Biology Faculty Articles
Bvr-1 is a dominant X-linked feline gene which restricts the replication of B-tropic murineleukemia virus (B-MuLV) in somatic cell hybrids between murine BALB/c-RAG cells and FL-74 feline cells. Since the hybrids were originally derived by the hypoxanthine aminopterin thymidine selection scheme, counter selection experiments on 6-thioguanine result in preferential survival of hybrid cells which have spontaneously lost the feline X-chromosome on which is located the structural gene for hypoxanthine guanine phosphoribosyl transferase (IMP: pyrophosphate phosphoribosyl transferase, E.C. 2.4.2.8) and Bvr-1. Back selected Bvr-1- cells express high parental levels of B-MuLV. Bvr-1- effectively restricts the IdU-mediated induction of …

The ampullariid Pomacea urceus uses its ctenidium and lung in respiration. The snails reach the surface with their siphon and ventilate their lung by withdrawing their head-foot in a pumping action. The lung gas also serves to change the buoyancy of the snails. After a ventilation a snail does not necessarily return to the same buoyancy level. They may achieve overall specific gravities of <1 to > 1 after a ventilation. They may remain submerged (s.g. > 1) or float (s.g.

Three Linked Enzyme Loci In Fishes: Implications In The Evolution Of Vertebrate Chromosomes, D C Morizot, D A Wright, M J Siciliano
Faculty, Staff and Student Publications
A three-point linkage group comprised of loci coding for adenosine deaminase (ADA), glucose-6-phosphate dehydrogenase (G6PDH), and 6-phospho-gluconate dehydrogenase (6PGD) is described in fish of the genus Xiphophorus (Poeciliidae). The alleles at loci in this group were shown to assort independently from the alleles at three other loci--isocitrate dehydrogenase 1 and 2, and glyceraldehyde-3-phosphate dehydrogenase 1. Alleles at the latter three loci also assort independently from each other. Data were obtained by observing the segregation of electrophoretically variant alleles in reciprocal backcross hybrids derived from crosses between either X. helleri guentheri or X. h. strigatus and X. maculatus. The linkage component …

Sex-Linked Mental Retardation Without Physical Stigmata (Martin-Bell Or Renpenning Type) : A Genetic And Psychometric Approach, Craig L. Hanis
Theses and Dissertations
Screening the Utah State Training School (a resident institution for the mentally retarded) for kindreds having at least two institutionalized sibs generated 54 sib groups. Of these, 20-male only sibships had histories compatible with sex-linked mental retardation without physical stigmata (Martin-Bell or Renpenning type). Affected males had no characteristic physical stigmata (an appreciable number did have speech problems and/or seizure disorders) and exhibited IQs ranging from 5 to 74 with a mean of 34.2. Obligate carrier females had a mean IQ score of 91.9 (range, 79 to 106), which is as would be predicted due to random X-inactivation. Carrier females …
Cytogenetic Test Of Captan And Triethylenemelamine In Mouse Bone Marrow, Stephen M. Fry
Cytogenetic Test Of Captan And Triethylenemelamine In Mouse Bone Marrow, Stephen M. Fry
Honors Theses
Following a single non-toxic i.p. injection with 0.5 or 1.0 mg triethylenemelamine/kig a significant increase in chromosome aberrations in mouse bone marrow was observed 6, 12, 30, but not 54 h after treatment. A single i.p. injection with 250 mg captan/kig caused up to 68% lethality above controls, but no significant increase in chromosome aberrations after the same time intervals. One metacentric chromosome was observed 6, 30, and 54 h after captan treatment among 300, 300, and 101 metaphase spreads respectively, whereas none appeared in 1495 control spreads.

