Nutrition and growth are strongly linked, but not much is known about how nutrition leads to growth. To understand the connection between nutrition through diet, growth and/or proliferation, we need to study the phenotypes resulting from the activation and inhibition of central metabolic pathways. Here I use the model organism Drosophila melanogaster (D. mel.) and three non-model Drosophila species with different dietary needs, Drosophila guttifera (D. gut.), Drosophila deflecta (D. def.), and Drosophila tripunctata (D. tri.), to study the effects of dietary amino acid availability in these diverse flies on fecundity and …

The vertebrate eye is a complex organ, responsible for the primary sense with which we interact with our environment: vision. Development of the eye is a tightly regulated process, controlled by a vast network of genes. This process begins with eye morphogenesis, when the eye structure is formed through a series of morphogenetic movements and culminates in the creation of the optic cup, lens, and presumptive optic stalk. Next, retinal differentiation creates the critical cell layers of the retina needed to process light waves that enter the eye, including rod and cone photoreceptors, interneurons, and support cells. Cellular expansion in plants is a complex process driven by the constraint of internal cellular turgor pressure by an expansible cell wall. The main structural element of the cell wall is cellulose. Cellulose is vital to plant fitness and the protein complex that creates it is an excellent target for small molecule inhibition to create herbicides. In the following thesis many small molecules (SMs) from a diverse library were screened in search of new cellulose biosynthesis inhibitors (CBI). Mitochondria are complex organelles that generate most of the energy required to sustain life and function in metabolic and signaling pathways required to maintain cellular homeostasis. MNRR1 (mitochondrial nuclear retrograde regulator 1 or CHCHD2) is a small, bi-organellar twin CX9C protein that is emerging as an important regulator of mitochondrial function, apoptosis, and cellular stress by participating in mitochondrial-nuclear crosstalk. Our lab has previously shown that in the mitochondria, MNRR1 regulates complex IV (Cytochrome c oxidase or COX) and is able to finetune the oxidase function through phosphorylation status. Vertebrate retinal development requires timely and precise fusion of the optic fissure. Failure of this event leads to congenital vision impairment in the form of coloboma. Recent studies have suggested hyaloid vasculature to be involved in OF fusion. In order to examine this link, we analyzed optic fissure fusion and hyaloid vasculogenesis in the zebrafish pax2a noi mutant line. We first determined that pax2a-/- embryos fail to accumulate F-actin in the optic fissure prior to basement membrane (BM) degradation. Asphondylia borrichaie is a small fly (Diptera: Cecidomyiidae) that is currently undergoing host-associated sympatric divergence. Asphondylia borrichaie is an ambrosia galler, these insects utilize a host plant for oviposition, but its offspring also rely on a symbiotic fungus (or fungal community) to promote the formation of the gall as well as serve as a food source for the developing larvae. DNA profiles do not provide as much evidentiary value when either all known samples can be excluded or when there is no known suspect. DNA phenotyping is a technique used for investigative purposes to predict externally visible characteristics (EVCs) based on specific markers found in the genome. การเพาะเลี้ยงสัตว์น้ำเพื่อเป็นแหล่งอาหาร มีผลต่อความมั่นคงทางอาหารและโภชนาการของมนุษย์ โดยกุ้งทะเลจัดเป็นสัตว์น้ำเศรษฐกิจอันดับหนึ่งของไทย ตั้งแต่ปี พ.ศ. 2534 เป็นต้นมา เนื่องจากเป็นสัตว์น้ำที่ได้รับความนิยมบริโภคทั้งในประเทศและต่างประเทศทั่วโลก แต่ในปี พ.