While the expression of circadian rhythms is nearly universal among multicellular eukaryotic organisms, demonstration of this phenomenon in prokaryotes has been largely restricted to photosynthetic cyanobacteria until very recently. Further, growing interest in gastrointestinal microbiomes has revealed a complex temporal relationship between the gastrointestinal clock and the bacterial microbiome within. At least one member of the gut microbiome, Klebsiella (née Enterobacter) aerogenes, responds to the indoleamine hormone melatonin, secreted by the gastrointestinal system itself. Further research revealed that K. aerogenes also expresses a circadian rhythm in motility and gene expression that is temperature compensated. Although rhythmicity is unaltered …

Under pathophysiological conditions, the microtubule protein tau (MAPT) forms neurofibrillary tangles that are the hallmark of sporadic Alzheimer’s disease as well as familial frontotemporal dementias linked to chromosome 17 (FTDP-17). In this work, I report that MAPT forms circular RNAs through backsplicing of exon 12 to either exon 10 or exon 7 (12→10; 12→7), and that these circular RNAs are translated into proteins.
Using stable cell lines overexpressing the circular tau RNAs 12→7 and 12→10, we have discovered that the tau circular RNA 12→7 is translated in a rolling circle, giving rise to multiple proteins. This circular RNA …

Background: A higher intake of dietary fiber is associated with a decreased risk of chronic inflammatory diseases such as cardiovascular disease and inflammatory bowel disease. This may function in part due to abrogation of chronic systemic inflammation induced by factors such as dysbiotic gut communities. Data regarding the detailed influences of long-term and recent intake of differing dietary fiber sources on the human gut microbiome are lacking.

Objective: Significant morbidity and mortality have occurred in the USA, Europe, and Asia due to severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), whereas the numbers of infections and deaths in sub-Saharan Africa (SSA) have remained comparatively low. It has been hypothesized that exposure of the population in SSA to other coronaviruses prior to the COVID-19 pandemic resulted in some degree of cross-protection against SARS-CoV-2 infection and pathogenesis. We evaluated this hypothesis by comparing SARS-CoV-2 cross-reactive antibodies in pre-pandemic plasma samples collected from SSA and the USA.

Due to the COVID-19 pandemic and multiple devastating forest fires, the 2020 meeting of the Rocky Mountain Virology Association was held virtually. The three-day gathering featured talks describing recent advances in virology and prion research. The keynote presentation described the measles virus paradox of immune suppression and life-long immunity. The structures of the four N-linked glycans from the prototype chlorovirus PBCV-1 major capsid protein do not resemble any other glycans in the three domains of life. All known chloroviruses and antigenic variants (or mutants) share a unique conserved central glycan core consisting of five sugars, except for antigenic mutant virus P1L6, which has four of the five sugars. A combination of ge- netic and structural analyses indicates that the protein coded by PBCV-1 gene a111/114r, conserved in all chloroviruses, is a glycosyltransferase with three putative domains of approximately 300 amino acids each. Influenza virus has significant viral diversity, both through antigenic drift and shift, which makes development of a vaccine challenging. Current influenza vaccines are updated yearly to in- clude strains predicted to circulate in the upcoming influenza season, however this can lead to a mismatch which reduces vaccine efficacy. Several strategies targeting the most abundant and im- munogenic surface protein of influenza, the hemagglutinin (HA) protein, have been explored. These strategies include stalk-directed, consensus-based, and computationally derived HA immunogens. Zika virus (ZIKV) infection to a pregnant woman can be vertically transmitted to the fetus via the placenta leading to Congenital Zika syndrome. This is characterized by microcephaly, retinal defects, and intrauterine growth retardation. ZIKV induces placental trophoblast apoptosis leading to severe abnormalities in the growth and development of the fetus. However, the molecular mechanism behind ZIKV-induced apoptosis in placental trophoblasts remains unclear. We hypothesize that ZIKV infection induces endoplasmic reticulum (ER) stress in the trophoblasts, and sustained ER stress results in apoptosis. The exploration of new and diverse animal groups in the study of sexual selection is both necessary and important to help better understand broad patterns and test sexual selection hypotheses regarding the evolutionary origins and maintenance of reproductive tactics and associated traits. Solifuges are, in this matter, an exceptional group and very little explored from the sexual selection point of view. At first glance, mating is