Time Course Of Pr Of Uv-Induced Chromosomal Aberrations And Lethal Damage In G1 Xenopus Cells, Jan Payne, H. Gaston Griggs
Journal of the Arkansas Academy of Science
Synchronous cultures of early G1 cells were exposed to UV and their ability to photoreactivate lethal and aberrational damage was determined as a function of time following UV exposure. Lesions leading to cell death were converted to a non-photoreactivable state before cells entered S phase, while lesions leading to chromosomal aberrations were converted to a non-photoreactivable state as the cells entered S phase. These results indicate that the intracellular mechanism which expresses photoreactivable UV-induced lesions in G1 cells as cell death is not identical to the mechanism which expresses such lesions as chromosomal aberrations, and the two mechanisms operate primarily …
Poultry Breeds Must Be Conserved : An Ideal Small Unit, R H. Morris
Poultry Breeds Must Be Conserved : An Ideal Small Unit, R H. Morris
Journal of the Department of Agriculture, Western Australia, Series 4
The Department of Agriculture is concerned at the rapid disapearance of some poultry breeds and strains over recent years. This trend has resulted from the disposal of flocks previously held by commercial poultry breeders and fanciers.
One way to stop the trend of disappearance of poultry breeds is for interested people to maintain a breeding flock of pure-bred poultry, making the progeny available to others.
This article describes a small unit suitable for housing breeding fowls. The unit is based on designs developed by the Department of Agriculture.

Inactivation Of Brain Hexokinase By An Adenosine 5'-Triphosphate Analog, Subbarao Bondada, Umakant W. Kenkare
Microbiology, Immunology, and Molecular Genetics Faculty Publications
An analog of ATP, 6-mercapto-9+nribofuranosylpurine 5’-triphosphate (SH-TP), functions as a phosphoryl donor for the reaction catalyzed by hexokinase. Incubation of the enzyme with millimolar concentrations of this reagent led to its rapid inactivation. However, the corresponding monophosphate derivative of this reagent was much less effective. A plot of the initial rate of inactivation versus the concentration of SH-TP exhibited saturation, suggesting the formation of a reversible complex between the enzyme and SH-TP prior to the inactivation reaction. The dissociation constant of this enzyme-inhibitor complex was the same as the K, of this reagent with respect to hexokinase in the phosphoryl …

Reaction Of Brain Hexokinase With Tetranitromethane: Oxidation Of Essential Thiol Groups, Subbarao Bondada, Umakant W. Kenkare
Microbiology, Immunology, and Molecular Genetics Faculty Publications
Treatment of bovine brain mitochondrial hexokinase with a fivefold molar excess of tetranitromethane (TNM) at pH 8.0 results in a complete loss of activity of the enzyme. Spectral measurements and amino acid analysis showed that only cysteinyl residues were modified by TNM under these conditions. The products of the reaction of TNM with the enzyme were cysteic acid and disulfides as found by amino acid analysis of the carboxymethylated hexokinase. Failure of 2-mercaptoethanol to reverse the effect of TNM suggested that the thiol group that is oxidized to cysteic acid is the one essential for enzyme activity. Substrates glucose and …

A Cytogenetic Study Of Xx/Xy Chimerism And Other Anomalies Of Bos Taurus In A Sampled Dairy Herd, Ferne M. Rogers
Masters Theses
The establishment of normal chromosomal patterns for a wide range of plants and animals has led to the recognition of abnormalities as well as their relationship to phenotypic irregularities. Research dealing with the expression of human karyotype is of prime interest; but, closely rivaling that is the genetic study of domestic animals which are of economic importance to man.
One observed anomaly which has come under scrutiny by cytogeneticists is that of the female member of a heterogeneous bovine twin pair, the freemartin. This is a sterile animal with internal morphology showing varying degrees of intersex. Cytogenetic studies reveal XX/XY …

Bvr-1, A Restriction Locus Of A Type C Rna Virus In The Feline Cellular Genome: Identification, Location, And Phenotypic Characterization In Cat X Mouse Somatic Cell Hybrids, Stephen J. O'Brien
Biology Faculty Articles
Somatic cell hybrids were constructed between BALB/c-RAG mouse cells and feline lymphoma cells by the hypoxanthine-aminopterin-thymidine selection scheme. RAG cells spontaneously produce an endogenous B-tropic type C virus. Cat-mouse hybrids preferentially segregate feline chromosomes and retain murine chromosomes, demonstrable by karyotypic and isozyme analyses. Despite the presence of the complete mouse genome, including the viral genome, virus production was diminished to 1-5% of the levels observed in RAG parents based upon particle-associated RNA-dependent DNA polymerase (reverse transcriptase) activity in the culture fluid. Thirty-seven hybrids made on four different occasions had suppressed virus levels, and no hybrids expressed parental virus levels. Reverse …
