Probiotics, live microorganisms that beneficially affect the health of their host, must undergo extensive research to ensure they are safe for consumption and possess certain functional properties. Antibiotic resistance in probiotics has raised concern due to the possibility of its transfer to pathogens. Acid and bile tolerance ensures that organisms will survive passage into the intestines. Prebiotic utilization indicates ability to ferment specific carbohydrates for enhanced growth. The objective of this study was to characterize a group of commercial probiotics for their suitability as probiotics.

Probiotics are viable nonpathogenic microbes that positively affect host health. Probiotics inhibit infection, activate immunity, and promote mucosal-barrier development. Many microbes have probiotic activity. Nonetheless, the selection of stable strains and their specific mechanism(s) of action are not fully elucidated. Bacteria from ‘Amabere amaruranu’ cultured milk from Kenya were isolated and identified by PCR sequence analysis of the 16S rRNA gene. Isolates were examined for stability to acid and bile, antimicrobial activity, mucin production, and degradation and sensitivity to antibiotics, hence their potential for probiotics. Lactobacillus isolates were acid unstable, bile-stable, nonmucinolytic, and presented antibacterial activity. L. rhamnosus cell fractions …

By-products from the dry-milling ethanol process can be used in cattle diets to replace corn. There is a significant interaction between corn processing methods as CCDS concentration increases in the diet. Improvements are observed for SFC in final BW, ADG, and G:F over DRC. An experiment was conducted to determine if greater concentrations of CCDS could be fed in SFC based diets and maintain or improve performance. Performance and carcass characteristics were evaluated with increasing concentrations of CCDS at 0, 9, 18, 27, or 36% in place of SFC in feedlot finishing diets. As CCDS concentration increased, DMI decreased quadratically. …

The purpose of this study was to examine participants in a National School Lunch Program (NSLP), and the effect of gender and/or participation in the NSLP on selection of high saturated fat content entrée items by the students. Nutrition information provided to parents often does not include a profile of the fats offered in school lunch items. This was true of this study's schools. However, new NSLP guidelines focus upon saturated and trans fat content in school lunches. Adherence is the first and one of the most important steps of bacterial pathogenesis. Natural derived components that inhibit the adherence of pathogens to the surface of epithelial cells have received considered interest. The goal of this research was to assess the anti-adherence activity of mannan oligosaccharides (MOS), pectic oligosaccharides (POS) and cranberry high molecular weight component (HMW) against Campylobacter jejuni and Campylobacter coli. First the anti-adherence activity of MOS and their purified fraction (pMOS) was tested against three strains of C. jejuni and two strains of C. coli. The anthocyanin composition and HPLC fingerprints of six small berries endemic of the VIII region of Chile were investigated using high resolution mass analysis for the first time (HR-ToF-ESI-MS). The antioxidant features of the six endemic species were compared, including a variety of blueberries which is one of the most commercially significant berry crops in Chile. The anthocyanin fingerprints obtained for the fruits were compared and correlated with the antioxidant features measured by the bleaching of the DPPH radical, the ferric reducing antioxidant power (FRAP), the superoxide anion scavenging activity assay (SA), and total content of phenolics, flavonoids and anthocyanins …

Metagenomic Identification Of A Novel Salt Tolerance Gene From The Human Gut Microbiome Which Encodes A Membrane Protein With Homology To A Brp/Blh-Family Beta-Carotene 15,15'-Monooxygenase, Eamonn P. Culligan, Roy D. Sleator, Julian R. Marchesi, Colin Hill

The human gut microbiome consists of at least 3 million non-redundant genes, 150 times that of the core human genome. Herein, we report the identification and characterisation of a novel stress tolerance gene from the human gut metagenome. The locus, assigned brpA, encodes a membrane protein with homology to a brp/blh-family β-carotene monooxygenase. Cloning and heterologous expression of brpA in Escherichia coli confers a significant salt tolerance phenotype. Furthermore, when cultured in the presence of exogenous β-carotene, cell pellets adopt a red/orange pigmentation indicating the incorporation of carotenoids in the cell membrane.

The current study is an attempt to better understand the motivations of consumers that engage in target shooting as a serious leisure activity. The instrument design was based on the Leisure Satisfaction Scale and the Leisure Motivation Scale. A total of 5502 usable surveys were collected utilising the Internet and a website related to gun ownership and target shooting. Exploratory in nature, results indicated that three variables in particular, Escapism, Social Interaction and Self-Actualisation, were primary motivators when it comes to engagement in target shooting as a serious leisure pursuit. Although regarded as a low-risk commodity, wheat flour-based mixes have been implicated in several food safety incidents. The present thesis is a compilation of five scientific manuscripts on the effect of weather variation, milling steps and implementation of pre-milling interventions on the microbiological quality and safety of wheat and milled products. The first manuscript is a review of the microbiological quality and safety of wheat-based products. There are five major mechanisms of water-solid interactions. The primary focus of this thesis was on two of these: deliquescence and hydrate formation. Many crystalline food ingredients are deliquescent compounds (e.g., NaCl, sucrose, and ascorbic acid) and some are both deliquescent and hydrate formers (e.g., glucose, thiamine HCl, citric acid). Deliquescence is the first order phase transformation of a crystalline solid to a solution above a critical relative humidity (RH) known as the deliquescence point (RH0). A crystalline hydrate is a pseudo-polymorph in which water is incorporated into the crystal structure, altering the molecular formula and the physical properties.^ To …

Pectin is an important polymer used in the food industry as a thickening and gelling agent. Though pectin is ubiquitous in plants, chemical and structural differences among pectin molecules prevent most from being viable for industrial use. Enzymes found naturally in fruit cell walls during the ripening process impair many desirable attributes of fruit pectins, rendering them unsuitable for industrial applications. Pectin methylesterase (PME) is one such enzyme whose expression can be altered during ripening through the use of recombinant genetic engineering. Shelled corn was collected from two sources - commercial grain elevators and corn harvested from Purdue University farms and then frozen until tested. Using a carbon dioxide (CO2) test kit manufactured by Woods End Laboratories, CO2 evolution measurements were conducted on shelled corn from both sources, after the samples were re-wetted to approximately 21% moisture content, placed in a 473 ml glass jar, and incubated at room temperature for 72 hours. The CO2 test kit uses a gel that changes color in response to a change in the level of CO2 in the surrounding air. …

The western corn rootworm, or WCR, (Diabrotica virgifera virgifera LeConte) is the most significant pest of field corn (Zea mays) in the United States, and has recently expanded its range into Europe. Large increases in visceral adipose tissue accumulation increase risk for metabolic disorders including insulin resistance, type 2 diabetes, and cardiovascular diseases. The global market for eggs is projected to reach 1.154 trillion by the year 2015. The number one risk to consumers from chicken eggs is contamination with Salmonella Enteritidis (SE). FDA estimates each year 142,000 illnesses in the U.S. are caused by SE contamination in chicken eggs. Chemical treatment and decreased particle size are methods used to improve digestibility and utilization of the available nutrients in low quality forages. Previous research has indicated that chemically treated corn residue can take the place of corn when included in finishing rations containing distillers grains. Also, decreasing particle size utilizing methods such as pelleting has been shown to improve DMI and ADG. This study looked at risk factors associated with vitamin D levels in the body among a representative sample of adults in the U.S., NHANES III (2001-2006) data were used to assess the relationship between several demographic and health risk factors and vitamin D levels in the body. The Baseline-Category Logit Model was used to test the association between vitamin D level and the potential risk factors age, education, ethnicity, poverty status, physical activity, smoking, alcohol, obesity, diabetes and total cholesterol with both genders.