apparently quite simple and conserved within this arachnid order, but solifuge reproductive behavior is unique among arachnids and more diverse than previously thought. Predator diets represent a potential interaction between local prey availability, prey antipredator defenses, and predator foraging behavior. Female spider-specialist muddauber wasps (Hymenoptera: Sphecidae) collect spiders and provision them intact, but paralyzed, to their developing larvae, providing a unique means of quantifying the diversity and abundance of prey that they capture. Mud-dauber wasps are hypothesized to be a major source of selection on antipredator defenses in web-building spiders, and the spiny and thickened abdomens of female spiny orb-weaving spiders (Araneae: Araneidae) are hypothesized to function as antiwasp defenses. Amblypygids, or whip spiders, are nocturnally active arachnids which live in structurally complex environments. Whip spiders are excellent navigators that can re-locate a home refuge without relying on visual input. Therefore, an open question is whether visual input can control any aspect of whip spider spatial behavior. In the current study, Phrynus marginemaculatus were trained to locate an escape refuge by discriminating between differently oriented black and white stripes placed either on the walls of a testing arena (frontal discrimination) or on the ceiling of the same testing arena (overhead discrimination). Loxosceles reclusa, or brown recluse spider, is a harmful household spider whose habitat extends throughout the Midwest in the USA and other regions in the world. The pheromones and other biomolecules that facilitate signaling for brown recluses and other spider species are poorly understood. Parasitoid wasps are effective biological control agents that have evolved intricate host immune suppression strategies to overcome host immune defenses. The most important anti-parasite defense mechanism is encapsulation, where the host’s blood cells surround the wasp egg. In the Drosophila/Leptopilina host-parasitoid system, extracellular vesicles (EVs) are oviposited along with the wasp egg. The EVs possess spikes and contain hundreds of proteins, some of which enable wasp eggs to escape encapsulation by changing the morphology and function of hemocytes, or killing them. Sexually differentiated behavior has been shown to be affected by both genes and hormones. The discovery of the SRY gene, which codes for the development of testes, lead to the development of the Four Core Genotypes model of mice, and the separation of sex chromosomes and its resulting gonadal hormones. DNA profiles do not provide as much evidentiary value when either all known samples can be excluded or when there is no known suspect. DNA phenotyping is a technique used for investigative purposes to predict externally visible characteristics (EVCs) based on specific markers found in the genome. RATIONALE: Recent studies have discovered several unique tumor response subgroups outside of response classification by Response Evaluation Criteria for Solid Tumors (RECIST), such as mixed response and oligometastasis. These subtypes have a distinctive property, lesion heterogeneity defined as diversity of tumor growth profiles in RECIST target lesions. Furthermore, many cancer clinical trials have been activated to evaluate various treatment options for heterogeneity-related subgroups (e.g., 29 trials so far listed in clinicaltrials.gov for cancer patients with oligometastasis). The Protein inference problem is becoming an increasingly important tool that aids in the characterization of complex proteomes and analysis of complex protein samples. In bottom-up shotgun proteomics experiments the metrics for evaluation (like AUC and calibration error) are based on an often imperfect target-decoy database. These metrics make the inherent assumption that all of the proteins in the target set are present in the sample being analyzed. When tissue samples are studied in bulk without consideration for different cell proportions and types, results can be biased due to the attenuation of unique cellular expressions. In order to study cell type specific RNA expression profiles within tissue, single cell RNA sequencing (scRNA-seq) is used. For scRNA-seq studies it is critical to have intact cells. Biomedical intelligence provides a predictive mechanism for the automatic diagnosis of diseases and disorders. With the advancements of computational biology, neuroimaging techniques have been used extensively in clinical data analysis. Attention deficit hyperactivity disorder (ADHD) is a psychiatric disorder, with the symptomology of inattention, impulsivity, and hyperactivity, in which early diagnosis is crucial to prevent unwelcome outcomes. A unique genotype of Rickettsia parkeri, designated R. parkeri strain Black Gap, has thus far been associated exclusively with the North American tick, Dermacentor parumapertus. The compete genome consists of a single circular chromosome with 1,329,522 bp and a G+C content of 32.5%.

