iLEARN Teaching Resources

In this lesson, students will build on the previous lesson (reading and analyzing the Three Sisters chapter from Braiding Sweetgrass) by experimenting on soil in their schoolyard. Students will collect soil samples from multiple locations around their school grounds and evaluate soil texture, structure, infiltration, color, organic matter, and pH. Students Through field observations and laboratory analysis, students examine how soil characteristics influence plant growth and ecosystem health. Genetic Determinants Associated With Persistence Of Listeria Species And Background Microflora From A Dairy Processing Environment, Vaishali Poswal, Sanjeev Anand, Jose L. Gonzalez-Hernandez, Brian Kraus

Listeria monocytogenes is a persistent foodborne pathogen capable of surviving in food processing environments, often in association with diverse environmental microflora. This study examines genomic determinants of persistence, specifically stress adaptation and biofilm-associated traits, in environmental Listeria species and other environmental microflora from a dairy processing facility by analyzing whole-genome sequences of 6 environmental Listeria isolates, 4 ATCC reference strains, and 22 air and floor swab cultures, annotated using the RAST platform. Subsystem analysis revealed that Listeria isolates carried a defined set of genes linked to biofilm formation, antimicrobial resistance, and stress response, though in lower abundance than environmental cultures. …

Enhancing The Biopreservative Effect Of Non-Starter Lactic Acid Bacteria Using Soluble Fiber During Cheese Ripening, Rakesh Kaushik, Sanjeev Anand

Cheddar cheese ripening creates favorable conditions for desired microbial changes but also allows survival and outgrowth of spores like Clostridium tyrobutyricum and Bacillus licheniformis, leading to late-blowing defects. In the first phase of the study, NSLAB dynamics were evaluated in the presence of spores, where pilot-scale cheeses (110 L) were produced in four treatments: control, T1 (BL), T2 (CT), and T3 (BL+CT), each inoculated at 2.0 Log10 CFU/mL with spores. Results showed that SLAB declined from 8.0 to 0.2 Log10 CFU/g, while NSLAB increased from 2.0 to 8.5 Log10 CFU/g by the third month and remained stable thereafter. Spore counts …

Evaluating The Effectiveness Of Individual Cleaning Steps Of A Cip Protocol In Membrane Biofilm Removal Under Dynamic Conditions, Diwaker Singh, Sanjeev Anand

This study evaluated the effectiveness of individual clean-in-place (CIP) steps in removing biofilms from reverse osmosis (RO) membranes under dynamic flow conditions using the Centers for Disease Control (CDC) biofilm reactor. Biofilms were developed in the laboratory under continuous flow, using mixed-species bacterial isolates obtained from 10-month-old RO membrane biofilms from a commercial facility. Individual CIP chemicals, representative of those used in commercial protocols, were tested against 24 h-old biofilms. Additionally, a complete six-step sequential CIP process was conducted under dynamic conditions, consisting of treatments with alkali, surfactant, acid, enzyme, a secondary surfactant, and sanitizer. Cheese ripening involves microbial changes, with starter lactic acid bacteria (SLAB) initiat- ing fermentation and nonstarter lactic acid bacteria (NSLAB) driving flavor and texture development. However, heat-resistant spores of Clostridium and Bacillus can survive pas- teurization and cause spoilage during ripening. This study evaluated NSLAB dynamics in the presence of spores during cheese ripening. Cheddar cheese samples at pilot-scale level (110 L) with four treatments, namely control, with spores of B. licheniformis (T1), with spores of Cl. tyrobutyricum (T2), and both spores (T3) at 2.0 Log10 CFU/mL, were ripened at 7 ◦C for six months. Listeria monocytogenes (Lm) may persist in food processing environments (FPEs) alongside diverse background microflora. While microbial communities in FPEs can in- fluence Lm survival, their role in supporting or suppressing its growth remains unclear. This study aimed to characterize the microflora in floor swabs and air samples collected from a dairy processing facility across three seasons and assess their potential impact on the growth of a Lm test strain previously isolated from a dairy processing environment. Plant-based meat products are a valued choice for vegetarians and others. Their production has been perceived as an innovation promoting animal welfare and sustainability. However, there are challenges in simulating desirable appearance, texture, and flavor using only plant-based protein sources. Polysaccharide addition during the formulation and texturization of plant proteins can improve the physi- cochemical and sensory properties of texturized protein products. Peas (Pisum sativum L.) and lentils (Lens culinaris) are nutritionally rich pulses valued for their high protein, fiber, and starch content. Although their composition and health advantages are widely known, little is known about the effects of long-term storage on their functional qualities and nutritional value. The purpose of this study was to investigate the impact of storage conditions and duration on the proximate composition, physical attributes, and techno-functional traits of lentils and peas. Globally, soybean is the most widely cultivated oilseed crops, and serve as the primary source of cooking oil, contributing to around 50% of global oilseed production. During the industrial processing of soybean oil, a significant amount of soybean hull (SH) is produced, which makes up about 8% of the grain yield, resulting in approximately 18–20 million tons of hulls, one of the largest by-products in the soybean industry. Fresh produce is traditionally labeled with plastic Price LookUp (PLU) stickers that are environmental contaminants and can easily detach during handling and disrupt traceability. The CO2 laser-labeling technology (LLT), an alternate method of produce labeling, has been gaining attention. However, the performance of this technology varies by produce, and consumer perception and acceptability remain understudied. This study evaluated the efficacy of five commercially available sanitizers to reduce Salmonella (sessile and biofilm forms) count on experimentally inoculated materials representative of harvesting bins and picking bags in the fresh produce industry. Sessile Salmonella cells were grown onto tryptic soy agar to create a bacterial lawn, while multistrain Salmonella biofilms were grown in a Centers for Disease Control and Prevention (CDC) reactor at 22 ± 2 °C for 96 h. In dairy-based imitation mozzarella cheese (IMC) formulations, intact casein is critical and imparts IMC with a firm and elastic, stringy, melted texture. Rennet casein (RCN) is the desired ingredient to provide intact casein in IMC and is preferred over milk protein concentrate (MPC) and micellar casein concentrate (MCC). Transglutaminase (TGase), a crosslinking enzyme, alters the physical properties of MPC or MCC and may change IMC functionality.
