Optimization Of Bioactive Compound Extraction From Rambutan Peels Using Pulsed Electric Field And Its Application, Robby Aji Prima

This study aimed to optimize the extraction of bioactive compounds from rambutan peel using Pulsed Electric Field (PEF) technology and evaluate its functionality as a natural antioxidant. A Central Composite Design (CCD) was employed to examine the effects of ethanol concentration, electric field intensity, and pulse number on extraction efficiency. Response Surface Methodology (RSM) analysis revealed that the optimization model achieved a high composite desirability (D = 0.982). The optimal extraction conditions were identified as 70% ethanol, an electric field intensity of 4,000 V/cm, and 120 pulses. The swine industry has witnessed a remarkable increase in average litter size in recent decades, driven by genetic advancements and breeding programs aimed at enhancing prolificacy. This rise in hyperprolificacy has presented both opportunities and challenges, particularly in tropical environments where factors such as ambient temperature and humidity can influence reproductive performance. This review synthesizes current knowledge on farrowing performance and piglet characteristics of highly prolific sows in tropical settings. This study examined the safety and productivity of a layer farm over a 3-year period following the implementation of a farm Hazard Analysis and Critical Control Points (HACCP) system in 2019. The critical control point was “automatic egg collection” and no deviations from critical limits were observed, indicating the eggs were safe. In addition, the egg production rate increased and the feed conversion rate decreased. In contrast, the viability after the introduction of a farm HACCP system, in which the replenishment of new hens was discontinued after forced molting, showed an increasing trend. The presence of extended-spectrum beta-lactamase and colistin resistance E. coli in food-producing animals is a public health concern. This study was conducted using a rigorous methodology to survey the prevalence of ESBL-producing and colistin-resistant E. coli from pig farms in the central part of Thailand. A total of 519 samples were collected from fecal, feed, and waste in 53 farms located in central Thailand. All samples were inoculated onto selective agar, and species identification was performed using biochemical assays and MAlDI-TOF MS. Antimicrobial susceptibility was determined by broth microdilution, and ESBL production was confirmed by the combination disc diffusion method. …