Alternative life-history strategies (ALHS) are genetic polymorphisms generating phenotypes differing in life histories that generally arise due to metabolic resource allocation tradeoffs. Although ALHS are often be limited to a single sex or populations of a species, they can, in rare cases, be found among several species across a genus. In the butterfly genus Colias, at least a third of the species have a female limited ALHS called Alba. While many females develop brightly pigmented wings, Alba females reallocate nitrogen resources used in pigment synthesis to reproductive development, producing white-winged, more fecund females. เชื้อแบคทีเรียแกรมลบ Acinetobacter baumannii เป็นเชื้อฉวยโอกาส ที่ก่อให้เกิดการติดเชื้อในโรงพยาบาล และมีการดื้อต่อยาปฏิชีวนะหลายชนิด จากหลายกลไกการดื้อยา รวมถึงการใช้ efflux pump ขับยาออกนอกเซลล์ การศึกษาครั้งนี้ผู้วิจัยได้ทดสอบหาค่าความเข้มข้นต่ำสุดที่ยับยั้งการเจริญเติบโตของเชื้อ (MIC) ต่อยา amikacin ในเชื้อ A. baumannii ทั้งหมด 8 ตัวอย่าง ที่ได้จากสิ่งส่งตรวจของผู้ป่วย และเชื้อสายพันธุ์มาตรฐาน ATCC 19606 พบว่าเชื้อ A. baumannii 7 ตัวอย่างมีค่า MIC อยู่ในช่วง 0.25 – 4 µg/mL (ไวต่อยา) และเชื้ออีก 2 ตัวอย่างมีค่า MIC เท่ากับ 64 และมากกว่า 256 µg/mL (ดื้อต่อยา) เมื่อเหนี่ยวนำเชื้อด้วยยา amikacin โดยเพาะเลี้ยงเชื้อในสภาวะที่มียา amikacin ความเข้มข้นเพิ่มขึ้นเป็นสองเท่า ทุก 48 ชั่วโมง ติดต่อกันเป็นเวลาหลายวัน พบว่าเชื้อ A. baumannii หลังเหนี่ยวนำด้วยยา amikacin ทั้ง 8 ตัวอย่าง มีค่า MIC เพิ่มขึ้นอยู่ในช่วง 32 ถึงมากกว่า 256 µg/mL ผลการศึกษาการใช้สารยับยั้ง efflux pump ได้แก่ สาร carbonylcyanide m-chlorophenylhydrazone (CCCP) พบว่าเชื้อ A. baumannii หลังเหนี่ยวนำด้วยยา amikacin ทั้ง 8 ตัวอย่าง มีค่า MIC ต่อยา amikacin เมื่อมีสาร CCCP ลดลงมากกว่าหรือเท่ากับ 4 เท่าของค่า MIC ต่อยา amikacin ที่ไม่มีสาร …

มะเร็งปากมดลูก เป็นโรคที่คร่าชีวิตสตรีเป็นลำดับต้นสามในประเทศไทย มีสาเหตุหลักมาจากการติดเชื้อเอชพีวี โดยหนึ่งในยีนที่มีส่วนสำคัญคือยีน E7 ซึ่งสามารถจับกับบริเวณโพรโมเตอร์ของยีน CCNA1 ซึ่งเป็นยีนต้านมะเร็ง ร่วมกับเอนไซม์ DNMT1 ชักนำให้เกิดการเติมหมู่เมทิล ทำให้การแสดงออกของยีน CCNA1 ลดลง นำไปสู่การเกิดมะเร็งปากมดลูก การศึกษาก่อนหน้าพบว่าในยีน ALDH3B1 ASAH2B และ FOXC1 มีการแสดงออกที่ลดลงเนื่องจากการเติมหมู่เมทิลที่บริเวณโพรโมเตอร์ในเซลล์สายพันธุ์ SiHa และ HEK293 ที่ถูกกทรานสเฟ็กด้วย E7 ในการศึกษานี้จึงมีวัตถุประสงค์เพื่อประเมินการเติมหมู่เมทิลของยีน ALDH3B1 ASAH2B และยีน FOXC1 