Vertebrate retinal development requires timely and precise fusion of the optic fissure. Failure of this event leads to congenital vision impairment in the form of coloboma. Recent studies have suggested hyaloid vasculature to be involved in OF fusion. In order to examine this link, we analyzed optic fissure fusion and hyaloid vasculogenesis in the zebrafish pax2a noi mutant line. We first determined that pax2a-/- embryos fail to accumulate F-actin in the optic fissure prior to basement membrane (BM) degradation. While the expression of circadian rhythms is nearly universal among multicellular eukaryotic organisms, demonstration of this phenomenon in prokaryotes has been largely restricted to photosynthetic cyanobacteria until very recently. Further, growing interest in gastrointestinal microbiomes has revealed a complex temporal relationship between the gastrointestinal clock and the bacterial microbiome within. At least one member of the gut microbiome, Klebsiella (née Enterobacter) aerogenes, responds to the indoleamine hormone melatonin, secreted by the gastrointestinal system itself. Further research revealed that K. aerogenes also expresses a circadian rhythm in motility and gene expression that is temperature compensated. Although rhythmicity is unaltered …

Under pathophysiological conditions, the microtubule protein tau (MAPT) forms neurofibrillary tangles that are the hallmark of sporadic Alzheimer’s disease as well as familial frontotemporal dementias linked to chromosome 17 (FTDP-17). In this work, I report that MAPT forms circular RNAs through backsplicing of exon 12 to either exon 10 or exon 7 (12→10; 12→7), and that these circular RNAs are translated into proteins.

Using stable cell lines overexpressing the circular tau RNAs 12→7 and 12→10, we have discovered that the tau circular RNA 12→7 is translated in a rolling circle, giving rise to multiple proteins. This circular RNA …

Parasitoid wasps are effective biological control agents that have evolved intricate host immune suppression strategies to overcome host immune defenses. The most important anti-parasite defense mechanism is encapsulation, where the host’s blood cells surround the wasp egg. In the Drosophila/Leptopilina host-parasitoid system, extracellular vesicles (EVs) are oviposited along with the wasp egg. The EVs possess spikes and contain hundreds of proteins, some of which enable wasp eggs to escape encapsulation by changing the morphology and function of hemocytes, or killing them. When tissue samples are studied in bulk without consideration for different cell proportions and types, results can be biased due to the attenuation of unique cellular expressions. In order to study cell type specific RNA expression profiles within tissue, single cell RNA sequencing (scRNA-seq) is used. For scRNA-seq studies it is critical to have intact cells. เชื้อแบคทีเรียแกรมลบ Acinetobacter baumannii เป็นเชื้อฉวยโอกาส ที่ก่อให้เกิดการติดเชื้อในโรงพยาบาล และมีการดื้อต่อยาปฏิชีวนะหลายชนิด จากหลายกลไกการดื้อยา รวมถึงการใช้ efflux pump ขับยาออกนอกเซลล์ การศึกษาครั้งนี้ผู้วิจัยได้ทดสอบหาค่าความเข้มข้นต่ำสุดที่ยับยั้งการเจริญเติบโตของเชื้อ (MIC) ต่อยา amikacin ในเชื้อ A. baumannii ทั้งหมด 8 ตัวอย่าง