This study aimed to develop rice starch nanoparticles with enhanced heat resistance through a sequential combination of acid hydrolysis, ball milling, and cross-linking modification using sodium trimetaphosphate (STMP), while investigating the effects of these modifications on the physicochemical properties of rice starch. Native rice starch (25.2% amylose) was hydrolyzed using 3.16 M sulfuric acid at 40°C for up to 24 hours. A significant yield reduction occurred within the first 16 hours, followed by a slower decline; therefore, starch hydrolyzed within 16 hours was selected to minimize crystalline structure damage. This thesis contains 5 chapters: an introduction, three study sections, and a conclusion. Chapter 1 highlights the rationale and significance of studying dysphagic drinks through rheology. It introduces new high-protein, plant-based formulations and literature review of hydrocolloid studies for dysphagia management, utilizing both shear and extensional rheology to explore fluid behavior during swallowing. Chapter 2 presents the first study, which developed six high-protein soy-based supplement formulations for dysphagia management with varying caloric densities. Formulations with higher caloric densities than 1.5 kcal/mL showed increased viscosity and visual cohesiveness, making them suitable for extensional flow analysis. In this study, butter was used to adsorb the aroma from dried lavender flowers (Lavandula angustifolia). This study aimed to prepare Konjac glucomannan hydrolysate (KGMH) and investigate its glycation with animal and plant proteins through the Maillard reaction, focusing on varying degrees of polymerization (DPs). Initially, KGMH was produced from high-content Konjac glucomannan (KGM) using beta-mannanase (200 to 300 U/g) within 60 minutes, resulting in KGMH with DPs ranging from 2 to 9 and reduced viscosity. Under optimized conditions (40% KGM and 250 U/g enzyme), KGMH samples with the widest DP range (4.20 to 6.11) were used to assess antioxidant and antiglycation activities. โคพีพอด Apocyclops royi เป็นแพลงก์ตอนสัตว์ในกลุ่มครัสเตเชียนที่มีบทบาทสำคัญต่อการอนุบาลสัตว์น้ำวัยอ่อนเนื่องจากมีคุณค่าทางโภชนาการที่โดดเด่นและสามารถสังเคราะห์กรดไขมันสายยาวไม่อิ่มตัวเชิงซ้อน (LC-PUFA) ในงานวิจัยนี้ได้ศึกษาหน้าที่และการแสดงออกของยีน Fatty acid-binding