ในหลายระยะของการเกิดมะเร็งปากมดลูก ตัวอย่างที่ใช้เป็นแปบสเมียร์และตัวอย่าง ASC-US บริเวณปากมดลูกที่ได้รับการวินิจฉัยแล้ว นำมาสกัดดีเอ็นเอ จากนั้นนำมาผ่านกระบวนการไบซัลไฟต์ทรีทเมนท์ และตรวจการเติมหมู่เมทิลด้วยเทคนิค MSP ผลการศึกษาพบว่ายีน ALDH3B1 มีจำนวนตัวอย่างที่มีการเติมหมู่เมทิลในเซลล์ปากมดลูกปกติมากกว่าเซลล์ที่ติดเชื้อเอชพีวี ในขณะที่ยีน ASAH2B และยีน FOXC1 พบการเติมหมู่เมทิลมากขึ้นตามความพัฒนาของเซลล์ที่ผิดปกติเมื่อเปรียบเทียบกับเซลล์ที่ไม่มีการติดเชื้อเอชพีวี และพบความแตกต่างอย่างมีนัยสำคัญระหว่างการติดเชื้อเอชพีวีและการเติมหมู่เมทิล อย่างไรก็ตาม จำนวนตัวอย่างที่มีการเติมหมู่เมทิลกลับลดลงในเซลล์มะเร็งปากทดลูกในทุกยีน จากผลการศึกษาแสดงให้เห็นว่ามีเพียงยีน ASAH2B และยีน FOXC1 ที่อาจพอจะนำมาเป็นเครื่องหมายโมเลกุลที่จำเพาะกับมะเร็งปากมดลูก เพื่อใช้ในการคัดกรองมะเร็งปากมดลูกในระยะก่อนเกิดมะเร็ง อีกทั้งควรที่จะติดตามข้อมูลและนัดหมายกลุ่มประชากรเสี่ยงที่อยู่ในกลุ่มระยะก่อนเป็นมะเร็งต่อไป

โรคข้อเข่าเสื่อมเป็นโรคเกี่ยวกับกระดูกที่พบได้มากในกลุ่มผู้สูงอายุ โดยผิวของข้อถูกทำลาย และมีปุ่มกระดูกงอก โรคข้อเข่าเสื่อมสามารถเกิดได้จากหลายสาเหตุ อายุ เพศ น้ำหนักและพันธุกรรม โดยปัจจัยพันธุกรรมเป็นบทบาทสำคัญในการเกิดโรคข้อเข่าเสื่อม งานวิจัยนี้ จึงศึกษาเครื่องหมายทางพันธุกรรมที่อาจใช้ทำนายการเกิดโรคข้อเข่าเสื่อมได้ โดยมีวัตถุประสงค์ที่จะศึกษายีน CALM1 ซึ่งมีความสัมพันธ์กับการสลายกระดูกอ่อนและการส่งสัญญาณของแคลเซียม โดยเก็บตัวอย่างเลือดจากประชากรไทยที่เป็นโรคข้อเข่าเสื่อมจำนวน 170 คน และประชากรไทยที่ไม่ได้เป็นโรคข้อเข่าเสื่อม จำนวน 150 คน ทำการสกัดดีเอ็นเอจากตัวอย่างเลือด ศึกษาภาวะพหุสัณฐานของยีน CALM1 ด้วยวิธี high-resolution melting (HRM) analysis และวิธี polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) โดยวิเคราะห์ข้อมูลทางสถิติด้วย odds ratio ผลการทดลองพบความสัมพันธ์ของภาวะพหุสัณฐานตำแหน่ง rs12885713 ในประชากรทั้งหมด และเพศหญิง พบว่า แอลลีล T (OR = 0.64, 95% CI = 0.49 – 0.87, P = 0.005), (OR = 0.65, 95% CI = 0.45 – 0.91, P = 0.015) ตามลำดับ และจีโนไทป์ TT (OR = 0.3, 95% CI = 0.14 – 0.63, P = 0.0014), (OR = 0.28, 95% CI = 0.12 – 0.66, P = 0.003) ตามลำดับ มีความสัมพันธ์กับการเกิดโรคข้อเข่าเสื่อมอย่างมีนัยสำคัญทางสถิติ ไม่พบความสัมพันธ์ของกับการเกิดโรคข้อเข่าเสื่อมของภาวะพหุสัณฐานตำแหน่ง …

