Optimization Of Pre-Enrichment And Screening Methods For The Detection Of Salmonella Enterica In Meat Analog Products, Kylie Sacapano May 2024

Optimization Of Pre-Enrichment And Screening Methods For The Detection Of Salmonella Enterica In Meat Analog Products, Kylie Sacapano

Food Science (MS) Theses

With the growing demand and variety of meat analogs on the commercial market, there is a need to revisit the methodology for the detection of foodborne pathogens in these products. The current Food and Drug Administration (FDA) regulatory methods for Salmonella enterica detection in meat analogs follow the Bacteriological Analytical Manual (BAM) and are time-consuming and costly, requiring pre-enrichment using lactose broth (LB) with a surfactant (e.g., Triton X-100). Therefore, this study aimed to optimize pre-enrichment and screening methods for the detection of Salmonella in meat analogs by comparing the current FDA reference methods to updated techniques. To achieve this, …

Disinfection Of Foodborne Bacteria Using The Contamination Sanitization Inspection And Disinfection (Csi-D) Device, Jennifer Mccoy Sanders Dec 2023

Disinfection Of Foodborne Bacteria Using The Contamination Sanitization Inspection And Disinfection (Csi-D) Device, Jennifer Mccoy Sanders

Food Science (MS) Theses

Foodborne pathogens, including Escherichia coli, Salmonella enterica, and Listeria monocytogenes, are major causes of gastrointestinal disease globally. The Contamination Sanitization Inspection and Disinfection (CSI-D) device is a new handheld fluorescence-based imaging system designed to disinfect food contact surfaces contaminated with microorganisms using ultraviolet-C (UVC) illumination. This study aimed to determine the optimal parameters for the disinfection of foodborne bacteria using the CSI-D device. The following bacterial strains were tested: generic E. coli; enterohemorrhagic E. coli (EHEC) O157:H7; enterotoxigenic E. coli (ETEC) O78:H11; S. enterica serotypes Enteritidis, Newport, Typhimurium, and Javiana; and L. monocytogenes serotypes 1/2a, 1/2b, and 4b …

Single-Laboratory Validation For The Detection Of Salmonella Enterica In Meat Analog Products, Amanda Tabb May 2023

Single-Laboratory Validation For The Detection Of Salmonella Enterica In Meat Analog Products, Amanda Tabb

Food Science (MS) Theses

Meat analogs are becoming increasingly popular for health, environmental, and ethical reasons. However, these products are at risk for contamination with Salmonella enterica. Therefore, it is important to ensure that detection methods for S. enterica in meat analog products are effective. Currently, the Bacteriological Analytical Manual (BAM) chapter on Salmonella detection calls for pre-enrichment of meat analogs using lactose broth (LB) with added surfactants, Tergitol Anionic 7 or Triton X-100. However, it remains to be determined whether surfactants are necessary for pre-enrichment of meat analogs. Following pre-enrichment, screening with VIDAS is typically performed. While VIDAS is an effective screening method, …

Evaluating Mislabeling Of Raw, Ready-To-Eat Seafood Products In Orange County, California, Courtney Kitch Dec 2022

Evaluating Mislabeling Of Raw, Ready-To-Eat Seafood Products In Orange County, California, Courtney Kitch

Food Science (MS) Theses

Raw, ready-to-eat (RTE) seafood products have become increasingly popular globally, but they are vulnerable to species mislabeling and substitution. DNA barcoding allows for fish species identification by extracting, amplifying, and sequencing a standardized gene target. A wide variety of fish products have been studied with DNA barcoding, but notably missing are ceviche and poke, particularly in the United States. Sushi is known to be a target of mislabeling but has not been extensively studied in Orange County, CA. The objective of this study was to investigate species labeling and use of acceptable market names for sushi, poke, and ceviche sold …

Assessing The Efficacy Of Bacteriological Analytical Manual (Bam) Enrichment Broths For Detection Of Salmonella Spp. In Meat Analogs, Georgia Sampson Aug 2021

Assessing The Efficacy Of Bacteriological Analytical Manual (Bam) Enrichment Broths For Detection Of Salmonella Spp. In Meat Analogs, Georgia Sampson

Food Science (MS) Theses

Plant-based meat alternatives are gaining popularity as the number of market options increases. However, the recent appearance of these products on the market post-dates the validation of the current approach for isolating Salmonella from meat analogs. Therefore, the objective of this study was to compare the efficacy of the currently used Bacteriological Analytical Manual (BAM) pre-enrichment broth (lactose broth) with two additional broths (universal pre-enrichment broth and buffered peptone water) for the detection of Salmonella in five plant-based burger products. Each burger product was spiked with a Salmonella serotype (S. Enteritidis or S. Agona) that has previously been linked …

Use Of Gamma Irradiation As An Intervention Treatment To Inactivate Escherichia Coli O157:H7 In Freshly Extracted Apple Juice, Dielle Aurelia Fernandes May 2019

Use Of Gamma Irradiation As An Intervention Treatment To Inactivate Escherichia Coli O157:H7 In Freshly Extracted Apple Juice, Dielle Aurelia Fernandes

Food Science (MS) Theses

Escherichia coli O157:H7 can contaminate dropped apples used for juicing via contact with manure or fecally tainted irrigation water and attach to the flesh of the apple through bruises and wounds where surface sanitizers are not effective. The goal of this project was to determine the efficacy of gamma irradiation at the maximum allowed dose of 1000 Gy to inactivate Escherichia coli O157: H7 in whole apples used for juicing. Whole apples were punctured to simulate wounds which were then inoculated with an outbreak strain of E.coli O157:H7 and subjected to gamma irradiation at doses upto 1000 Gy. The D-value …

Antimicrobial Susceptibility Of Listeria Monocytogenes To Bacteriophage Listex™ P100 In Alfalfa Sprouts (Medicago Sativa), Tushar Sawant May 2015

Antimicrobial Susceptibility Of Listeria Monocytogenes To Bacteriophage Listex™ P100 In Alfalfa Sprouts (Medicago Sativa), Tushar Sawant

Food Science (MS) Theses

The seed germination process during sprout production provides suitable environmental conditions for the growth of pathogenic bacteria, such as Listeria monocytogenes. A potential way to control this bacterial growth is through the use of bacteriophages, which are naturally occurring viruses that specifically attack bacterial targets and have been shown to be effective antimicrobials in some foods. Therefore, the objective of this study was to evaluate the antimicrobial susceptibility of L. monocytogenes to bacteriophage on alfalfa sprouts during seed germination and subsequent refrigerated storage at 4 °C. Alfalfa sprout seeds were dip-inoculated with 5.5 x 105 CFU/ml L. monocytogenes serogroups 1 …
