Context: Low current intensity iontophoresis treatments have shown an increase in skin perfusion over 700% from baseline potentially increasing drug clearance from the targeted area. Objective: To determine the effects of a 10-minute ice massage on subcutaneous dexamethasone sodium phosphate (Dex-P) concentration and skin perfusion during and after a 4 mA iontophoresis treatment. Design: Controlled laboratory study. Setting: Research Laboratory. Patients or Other Participants: 26 individual participants (Males = 15, Females = 11, age = 25.6 ± 4.5 y, height = 173.9 ± 8.51 cm, mass = 76.11 ± 16.84 kg). CONTEXT: Nitric Oxide (NO) is an endothelial-derived vasoactive molecule that causes an increase in blood flow and oxygen delivery to tissue. A reduction in NO bioavailability has been found to occur in adults over the age of 60 and can be reversed pharmacologically by improving NO synthase (NOS) activity. Reversing these age-related changes with alternative interventions, such as aerobic exercise, has shown some promising results. OBJECTIVE: To quantify blood NO-bioavailability (as measured by blood nitrite levels) in a population of aerobically trained elderly men and compare these data to a group of age-matched, inactive individuals. Study Design: Controlled laboratory study. Objectives: To determine the time course of dexamethasone sodium phosphate (Dex-P) iontophoresis delivery to underlying tissues using microdialysis. Background: The efficacy of iontophoresis at delivering Dex-P through the skin is unknown in humans because of the lack of minimally invasive measurement techniques. Context: Transdermal delivery of hydrocortisone by phonophoresis is used for the treatment of musculoskeletal conditions. Research shows hydrocortisone and other white or opaque topical preparations transmit ultrasound energy poorly. Effective transmission of ultrasound is important in phonophoresis. Main Outcome measured: Samples of subcutaneous interstitial fluid were collected during and for 20 minutes following phonophoresis treatment. Cortisol concentrations were analyzed by an enzyme linked immune-assay (ELISA) test. Objective: Determine the subcutaneous cortisol concentration after two different phonophoresis treatments using a 2.5% hydrocortisone preparation. The purpose of this study was to characterize a novel technique to study neurogenic control of cutaneous vasodilation. We monitored skin blood flow (SkBF) during in-vivo electrical stimulation (e-stim) intended to activate cutaneous nerves and used intradermal microdialysis to deliver receptor antagonists to characterize their contribution to cutaneous vasodilation. We examined the role of acetylcholine receptors (RACh), nitric oxide (NO), and vasoactive intestinal peptide receptors (RVIP) on the cutaneous vasodilation induced by e-stim in the absence of the sympathetic adrenergic nervous system. Objective: Determine if varying ultrasound frequency affects the delivery of 10% hydrocortisone concentrations during phonophoresis. Utilize intramuscular microdialysis probe for drug collection, thus improving the experimental model. Methods: Thirty one (10 in groups 1 and 2, 11 in group 3) healthy subjects participated in this study. Interventions: Subjects were randomly assigned to one of three treatment groups receiving 10 minute ultrasound treatments applied to a standardized area of the gastrocnemius muscle of the right leg. The ultrasound was performed over the treated area using a 10% hydrocortisone compound mixed with standard ultrasound gel. The contralateral limb served as the control …
