Bedbug Education, Treatment, And Prevention, Elizabeth Bedford Jan 2021

Bedbug Education, Treatment, And Prevention, Elizabeth Bedford

Department of Entomology: Distance Master of Science Projects

Cimex lectularius (bedbug) has become a severe issue in more recent years due to travel, immigration, and importation of products. Unfortunately, most people do not know their biology hence do not treat these arthropods correctly. Many myths and misinformation are currently available to people on the internet and with pest professionals. Education and proper treatment must eradicate these pests in homes, commercial buildings, and other places. To prove whether or not education with treatment garners better results, ten homes were treated with the same heating and chemical process, with only five being educated before, during and after. All homes were inspected …

Macroinvertebrate Responses To Hydrological Variation In Experimental Wetlands., Sergio A. Sabat-Bonilla Jan 2021

Macroinvertebrate Responses To Hydrological Variation In Experimental Wetlands., Sergio A. Sabat-Bonilla

College of Graduate Studies: Theses & Dissertations

Predicted increases in the frequency of intense storms and periods of severe drought due to climate change represent a threat to wetland macroinvertebrate communities through alterations to the hydrological regime. I used experimental ponds to assess the effects of water permanence (i.e., duration of flooding) on the communities of aquatic macroinvertebrates. I predicted that permanent ponds would harbor higher diversity of longer-lived taxa whereas temporary ones will favor colonization by quick turnover, short-lived taxa and support lower consumer diversity. Results show differences in macroinvertebrate communities between permanent and temporary ponds can be mostly explained by hydrology and the amount of …

Evaluation Of Biotype 4 Aphis Glycines (Hemiptera: Aphididae) Induced Susceptibility On Soybean And Soja, Jakob David Hicks Jan 2021

Evaluation Of Biotype 4 Aphis Glycines (Hemiptera: Aphididae) Induced Susceptibility On Soybean And Soja, Jakob David Hicks

Electronic Theses and Dissertations

Soybean aphids have been a significant pest of soybeans in North America since 2000. Before 2000, soybeans did not face significant insect pest pressure from any arthropods with piercing-sucking mouthparts. It is estimated that economic damage from soybean aphids range from $1billion to $4.7 billion annually. Research efforts focused on the identification of host plant resistance genes in soybean and discovered many resistant to Aphis glycines genes (i.e., Rag genes) in soybean. However, the adoption of commercially released Rag soybean cultivars has been limited. The prospect of management with Rag genes was further complicated by the identification of three virulent …

The Reconstruction Of Hatchery Creek: Effects On Macroinvertebrate And Fish Community Dynamics, Patrick Vrablik Jan 2021

The Reconstruction Of Hatchery Creek: Effects On Macroinvertebrate And Fish Community Dynamics, Patrick Vrablik

Murray State Theses and Dissertations

Hatchery Creek is a restored stream in Jamestown, KY that drains Wolf Creek National Fish Hatchery. The previous degraded channel of Hatchery Creek caused large sediment plumes in the Lower Cumberland River and was restored to decrease sediment loss and provide the opportunity for a self-sustaining trout population. I predicted that the increased amount of habitat would increase taxa richness, abundance and biomass of macroinvertebrates. Macroinvertebrates were monitored in three periods; the degraded period, a one-year recovery period, and a restored period using surber and kicknet samples in order to determine biomass, abundance, diversity, taxa richness, and macroinvertebrate biotic index. …

Use Of Lymesim 2.0 To Assess The Potential For Single And Integrated Management Methods To Control Blacklegged Ticks (Ixodes Scapularis; Acari: Ixodidae) And Transmission Of Lyme Disease Spirochetes, Shravani Chitineni, Elizabeth R. Gleim, Holly D. Gaff Jan 2021

