The natural flow regime of the majority of rivers in the United States has been altered by anthropogenic structures. This loss of connectivity plays a strong role in shaping river ecosystems by altering physical habitat characteristics and shaping fish community assemblages. Although the impacts of large dams on river systems are well documented and often easy to observe, there are fewer studies on the effects of smaller low-head dams. As totipotent cells differentiate to specific cell types in multicellular organisms, certain sets of genes are turned off (gene silencing/gene extinction) while others are turned on (gene activation). Cell type specific gene silencing and gene activation is the basis of cell differentiation. Somatic cell hybrids which are produced by fusing different types of somatic cells from two different tissues have been used as a valuable resource to discern the phenomenon of lineage- specific gene extinction. Chronically high blood glucose levels lead to many problems, such as insulin resistance, the hallmark of Type II diabetes. Increased flux through the hexosamine biosynthesis pathway is one mechanism by which high glucose as well as glucosamine has been shown to induce insulin resistance. This study tests the effects of glucosamine induced insulin resistance on insulin regulation of the metabolic genes glucose-6-phosphate dehydrogenase (G6PDH) and fatty acid synthase (FAS) as well as insulin responsive proteins tribbles homolog (TRIB3) and sterol regulatory element binding protein (SERBP-1c) 1c.

Genetic population substructure is often overlooked because of discontinuities between management and actual population structure as in the case of yellow perch, an ecologically and economically important indigenous fish species in the Laurentian Great Lakes. A knowledge gaps pertaining to the natural history of yellow perch relates to the biological connectivity between nearshore Lake Michigan and drowned river mouth (DRM) lakes, where it remains unclear whether resident yellow perch from Lake Michigan use DRM lakes for spawning or whether DRM lakes contribute to nearshore yellow perch populations in Lake Michigan. Gene duplication is believed to be the major source for providing genetic materials for the innovation and diversification of protein functions; natural selection and/or neutral drift then works on these genetic materials to guide their evolutionary directions. Here, I used the salicylic acid/benzoic acid/theobromine (SABATH) multigene family to study how natural selection acted on duplicated genes to prompt functional diversification. Members in this family methylate plant secondary metabolites by transferring the methyl group from S-adenosyl-L-methionine (SAM) to the carboxyl group or ring nitrogen of the substrates. The question of whether and how apparently "altruistic" behaviors can evolve in social animals has received an enormous amount of attention from evolutionary biologists and has been termed "the central theoretical problem of sociobiology". Phylogeography is defined as the spatial distribution of taxa with respect to geologic and geographic events. It is well documented that the distributions of many taxa have been affected by glacial events during the Pleistocene Era. The patterns generated can be very complex and result from shifts in climate and/or vegetation.

Dollar sunfish (Lepomis marginatus) at the Savannah River Site were sampled from two radiocesium contaminated ponds with a history of thermal elevation and from reference ponds with no history of radioactive or thermal contamination. Fluorescent sequencing of a portion of the mitochondrial control region showed no genetic variation within or between populations. Centrarchidae microsatellite loci Lma 120 (6 alleles, N=124 from 5 populations), Lma 20 (10 alleles, N=37 from 3 populations) and Rb 7 (14 alleles. N=17 from 2 populations) were variable for dollar sunfish. An additional 9 microsatellite primer pairs were designed for dollar sunfish. Testing of the designed …

The study was conducted to determine the effects of lowered genetic variance in the Western mosquitofish (Gambusia affinis) on the physiological parameters of critical thermal maximum and minimum pH. Mosquitofish were obtained from two small central Illinois ponds, Lost Pond and Shadow Pond, by seines and dip nets. Once caught, fish were returned to the laboratory and acclimated for a minimum of five days at a temperature of 27°C and a pH of 8.25. At the beginning of each experimental run, fish were subjected to the physiological experiments for determination of critical thermal maximum and minimum pH, and then measured …

As part of a biosystematic study of Panus conchatus (Bull.: Fr.) Fr., North American and European herbarium specimens from the Field Museum and the University of Michigan were examined and a description of the taxon prepared. Tissue and single spore isolates were obtained from basidiocarps collected in Illinois. Single and polyspore isolates were obtained from a collection made in Switzerland by R. H. Petersen. Culture mat analyses were completed on several media and the taxonomic value of cultural characters assessed. Cloning And Sequencing Of The Eco Rii Monomer Fragment Of Satellite Dna From The Mouse Mus Musculus, Timothy Patrick Boyle

Total mouse satellite DNA was isolated in cesium chloride gradients in the presence of the dye Hoechst 33258. This satellite fraction was digested with Bst Nl, an isoschizomer of the restriction enzyme Eco RII, and the 234 base pair monomer fragment was cloned into the plasmid vector pBS. Dideoxynucleotide sequencing of the cloned DNA yielded significant homology with the published consensus sequence of mouse satellite DNA (92%).