protein (FABP) ซึ่งเป็นโปรตีนที่มีหน้าที่สำคัญในการขนส่งกรดไขมัน ผลจากการศึกษาลักษณะสมบัติของยีน ArFABP ของโคพีพอดสายพันธุ์ไทย A. royi-TH พบว่าประกอบด้วยบริเวณ open reading frame จำนวน 465 คู่เบส ซึ่งสามารถแปลรหัสเป็นโปรตีนขนาด 154 กรดอะมิโน โดยโปรตีน ArFABP ประกอบด้วยบริเวณ Lipocalin_7 domain ซึ่งมีความเหมือนกับโปรตีน FABP ในโคพีพอด Caligus rogercresseyi 55% จากการศึกษาหน้าที่ของยีน ArFABP ต่อความเกี่ยวข้องในการสังเคราะห์กรดไขมัน PUFA ในโคพีพอด ได้ทำการตรวจสอบการแสดงออกของยีน ArFABP ในตัวอย่างโคพีพอด A. royi-TH กลุ่มที่เลี้ยงด้วย Tetraselmis sp. (TET) เปรียบเทียบกับ Thalassiosira sp. (THA) พบว่าโคพีพอดกลุ่มที่เลี้ยงด้วย THA มีระดับการแสดงออกของยีน ArFABP สูงกว่าโคพีพอดกลุ่มที่เลี้ยงด้วย TET อย่างมีนัยสำคัญทางสถิติ ซึ่งสอดคล้องกับผลการวิเคราะห์องค์ประกอบของกรดไขมัน พบว่าโคพีพอดกลุ่มที่เลี้ยงด้วย THA มีสัดส่วนของกรดไขมัน LC-PUFA และกรดไขมัน arachidonic acid (ARA) สูงกว่ากลุ่มที่เลี้ยงด้วย TET อย่างมีนัยสำคัญทางสถิติ นอกจากนี้ผลจากงานวิจัยยังพบว่าระดับการแสดงออกของยีน ArFABP มีการแสดงออกสูงในโคพีพอดที่เลี้ยงในสภาวะความเค็ม 15 พีพีที (ความเค็มต่ำ) เปรียบเทียบกับกลุ่มโคพีพอดที่เลี้ยงในสภาวะความเค็ม 25 พีพีที (กลุ่มควบคุม) โดยพบว่าโคพีพอดกลุ่มความเค็ม 15 พีพีที มีสัดส่วนของกรดไขมัน ARA สูงกว่ากลุ่มความเค็ม 25 พีพีที อย่างมีนัยสำคัญทางสถิติ โดยจากผลการศึกษานี้แสดงให้เห็นว่ายีน ArFABP อาจมีบทบาทเกี่ยวข้องกับการขนส่งกรดไขมันในการสังเคราะห์กรดไขมัน LC-PUFA ในโคพีพอด และจากการศึกษาไมโครไบโอมพบว่าการเพาะเลี้ยงโคพีพอด A. …

อาหารที่มีคุณค่าทางโภชนาการสูงเป็นสิ่งสำคัญสำหรับการอนุบาลลูกสัตว์น้ำแรกเกิด ซึ่งจะส่งผลทำให้สิ่งมีชีวิตเติบโตได้อย่างมีประสิทธิภาพและช่วยเพิ่มอัตราการรอดของสัตว์น้ำวัยอ่อน โคพีพอด Apocyclops royi จัดอยู่ในกลุ่มของครัสเตเชียนขนาดเล็ก เป็นอาหารของสัตว์น้ำ ที่อุดมไปด้วยคุณค่าทางโภชนาการที่ดี เนื่องจากสามารถสังเคราะห์กรดไขมันไม่อิ่มตัวเชิงซ้อนสายยาว (LC-PUFA) ได้สูง โดยงานวิจัยมีวัตถุประสงค์เพื่อศึกษาลักษณะสมบัติและบทบาทของยีน Sterol regulatory element-binding protein 1 (ArSREBP-1), ยีน Carbohydrate-responsive element-binding protein (ArChREBP) และยีน Max-like factor X (ArMLX) ในโคพีพอด Apocyclops royi สายพันธุ์ไทย (A. royi-TH) และศึกษาอิทธิพลขององค์ประกอบไขมันในอาหารต่างชนิดกันที่ส่งผลต่อปริมาณกรดไขมันในโคพีพอด A.royi-TH จากการศึกษาพบว่ายีน ArSREBP-1 มีความเหมือนกับยีน SREBP-1 ของ Tigriopus kingsejongensis เท่ากับ 64%, ยีน ArChREBP มีความเหมือนกับยีน ChREBP ของParacyclopina nana เท่ากับ 58% และ ยีน ArMLX มีความเหมือนกับยีน MLX ของ Paracyclopina nana เท่ากับ 62% จากผลการศึกษาการแสดงออกของยีน Delta 