Gaff

Undergraduate Honors Theses

Annual Lyme disease cases continue to rise in the U.S. making it the most reported vector-borne illness in the country. The pathogen (Borrelia burgdorferi) and primary vector (Ixodes scapularis; blacklegged tick) dynamics of Lyme disease are complicated by the multitude of vertebrate hosts and varying environmental factors, making models an ideal tool for exploring disease dynamics in a time- and cost-effective way. In the current study, LYMESIM 2.0, a mechanistic model, was used to explore the effectiveness of three commonly used tick control methods: habitat-targeted acaricide (spraying), rodent-targeted acaricide (bait boxes), and white-tailed deer targeted acaricide (4-poster …

High Plains Wheat Mosaic Virus: An Enigmatic Disease Of Wheat And Corn Causing The High Plains Disease, Satyanarayana Tatineni, Gary Hein Jan 2021

High Plains Wheat Mosaic Virus: An Enigmatic Disease Of Wheat And Corn Causing The High Plains Disease, Satyanarayana Tatineni, Gary Hein

Department of Entomology: Faculty Publications

Brief history: In 1993, severe mosaic and necrosis symptoms were observed on corn (maize) and wheat from several Great Plains states of the USA. Based on the geographical location of infections, the disease was named High Plains disease and the causal agent was tentatively named High Plains virus. Subsequently, researchers renamed this virus as maize red stripe virus and wheat mosaic virus to represent the host and symptom phenotype of the virus. After sequencing the genome of the pathogen, the causal agent of High Plains disease was officially named as High Plains wheat mosaic virus. Hence, High Plains virus, …

Landscape Characterization Of Floral Resources For Pollinators In The Prairie Pothole Region Of The United States, Autumn H. Smart, Clint R.V. Otto, Alisa L. Gallant, Michael P. Simanonok

Department of Entomology: Faculty Publications

Across agricultural areas of the Prairie Pothole Region (PPR), floral resources are primarily found on public grasslands, roadsides, and private grasslands used as pasture or enrolled in federal conservation programs. Little research has characterized the availability of flowers across the region or identified the primary stakeholders managing lands supporting pollinators. We explored spatial and temporal variability in flower abundance and richness across multiple grassland categories (i.e. general grassland, conservation grassland, and engineered pollinator habitat) in the PPR from 2015 to 2018 and used these data to estimate the number of flowering stems present across the region on private and public …

Characterizing The Relationship Between Western Corn Rootworm (Coleoptera: Chrysomelidae) Larval Survival On Cry3bb1-Expressing Corn And Larval Development Metrics, Jordan D. Reinders, David S. Wangila, Emily Robinson, B. Wade French, Lance Meinke

Department of Entomology: Faculty Publications

The western corn rootworm, Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), is a significant pest of field corn, Zea mays L. (Poales: Poaceae), across the United States Corn Belt. Widespread adoption and continuous use of corn hybrids expressing the Cry3Bb1 protein to manage the western corn rootworm has resulted in greater than expected injury to Cry3Bb1-expressing hybrids in multiple areas of Nebraska. Single-plant bioassays were conducted on larval western corn rootworm populations to determine the level of resistance present in various Nebraska counties. The results confirmed a mosaic of susceptibility to Cry3Bb1 across Nebraska. Larval development metrics, including head capsule width …

Transcriptional Regulation Of Duox-Dependent Reactive Oxygen Species Production Against Bacterial Infection In The Gut Of Cat Fleas (Ctenocephalides Felis)., Ryne W. Maness Jan 2021

Transcriptional Regulation Of Duox-Dependent Reactive Oxygen Species Production Against Bacterial Infection In The Gut Of Cat Fleas (Ctenocephalides Felis)., Ryne W. Maness

College of Graduate Studies: Theses & Dissertations

Fleas (Order Siphonaptera) are opportunistic blood feeders that parasitize a wide variety of mammals and birds. They also transmit bacterial pathogens that cause diseases in humans (e.g., murine typhus, flea-borne spotted fever, cat scratch disease, and plague). Because they acquire infectious pathogens while blood feeding, the flea gut is considered to be the initial site of infection. While immune responses have been well documented in other disease vectors, few studies have identified the immune mechanisms involved in defense of the flea gut. In other hematophagous insects, the synthesis of reactive oxygen species (ROS) is the immediate immune defense mechanism against …

