Dispermic Origin And Clinical Outcome Of Three Complete Hydatidiform Moles With 46,Xy Karyotype, U. Surti, A. E. Szulman, S. O'Brien

Three new cases of complete, classic hydatidiform mole with a 46,XY karyotype are described. They originated by dispermy as demonstrated by chromosome and enzyme analyses. Levels of human chorionic gonadotropin decreased to normal spontaneously within a short time, indicating a benign course in these three cases. Of a total of 18 cases reported to date, postoperative clinical information was available for 10 patients. Two of these 10 patients had a malignant course with lung metastases. It is of theoretical and clinical importance to establish the magnitude of the risk of malignancy for hydatidiform mole with a 46,XY karyotype. © 1982.

A deletion mutant of the colicin E1-derived plasmid, pDMS6642, exhibited an approximately fourfold increase in copy number. We subsequently isolated hydroxylamine-induced mutants of that plasmid that had a further increase in copy number. Analysis of them suggests that the increased copy number of pDMS6642 is associated with transcriptional readthrough from a Tn3 transposon into the region of ColE1 containing information that influences plasmid replication. The hydroxylamine mutation in one copy number mutant appeared to increase the plasmid copy number by stimulating readthrough transcription from the Tn3 transposon into the ColE1 replication control region, whereas the other hydroxylamine mutation acts by …

Several recent reports have established the biological and molecular genetic similarity between the endogenous AKV virus of strain AKR, and an N-ecotropic endogenous virus found in the genome of feral Japanese mice, Mus musculus molossinus. The similarities are so striking as to suggest a common origin of these viruses, which are present in some, but not all, inbred mouse strains. The virogenes of AKR mice may have been acquired by either: (a) common descent of AKR (and other AKV+ strains) from a common ancestor of AKR and molossinus animals, or (b) horizontal germ line infection of the AKR …

Treatment of Schneider's line 3 Drosophila cells with the steroid hormone ecdysterone rapidly stimulated the synthesis and accumulation of the polypeptide previously designated p7 [Berger, E. M., Ireland, R. C. & Wyss, C. (1980) Somatic Cell Genet. 6, 119-129]. In this report, p7 is identified as the 23,000-dalton heat shock polypeptide (hsp23). In addition to hsp23, the synthesis of the low molecular weight heat shock polypeptides hsp22, hsp26, and hsp27 was also stimulated by ecdysterone, although to different extents. Hybridization of a nick-translated genomic clone containing the hsp23 gene to a total RNA blot showed that ecdysterone stimulation of hsp23 …

In this essay I outline the processes of adaptation of animals and of animal populations and discuss their relevance to the problem of animal welfare. Because "animal welfare" has many different aspects including philosophical, ethical, and biological, it is important to examine some of the fundamental issues that underly the concept. Hence, in this essay, I comment on how people come to "know," how information accumulates, and how what we know influences our actions. Drosophila Alcohol Dehydrogenase: Developmental Studies On Cryptic Variant Lines, Gurbachan Miglani, Franklin Ampy

Thirty-five cryptic variant lines were used to examine the mechanisms involved in genetic modulation of alcohol metabolism in Drosophila. Late third-instar larval, preemergence pupal, and adult stages cultured at 18 and 28 C were examined. Spectrophotometric analyses for native alcohol dehydrogenase (ADH) activity and residual ADH activity after treatment with guanidine hydrochloride and heat were performed. Differential response of cryptic variants to treatment with the denaturants during development suggested that this variation may have an adaptive significance.

Crude extracts of triple-cloned, purified cultures of 22 species of Mycoplasma and Acholeplasma were examined for expression of 21 isozyme systems routinely used to type mammalian cells. Nine previously described enzymes (purine nucleoside phosphorylase, adenylate kinase, dipeptidase, esterase, glyceraldehyde-3-phosphate dehydrogenase, glucose phosphate isomerase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and superoxide dismutase) and three enzymes not previously reported in mycoplasma (triose phosphate isomerase, inorganic pyrophosphatase, and acid phosphatase) were detected in some or all of the species examined. These findings provide new information on the enzymatic expressions of these organisms. Interpopulation Variation In Calcareous And Proteinaceous Shell Components In The Stream Limpet, Ferrissia Rivularis, Carl M. Way, Daniel J. Hornbach, Albert J. Burky

Natural populations of the North American stream limpet, F. rivularis, were studied in upstate New York [USA], in a set of localities whose waters have a 15-fold range of dissolved Ca (4.6-67.6 mg/liter) and also range from oligotrophy to eutrophy. Shell component analyses (CaCO3, total organic C and total N) are reported as component mass-fractions (mg/g or .mu.g/g dry weight) and as values for a standard limpet shell of 35 mm aperture length (AL). Seasonal metabolic rates were examined for the sphaeriid clam, Musculium partumeium (Say) from a permanent and a temporary pond. For the fall-born generation from the permanent pond and for the single generation from the temporary pond, metabolic rates peak during periods of greatest growth and reproduction. Metabolic rates were consistently higher for the permanent pond population. These rates for Musculium partumeium extend the available information on the family Sphaeriidae to the genus Musculium.

Clarke (1973) states that Pisidium casertanum (Poli) is perhaps the most common species of Pisidium and is truly cosmopolitan, occurring throughout Eurasia, Africa, Australia, and South, Central and North America. A group of Escherichia coli isolates from nature were compared with one another and with laboratory strains of E. coli with respect to size distribution of chromosomal restriction endonuclease fragments and differences in nucleotide sequences in selected small portions of the genomes. The estimated frequency of base substitutions in nucleotide sequences in and near the trp operons of 26 of the 28 E. coli strains examined ranged from 0.008 to 0.066. Nucleotide sequences in or near λ prophage homologs were significantly more variable than the sequences in or near trp, tnaA, and thyA genes. Thus, the λ-homologous regions …

Muskrat (Ondatra zibethicus) populations are located in extensive marshes that are managed either privately or by state or federal refuge personnel. Maintenance of a balanced muskrat population is important to marsh management, therefore necessitating accurate density estimates. An objective of the NC-103 regional swine breeding project is to evaluate inter-population and intra-population performance of domestic and exotic strains of swine. Several cooperating stations have conducted experiments relative to this objective. Projects were not exact replicates, but sufficient overlap allowed combining the information to yield more precise estimates of heterosis and breed effects than was possible from the analyses of data from any single experiment. This publication summarizes data available from NC-103 cooperating stations on breed and heterosis effects in swine. For the past decade, numerous investigators have attempted to explain the plethora of genetic polymorphism found in nature (see Ayala 1976). However, few researchers have been able to document how polymorphisms have been maintained, whether the polymorphisms were genic or chromosomal. Our investigations into the maintenance of a Robertsonian chromosomal polymorphism in the plains pocket goher, Geomys bursarius major, have revealed evidence that this polymorphism is maintained by differential viabilities of the three chromosomal morphs, with the heterozygote being favored.

Lateral root cuttings were collected at five quaking aspen (Populus tremuloides Michx.) clones in Larimer County, Colorado on October 2, 1977 and May 30, 1978. The October 1977 root cuttings were stored eight months in air-tight plastic bags at 0 to 4°C. On June 1, 1978 the stored (October 1977) and the fresh (May 1978) root cuttings were planted in a greenhouse propagation bench. All sprouts that developed were allowed to grow on half of the 10-cm long root cuttings randomly selected as controls.