ที่ได้จากสิ่งส่งตรวจของผู้ป่วย และเชื้อสายพันธุ์มาตรฐาน ATCC 19606 พบว่าเชื้อ A. baumannii 7 ตัวอย่างมีค่า MIC อยู่ในช่วง 0.25 – 4 µg/mL (ไวต่อยา) และเชื้ออีก 2 ตัวอย่างมีค่า MIC เท่ากับ 64 และมากกว่า 256 µg/mL (ดื้อต่อยา) เมื่อเหนี่ยวนำเชื้อด้วยยา amikacin โดยเพาะเลี้ยงเชื้อในสภาวะที่มียา amikacin ความเข้มข้นเพิ่มขึ้นเป็นสองเท่า ทุก 48 ชั่วโมง ติดต่อกันเป็นเวลาหลายวัน พบว่าเชื้อ A. baumannii หลังเหนี่ยวนำด้วยยา amikacin ทั้ง 8 ตัวอย่าง มีค่า MIC เพิ่มขึ้นอยู่ในช่วง 32 ถึงมากกว่า 256 µg/mL ผลการศึกษาการใช้สารยับยั้ง efflux pump ได้แก่ สาร carbonylcyanide m-chlorophenylhydrazone (CCCP) พบว่าเชื้อ A. baumannii หลังเหนี่ยวนำด้วยยา amikacin ทั้ง 8 ตัวอย่าง มีค่า MIC ต่อยา amikacin เมื่อมีสาร CCCP ลดลงมากกว่าหรือเท่ากับ 4 เท่าของค่า MIC ต่อยา amikacin ที่ไม่มีสาร …

การเพาะเลี้ยงสัตว์น้ำเพื่อเป็นแหล่งอาหาร มีผลต่อความมั่นคงทางอาหารและโภชนาการของมนุษย์ โดยกุ้งทะเลจัดเป็นสัตว์น้ำเศรษฐกิจอันดับหนึ่งของไทย ตั้งแต่ปี พ.ศ. 2534 เป็นต้นมา เนื่องจากเป็นสัตว์น้ำที่ได้รับความนิยมบริโภคทั้งในประเทศและต่างประเทศทั่วโลก แต่ในปี พ.ศ. 2555-2556 การเลี้ยงกุ้งเริ่มประสบปัญหาโรคระบาดอย่างหนักจากอาการตายด่วนเนื่องจากตับและตับอ่อนวายเฉียบพลัน (Early mortality syndrome/ acute hepatopancreatic necrosis disease : EMS/AHPND) ทําให้เกิดความเสียหายต่อเกษตรกรและส่งผลกระทบต่อภาคอุตสาหกรรม รวมไปถึงสูญเสียส่วนแบ่งการตลาดและมูลค่าการส่งออกกุ้งทะเลเป็นอย่างมาก ต่อมาได้มีการพัฒนาจุลินทรีย์ ปม.1 ที่เป็นโพรไบโอติกในกลุ่มบาซิลัส เพื่อลดอัตราการเกิดโรค ซึ่งเชื้อ B. licheniformis ที่รวมในกลุ่มโพรไบโอติกนี้ มีความสามารถสร้างแบคทีริโอซินที่ยับยั้งการเจริญของเชื้อก่อโรคได้ มีคุณสมบัติทนความร้อน 100 °C ทำงานได้ดีในค่าความเป็นกรด-ด่างกว้าง จึงเป็นที่น่าสนใจในการศึกษาสภาวะที่เหมาะสมเพื่อเพิ่มปริมาณการสร้างแบคทีริโอซินของเชื้อ B. licheniformis และประสิทธิภาพในการยับยั้งเชื้อก่อโรค จากการทดสอบเพาะเลี้ยงเชื้อ B. licheniformis ในอาหารเหลว Tryptic Soy Broth มีความเข้มข้นของเชื้อเป็น 106 CFU/มิลลิลิตร ในสภาวะที่ปรับเปลี่ยนอุณหภูมิ ช่วงเวลา และความเข้มข้นของเกลือที่ต่างกัน ผลการทดสอบประสิทธิภาพการยับยั้งเชื้อก่อโรคในกุ้งด้วยวิธี Total plate count พบว่า ในน้ำเลี้ยงเชื้อ (Cell Free Supernatant: CFS) สามารถยับยั้งเชื้อทดสอบได้แก่ เชื้อ Vibrio alginolyticus และ Vibrio parahaemolyticus ซึ่งเป็นสายพันธุ์ที่เป็นสาเหตุของโรคตับและตับอ่อนวายเฉียบพลันในกุ้งได้ดี เมื่อเพาะเลี้ยงเชื้อ B. licheniformis ที่อุณหภูมิ 30 องศาเซลเซียสเป็นเวลา 72 ชั่วโมง และเลี้ยงในอาหารเหลว TSB ที่มีเกลือความเข้มข้น 2.5% และเมื่อเลี้ยง B. licheniformis ร่วมกับเซลล์ของเชื้อทดสอบที่ถูกทำให้ตายด้วยความร้อน พบว่าน้ำเลี้ยงเชื้อยังคงมีประสิทธิภาพในการยับยั้งเชื้อทดสอบ จากนั้นตรวจวิเคราะห์ประสิทธิภาพการทำลายเซลล์ของแบคทีเรียก่อโรคของน้ำเลี้ยงเชื้อด้วยกล้องจุลทรรศน์อิเลคตรอนพบว่าส่วนที่ห่อหุ้มเซลล์ของเชื้อทดสอบมีลักษณะเป็นรู นอกจากนี้พบว่าสารยับยั้งในน้ำเลี้ยงเชื้อที่เกิดจากการเพาะเลี้ยงและจาก B. licheniformis ร่วมกับเซลล์ V. alginolyticus ที่ถูกทำให้ตายด้วยความร้อน มีความสามารถทำให้เซลล์ของเชื้อทดสอบมีขนาดเล็กลง และลดการเกาะกลุ่มของเชื้อทดสอบได้ดี ผลจากการวิเคราะห์ลำดับกรดอะมิโนในน้ำเลี้ยงเชื้อ (Cell-Free …

