaching the Earth's surface, posing significant challenges for plant health and development. This study investigates the oxidative stress responses in Arabidopsis thaliana when exposed to UV-B irradiation. Arabidopsis thaliana, a well-established model organism, is particularly sensitive to UV-B, making it ideal for understanding the effects of UV-B-induced oxidative stress. UV-B radiation, a mutagenic component of sunlight, disrupts key metabolic, molecular, and developmental processes, leading to DNA damage, impaired metabolism, and morphological changes. The Louisiana Graduate Network in Applied Evolution (hereinafter; LAGNiAppE) was funded by a grant awarded through the National Science Foundation (NSF 22-506), as part of the Research and Mentoring for Postbaccalaureates in Biological Sciences (RaMP) grant program. The LAGNiAppE program provides training and mentorship to cohorts of recent college graduates from underrepresented groups interested in advancing their knowledge and research skills in evolutionary biology. The association between sleep and the immune-endocrine system is well recognized, but the nature of that relationship is not well understood. Sleep fragmentation induces a pro-inflammatory response in peripheral tissues and brain, but it also activates the hypothalamic-pituitary-adrenal (HPA) axis, releasing glucocorticoids (GCs) (cortisol in humans and corticosterone in mice). It is unclear whether this rapid release of glucocorticoids acts to potentiate or dampen the inflammatory response in the short term. Cranial radiation therapy plays an integral role in the treatment of brain tumors but can lead to progressive cognitive deficits in survivors by mechanisms that are poorly understood. To develop preventive or mitigative strategies, it is crucial to better understand the underlying pathogenesis of radiation-induced cognitive impairments. The study investigated single-cell transcriptomics and DNA methylation changes as potential drivers of persistent cellular dysfunction after radiation exposure, specifically concentrating on the CA1-3 regions of the hippocampus and the prefrontal cortex due to their role in cognitive functions. During asymmetric cell division, cell polarity and cell cycle are tightly coupled to ensure robust segregation of cell fate determinants and generation of cellular diversity. In the one-cell C. elegans embryo, multiple cell cycle kinases regulate cell polarization and posterior segregation of germline fate determinants (germplasm). For example, PLK-1 kinase inhibits the retention of its germplasm substrate POS-1 in the anterior, driving POS-1 segregation to the posterior (Han et al., 2018). In this study, we investigated the color-changing capabilities of Trinidadian killifish, Anablepsoides hartii, through exposure to varied color conditions over defined periods of time. Our experiment aims to understand adaptive responses when killifish are subjected to different color backgrounds, exploring their mechanisms to change color. Mangroves serve as important nursery habitats for several species of subtropical fishes and invertebrates. Southeastern Florida mangrove environments have experienced an increasing frequency of strong hot and cold weather front systems, but little information is known about the temperature thresholds for mangrove-associated prey species that lethally disrupts homeostasis. To assess the vulnerability of mangrove-associated prey species to these short-duration temperature fluctuations, critical thermal methodology (CTM) was used to determine thermal tolerances for two fishes and two invertebrates, using loss of equilibrium (LOE) as the endpoint to determine CTmax and CTmin values as a survivable proxy for death. Recent work has shown that bats in Colorado and Kansas are more active over prairie dog colonies. It is hypothesized that the reason for this is increased abundance and richness of insects found on colonies versus off colonies. Sampling from 2022 indicated that insect biomass was not different between location types, but suggested that the relative abundance of insect orders may differ. Anaplastic thyroid cancer is a rare, fatal cancer with a five-year survival of 4%. Universally diagnosed at stage IV, anaplastic thyroid cancer is characterized by its lack of differentiation, rapid proliferative rate, highly inflammatory tumor microenvironment, and metabolic dysregulation. Refractory to all established therapies, anaplastic thyroid cancer requires a novel therapeutic approach that targets all of these drivers of anaplastic thyroid cancer carcinogenesis. We propose natural alkaloid berberine as a therapeutic with multitarget efficacy to alter mitochondrial metabolism and reprogram anaplastic thyroid cancer’s aggressive phenotype. The endangered Blanding’s turtle (Emydoidea blandingii) is a species with a generally diverse diet in their freshwater wetland habitats; they consume prey such as crayfish and snails. In captivity, these options often become more limited due to logistical reasons. As ex situ conservation efforts such as captive rearing and head-starting are becoming more popular for this species, it will be important to gain more insight on how these more limited diets might affect these turtles. Methamphetamine (METH) abuse remains a global health concern, with emerging evidence highlighting its genotoxic potential. In the central nervous system METH enters dopaminergic cells primarily through the dopamine transporter (DAT), which controls the dynamics of dopamine (DA) neurotransmission by driving the reuptake of extracellular DA into the presynaptic neuronal cell. Additional effects of METH on the storage of DA in synaptic vesicles lead to the dysregulated cytosolic accumulation of DA. Previous studies have shown that after METH disrupts intracellular vesicular stores of DA, the excess DA in the cytosol is rapidly oxidized. This generates an abundance of reactive oxygen species …

Adfluvial Brook Trout Salvelinus fontinalis are known to typically grow larger than riverine Brook Trout. The Kabekona River in Hubbard County, Minnesota exhibits suitable habitat for both fluvial and adfluvial Brook Trout populations. The objective of this study was to analyze the relationships between depth, stream width, canopy cover, water temperature, sediment size, and distance from Kabekona Lake on Brook Trout size distribution in the Kabekona River. Walleye Sanders vitreus for many states, is a key attractant of anglers from around North America. Due to immense popularity, management efforts have been conducted to maintain Walleye populations, especially in the form of stocking. The purpose of this study is to analyze the effect of Walleye fry stocking on gillnet catch per unit effort (CPUE). A total of 90 lakes were analyzed for this study within three defined regions of Minnesota, 30 from each region.