ศ. 2555-2556 การเลี้ยงกุ้งเริ่มประสบปัญหาโรคระบาดอย่างหนักจากอาการตายด่วนเนื่องจากตับและตับอ่อนวายเฉียบพลัน (Early mortality syndrome/ acute hepatopancreatic necrosis disease : EMS/AHPND) ทําให้เกิดความเสียหายต่อเกษตรกรและส่งผลกระทบต่อภาคอุตสาหกรรม รวมไปถึงสูญเสียส่วนแบ่งการตลาดและมูลค่าการส่งออกกุ้งทะเลเป็นอย่างมาก ต่อมาได้มีการพัฒนาจุลินทรีย์ ปม.1 ที่เป็นโพรไบโอติกในกลุ่มบาซิลัส เพื่อลดอัตราการเกิดโรค ซึ่งเชื้อ B. licheniformis ที่รวมในกลุ่มโพรไบโอติกนี้ มีความสามารถสร้างแบคทีริโอซินที่ยับยั้งการเจริญของเชื้อก่อโรคได้ มีคุณสมบัติทนความร้อน 100 °C ทำงานได้ดีในค่าความเป็นกรด-ด่างกว้าง จึงเป็นที่น่าสนใจในการศึกษาสภาวะที่เหมาะสมเพื่อเพิ่มปริมาณการสร้างแบคทีริโอซินของเชื้อ B. licheniformis และประสิทธิภาพในการยับยั้งเชื้อก่อโรค จากการทดสอบเพาะเลี้ยงเชื้อ B. licheniformis ในอาหารเหลว Tryptic Soy Broth มีความเข้มข้นของเชื้อเป็น 106 CFU/มิลลิลิตร ในสภาวะที่ปรับเปลี่ยนอุณหภูมิ ช่วงเวลา และความเข้มข้นของเกลือที่ต่างกัน ผลการทดสอบประสิทธิภาพการยับยั้งเชื้อก่อโรคในกุ้งด้วยวิธี Total plate count พบว่า ในน้ำเลี้ยงเชื้อ (Cell Free Supernatant: CFS) สามารถยับยั้งเชื้อทดสอบได้แก่ เชื้อ Vibrio alginolyticus และ Vibrio parahaemolyticus ซึ่งเป็นสายพันธุ์ที่เป็นสาเหตุของโรคตับและตับอ่อนวายเฉียบพลันในกุ้งได้ดี เมื่อเพาะเลี้ยงเชื้อ B. licheniformis ที่อุณหภูมิ 30 องศาเซลเซียสเป็นเวลา 72 ชั่วโมง และเลี้ยงในอาหารเหลว TSB ที่มีเกลือความเข้มข้น 2.5% และเมื่อเลี้ยง B. licheniformis ร่วมกับเซลล์ของเชื้อทดสอบที่ถูกทำให้ตายด้วยความร้อน พบว่าน้ำเลี้ยงเชื้อยังคงมีประสิทธิภาพในการยับยั้งเชื้อทดสอบ จากนั้นตรวจวิเคราะห์ประสิทธิภาพการทำลายเซลล์ของแบคทีเรียก่อโรคของน้ำเลี้ยงเชื้อด้วยกล้องจุลทรรศน์อิเลคตรอนพบว่าส่วนที่ห่อหุ้มเซลล์ของเชื้อทดสอบมีลักษณะเป็นรู นอกจากนี้พบว่าสารยับยั้งในน้ำเลี้ยงเชื้อที่เกิดจากการเพาะเลี้ยงและจาก B. licheniformis ร่วมกับเซลล์ V. alginolyticus ที่ถูกทำให้ตายด้วยความร้อน มีความสามารถทำให้เซลล์ของเชื้อทดสอบมีขนาดเล็กลง และลดการเกาะกลุ่มของเชื้อทดสอบได้ดี ผลจากการวิเคราะห์ลำดับกรดอะมิโนในน้ำเลี้ยงเชื้อ (Cell-Free …

Combined Oxidative Phosphorylation Deficiency 23 (COXPD23) caused by mutations in GTPBP3 gene is a rare mitochondrial disease, and this disorder identified from the Chinese population has not been described thus far. Here, we report a case series of three patients with COXPD23 caused by GTPBP3 mutations, from a severe to a mild phenotype. The main clinical features of these patients include lactic acidosis, myocardial damage, and neurologic symptoms. Genetic analysis of reintroduced populations is vital to assess genetic diversity as an indicative of population viability. Fungus-growing ants (Tribe Attini) and their fungal cultivars share a 50-million-year coevolutionary history. Large scale phylogenetic analyses depict a strong co-phyletic signal among ants and their farmed fungi yet fungus sharing among unrelated ant lineages is somewhat widespread. An overview of sharing has been hampered by a lack of genetic markers that exhibit intraspecific variation and surveys across geographic regions. An ancient and ubiquitous set of enzymes known as the aminoacyl-tRNA synthetases are required for the viability of all organisms. The aminoacyl-tRNA synthetases catalyze the attachment of amino acids onto tRNA molecules. The aminoacylated, or charged, tRNA is then transported and utilized at the ribosome for the synthesis of proteins. The genome of the hyperthermophilic microorganism Sulfolobus islandicus (S. islandicus) harbors a unique leucyl-tRNA synthetase paralog, LeuRS-I, of unknown function. Genome integrity is necessary to prevent mutations and disease. During eukaryotic DNA replication, DNA polymerases ε (Polε) and δ (Polδ) synthesize the leading and lagging strand, respectively. Polε and Polδ also have exonuclease activity that acts in series with post-replicative mismatch repair (MMR) to remove replication errors. Defects in proofreading and MMR lead to an increase in mutations and cause cancer in humans. This dissertation focuses on several unresolved issues involving the relationship between Polε and Polδ in replication error avoidance. Mechanotransduction is the process by which a mechanical stimulus is converted to a cellular signal. This process is heavily influential of cell