วัณโรคดื้อยาหลายขนาน (Multidrug-resistant tuberculosis: MDR-TB) เป็นหนึ่งในสาเหตุหลักที่ทำให้การรักษาวัณโรคล้มเหลว และส่งผลให้การแพร่ระบาดของโรคยังคงมีอัตราที่สูง วัณโรคดื้อยาหลายขนานเกิดจากเชื้อ Mycobacterium tuberculosis ที่ดื้อต่อยา Rifampicin และยา Isoniazid พร้อมกัน โดยมากกว่า 90% ของวัณโรคที่ดื้อยา Rifampicin มักเกิดการดื้อยา Isoniazid ร่วมด้วยภายหลัง ดังนั้นการตรวจวินิจฉัยการดื้อยา Rifampicin ของเชื้อ M. tuberculosis จึงสามารถใช้เป็นตัวแทนในการตรวจหาวัณโรคดื้อยาหลายขนานได้ วิธีทางอณูชีววิทยาที่ใช้วินิจฉัยวัณโรคดื้อยาหลายขนานในปัจจุบันต้องอาศัยเครื่องมือที่มีราคาแพงและการดูแลซ่อมบำรุงอย่างสม่ำเสมอ การเพิ่มปริมาณสารพันธุกรรมภายใต้อุณหภูมิคงที่ได้เข้ามามีบทบาทอย่างมากในการวินิจฉัยโรคทางห้องปฏิบัติการและเป็นทางเลือกสำหรับการทดสอบที่ไม่ต้องอาศัยเครื่องมือที่จำเพาะ สามารถให้ผลการทดสอบที่ถูกต้อง รวดเร็ว และนำไปประยุกต์ใช้ ณ งานภาคสนามได้ การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิค Multiplex allele-specific recombinase polymerase amplification ร่วมกับ dipstick chromatography (MAS-RPA-DC) สำหรับตรวจหาการกลายพันธุ์ภายในยีน rpoB ณ ตำแหน่งโคดอน 516 526 และ 531 ซึ่งสัมพันธ์กับการดื้อยา Rifampicin ของเชื้อ M. tuberculosis ในการทดสอบภายใต้ปฏิกิริยาเดียวกันและแบบอ่านผลด้วยตาเปล่า ภายหลังนำเทคนิค MAS-RPA-DC มาทดสอบกับ DNA ที่สกัดได้จากโคโลนีของเชื้อ M. tuberculosis สายพันธุ์คลินิกจำนวน 141 ตัวอย่าง พบว่าเทคนิค MAS-RPA-DC มีความไวและความจำเพาะที่สูงเมื่อเปรียบเทียบกับเทคนิค Sanger DNA sequencing โดยมีความไวในการตรวจการกลายพันธุ์ของยีน rpoB516 rpoB526 และ rpoB531 มีค่าเท่ากับ 100 % 70 % และ 98.2 % ตามลำดับ มีค่าความจำเพาะในการตรวจการกลายพันธุ์ของยีน rpoB516 rpoB526 และ rpoB531 เท่ากับ 95.5 % 97.5 % และ 97.7 …

Fish assemblages are often assessed as a biological proxy for environmental health. While humans value healthy environments for the ecosystem services and recreational opportunities they provide, it is increasingly evident that such resources can be paradoxically degraded by anthropogenic activities. In this investigation, we studied the relationship between different intensities of anthropogenic land-use change and habitat-driven fish assemblage response across multiple spatiotemporal scales. Current presumptive tests lack the sensitivity and specificity molecular techniques can achieve; therefore, molecular methods, such as microRNA and microbial signatures, have been extensively researched in the forensic community. Neuromodulation is a nearly ubiquitous process that endows the nervous system with the capacity to alter neural function at every level (synaptic, circuit, network, etc.) without necessarily adding new neurons. Through the actions of neuromodulators, the existing neural circuitry can be adaptively tuned to achieve flexible network output and similarly dynamic behavioral output. A critical barrier to achieving a functional cure for infection by human immunodeficiency virus type one (HIV-1) rests in the presence of latent proviral DNA integrated in the nuclei of host CD4+ T cells and mononuclear phagocytes. Accordingly, HIV-1-infected patients must adhere to lifelong regimens of antiretroviral therapy (ART) to prevent viral rebound, CD4+ T cell decline, and progression to acquired immunodeficiency syndrome (AIDS). Abstract: In 2013, we observed the presence of an undocumented Ammocrypta species in the lower Flint River, Georgia. The occurrence represents the first record of the genus in Georgia. Subsequent surveys at 24 sites, using seining or snorkeling, documented additional specimens from mainstem sites (n = 3) between