การเพาะเลี้ยงสัตว์น้ำเพื่อเป็นแหล่งอาหาร มีผลต่อความมั่นคงทางอาหารและโภชนาการของมนุษย์ โดยกุ้งทะเลจัดเป็นสัตว์น้ำเศรษฐกิจอันดับหนึ่งของไทย ตั้งแต่ปี พ.ศ. 2534 เป็นต้นมา เนื่องจากเป็นสัตว์น้ำที่ได้รับความนิยมบริโภคทั้งในประเทศและต่างประเทศทั่วโลก แต่ในปี พ.ศ. 2555-2556 การเลี้ยงกุ้งเริ่มประสบปัญหาโรคระบาดอย่างหนักจากอาการตายด่วนเนื่องจากตับและตับอ่อนวายเฉียบพลัน (Early mortality syndrome/ acute hepatopancreatic necrosis disease : EMS/AHPND) ทําให้เกิดความเสียหายต่อเกษตรกรและส่งผลกระทบต่อภาคอุตสาหกรรม รวมไปถึงสูญเสียส่วนแบ่งการตลาดและมูลค่าการส่งออกกุ้งทะเลเป็นอย่างมาก ต่อมาได้มีการพัฒนาจุลินทรีย์ ปม.1 ที่เป็นโพรไบโอติกในกลุ่มบาซิลัส เพื่อลดอัตราการเกิดโรค ซึ่งเชื้อ B. licheniformis ที่รวมในกลุ่มโพรไบโอติกนี้ มีความสามารถสร้างแบคทีริโอซินที่ยับยั้งการเจริญของเชื้อก่อโรคได้ มีคุณสมบัติทนความร้อน 100 °C ทำงานได้ดีในค่าความเป็นกรด-ด่างกว้าง จึงเป็นที่น่าสนใจในการศึกษาสภาวะที่เหมาะสมเพื่อเพิ่มปริมาณการสร้างแบคทีริโอซินของเชื้อ B. licheniformis และประสิทธิภาพในการยับยั้งเชื้อก่อโรค จากการทดสอบเพาะเลี้ยงเชื้อ B. licheniformis ในอาหารเหลว Tryptic Soy Broth มีความเข้มข้นของเชื้อเป็น 106 CFU/มิลลิลิตร ในสภาวะที่ปรับเปลี่ยนอุณหภูมิ ช่วงเวลา และความเข้มข้นของเกลือที่ต่างกัน ผลการทดสอบประสิทธิภาพการยับยั้งเชื้อก่อโรคในกุ้งด้วยวิธี Total plate count พบว่า ในน้ำเลี้ยงเชื้อ (Cell Free Supernatant: CFS) สามารถยับยั้งเชื้อทดสอบได้แก่ เชื้อ Vibrio alginolyticus และ Vibrio parahaemolyticus ซึ่งเป็นสายพันธุ์ที่เป็นสาเหตุของโรคตับและตับอ่อนวายเฉียบพลันในกุ้งได้ดี เมื่อเพาะเลี้ยงเชื้อ B. licheniformis ที่อุณหภูมิ 30 องศาเซลเซียสเป็นเวลา 72 ชั่วโมง และเลี้ยงในอาหารเหลว TSB ที่มีเกลือความเข้มข้น 2.5% และเมื่อเลี้ยง B. licheniformis ร่วมกับเซลล์ของเชื้อทดสอบที่ถูกทำให้ตายด้วยความร้อน พบว่าน้ำเลี้ยงเชื้อยังคงมีประสิทธิภาพในการยับยั้งเชื้อทดสอบ จากนั้นตรวจวิเคราะห์ประสิทธิภาพการทำลายเซลล์ของแบคทีเรียก่อโรคของน้ำเลี้ยงเชื้อด้วยกล้องจุลทรรศน์อิเลคตรอนพบว่าส่วนที่ห่อหุ้มเซลล์ของเชื้อทดสอบมีลักษณะเป็นรู นอกจากนี้พบว่าสารยับยั้งในน้ำเลี้ยงเชื้อที่เกิดจากการเพาะเลี้ยงและจาก B. licheniformis ร่วมกับเซลล์ V. alginolyticus ที่ถูกทำให้ตายด้วยความร้อน มีความสามารถทำให้เซลล์ของเชื้อทดสอบมีขนาดเล็กลง และลดการเกาะกลุ่มของเชื้อทดสอบได้ดี ผลจากการวิเคราะห์ลำดับกรดอะมิโนในน้ำเลี้ยงเชื้อ (Cell-Free …

วัณโรคดื้อยาหลายขนาน (Multidrug-resistant tuberculosis: MDR-TB) เป็นหนึ่งในสาเหตุหลักที่ทำให้การรักษาวัณโรคล้มเหลว และส่งผลให้การแพร่ระบาดของโรคยังคงมีอัตราที่สูง วัณโรคดื้อยาหลายขนานเกิดจากเชื้อ Mycobacterium tuberculosis ที่ดื้อต่อยา Rifampicin และยา Isoniazid พร้อมกัน โดยมากกว่า 90% ของวัณโรคที่ดื้อยา Rifampicin มักเกิดการดื้อยา Isoniazid ร่วมด้วยภายหลัง ดังนั้นการตรวจวินิจฉัยการดื้อยา