Southern Pine Beetle-Specific Rna Interference Demonstrates No Effects On Nontarget Insects, Hannah Marie Hollowell Jan 2021

Southern Pine Beetle-Specific Rna Interference Demonstrates No Effects On Nontarget Insects, Hannah Marie Hollowell

Theses and Dissertations--Entomology

The endemic southern pine beetle (SPB), Dendroctonus frontalis Zimmermann, is a highly destructive bark beetle that has devastated coniferous forests of the southern United States for decades. Traditional management strategies have been unable to respond effectively to largescale SPB outbreaks, prompting exploration of novel techniques including RNA interference (RNAi). Double-stranded RNAs (dsRNAs) are effective at triggering gene silencing and inducing mortality in SPB, but determining delivery methods and demonstrating specificity of the treatments is necessary before this technology may progress to deployment.

I evaluated three SPB-specific dsRNAs on model nontarget insects to assess potential lethal effects, sublethal effects such as …

Distribution And Diversity Of Heliothine And Other Lepidopteran Nudiviruses, Emrah Ozel Jan 2021

Distribution And Diversity Of Heliothine And Other Lepidopteran Nudiviruses, Emrah Ozel

Theses and Dissertations--Entomology

Helicoverpa zea nudivirus 2 (HzNV-2) is the only known sterilizing and sexually-transmitted insect virus and causes pathological symptoms in H. zea reproductive tissues. HzNV-2 has features that make it a candidate as a H. zea (corn earworm) control agent, such as the ability to cause asymptomatic (latent) and symptomatic (lytic) infections and the ability to influence mating behavior of its host to favor virus spread. HzNV pathology has been studied and its genome sequenced, however, its prevalence in natural populations is largely unknown. In this study, we developed and used a low-cost PCR-based molecular survey to investigate HzNV-2 prevalence and …

Spatial And Temporal Variation In Aedes Albopictus Prevalence Across Arkansas, Alyssa N. Jones, Eric C. Lovely, Douglas G. Barron Jan 2021

Spatial And Temporal Variation In Aedes Albopictus Prevalence Across Arkansas, Alyssa N. Jones, Eric C. Lovely, Douglas G. Barron

Journal of the Arkansas Academy of Science

Aedes albopictus is a well-known vector species of mosquito that is responsible for the transmission of many arboviruses such as Zika, chikungunya, and dengue. The objective of this study was to quantify spatial and temporal variation of Ae. albopictus prevalence in Arkansas. We used egg abundance as a proxy for mosquito prevalence. Across 2 years, we worked with the Arkansas Department of Health to collect mosquito eggs using oviposition traps. Eggs were desiccated, counted, and later rehydrated in rearing chambers and raised through adulthood for species determination (>99% Ae. albopictus). We determined mean egg abundance by month, year, …

Quantitation Of Fentanyl And Metabolites From Blow Fly Tissue And Development Effects Of Fentanyl On Lucilia Sericata, Joseph Allen Cox

Graduate Theses, Dissertations, and Problem Reports (ETD)

Death investigations often involve collaborative work between different forensic disciplines, especially with regards to pathology, toxicology, and entomology. This dissertation investigates the effects of designer opioids on the development of blow fly (Lucilia sericata) larvae and the ability to detect and quantify the drugs and/or metabolites present in each stage of the fly’s life cycle. Therefore, we evaluated human tissue fortified with fentanyl at four treatment concentrations, and blow fly larvae were placed to feed on the drug infused tissue. A quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction method was developed, validated, and applied to extract …

Insect Community Associated With Black Cherry (Prunus Serotina) In The Allegheny National Forest, Craig James Larcenaire Jan 2021

Insect Community Associated With Black Cherry (Prunus Serotina) In The Allegheny National Forest, Craig James Larcenaire

Graduate Theses, Dissertations, and Problem Reports (ETD)

Black cherry (Prunus serotina Ehrh.) is valuable species managed for its economic and ecologic benefits. This species grows best in the environment of the Allegheny Plateau region in northwestern Pennsylvania. Land managers on the Allegheny National Forest (ANF) have been managing black cherry for a variety of goals and purposes. Over the last few decades, the land managers have noticed a decline in the natural regeneration of black cherry in the forest. Because the black cherry flowers are self-incompatible and require animal pollinators to transfer the pollen from one tree to another one hypothesis for this decline could be …

