My MS project is a practical and comprehensive training module for the advanced breeding technique of instrumental insemination of honey bee queens. Instrumental insemination allows for control over mating. Honey bee queens typically mate with 15-20 different drones in the air, making specific trait selection quite difficult. II is invaluable for concentrating specific traits in breeding stock, especially host pest resistance. The commercial bee industry has become overly reliant on chemical solutions to control varroa mites in their IPM programs. With recent evidence of mite resistance to popular miticides, breeding for host resistance is critically important to sustain the industry. …

Pyrethrum is an insecticide created from the dried flower heads of plants in the genus Chrysanthemum. The six compounds responsible for the insecticidal activity are collectively known as pyrethrins, and are known individually as Pyrethrin-I, Pyrethrin-II, Cinerin-I, Cinerin-II, Jasmolin-I, and Jasmolin-II. Of these compounds, Pyrethrin-I is best known for mortality, and Pyrethrin-II is best known for knockdown activity (Sisay et al. 2001). These compounds are voltage gated sodium-ion channel disruptors acting on nerve cells. The inside of nerve cells is negative relative to the outside at rest. Invasive species can be very disruptive to established integrated pest management programs. They can cause increases in broad-spectrum insecticide applications with variable effectiveness to mitigate damage. However, not all introduced species develop invasive characteristics; often, many introduced species go undetected. It is not until a species begins to cause economic damage to commodities essential to humans that it becomes classified as an invasive pest. Phenotypic plasticity is the ability of an organism to integrate information from environmental cues to inform the development of its phenotype and remains understudied in biology. Models of plasticity are needed because evolution in the presence of plasticity is poorly understood. Jadera haematoloma, a hemimetabolous true bug, is an excellent animal model of plasticity, exhibiting a non-linear plastic response to juvenile nutrition that biases adult development into groups with differences in flight capability, wing shape, and fecundity. Environmental factors such as temperature are substantial determinants of the spongy moth, L. dispar, distribution, reproduction, and growth. Accumulating energy reserves at the larval stage is particularly important to L. dispar, since the larvae metamorphose into a fully-grown, non-feeding adult. As non-feeding adults, the energy balance of the pupae must be adequate, to ensure enough energy reserves for adult dispersal, egg maturation and overall persistence of the species in the region. Wetlands improve the quality of our nation’s streams, rivers, and lakes, and they support a diverse assemblage of plant and animal species. The U.S. Department of Agriculture Natural Resources Conservation Service (USDA-NRCS) is responsible for administering the Wetland Reserve Program (WRP), a multi-billion-dollar effort to restore wetlands throughout the nation. Each year, WRP enrolls thousands of acres of private farmland into conservation easements with the goal of improving water quality and creating wildlife habitat. The western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte, is considered one of the most economically important pests of maize (Zea mays L.) in the United States (U.S.) Corn Belt with costs of management and yield losses exceeding USD ~1–2 billion annually. Nasonia is a parasitoid wasp genus that serves as an emerging model for studying speciation due to an incompatibility between nuclear and mitochondrial genomes between sister taxa. Short generation times, easy rearing in a lab setting, producing large amounts of progeny, and whole genome sequencing make Nasonia is an excellent candidate for studying incomplete reproductive isolation. Nasonia have five chromosomes and exhibit haplo-diploid sex determination in which fertilized eggs develop into diploid females and unfertilized eggs develop into haploid males. Massive economic damage by spotted-wing drosophila (SWD), Drosophila suzukii, results from their unique egg laying behavior. Gravid SWD flies employ a complex suite of behaviors to find fruit with the optimum firmness and chemistry. We tested for SWD attraction to various fruits and found overripe raspberries to be most attractive to gravid flies which was unexpected since they oviposit in ripe fruit. We next tested the same choice assay in a T-maze and found that the results were comparable. We also found discrete odor profiles among the ripening stages. Agricultural intensification and expansion have reduced biodiversity in agroecosystems, jeopardizing the ecosystem services that nature provides to humans in these landscapes including wildlife-mediated pest control. Among these purveyors of ecosystem services are birds, who can provide services to agroecosystems by consuming arthropod pests. Yet some bird species also act as pests by consuming crops. Herein, I use molecular diet analysis through high-throughput sequencing on DNA extracted from bird fecal samples to examine birds’ diet and classify bird species as consumers of major strawberry pests (Lygus spp.). Remodeling of larval tissues including the midgut is essential for the completion of metamorphosis in the yellow fever mosquito, Aedes aegypti (Diptera: Culicidae). The midgut structure and function have been extensively studied in adult Ae. aegypti, because adult females need to blood feed to reproduce, and the alimentary canal is the first barrier that encounters pathogens ingested through the blood meal. The midgut of Ae. aegypti undergoes remodeling during the larval-pupal metamorphosis. The emerald ash borer (EAB), Agrilus planipennis (Coleoptera: Buprestidae), is a highly invasive phloem-feeding insect native to Asia. It has killed millions of ash trees (Fraxinus spp.) since its accidental introduction into North America, causing profound economic and ecological impacts. RNA interference (RNAi) or double-stranded RNA (dsRNA)-mediated gene silencing is an emerging biotechnology that is developing as an innovative tool for pest management. Dendroctonus bark beetles are among the most economically and ecologically significant forest pests in North America and play a critical role in the overall health of conifer forest ecosystems. Dendroctonus bark beetles influence ecosystem benefits and biodiversity and drive forest succession, and adversely affect timber production, forest management, and recreation. As temperatures surge and climatic fluctuations become more extreme, catastrophic bark beetle outbreaks are increasing in frequency, escalating pressures on highly vulnerable conifer forests already compromised by heat and drought. Insects living in temperate and polar environments have developed numerous adaptations to increase survival at low temperatures. A majority of insects are freeze-intolerant and die from internal ice formation, but some are freeze-tolerant and can survive ice formation in extracellular spaces. Both categories of insects prepare for winter with a combination of seasonal and rapid acclimation responses, which differ both in time course and in underlying mechanisms. Leaf decomposition in streams is an important ecological function facilitated by bacteria, fungi, and macroinvertebrates. Metal contamination can decrease leaf decomposition rates and reduce macroinvertebrate abundance and diversity. In the current study, I focused on mining-contamination in well-buffered streams in two different Missouri mining districts with varying extents of mining contamination, Big River and Pierson Creek. I measured decomposition rates of sycamore leaves, and abundance and diversity of macroinvertebrate shredders in leaf pack experiments with a full-factorial design to determine the effects of metal contamination of leaves and stream substrates. In male blue gourami fish (Trichogaster trichopterus), signaled presentation of a male rival produces an aggressive conditioned response that results in an increased likelihood of the male winning the contest (Hollis, 1984). I replicated this classic study in a species that also shows territorial aggression: the Madagascar hissing cockroach (MHC; Gromphadorhina portentosa). Fusarium graminearum (Fg) is an important fungal pathogen of small grain cereals that can also infect Arabidopsis thaliana. In Arabidopsis, jasmonic acid (JA) signalling involving JASMONATE RESISTANT 1 (JAR1), which synthesizes JA-isoleucine, a signalling form of JA, promotes susceptibility to Fg. Here we show that Arabidopsis MYZUS PERSICAE-INDUCED LIPASE 1 (MPL1), via its influence on limiting JA accumulation, restricts Fg infection. MPL1 expression was up-regulated in response to Fg infection, and MPL1-OE plants, which overexpress MPL1, exhibited enhanced resistance against Fg. In comparison, disease severity was higher on the mpl1 mutant than the wild type. JA content was …