6 desaturase (ArD6D), ยีน ArSREBP-1, ยีน ArChREBP และ ยีน ArMLX ต่อความสำคัญในการสังเคราะห์กรดไขมันในโคพีพอด A. royi-TH โดยเลี้ยงโคพีพอดด้วยอาหารที่แตกต่างกัน 2 ชนิด คือ สาหร่าย Tetraselmis sp. The objective of this study was to optimize the ultrasound-assisted extraction (UAE) conditions for tamarind seed extract (TSE) using response surface methodology (RSM). Subsequently, microcapsules were produced via spray drying using various wall materials and ratios. The extraction conditions were temperature levels (30, 40, and 50 °C), exposure periods (5, 10, and 15 minutes), and ethanol concentrations (30, 40, and 50 % v/v). Thua nao is a fermented soybean product that is widely consumed, especially in the northern part of Thailand. Thua nao can be used as a condiment to enhance the aroma and flavor of foods. In this study, stir bar sorptive extraction (SBSE) coupled with gas chromatography‒mass spectrometry/olfactometry (GC‒MS/O) was used to analyze volatile aroma compounds in thua nao products. Thua nao fermented with commercial Bacillus subtilis under controlled conditions exhibited the presence of compounds such as 2-methyl butanoic acid, phenylethyl alcohol, benzaldehyde, 2,5-dimethylpyrazine, 2,6-dimethylpyrazine, and 2-methoxy phenol. Se’i sapi is a traditional Indonesian smoked meat product that is normally degraded by both microorganisms and lipid oxidation. The aim of this study was to determine the effect of essential oils on the quality of sterilized se’i sapi. Firstly, antioxidant activity and total phenolic compounds of cinnamon (CEO) and nutmeg (NEO) essential oils, and liquid smoke were investigated. It was found that CEO and NEO had good potential on their antioxidant activities, while liquid smoke exhibited very low antioxidant activity. Total phenolic content of CEO and NEO showed a high value, which were 110.64 and 109.13 mg GAE/g, respectively. …

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาผลของเอนไซม์ Pectinex® Ultra SL-L ต่อการลดปริมาณน้ำตาลซูโครสในน้ำทุเรียนเข้มข้น 15% w/w โดยศึกษาที่อุณหภูมิ 55 องศาเซลเซียส และแปรระยะเวลาในการบ่ม 2, 4, 8, 24 และ 48 ชั่วโมง ศึกษาผลของการเติมนมต่อความคงตัวของฤทธิ์การต้านอนุมูลอิสระในน้ำทุเรียนที่เก็บรักษาที่อุณหภูมิ 4 องศาเซลเซียส เป็นเวลา 15 วัน และศึกษาหาภาวะที่เหมาะสมต่อการผลิตไมโครแคปซูลน้ำทุเรียนด้วยการทำแห้งแบบพ่นฝอย โดยแปรชนิดสารห่อหุ้ม (มอลโตเด็กซ์ตรินต้านทานการย่อย, มอลโตเด็กซ์ตรินต้านทานการย่อยผสมกับกัมอาระบิก และกัมอาระบิก) และอุณหภูมิขาเข้า (140 และ 160 องศาเซลเซียส) โดยผลการทดลองพบว่าการใช้เอนไซม์ Pectinex® Ultra SP-L ส่งผลทำให้ปริมาณน้ำตาลซูโครสในน้ำทุเรียนมีปริมาณลดลงและมีปริมาณ fructooligosaccharide (FOS) เพิ่มขึ้น โดยระยะเวลาการบ่มไม่ส่งผลต่อปริมาณ FOS อย่างมีนัยสำคัญ (p>0.05) และเลือกระยะการบ่มที่เวลา 2 ชั่วโมง ไปใช้ในขั้นตอนต่อไป และจากผลการศึกษาผลของการเติมนมต่อความคงตัวของฤทธิ์การต้านอนุมูลอิสระในน้ำทุเรียนพบว่าน้ำทุเรียน น้ำทุเรียนเติมเอนไซม์ น้ำทุเรียนผสมนม และน้ำทุเรียนผสมนมเติมเอนไซม์ มีปริมาณน้ำตาลซูโครสและ FOS ปริมาณสารประกอบฟีนอลิกทั้งหมด และฤทธิ์การต้านอนุมูลอิสระที่ลดลงอย่างมีนัยสำคัญทางสถิติ (p≤0.05) เมื่อมีระยะเวลาการเก็บรักษาที่เพิ่มขึ้น ในขณะที่ปริมาณจุลินทรีย์ทั้งหมดและปริมาณยีสต์และรามีปริมาณเพิ่มขึ้นน้อยกว่า 4 log CFU/mL โดยน้ำทุเรียนผสมนมเติมเอนไซม์มีค่าพรีไบโอติกแอกทิวิตีมากที่สุด และเลือกน้ำทุเรียนเติมเอนไซม์ไปใช้ในการศึกษาหาภาวะที่เหมาะสมต่อการผลิตไมโครแคปซูลน้ำทุเรียนด้วยการทำแห้งแบบพ่นฝอย จากการทดลองพบว่าทุกปัจจัยในภาวะการผลิตผงไมโครแคปซูลน้ำทุเรียนส่งผลต่อสมบัติทางกายภาพ เคมี และฤทธิ์การต้านอนุมูลอิสระของไมโครแคปซูลน้ำทุเรียนอย่างมีนัยสำคัญทางสถิติ (p≤0.05) และพบว่าผงไมโครแคปซูลมีสีเหลืองสว่างปนเขียวอ่อน มีร้อยละผลผลิตอยู่ในช่วง 48.94±0.90% ถึง 64.50±1.86% มีความชื้นและค่ากิจกรรมของน้ำต่ำกว่า 6% และ 0.3 ตามลำดับ ซึ่งเป็นไปตามมาตรฐานผลิตภัณฑ์อาหารแห้ง ความสามารถในการละลายอยู่ในช่วง 81.53±4.65% ถึง 94.21±5.58% มีประสิทธิภาพในการกักเก็บสารต้านอนุมูลอิสระอยู่ในช่วง 88.63±0.20% ถึง 97.28±0.14% และผลจากกล้องจุลทรรศอิเล็กตรอนแบบส่องกราดพบว่าผงไมโครแคปซูลที่ใช้มอลโตเด็กซ์ตรินต้านทานการย่อยเป็นสารห่อหุ้มมีลักษณะรูปร่างทรงกลม พื้นผิวเรียบมากกว่าสารห่อหุ้มชนิดอื่น ในขณะที่ผงไมโครแคปซูลที่ใช้มอลโตเด็กซ์ตรินต้านทานการย่อยผสมกัมอาระบิกและกัมอาระบิกเป็นสารห้อหุ้มมีลักษณะพื้นผิวไม่เรียบและมีรอยบุบโดยรอบ เมื่อพิจารณาปริมาณสารประกอบฟีนอลิกและฤทธิ์การต้านอนุมูลอิสระพบว่าผงไมโครแคปซูลน้ำทุเรียนที่ใช้กัมอาระบิกเป็นสารห่อหุ้มและมีอุณหภูมิขาเข้า 140 องศาเซลเซียส มีปริมาณสารประกอบฟีนอลิกและฤทธิ์การต้านอนุมูลอิสระด้วยวิธี DPPH และ FRAP …

โคพีพอด Apocyclops royi เป็นแพลงก์ตอนสัตว์ขนาดเล็กกลุ่มครัสเตเชียน ที่นิยมนำมาเป็นอาหารมีชีวิตในการอนุบาลลูกสัตว์น้ำ โคพีพอดมีความสามารถในการสังเคราะห์กรดไขมันสายยาวไม่อิ่มตัวเชิงซ้อน (Long chain polyunsaturated fatty acid; LC-PUFA) ผ่านการทำงานของเอนไซม์ดีเซสทูเรส (Desaturase) และอีลองเกส (Elongase) งานวิจัยนี้ได้ศึกษาลักษณะสมบัติและบทบาทของยีน desaturase-like (ArCb5D6D) ในโคพีพอด Apocyclops royi สายพันธุ์ไทย (A. royi-TH) จากการศึกษาพบว่ายีน ArCb5D6D ประกอบด้วยบริเวณ open reading