Trends And Opportunities In Tick-Borne Disease Geography, Catherine A. Lippi, Sadie J. Ryan, Alexis L. White, Holly D. Gaff, Colin J. Carlson

Biological Sciences Faculty Publications

Tick-borne diseases are a growing problem in many parts of the world, and their surveillance and control touch on challenging issues in medical entomology, agricultural health, veterinary medicine, and biosecurity. Spatial approaches can be used to synthesize the data generated by integrative One Health surveillance systems, and help stakeholders, managers, and medical geographers understand the current and future distribution of risk. Here, we performed a systematic review of over 8,000 studies and identified a total of 303 scientific publications that map tick-borne diseases using data on vectors, pathogens, and hosts (including wildlife, livestock, and human cases). We find that the …

Lepidopteran Granivory Reduces Seed Counts In A Rare Species Of Riparian Scour Prairies, Cheyenne Moore, Angela J. Mcdonnell, Scott Schuette, Christopher T. Martine Jan 2021

Lepidopteran Granivory Reduces Seed Counts In A Rare Species Of Riparian Scour Prairies, Cheyenne Moore, Angela J. Mcdonnell, Scott Schuette, Christopher T. Martine

Faculty Journal Articles

In Pennsylvania Baptisia australis var. australis is found along only four waterways: the Allegheny River, Youghiogheny River, Clarion River, and Red Bank Creek. Because of its limited distribution and small number of extant populations, the species is considered state-threatened in Pennsylvania. In addition, the riparian prairie habitat that Pennsylvania Baptisia australis var. australis is restricted to is also in decline and considered vulnerable. Because of these conservation concerns, insights into the natural history of the taxon in the state is valuable and will inform conservation efforts. Field surveys and fruit collections along the Allegheny River and herbarium collections were used …

Scoping Review Of Distribution Models For Selected Amblyomma Ticks And Rickettsial Group Pathogens, Catherine A. Lippi, Holly D. Gaff, Alexis L. White, Sadie J. Ryan

Biological Sciences Faculty Publications

The rising prevalence of tick-borne diseases in humans in recent decades has called attention to the need for more information on geographic risk for public health planning. Species distribution models (SDMs) are an increasingly utilized method of constructing potential geographic ranges. There are many knowledge gaps in our understanding of risk of exposure to tick-borne pathogens, particularly for those in the rickettsial group. Here, we conducted a systematic scoping review of the SDM literature for rickettsial pathogens and tick vectors in the genus Amblyomma. Of the 174 reviewed articles, only 24 studies used SDMs to estimate the potential extent …

Department of Entomology: Faculty Publications

The horn fly, Haematobia irritans irritans (L.), is one of the most important external parasites of cattle in North America and elsewhere. Horn fly adults have an intimate association with cattle, their primary host. With their often-high numbers and by feeding up to 38 times per day per fly, horn flies stress cattle. The resulting productivity loss is valued at more than 2.3 billion USD in the United States. Insecticides are commonly used to mitigate direct injury from feeding and indirect injury from disease transmission. This paper discusses horn fly biology, distribution, and management. Emphasis is on promising new approaches …

Pyrethroid Exposure Reduces Growth And Development Of Monarch Butterfly (Lepidoptera: Nymphalidae) Caterpillars, Annie Krueger, Kathryn J. Hanford, Thomas Weissling, Ana M. Vélez, Troy D. Anderson

Department of Entomology: Faculty Publications

Insecticide exposure has been identified as a contributing stressor to the decline in the North American monarch butterfly Danaus plexippus L. (Lepidoptera: Nymphalidae) population. Monarch toxicity data are currently limited and available data focuses on lethal endpoints. This study examined the 72-h toxicity of two pyrethroid insecticides, bifenthrin and β-cyfluthrin, and their effects on growth and diet consumption. The toxicity of bifenthrin to caterpillars was lower than β-cyfluthrin after 72 h. Survival was the most sensitive endpoint for bifenthrin, but diet consumption and caterpillar growth were significantly reduced at sublethal levels of β-cyfluthrin. Using AgDRIFT spray drift assessment, the aerial …

