Life history morph, sex, and body condition are traits that may influence stress within salamander populations because of differences in physiology and environmental conditions. Given widespread declines and the effects chronic stress can have on amphibian health, it is important to understand within-population drivers of stress and how population level variation may influence population viability. The 14-3-3 (YWHA) proteins are homologous, ubiquitous, and conserved in most organisms ranging from plants to animals and play important roles in regulating key cellular events such as cell signaling, development, apoptosis, etc. These proteins consist of seven isoforms in mammals, termed under Greek alphabetization: beta (β), gamma (γ), epsilon (ε), eta (η), tau/theta (τ), sigma (σ), and zeta (ζ). Each of these isoforms can interact with a plethora of binding partners and has been shown to serve a distinct role in molecular crosstalk, biological processes, and disease susceptibility. Largemouth Bass Micropterus salmoides and Smallmouth Bass Micropterus dolomieu are two of the most popular species of fish for anglers to target in Minnesota. In this study, Largemouth Bass and Smallmouth Bass stress was characterized by handling time and recovery time. This study compares handling time and recovery time between barbed and barbless hooks. For Largemouth Bass, there was a significant relationship between recovery time and time out of the water for barbed (P = 0.02, R2 = 0.86) and barbless hooks (P < 0.01, R2 = 0.74). For Smallmouth Bass, there was no significant relationship between recovery time and time …

Aquatic macroinvertebrates bridge the gap in part between primary and secondary consumers, making them ecologically important for most aquatic systems. They are used as biotic indicators for water quality due to their sensitivity to pollutants and may be affected, specifically, by chloride ion concentrations in the water. Water quality can be evaluated by looking at species compositions and their respective tolerance values. Amphipod occurrence and abundance has declined in the Prairie Pothole Region (PPR). Wildlife managers have stocked prairie pothole wetlands with Gammarus lacustris since 2018 to establish self-sustaining populations. G. lacustris have been detected in only 2 of 22 stocked basins, suggesting that they have died or failed to reproduce in most stocking attempts. Aquatic plant surveys were conducted in stocked wetlands and compared to data from surveys of wetlands with naturally occurring populations to understand if G. lacustris would be expected to survive in stocked wetlands based on characteristics of the aquatic vegetation communities. This study seeks to establish a relationship between a fish’s total length to the size of various bony structures of prey fishes. Three important prey fish species in Minnesota were selected for examination in this study, including Black Crappie Pomoxis nigromaculatus, Bluegill Lepomis macrochirus, and Yellow Perch Perca flavescens. Length and weight dimensions of each fish’s otoliths, cleithra, and vertebrae were associated to total lengths of the fish they came from. In many dietary studies, the need for these relationships is strongly related to the disfigurement or degradation of prey fish in stomach samples. Burbots have become an increasingly popular game fish among anglers and have become a strong predator within the systems, consuming a wide variety of organisms. Therefore, the objective of this study was to compare male and female burbots diets post-spawn in a system. Burbots angled from Bad Medicine Lake were sacrificed and the stomach contents were observed, categorized by the lowest taxonomic key, and then weighed in grams. Effectiveness Of Endothall Treatment In Reducing Potamogeton Crispus In Lake Julia, Jake Sorensen

Curly leaf pondweed Potamogeton crispus, is a submerged aquatic plant non-native to North America. It was most likely introduced when common carp Cyprinus carpio were intentionally brought to the Midwest. Endothall is an aquatic herbicide commonly used to control curlyleaf pondweed in lakes. This study focused on collecting data from Lake Julia in Sherburne County, MN. Lake Julia is treated annually in the spring to control curlyleaf pondweed. The results show treatment areas saw a significant increase in vegetation density compared to control areas (P < 0.01). Changes in percent vegetation coverage by date were insignificant (P = 0.24). No significant signs of fish kills were observed. Because this study looked at overall vegetation coverage instead of specifically curlyleaf pondweed, we cannot conclude the increase in vegetation coverage was from curlyleaf pondweed alone. Based on the results from Poovey et al. (2002) we would expect lower vegetation coverage in treatment areas instead of the increased vegetation coverage observed in our study. This suggests the possibility that the endothall treatment successfully killed curlyleaf pondweed in treated areas, which allowed for new growth of submerged aquatic plants to take place as water temperatures increased.

