During a long-term survey of the parasites of mammals from all over Bolivia, 44 individuals of Oxymycterus inca and 6 of Oxymycterus paramensis were collected and examined for parasites from the foothills and Eastern Cordillera (Los Yungas) of the Andes of Bolivia. Three species of nematode were found including, from the cecum, a previously unknown genus and species of pinworm, Caroloxyuris boliviensis n. gen, n. sp., aspidoderids representing Nematomystes rodentophilus, and from the stomach, Protospirura numidica criceticola. These helminths occurred in prevalences of 25, 9, and 14%, respectively, in O. paramensis. Caroloxyuris boliviensis resembles species included in …

The BCL-2 family member BAX plays a critical role in regulating apoptosis. Surprisingly, bax-deficient mice display limited phenotypic abnormalities. Here we investigate the effect of BAX on infection by the sexually transmitted pathogen,Chlamydia muridarum (the mouse pneumonitis strain ofChlamydia trachomatis). Bax −/−cells are relatively resistant to Chlamydia-induced apoptosis, and fewer bacteria are recovered after two infection cycles from Bax −/− cells than from wild-type cells. These results suggest that BAX-dependent apoptosis may be used to initiate a new round of infection, most likely by releasingChlamydia-containing apoptotic bodies from infected cells that could be internalized by neighboring uninfected cells. Nonetheless, infected …

Cyclosporin A (CSA) is an immunosuppressor used in organ transplantation. A recent proteomic analysis has revealed that activation of T cells in the presence of CSA induces the synthesis of hundreds of new proteins. Here we used representational difference analysis to characterize some of the corresponding induced genes. After cDNA bank screening we focused on one of these genes, which we named CSA-conditional, T cell activation-dependent (CSTAD) gene. This gene produces two mRNAs resulting from alternative splicing events. They encode two proteins of 104 and 141 amino acids, CSTADp-S and CSTADp-L, for the short and long forms, respectively. FK506 had …

Host Specificity Among Ancyrocephalinae (Monogenoidea) Of Nebraska Sunfish, Megan R. Collins, John J. Janovy Jr.
John J. Janovy, Jr. Publications
This study addressed the problem of local patterns of host specificity among Ancyrocephalinae (Monogenoidea) on bass and sunfish species, when the hosts occur in different species combinations in separate ponds. One hundred fifty-three fish of the Centrarchidae, from four study sites in Nebraska, were collected. Host species included bluegill (Lepomis macrochirus), green sunfish (L. cyanellus), largemouth bass (Micropterus salmoides), black crappie (Pomoxis nigromaculatus), white crappie (P. annularis), and rock bass (Ambloplites rupestris). These fish occurred in different species combinations, depending on the pond sampled. Results indicated that several …

A molecular paleoparasitological diagnostic approach was developed for Enterobius vermicularis. Ancient DNA was extracted from 27 coprolites from archaeological sites in Chile and USA. Enzymatic amplification of human mtDNA sequences confirmed the human origin. We designed primers specific to the E. vermicularis 5S ribosomal RNA spacer region and they allowed reproducible polymerase chain reaction identification of ancient material. We suggested that the paleoparasitological microscopic identification could accompany molecular diagnosis, which also opens the possibility of sequence analysis to understand parasite-host evolution.

The present study was undertaken to analyze the regulatory T cells generated in response to class I derived self-I-A beta(g7) (54-76) peptide. It was observed T cells from young unprimed type 1 diabetes (T1D) prone NOD mice did not respond to self-I-A beta(g7) (54-76) peptide although T cells from primed young NOD mice showed a strong response. T cells from young unprimed BALB/c mice responded to self-I-A beta(d) (62-78) peptide. However, a breakdown of tolerance to these peptides was observed with age in both the strains. Culture supernatant from I-A beta(g7) (54-76) peptide-primed cells secreted large amounts of TGF-beta and …

The ability to regulate cell volume is an ancient conserved trait present in essentially all species through evolution. The maintenance of a constant cell volume is a homeostatic imperative in animal cells. Changes in cell water content affecting the concentration of intracellular messenger molecules impair the complex signaling network, crucial for cell functioning and intercellular communication. Although the renal homeostatic mechanisms exert a precise control of extracellular fluid osmolarity, this is challenged in a variety of pathological situations. The intracellular volume constancy is continuously compromised by the generation of local and transient osmotic microgradients, associated with nutrients uptake, secretion, cytoskeleton …

The northern biotype of Echinococcus granulosus occurs throughout the holarctic zones of tundra and taiga, from eastern Fennoscandia to the Bering Strait in Eurasia and in North America from arctic Alaska approximately to the northern border of the United States. The cycle of the cestode is complex in taiga at lower latitudes, because of the greater diversity of potential hosts. In the Arctic and Subarctic, however, four patterns of predator/prey relationships may be discerned. Two natural cycles involve the wolf and wild reindeer and the wolf and elk (moose), respectively. Where deer of the two species coexist, both are prey …

We report the use of six oligoprobes designed from intergenic spacer region sequences to identify fourth-stage larvae (L4) of the tribe Cyathostominae. Oligoprobes were designed for identification of the following species: Cylicocyclus ashworthi, Cylicocyclus nassatus, Cylicocyclus insigne, Cyathostomum catinatum, Cylicostephanus goldi, and Cylicostephanus longibursatus. A seventh probe was designed as a positive control to identify all these members of the Cyathostominae. The intergenic spacer region was amplified by PCR using conserved primers. Initially, three oligoprobes were used in Southern blot analysis. To facilitate high-throughput identification, these and a further four oligoprobes were developed for use in a PCR–ELISA. …