Albany and Bainbridge and from Ichawaynochaway Creek (n = 5 sites), a large tributary to the Flint River. We used morphological and genetic data to identify specimens to species. Mechanotransduction is the process by which a mechanical stimulus is converted to a cellular signal. This process is heavily influential of cell morphology, differentiation, and behavior. However, altered levels of mechanical stimuli are also found in many pathological contexts. Sex-determining region Y (SRY) protein is the master switch in the initiation of male sex differentiation. Mutation in SRY gene results in ambiguous genitalia and abnormalities in reproductive organs. Its function is mainly controlled by its high mobility group (HMG) box. Damage to the HMG box may cause dysfunction of the SRY protein, which may, in turn, lead to sex reversal. This study was conducted to prioritize the deleterious effects of the non-synonymous single nucleotide polymorphisms (nsSNPs) on SRY protein. The normal flora community plays a significant role in both healthy and patient individual’s bodies. It has improved its ability to change from beneficial to opportunistic organisms causing crucial infections in immunocompromised patients. This study was conducted to identify the community of yeast species, and to investigate potential virulence factors. Saliva samples were collected from healthy individuals, yeast species were isolated and identified using both traditional and advance molecular genetics technique. The results revealed that Candida albicans was the dominant isolate, while Meyerozyma caribbica was the least. Tauopathies are neurodegenerative diseases, including Alzheimer’s disease, that are associated with pathological accumulation of the microtubule associated protein tau (MAPT, or tau) (Lee et al., 2001). Abnormal hyperphosphorylated tau (pTau) strongly correlate with cognitive impairment (Nelson et al., 2012). Neuroinflammation is also associated with tauopathies (Gerhard et al., 2006b; Edison et al., 2008) and is implicated in driving tau pathology (Yoshiyama et al., 2007, Maphis et al., 2015b). Turtles are iconic reptiles that inhabit a range of ecosystems from oceans to deserts and climates from the tropics to northern temperate regions. Yet, we have little understanding of the genetic adaptations that allow turtles to survive and reproduce in such diverse environments. Studies on population genetics examine the relationship and effects of population structure, migration, gene flow and demographic history, and are therefore important in the conservation of endangered species. Astragalus jaegerianus, a critically federally endangered species found in a geographically restricted range is investigated to determine population structure and genetic variation. Previous research on A. jaegerianus focused on DNA sequence data for cpDNA and nrDNA showed no variation. Further research on A. jaegerianus utilizing AFLP’s on the whole genome indicated substantial gene diversity and population structure consistent with geographically widespread species. Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy with a very poor patient prognosis (5-year survival of ≤ 7%). While transcriptional profiling has aided in the classification of this disease into at least two broader subtypes, this alone has so far been insufficient to inform on more nuanced patterns of oncogenic dependency. We hypothesized that a more comprehensive and granular characterization of PDAC disease diversity is required to establish relevant context for targeted therapy. Cell stress and DNA damage activate the tumor suppressor p53, triggering transcriptional activation of a myriad of target genes. The molecular, morphological, and physiological consequences of this activation remain poorly understood in vivo. We activated a p53 transcriptional program in mice by deletion of Mdm2, a gene which encodes the major p53 inhibitor. By overlaying tissue-specific RNA-sequencing data from pancreas, small intestine, ovary, kidney, and heart with existing p53 ChIP-sequencing, we identified a large repertoire of tissue-specific p53 genes and a common p53 transcriptional signature of seven genes which included Mdm2 but not p21. Accurate and cost-effective PCR based sex identification is important in animal production because it gives producers the ability to determine the sex of embryos prior to transfer, saving time and money. The skin color of muscadine grapes (Vitis rotundifolia) is typically classified as black or bronze. A glutathione S-transferase, VrunGST4, has been identified as a candidate gene for berry skin color in muscadine grapes. A molecular marker was developed within VrunGST4 to distinguish between muscadine genotypes (cultivars and selections) with bronze (T:T), heterozygote black (C:T), and homozygote black (C:C) berries. The growing prevalence and diversity of carbapenemase producers among carbapenem-resistant Pseudomonas aeruginosa (CRPA) isolates warrants an expansion of detection capabilities. The purpose of this study was to evaluate the performance of the commercially available Xpert Carba-R (Carba-R) and the research-use-only Xpert Carba-R NxG (Carba-R NxG) in a global collection of P. aeruginosa. The challenge set included 123 P. aeruginosa clinical isolates from 12 countries. Isolates were previously categorized via PCR or whole-genome sequencing. Carbapenemase classes tested include VIM, IMP, NDM, SPM, KPC, and GES. Non-carbapenemase (non-CP)-harboring isolates were also tested (negative control). Fungi perform several critical functions in the environment. Spatiotemporal distributions of fungal communities will mediate when and where these functions happen and how they vary across the landscape. I first explored tropical tree canopy fungal community variation at small spatial scales and documented near total turnover of fungi across sub-meter distances and among adjacent substrates. The second chapter analyzed fungal turnover over the course of three years, where community stability was driven primarily by abundant fungi. RNA interference (RNAi) is a wide-spread gene silencing mechanism that control diverse biological functions and triggered by small interfering RNAs (siRNAs) processed from the viral genome or its replication intermediates. Mechanistic studies of antiviral RNAi in Caenorhabditis elegans has led to the identification of several critical factors involved in the process.