Rifampicin ของเชื้อ M. tuberculosis จึงสามารถใช้เป็นตัวแทนในการตรวจหาวัณโรคดื้อยาหลายขนานได้ วิธีทางอณูชีววิทยาที่ใช้วินิจฉัยวัณโรคดื้อยาหลายขนานในปัจจุบันต้องอาศัยเครื่องมือที่มีราคาแพงและการดูแลซ่อมบำรุงอย่างสม่ำเสมอ การเพิ่มปริมาณสารพันธุกรรมภายใต้อุณหภูมิคงที่ได้เข้ามามีบทบาทอย่างมากในการวินิจฉัยโรคทางห้องปฏิบัติการและเป็นทางเลือกสำหรับการทดสอบที่ไม่ต้องอาศัยเครื่องมือที่จำเพาะ สามารถให้ผลการทดสอบที่ถูกต้อง รวดเร็ว และนำไปประยุกต์ใช้ ณ งานภาคสนามได้ การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิค Multiplex allele-specific recombinase polymerase amplification ร่วมกับ dipstick chromatography (MAS-RPA-DC) สำหรับตรวจหาการกลายพันธุ์ภายในยีน rpoB ณ ตำแหน่งโคดอน 516 526 และ 531 ซึ่งสัมพันธ์กับการดื้อยา Rifampicin ของเชื้อ M. tuberculosis ในการทดสอบภายใต้ปฏิกิริยาเดียวกันและแบบอ่านผลด้วยตาเปล่า ภายหลังนำเทคนิค MAS-RPA-DC มาทดสอบกับ DNA ที่สกัดได้จากโคโลนีของเชื้อ M. tuberculosis สายพันธุ์คลินิกจำนวน 141 ตัวอย่าง พบว่าเทคนิค MAS-RPA-DC มีความไวและความจำเพาะที่สูงเมื่อเปรียบเทียบกับเทคนิค Sanger DNA sequencing โดยมีความไวในการตรวจการกลายพันธุ์ของยีน rpoB516 rpoB526 และ rpoB531 มีค่าเท่ากับ 100 % 70 % และ 98.2 % ตามลำดับ มีค่าความจำเพาะในการตรวจการกลายพันธุ์ของยีน rpoB516 rpoB526 และ rpoB531 เท่ากับ 95.5 % 97.5 % และ 97.7 …

While humans value healthy environments for the ecosystem services and recreational opportunities they provide, it is increasingly evident that such resources can be paradoxically degraded by anthropogenic activities. In this investigation, we studied the relationship between different intensities of anthropogenic land-use change and habitat-driven fish assemblage response across multiple spatiotemporal scales. Body fluid identification is essential in the forensic biology workflow that assists DNA analysts in determining where to collect DNA evidence. Current presumptive tests lack the sensitivity and specificity molecular techniques can achieve; therefore, molecular methods, such as microRNA and microbial signatures, have been extensively researched in the forensic community. Limitations of each method suggest combining molecular markers to increase discrimination efficiency of multiple body fluids from a single assay. While microbial signatures have been successful in identifying fluids with high bacterial abundances, microRNAs have shown promise in fluids with low microbial abundance.