frame 1323 คู่เบส ซึ่งสามารถแปลรหัสเป็นโปรตีนขนาด 440 กรดอะมิโน โดยโปรตีน ArCb5D6D ประกอบด้วยบริเวณ Cytochrome b5-like heme/steroid binding domain และบริเวณ Fatty acid desaturase domain ซึ่งมีความเหมือนกับโปรตีน desaturase-like ในกุ้งและปู 61-65% และจากการศึกษาการแสดงออกของยีน ArCb5D6D ในโคพีพอด A. royi-TH แต่ละระยะพัฒนาการ (NP: ระยะนอเพลียส, CD: ระยะโคพีโพดิด, AD: ระยะโตเต็มวัย) พบว่ายีน ArCb5D6D มีการแสดงออกสูงในโคพีพอดระยะ CD และโคพีพอดระยะ AD ซึ่งสอดคล้องกับปริมาณกรดไขมัน PUFA ในโคพีพอด โดยพบปริมาณกรดไขมัน PUFA สูงสุดในโคพีพอดระยะ AD รองลงมาคือ ระยะ CD และระยะ NP ตามลำดับ จากผลการศึกษานี้สามารถบ่งชี้ได้ว่ายีน ArCb5D6D เป็นยีนในกลุ่ม desaturase-like ซึ่งมีบทบาทในชีวสังเคราะห์กรดไขมันในสิ่งมีชีวิตครัสเตเชียน นอกจากนี้งานวิจัยนี้ยังได้ศึกษาอิทธิพลของความเค็มต่อคุณค่าทางโภชนาการของโคพีพอด A. royi-TH โดยเลี้ยงโคพีพอดที่สภาวะความเค็ม 15 พีพีที (ความเค็มต่ำ) และ 25 พีพีที (กลุ่มควบคุม) …

This study aimed to investigate the effects of heat curing and UV-C curing on properties of soy protein film. Both the heat and UV-C treatments were applied to either film-forming solution or pre-formed film. In the first part, film-forming solution or pre-formed film was cured at 60, 70, or 80°C for 2, 4, or 6 h. Heat curing had no effect on film thickness (p>0.05), but it did improve tensile strength of the films. งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาผลของนาโนอิมัลชันน้ำมันหอมระเหยวานิลลา (NE) และพลาสมาที่ไม่ใช้ความร้อน (PT) ต่อคุณภาพและอายุการเก็บของกะทิ เริ่มจากเตรียม NE โดยแปรอัตราส่วนของน้ำมันหอมระเหยวานิลลา (EO) และสารให้ความคงตัว (Tween 80) เป็น 3 ระดับ คือ 1:1 (NE1:1), 4:1 (NE4:1) และ 5:1(NE5:1) โดยวัดขนาดของอนุภาค (particle size), การกระจายตัวของอนุภาค (polydispersity index; PDI), สมบัติการต้านออกซิเดชัน ด้วยวิธี Ferric reducing antioxidant power (FRAP) และวิธี 2,2-diphenyl-1-picrylhydrazyl (DPPH) และสมบัติการยับยั้งจุลินทรีย์ ด้วยการหาความเข้มข้นต่ำสุดที่สามารถยับยั้งการเจริญของจุลินทรีย์ (Minimal inhibitory concentration: MIC) และความเข้มข้นต่ำสุดที่สามารถฆ่าจุลินทรีย์ (Minimal bactericidal concentration: MBC) โดยทดสอบกับจุลินทรีย์ 4 สายพันธุ์ ได้แก่ จุลินทรีย์แกรมบวก (S.aureus และ B. licheniformis) และจุลินทรีย์แกรมลบ (E.coli และ S. typhimurium) พบว่า NE1:1 มีขนาดอนุภาคเล็กกว่าและการกระจายตัวของอนุภาคดีกว่า NE4:1 และ NE5:1 ในระหว่างการเก็บรักษาที่อุณหภูมิ 30 ºC เป็นเวลา 30 วัน และพบว่า NE1:1 มีค่าการต้านออกซิเดชัน FRAP สูงกว่าสูตรอื่น แต่ค่า DPPH ต่ำที่สุด อาจเนื่องมาจากการเกิดปฏิกิริยาย้อนกลับของอนุมูล DPPH และสารฟีนอลิกบางชนิดที่อยู่ในวานิลลา และมีค่า MIC และ MBC ต่ำกว่า NE4:1 และ NE5:1 ดังนั้น NE1:1 จึงเป็นสูตรที่เหมาะสมสำหรับ NE การทดลองขั้นที่สองเป็นการหาปริมาณที่เหมาะสมของ …

งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาเครื่องดื่มเยลลี่โปรตีนสูงจากโปรตีนเวย์ โดยใช้โปรตีนเวย์ไอโซเลท (whey protein isolate; WPI) เป็นแหล่งของโปรตีน ใช้เจลาติน (gelatin; GT) และบุก (konjac glucomannan; KGM) เป็นสารที่ทำให้เกิดเจล ในขั้นต้น พัฒนาสูตรเครื่องดื่มเยลลี่ที่มีโปรตีน 2 ระดับ คือร้อยละ 6 และร้อยละ 10 (โดยน้ำหนัก) โดยมีส่วนประกอบของสารปรับกรด (กรดซิตริก:กรดมาลิก; 1:1) สารให้ความหวาน (ซูคราโลส:อะซิซัลเฟม โพแทสเซียม; 1:1) และสารให้กลิ่นรส ร้อยละ 0.025 (โดยน้ำหนัก) ในสูตรเครื่องดื่มเยลลี่ทั้งสอง 2 ความเข้มข้น จากนั้นแปรอัตราส่วนของสารที่ทำให้เกิดเจล (GT:KGM) ที่ความเข้มข้นร้อยละ 1.5 (โดยน้ำหนัก) เป็น 5 อัตรส่วน ได้แก่ 100:0 75:25 50:50 25:75 และ 0:100 เพื่อวิเคราะห์สมบัติทางเคมีกายภาพและลักษณะทางประสาทสัมผัส ในการคัดเลือกตัวอย่างเครื่องดื่มเยลลี่โปรตีนสูง 2 ระดับ พบว่าเครื่องดื่มเยลลี่ที่มีโปรตีนร้อยละ 6 และ 10 (โดยน้ำหนัก) ถูกเลือกจากคะแนนความชอบโดยรวมของผลิตภัณฑ์สูงสุดที่มีนัยสำคัญ (p £ 0.05) จากผู้ทดสอบจำนวน 30 คน ซึ่งได้อัตราส่วนที่เหมาะสมของ GT:KGM ที่ 75:25 และ 25:75 ตามลำดับ โดยมีปริมาณของแข็งทั้งหมดเท่ากับ 9.10 และ 12.50 °Brix ค่า pH เท่ากับ 3.41 และ 3.43 ค่าความแข็งแรงของเจลเท่ากับ 58.02 และ 35.00 g (force) และความสามารถในการอุ้มน้ำเท่ากับร้อยละ 93.00 และ 83.90 ตามลำดับ ทั้งนี้สมบัติทางการไหลของเครื่องดื่มเยลลี่ที่มีโปรตีนร้อยละ 6 …

งานวิจัยนี้มีวัตถุประสงค์ เพื่อพัฒนาแนวทางการใช้สับปะรดตกเกรดผลิตเซลลูโลสจากแบคทีเรียที่อุดมด้วยจุลินทรีย์โพรไบโอติก เพื่อเป็นเครื่องดื่มฟังก์ชั่น งานวิจัยประกอบด้วย (i) ประเมินความเป็นไปได้ในการใช้น้ำสับปะรดเต็มส่วน (pH 4.0) เพื่อผลิต Bacterial cellulose (BC) โดยเติมเพียงแอมโมเนียมไดไฮโดรเจนฟอสเฟต (0.6 % (w/v)) สำหรับชักนำการทำงานของเอนไซม์ cellulose synthase โดยทดลองหมักด้วยแบคทีเรียมาตรฐาน 4 สายพันธุ์ ได้แก่ Gluconacetobacter xylinus TISTR 1064 (ATCC 23767), Komagataeibacter xylinus TISTR 086, K. xylinus TISTR 428 และ K. xylinus TISTR 1061 พบว่า แต่ละสายพันธุ์ผลิต BC ในน้ำสับปะรดได้แตกต่างกัน โดย TISTR 428 ผลิต BC ได้มากที่สุด (105.10±0.08 g/L) (p<0.05) จึงเลือกมาประเมินสภาวะการหมัก โดยแปรพื้นที่ผิวสัมผัสอากาศ และปริมาตรของน้ำสับปะรด พบว่า ค่า BC yield (y) มีความสัมพันธ์เชิงเส้นตรงกับอัตราส่วนระหว่างพื้นที่ผิวสัมผัสอากาศต่อปริมาตรน้ำหมัก (x) ดังสมการ y =19.422x + 25.936 (R2 = 0.9346) และพบว่า เมื่ออัตราส่วนระหว่างพื้นที่ผิวสัมผัสอากาศต่อปริมาตรของน้ำหมักเพิ่มขึ้น