The Utility Of A Bumble Bee (Bombus Spp. [Hymenoptera: Apidae]) Brood Test For Evaluating The Effects Of Pesticides, Annie Krueger, Tara M. Early, Randy J. Ripperger, Ana R. Cabrera, Daniel R. Schmehl Jan 2021

The Utility Of A Bumble Bee (Bombus Spp. [Hymenoptera: Apidae]) Brood Test For Evaluating The Effects Of Pesticides, Annie Krueger, Tara M. Early, Randy J. Ripperger, Ana R. Cabrera, Daniel R. Schmehl

Department of Entomology: Faculty Publications

Risk assessment for chemicals in the United States relies upon the honey bee (Apis meliffera L. [Hymenoptera: Apidae]) as a surrogate for other bee species. There is uncertainty in extrapolating honey bee toxicity data to bumble bees due to differences in life history strategies, food consumption, and nest structure. Here we evaluated the design of a queenless bumble bee microcolony test that could be considered for generating larval toxicity data. Three microcolony studies were conducted with Bombus impatiens to evaluate the effects of exposure to 1) diflubenzuron in pollen, 2) dimethoate in pollen, and 3) dimethoate in sucrose. Immature drone …

Toxicological Analysis Of Stilbenes Against The Fall Armyworm, Spodoptera Frugiperda, Sarah E. Mccomic, Leslie C. Rault, Troy D. Anderson, Daniel R. Swale

Department of Entomology: Faculty Publications

The fall armyworm (FAW), Spodoptera frugiperda, is a global pest of multiple economically important row crops and the development of resistance to commercially available insecticidal classes has inhibited FAW control. Thus, there is a need to identify chemical scaffolds that can provide inspiration for the development of novel insecticides for FAW management. This study aimed to assess the sensitivity of central neurons and susceptibility of FAW to chloride channel modulators to establish a platform for repurposing existing insecticides or designing new chemicals capable of controlling FAW. Potency of select chloride channel modulators were initially studied against FAW central neuron …

Transcriptomic And Volatile Signatures Associated With Maize Defense Against Corn Leaf Aphid, Lise Pingault, Suresh Varsani, Nathan Palmer, Swayamjit Ray, W. Paul Williams, Dawn S. Luthe, Jared G. Ali, Gautam Sarath, Joe Louis

Department of Entomology: Faculty Publications

Background: Maize (Zea mays L.) is a major cereal crop, with the United States accounting for over 40% of the worldwide production. Corn leaf aphid [CLA; Rhopalosiphum maidis (Fitch)] is an economically important pest of maize and several other monocot crops. In addition to feeding damage, CLA acts as a vector for viruses that cause devastating diseases in maize. We have shown previously that the maize inbred line Mp708, which was developed by classical plant breeding, provides heightened resistance to CLA. However, the transcriptomic variation conferring CLA resistance to Mp708 has not been investigated.

Results: In this study, we contrasted …

Aboveground Herbivory Influences Belowground Defense Responses In Maize, Lise Pingault, Saumik Basu, Prince Zogli, W. Paul Williams, Nathan Palmer, Gautam Sarath, Joe Louis

Department of Entomology: Faculty Publications

The European corn borer (ECB; Ostrinia nubilalis) is an economically damaging insect pest of maize (Zea mays L.), an important cereal crop widely grown globally. Among inbred lines, the maize genotype Mp708 has shown resistance to diverse herbivorous insects, although several aspects of the defense mechanisms of Mp708 plants are yet to be explored. Here, the changes in root physiology arising from short-term feeding by ECB on the shoot tissues of Mp708 plants was evaluated directly using transcriptomics, and indirectly by monitoring changes in growth of western corn rootworm (WCR; Diabrotica virgifera virgifera) larvae. Mp708 defense responses …