Both soluble and cell-mediated components are involved in the innate immune response of arthropods. Injection of Borrelia burgdorferi, the Lyme disease agent, results in the secretion of defensin into the hemolymph of the ixodid tick, Dermacentor variabilis. The presence of the peptide is observed as early as 15 min post-challenge and remains present through 18 h post-challenge. As observed in insects and soft ticks, the transcript for defensin is detected as early as 1 h post-challenge in D. variabilis. RT-PCR resulted in an amplicon of 624 bp with a 225 bp region that translates to a 74 …

Use Of Irradiated And Formalin-Fixed Trichomonas Vaginalis To Examine Protective Immune Responses In The Mouse Intraperitoneal Model, Terryl L. Hostetler, James J. Daly, Timothy J. O'Brien, Max L. Baker
Journal of the Arkansas Academy of Science
Gamma irradiated Trichomonas vaginalis was used to initiate a protective immune response in mice by intraperitoneal inoculation using host mortality as the measure of virulence. Irradiated trichomonads of a virulent strain gave some immune protection but no more so than the use of formalin-fixed, virulent trichomonads. A formalin-fixed virulent strain combined with complete Freund's adjuvant (CFA) gave complete protection. Metabolically produced antigens do not appear to be important in conferring protective immunity in these experiments. A common laboratory strain of T. vaginalis (ATCC 30001) was used as an avirulent control. It gave no protection against a virulent strain. Combining CFA …

Swelling subsequent to hyposmotic conditions activates a process of volume regulation present in most cell types. This volume adjustment is accomplished by osmolyte translocation toward the extracellular space to reach a new osmotic equilibrium. Molecules involved in this homeostatic mechanism have been broadly classified into two categories: organic and inorganic osmolytes. Inorganic osmolytes comprise mainly the intracellular ions K+ and Cl–. Cell swelling–induced activation of separate K+ and Cl– channels has been described in most preparations. Organic osmolytes are grouped in three categories: amino acids, polyalcohols, and methylamines. These osmolytes, particularly taurine, are present in …

The cytochrome P450 (CYP) 2J2 arachidonic acid epoxygenase gene was down-regulated at a pre-translational level in human hepatoma-derived HepG2 cells incubated in a hypoxic environment; under these conditions, the expression of c-Jun and c-Fos mRNA and protein was increased. The 5′-upstream region of the CYP2J2 gene was isolated by amplification of a 2341 bp fragment and putative regulatory elements that resembled activator protein-1 (AP-1)-like sequences were identified. From transient transfection analysis, c-Jun was found to strongly activate a CYP2J2–luciferase reporter construct, but co-transfection with plasmids encoding c-Fos or c-Fos-related antigens, Fra-1 and -2, abrogated reporter activity. Using a series …

Tur megistoproctus, a new species of Laelapinae, is described from the pelage of the echimyid rodent Proechimys dimidiatus from the Atlantic forests of Ilha Grande, south of Rio de Janeiro. Measurements and illustrations are included for females and males. Another laelapine mite species, Tur turki Fonseca, co-occurred with T.megistoproctus in our studies and was recorded from the same host individuals and localities. These 2 laelapine mite species appear to be exclusively associated with a complex of echimyid rodent species (subgenus Trinomys) in the Atlantic forests of southeastern Brazil.

Phylogenetic Analysis Of The Metastrongyloidea (Nematoda: Strongylida) Inferred From Ribosomal Rna Gene Sequences, Ramon A. Carreno, Steven A. Nadler
Harold W. Manter Laboratory of Parasitology: Faculty Publications
Phylogenetic relationships among nematodes of the strongylid superfamily Metastrongyloidea were analyzed using partial sequences from the large-subunit ribosomal RNA (LSU rRNA) and small-subunit ribosomal RNA (SSU rRNA) genes. Regions of nuclear ribosomal DNA (rDNA) were amplified by polymerase chain reaction, directly sequenced, aligned, and phylogenies inferred using maximum parsimony. Phylogenetic hypotheses inferred from the SSU rRNA gene supported the monophyly of representative taxa from each of the 7 currently accepted metastrongyloid families. Metastrongyloid taxa formed the sister group to representative trichostrongyloid sequences based on SSU data. Sequences from either the SSU or LSU RNA regions alone provided poor resolution for …

Three of 6 female long-clawed shrews, Sorex unguiculatus Dobson, 1890, collected on the island of Hokkaido, Japan, were found to have unsporulated oocysts and sexual stages (both macro- and microgamonts) in varying stages of development of an unidentified coccidium in both lactating and non lactating mammary glands. Gamonts developed in the alveoli of the mammary glands, and oocysts were found in the lactiferous ducts and in pools of milk. Mature macrogamonts were 11.9 × 15.2 μm (10-14 × 14-20 μm), whereas completely developed microgamonts with many gametes were 14.8 × 16.8 μm (10-18 × 13-20 μm). Oocysts in tissue sections …

The present invention provides a method to detect the presence of a microorganism or agent in an animal. The method encompasses placement of devices at various locations where the animal resides so as to induce the animal to initiate contact with the device. As a result of this contact, the animal deposits various microorganisms and agents on the device. The device is then tested for the presence of the particular microorganism or agent of interest.

Nearly all human immunodeficiency virus (HIV) infections are acquired mucosally, and the gut-associated lymphoid tissues are important sites for early virus replication. Thus, vaccine strategies designed to prime virus-specific cytotoxic T lymphocyte (CTL) responses that home to mucosal compartments may be particularly effective at preventing or containing HIV infection. The Salmonella type III secretion system has been shown to be an effective approach for stimulating mucosal CTL responses in mice. We therefore tested ∆phoP-phoQ attenuated strains of Salmonella enterica serovar Typhimurium and S. enterica serovar Typhi expressing fragments of the simian immunodeficiency virus (SIV) Gag protein fused to the …