morphology, differentiation, and behavior. However, altered levels of mechanical stimuli are also found in many pathological contexts. Typified by their branching pattern, headwaters are numerically abundant as the density of these habitats increases with increasing distance from the base of a dendritic river system. Connectivity among headwaters is complex, resulting in the spatial isolation of populations. Headwater specialists have evolved a suite of traits that permit these species to permanently reside within these habitats. The spatial configuration and connectivity of headwaters has repercussions for metapopulations and meta-assemblages. Neurofibromatosis Type 2 (NF2) is a rare genetic disorder, classified as a neurocutaneous syndrome inherited through an autosomal dominant genetic mutation or deletion of the NF2 gene in humans. The tumor suppressor gene NF2 encodes the cytoskeletal protein MERLIN to promote apoptosis and inhibit abnormal cellular proliferation. Mutations in NF2 lead to a deficiency of functional MERLIN in humans and can result in hearing loss, disability, and premature death. The observed overlap between genetic variants associated with both adiposity and inflammatory markers suggests that changes in both adiposity and inflammation could be partially mediated by common pathways. The pervasive but sparsely characterized “pleiotropic” genetic variants associated with both adiposity and inflammation have been hypothesized to provide insight into the shared biology. This study explored and characterized the genetic pleiotropy underpinning adiposity and inflammation using genetic and phenotypic observations from the Cameron County Hispanic Cohort (CCHC). Studies on population genetics examine the relationship and effects of population structure, migration, gene flow and demographic history, and are therefore important in the conservation of endangered species. Astragalus jaegerianus, a critically federally endangered species found in a geographically restricted range is investigated to determine population structure and genetic variation. Previous research on A. jaegerianus focused on DNA sequence data for cpDNA and nrDNA showed no variation. Further research on A. jaegerianus utilizing AFLP’s on the whole genome indicated substantial gene diversity and population structure consistent with geographically widespread species. Multiple Myeloma (MM) constitutes 10% of diagnosed hematologic malignancies in the US, with over 12,000 deaths recorded each year. Race/ethnicity is a well-known MM risk factor, where individuals of African descent have over 2- to 3-fold increased risk of incidence compared to those of European descent. Additionally, Hispanics are diagnosed approximately three years younger than white American counterparts, for unknown reasons. Differences in clinical phenotype are also present for MM patients by ancestry, including varying rates of common initiation mutations such as IgH translocations and TP53 mutation between patients of European and African descent. Cell stress and DNA damage activate the tumor suppressor p53, triggering transcriptional activation of a myriad of target genes. The molecular, morphological, and physiological consequences of this activation remain poorly understood in vivo. We activated a p53 transcriptional program in mice by deletion of Mdm2, a gene which encodes the major p53 inhibitor. By overlaying tissue-specific RNA-sequencing data from pancreas, small intestine, ovary, kidney, and heart with existing p53 ChIP-sequencing, we identified a large repertoire of tissue-specific p53 genes and a common p53 transcriptional signature of seven genes which included Mdm2 but not p21. Lineage transcription factors mark, promote, and maintain multiple distinct cell types originating from a common progenitor. Despite their essential role, how such factors function and bind genome wide to orchestrate the epigenetic changes necessary to form and maintain these identities in vivo is unclear.
