Background
Human milk is considered the gold standard for infant nutrition; however, it is often discontinued in chylothorax and severe very long-chain acyl-CoA dehydrogenase (VLCAD) deficiency. As an alternative to complete human milk restriction, defatted human milk may be fed, though it may not fully support optimal infant growth and development. This study aimed to assess the micronutrient profile of defatted human milk.
Methods
Human milk was obtained from UCSD Human Milk Research Biorepository (San Diego, CA). Thirty samples representing 10 independent expressions in the morning, afternoon, and evening were pooled for analysis. Fat removal was performed by centrifugation at …

Alkaline And Acid Extraction Of An L-Tryptophan–Sunflower Flour Red Colorant: Production, Characterization, And Utilization, Spencer Nguyen, Marissa Wong, Lilian Were Senger
Food Science Faculty Articles and Research
The objective was to produce and characterize a red colorant resulting from interaction of sunflower-derived chlorogenic acid with L-tryptophan/Trp and to evaluate its applications. Yield, intensity, solubility, stability, protein, and metal content of colorants from pH 9 and pH 11 lyophilized sunflower flour- Trp colorants were measured. Red color formation took < 3 h with alkalization and subsequent acidification. The sunflower flour: Trp mixture ratio that produced the greatest redness was 10:2. The highest red pigment was in supernatants and not the precipitates. The red colorant intensity and solubility were highest at neutral pH when compared to acidic pH solutions. The color stability of the new red colorant in icing and refrigerated whole milk matrices, when assessed weekly for a month, showed no color change in refrigerated milk. After 30 min of incubation at 63 °C, pinkness in milk remained unchanged or increased. The pink color, however, faded in icing over time due to exposure to light. The present study established a new natural red colorant developed from sunflower protein-Trp mixtures at alkaline pH, as well as its application in icings and whole milk for commercial strawberry milk products. This colorant could serve as a replacement for the synthetic petroleum-based Red 3.

Ginseng (Panax sp.) is a medicinal plant used for its purported health benefits, primarily in East Asian countries. The COVID-19 pandemic led to a significant increase in sales of dietary supplements, including ginseng supplements, to purportedly support immune health and provide other health benefits. However, this heightened demand has subsequently increased the risk of adulteration in these dietary supplements. This study aimed to determine the efficacy of a novel combination of DNA barcoding and multiplex PCR to identify species in ginseng supplements. A total of 50 commercial ginseng supplements containing Panax ginseng, Panax quinquefolius, or Panax notoginseng were obtained …

Sunflower meal, a byproduct of sunflower oil pressing, is not commonly used in alkaline baking applications. This is because chlorogenic acid, the main phenolic antioxidant in sunflower seeds, reacts with protein, giving the baked product a green discoloration. Our group previously demonstrated that a chlorogenic acid esterase from Lactobacillus helveticus hydrolyzes chlorogenic acid in sunflower dough cookie formulations, resulting in cookies that were brown instead of green. This study presents a sensory analysis to determine the acceptability of enzymatically upcycled sunflower meal as an alternative protein source for those allergic to meals from legumes or tree nuts. We hypothesized that …

Sunflower seeds contain a high concentration of chlorogenic acid (CGA), which reacts with amino acids to form green pigments under alkaline conditions during food processing. Here, we present two approaches to prevent green pigment formation in sunflower cookies by (A) Addition of free thiols from cysteine and glutathione to sunflower cookie dough and (B) hydrolyzing CGA into caffeic acid and quinic acid with a CGA esterase from Lactobacillus helveticus. Greening occurred more slowly with cysteine; however, neither cysteine nor glutathione prevented greening in the cookies during storage. Chlorogenic acid esterase hydrolyzed CGA in both sunflower butter and flour, resulting …

Chlorogenic acid (CGA) is an ester between caffeic and quinic acid. It is found in many foods and reacts with free amino groups in proteins at alkaline pH, leading to the formation of an undesirable green pigment in sunflower seed-derived ingredients. This paper presents the biochemical characterization and application of a highly active chlorogenic acid esterase from Lactobacillus helveticus. The enzyme is one of the most active CGA esterases known to date with a Km of 0.090 mM and a kcat of 82.1 s−1. The CGA esterase is easily expressed recombinantly in E. coli in …

Thiols (cysteine and glutathione) were explored as potential decolorization agents to mitigate green pigment formation in chlorogenic acid quinone-lysine solutions. Reparameterizations of the Weibull cumulative distribution function were applied to describe the time-dependence of greening under varying pH conditions. Repeated fitting of 3-parameter models (RMSE = 0.0111, CVRMSE = 1.55%) indicated the linear dependence of model parameters on thiol concentration. A 6-parameter Weibull model incorporating time and initial thiol concentration (RMSE = 0.0255, CVRMSE = 3.56%) accurately predicted green color development. Calculated model parameters descriptive of greening rate, terminal greening magnitude, and lag time before greening onset facilitated comparison of …

Relative to standard red/green/blue (RGB) imaging systems, hyperspectral imaging systems offer superior capabilities but tend to be expensive and complex, requiring either a mechanically complex push-broom line scanning method, a tunable filter, or a large set of light emitting diodes (LEDs) to collect images in multiple wavelengths. This paper proposes a new methodology to support the design of a hypothesized system that uses three imaging modes—fluorescence, visible/near-infrared (VNIR) reflectance, and shortwave infrared (SWIR) reflectance—to capture narrow-band spectral data at only three to seven narrow wavelengths. Simulated annealing is applied to identify the optimal wavelengths for sparse spectral measurement with a …

Superficial scald is a postharvest disorder of apples related to increased ethylene production in stored fruit which leads to α-farnesene accumulation and oxidation. Ionizing irradiation inhibits ethylene production and has been shown to reduce superficial scald, but this phenomenon has not been explored at the molecular level. The goal of this study was to determine the effect of irradiation on gene expression of enzymes related to ethylene and α-farnesene in ‘Granny Smith’ apples. Irradiation at 310 Gy controlled scald severity and incidence and inhibited the rise in AFS1 expression up to 90 d of cold storage followed by 7 d …

In fresh milk, plasminogen, the zymogen form of plasmin (PL), is the predominant form. Therefore, plasminogen activators (PA) can contribute significantly to PL activity in milk. Both tissue-type PA (tPA) and urokinase-type PA (uPA) exist in milk; however, contradictory findings have been reported for which type of PA is most closely associated with the casein micelles. Little is known about the factors that might lead to variations in the individual activities of the PA. The objective of this work was therefore to investigate possible factors that might affect the association of tPA and uPA with the casein micelle and their …