The lipopolysaccharides (LPS) from Rhizobium fredii USDA 205 and its nod- mutant, HC 205, which has been cured of the 192-MDa symbiotic plasmid, were isolated by the phenol-water extraction method. The LPS from the wild-type strain which was grown in association with host-plant root extract or a flavone analog to a constituent of the root extract, which is required for the induction of at least a portion of the nodulation genes, was also isolated. Strain Differences And Mutagen Treatment Influence Lectin Binding To Mouse Germ Cells, Marilyn Schmalz Holm

Carbohydrate moieties on mouse germ cell surfaces were characterized by investigating the binding of FITC-labeled lectins to sperm and testis cells. Differences in lectin binding between different mouse strains, including T/t mice, were found. However, there was no evidence for the involvement of sialic acid or L-fucose in the T/t locus antigens. Antiserum to the t0, t6 haplotype was equally reactive to both ICR and T/t mouse testis cells in both immunofluorescence and cytotoxicity assays. Trypsinization of testis cells increased binding of soybean agglutinin and decreased binding of wheat germ agglutinin.

Groups of 5 male HA(ICR) mice were injected intraperitoneally with 60, 150, 300, or 600 mg/kg body weight of ethylmethanesulfonate or with a saline control. Each male was mated to two untreated females at two weeks after treatment and then again at five weeks after treatment. The treatment groups were labelled post-meiotic and pre-meiotic relative to the stage of spermatogenesis at time of treatment. Progeny were evaluated by observations of litter size, body weight, negative geotactic response, swimming patterns, limb use, water escape time and open field motor coordination activity. The compositions and sugar linkages of the acidic extracellular polysaccharide (EPS) from several Rhizobium trifolii mutants were compared. These mutants were derived from a wild type Rhizobium trifolii, ANU843, and are unable to cause normal root hair curling, Hac-, or nodulate clover, Nod-. Three of the mutants contain a Tn5 insertion in a 5.3 kb BglII fragment of the symbiotic plasmid (pSym). A fourth mutant is missing the pSym, which includes much of the 5.3 kb BglII fragment, both of these deletions result in Hac-Nod- phenotypes. Thirty-one mass collections of Claytonia virginica were made in eight counties in the eastern half of Illinois. Chromosome counts of pollen mother cells were determined for plants from Richland, Coles, Clark, Douglas, Cook and Moultrie counties. A range of 2n = 8 to 48 was determined with only 4 of the 89 counts being greater than 2n = 30. For all the populations, counts of 2n = 8, 10, 12, 14, 15, 16, 17, 18, 19, 20, 21, 22, 24, 27, 28, 29, 30, 37, 38, 44, 48 were reported. The two most common counts were 2n = 18 and …

The establishment of normal chromosomal patterns for a wide range of plants and animals has led to the recognition of abnormalities as well as their relationship to phenotypic irregularities. Research dealing with the expression of human karyotype is of prime interest; but, closely rivaling that is the genetic study of domestic animals which are of economic importance to man.

The development of a method for karyotype determination of cercaria provides a basis for comparative karyotype analysis between cercaria and adult, possibly eliminating the need for infection experiments for positive identification of cercaria. Snails (Goniobasis livescens) were collected from below the spillway of Lake Charleston, on the Embarras River, near Charleston, Illinois. Of a total of 196 snails collected 42 (21.5%) were found to be infected with undetermined furcocyticercous cercariae. Infected snails were detected through isolation procedure. This investigation involves the cytogenetics of three strains of field-collected, wild-type Illinois Drosophila melanogaster. These three strains were collected from Charleston, Channahon, and Olney, Illinois, in 1967.

The purpose of this study was to compare cytogenetically these three strains and to determine if there exists differences among them. First, the chromosomes themselves were studied for the purpose of identifying the four chromosomes of D. melanogaster by their characteristic banding patterns. Then, the chromosomes of the three strains were examined for chromosomal aberrations, principally inversions. The cytogenetics of the chromosomes of the three strains were then compared and contrasted.