Increasing global temperatures have been linked to shifts in the distributions and preferred microhabitats of terrestrial species. This trend is predicted to impact tropical forest insects, which may already be exposed to conditions that bring them close to their thermal maxima. Tropical microhabitats are linked to vertical stratification, because the canopy is hotter, winder, and less humid than the ground. In ectothermic organisms, such as beetles, thermoregulation is dependent on morphological and behavioral traits. Sandusky Bay is largely populated by cyanobacterial algal blooms, mainly formed by Planktothrix. Fertilizers containing nitrogen and phosphorus run from agricultural lands into the bay, building up excess nutrients forming eutrophic waters. The Planktothrix feed off these nutrients and grow into algal blooms. To determine a potential solution to the growth of these blooms, I implemented a hydroponics system involving Lactuca Sativa for analysis of Planktothrix growth and productivity. A powerful way to evaluate scientific explanations (hypotheses) is to test the predictions that they make. In this way, predictions serve as an important bridge between abstract hypotheses and concrete experiments. Experimental biologists, however, generally receive little guidance on how to generate quality predictions. Here, we identify two important components of good predictions – criticality and persuasiveness – which relate to the ability of a prediction (and the experiment it implies) to disprove a hypothesis or to convince a skeptic that the hypothesis has merit. This research assessed plankton dynamics in two urban, subtropical lakes (University Lake and City Park Lake) shortly before a proposed and approved lake restoration project. Plankton dynamics were determined in University Lake by the dilution method to attempt to quantify growth rates of phytoplankton and grazing rates of zooplankton. Landry and Hassett’s (1982) dilution method has been widely used in marine systems to estimate rates of growth and grazing. In many marine systems, nutrients must be added to prevent nutrient limitation. However, it is assumed, due to the hypereutrophic conditions, that nutrients would not be a limitation in University Lake. …

Eukaryotic cell division is an essential process that is carried out by the cell cycle, a tightly controlled process that has been extensively studied in the budding yeast Saccharomyces cerevisiae. The cell cycle is driven by Cyclin Dependent Kinase (Cdk1) activity. Protein phosphatase 2A-Cdc55 (PP2ACdc55) reverses Cdk1 phosphorylation events during late stages of the cell cycle to ensure the correct order of events. This thesis presents evidence that the anaphase inhibitor Pds1 is a PP2ACdc55 target. Pds1 binds to and inhibits separase (Esp1). Esp1 triggers sister chromatid segregation by cleaving the cohesin complex that holds the …

Canine heartworm disease is caused by the filarial nematode Dirofilaria immitis, which is transmitted by at least 25 known species of mosquito vectors. This study sought to understand which species of mosquitoes are present in Bulloch County, Georgia, and which species are transmitting canine heartworm disease. This study also investigated whether particular canine demographics correlated with a greater risk of heartworm disease. Surveillance of mosquitoes was conducted in known heartworm-positive canine locations using traditional gravid trapping and vacuum sampling. Microglia are the macrophages of the brain and spinal cord, playing an important role in the immune response to disease states of the nervous system. This study conducts an investigation on the activity of microglia in response to low grade neuroepithelial tumors. Using mouse models and microglial markers, a qualitative and quantitative analysis of microglia activation, migration, and invasion within the brain cortex during early stages of tumor development was conducted. The mammalian lung is a complex system of specialized cell types with precise spatial organization designed to cooperate to perform gas exchange. The classic genetics experiment by Salvador Luria and Max Delbruck showed that mutations are present prior to selection rather than arising in response to selective pressures. In this experiment, we used Escherichia coli as our model organism and the antibiotics streptomycin and rifampicin to show students in a laboratory setting how this hypothesis can be applied. Specifically, we optimized antibiotic concentrations and the amount of bacterial cells to use in order for the activity to run smoothly and consistently in a student laboratory. Plant secondary metabolites such as terpenes, phenolics, glycosides, and alkaloids play various functional roles including pigmentation, foliar and floral volatile synthesis, hormonal regulation, and direct and indirect defenses. Among these, phenolic compounds are commonly found in plants, but vary in the distribution of their specific compounds among plant families. Polyphenols, including anthocyanins and tannins, are widely distributed and have been well documented for their roles- primarily in plant pigmentation and also in plant defenses. Undergraduate biology education has changed over the past decade, incorporating an iterative and evidence-based approach. Many educational assessments have confirmed the effectiveness of integrating authentic research and open-ended inquiry into introductory biology courses, demonstrating a significant positive impact on students’ learning and attitude towards STEM majors. Despite these findings, only a handful of Biology instructors in 2-year colleges adopt this approach, and when adopted, most activities constitute a small fraction of these courses. I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

Nanotechnology takes advantage of cellular biology’s natural nanoscale operations by interacting with biomolecules differently than soluble or bulk materials, often altering normal cellular processes such as metabolism or growth. To gain a better understanding of how copper nanoparticles hybridized on cellulose fibers called carboxymethyl cellulose (CMC) affected growth of Saccharomyces cerevisiae, the mechanisms of toxicity were explored. Multiple methodologies covering genetics, proteomics, metallomics, and metabolomics were used during this investigation. The work that lead to this dissertation discovered that these cellulosic copper nanoparticles had a unique toxicity compared to copper. Acute lung injury affects around 200,000 critically ill patients annually, with a mortality rate near 33%. With a lack of targeted therapies, it is vital to discover and understand the fundamental cellular processes that drive macrophage inflammation. In the field of macrophage immunometabolism, it is accepted that macrophages upregulating glycolysis indicates a role in proinflammatory processes, however, there is a lack of knowledge surrounding the intermediate pathways in glycolysis. This study examined the effects of serine, glycine, one-carbon metabolism inhibition on LPS-induced inflammation in bone marrow-derived macrophages.