Hardness และ Cohesiveness มีค่าลดลง (p<0.05) ในขณะที่ Springiness, Gumminess และ Chewiness มีค่าเพิ่มขึ้น (p<0.05) (ii) คัดแยกแบคทีเรียผลิต BC เพื่อพัฒนาเป็นสายพันธุ์จำเพาะในการหมักน้ำสับปะรด โดยคัดแยกจากน้ำหมักสมุนไพรที่มีส่วนผสมของน้ำและ/หรือเนื้อสับปะรดที่มีอายุการหมัก 8 ปี 5 ปี และ 10 เดือน พบว่า สามารถคัดแยกแบคทีเรียที่มีสมบัติสร้างสารคล้าย BC ในน้ำสับปะรดทั้งหมด 4 ไอโซเลท จากน้ำหมักอายุ 10 เดือน เมื่อระบุชนิดด้วยการวิเคราะห์ลำดับนิวคลีโอไทด์ที่ตำแหน่ง 16s rDNA/RNA พบว่า ไอโซเลท K1, K4, K8 และ K3 ที่สร้างสารคล้าย BC คือ Kodamaea ohmeri (98.33%) Acinetobacter sp. (73.97%) Enterobacter cloacae (82.31%) และ G. xylinus (80.16%) ตามลำดับ เลือก K3 ที่มีสมบัติทางจีโนไทป์ที่ตรงกับสายพันธุ์ทางการค้า และเมื่อนำมาศึกษาสมบัติการสร้าง BC ในน้ำสับปะรด พบว่า สามารถสร้าง BC ได้มากกว่าและเร็วกว่าสายพันธุ์ TISTR ถึง 2 เท่า จึงนำไอโซเลท K3 มาพัฒนาเป็นสายพันธุ์จำเพาะในการผลิต BC ในน้ำสับปะรด โดยเพาะเลี้ยงภายใต้สภาวะที่ประเมินได้จาก (i) และแปรปริมาณแหล่งคาร์บอน (ซูโครส) ปริมาณแหล่งไนโตรเจน (เพพโทน) และปริมาณสารชักนำ (โซเดียมไดไฮโดรเจนฟอสเฟต) โดยใช้แผนการทดลองแบบ 3x2x2 แฟคทอเรียล พบว่า สภาวะที่มีปริมาณซูโครส เพพโทน และโซเดียมไดไฮโดรเจนฟอสเฟตที่ความเข้มข้น 5, 0.5 และ 0 % (w/v) ตามลำดับ มีปริมาณ BC ภายหลังการหมักสูงที่สุด (188.27±0.50 g/L) BC ที่ได้มีปริมาณความชื้นและค่า Aw สูงกว่าสภาวะอื่น (p<0.05) และเมื่อวิเคราะห์เนื้อสัมผัส (Texture profile analysis) พบว่า มีค่า Hardness ต่ำที่สุด และค่า Springiness สูงที่สุด น้ำสับปะรดหลังหมักประกอบด้วย กรดอินทรีย์หลักมากที่สุด คือ กรดซิตริก กรดมาลิก และกรดแอซิติก ตามลำดับ น้ำตาลประกอบด้วยฟรักโทสมากที่สุด รองลงมา คือ กลูโคส และซูโครส ตามลำดับ อีกทั้งมีปริมาณสารประกอบฟีนอลิกสูงที่สุด (p<0.05) และมีฤทธิ์ต้านออกซิเดชั่นสูงที่สุด (p<0.05) (iii) นำสภาวะที่ประเมินได้จาก (ii) มาใช้ในการผลิต BC ที่อุดมด้วยโพรไบโอติก โดยการเพาะเลี้ยง K3 ร่วมกับจุลินทรีย์ Lactobacillus plantarum FTCU10621 พบว่า BC ที่ได้มีปริมาณน้อยลง (26.40±0.80 g/L) และเป็น young BC และตรวจวัด L. plantarum FTCU10621 ในส่วน BC พบจำนวน 8.42±0.52 log CFU/g BC และในน้ำสับปะรดจำนวน 8.33±0.03 log CFU/ml ภาพถ่ายทางสัณฐานวิทยาของผิวหน้า BC พบเซลล์โพรไบโอติกถูกตรึงอยู่ใน BC คิดเป็น 7.08 log cell/g BC เมื่อวิเคราะห์โครงสร้างทางเคมีของ BC ด้วยเทคนิค FTIR พบว่า สเปกตรัม IR ของหมู่ไฮดรอกซิลมี % transmittance ต่ำ ส่งผลให้ BC มีลักษณะอ่อนนุ่ม และน้ำสับปะรดหลังหมักประกอบด้วยกรดอินทรีย์หลักมากที่สุด คือ กรดแลคติก กรดซิตริก กรดซัคซินิค และกรดมาลิก ตามลำดับ น้ำตาลประกอบด้วยฟรักโทสมากที่สุด รองลงมา คือ กลูโคส และซูโครส ตามลำดับ
