This dissertation explores the role that physical connections among and within habitats play in determining the local diversity and behaviors of cursorial organisms. Understanding the processes that regulate local community assembly and that contribute to the maintenance of species diversity is a cornerstone of community ecology. Ants are a useful model for studies of local communities because they are abundant and fulfill a variety of ecological roles. Chapter one of the dissertation examines the how lianas (woody vines) function as connective structures among tree crowns and how these connections affect arboreal ant community structure within a tropical forest canopy. Aedes albopictus (Asian tiger mosquito) and Aedes aegypti (yellow fever mosquito) are two mosquito species that are non-native to North America and are of medical importance. Both species can be found in aquatic container habitats, such as discarded tires and tree holes. In container habitats, the influx of detritus from the outside environment is essential to mosquito production and can include a variety of plant and animal material. Seeds from oak trees (acorns) can seasonally contribute to the detritus of these container habitats. Parasitic wasps act as keystone species in natural ecosystems. Adept at suppressing immunity of their insect hosts, these natural enemies of insect pests are used for biocontrol in many parts of the world. Female parasitic wasps of the closely-related species Leptopilina heterotoma (Lh), a generalist of many Drosophilia flies, and Leptopilina boulardi (Lb), a specialist on flies of the melanogaster subgroup, produce venom and virus-like particles (VLPs) in their long gland-reservoir complexes, a secretory organ connected to ovipositors. When an organism dies, the process of decomposition begins and many insects are attracted to the decomposing remains, or carrion, as a prospective food source for themselves, or their offspring. Blow flies (Family: Calliphoridae), in particular, are one of the first insects to discover and colonize a dead body due to the ability to detect a decomposing resource from large distances. As a result, forensic entomologists take advantage of the behavior and developmental cycles of early colonizers to calculate the post-mortem interval (PMI) in order to determine the time since death. As earthworms feed, they can egest soil and nutrient-rich aggregates (casts) on the soil surface. In low-cut turfgrass systems, such as golf course greens, tees, and fairways, surface casting can result in a muddy playing surface, ball roll issues, weed and pest invasion, reduced aesthetics, surface softening, and reduced photosynthesis. Because the use of pesticides for earthworm control is illegal in the U.S., earthworm casting must be managed through cultural practices. Worldwide, blackberries (Rubus L. subgenus Rubus) are an economically important crop. In 2007, Polyphagotarsonemus latus (Banks) (broad mites), were first reported damaging primocane-fruiting blackberries in Fayetteville, Arkansas. Since this time, broad mite damage to blackberries and yield loss has been reported in many states and countries. Despite the increasing reports of this blackberry pest, little is known about their population dynamics, and few treatments are available. Growers with broad mite populations need a pest management program to minimize yield loss. Screening Of Multimeric Β-Xylosidases From The Gut Microbiome Of A Higher Termite, Globitermes Brachycerastes, Chunyan Liu, Gen Zou, Xing Yan, Xuguo Zhou

Termite gut microbiome is a rich reservoir for glycoside hydrolases, a suite of enzymes critical for the degradation of lignocellulosic biomass. To search for hemicellulases, we screened 12,000 clones from a fosmid gut library of a higher termite, Globitermes brachycerastes. As a common Southeastern Asian genus, Globitermes distributes predominantly in tropical rain forests and relies on the lignocellulases from themselves and bacterial symbionts to digest wood. In total, 22 positive clones with β-xylosidase activity were isolated, in which 11 representing different restriction fragment length polymorphism (RFLP) patterns were pooled and subjected to 454 pyrosequencing. BACKGROUND: Small targets comprising panels of blue and insecticide-treated black netting material each 0.25 × 0.25 m have been shown to attract and kill Glossina fuscipes fuscipes Newstead, 1910 (Diptera: Glossinidae) thereby reducing its population density by over 90% in field trials. However, their attractive ability has not been fully exploited for sampling purposes. Therefore, in this study we assessed the effectiveness of using sticky small targets as sampling tools for G. f. fuscipes in western Kenya. We also determined the influence of colour on the landing response of female and male flies on sticky small targets.

Arboreal ants in the Brazilian Cerrado rely on cavities in living trees as nest sites. These cavities are created by a community of, wood-boring beetles, which act as ecosystem engineers. Despite the importance of these cavities as a resource, little is known about their natural abundance and heterogeneity, how ants use and modify them as nest sites, and how this interaction between cavities and their ant occupants influences trophic interactions on cerrado trees. Dermacentor variabilis, more commonly known as the wood tick, or the American dog tick, can be found all over North America. D. variabilis is a common tick in Bulloch county, Georgia, USA, and little is known about the population structure of this population of the species. Microsatellite markers are able to locate sections of the genome and can aid in estimating relatedness among individuals in a population. Six different oligonucleotide primer pairs were published for D. variabilis, however, published and non-published works indicated that not all of those primer pairs were working equally well for PCR. The activity of enzymes associated with digestion can reflect food availability and feeding preferences of invertebrates in a particular habitat. Caves are mostly nutrient-poor habitats lacking primary production. In the present study the enzymatic activity of cellulases, trehalases and chitinases was measured in eight collembolan species differently associated with the cave environment: the troglobionts (obligate cave species) Pseudacherontides spelaeus and Protaphorura janosik; the eutroglophiles Ceratophysella denticulata, Folsomia candida and Heteromurus nitidus; the subtroglophiles Hypogastrura aequepilosa and Orthonychiurus rectopapillatus; and the trogloxene (not associated with caves) Megaphorura arctica. Novel devices and methods of capturing, controlling and preventing infestation of insects using microfabricated surfaces are provided. In particular, a mechanism of insect capture inspired by the microstructures of the leaf surfaces of plants and the key features of those surfaces with respect to the capture and control of pests have been determined and engineered into a variety of microfabricated surfaces capable of reproducing the effectiveness of these physical capture methods.

Argentine ants, Linepithema humile (Mayr) (Hymenoptera: Formicidae), form mutualisms with hemipteran pests in crop systems. In vineyards, they feed on honeydew produced by mealybugs and soft scales, which they tend and protect from natural enemies. Few options for controlling Argentine ants are available; one of the more effective approaches is to use liquid baits containing a low dose of an insecticide. Knowledge of ant foraging patterns is required to estimate how many bait stations to deploy per unit area. The sugarcane aphid, Melanaphis sacchari (Zehntner) (Hemiptera: Aphididae), is an invasive pest of grain sorghum, Sorghum bicolor (L.) Moench. Since its first outbreak in sorghum in 2013, severe infestations have spread throughout the southern U.S. causing high economic losses. While insecticidal control has mitigated some of the impacts of this pest, a sustainable ecology-based management program is needed to reduce reliance on chemical control. This study investigates how human disturbance of ecosystems alters insect diversity and abundance, specifically exploring how insect communities inside Mazumbai Forest Reserve in Tanzania differ from insect communities in agricultural areas near the reserve. Following methods of previous studies on the effect of disturbance on insect populations (Bellamy et al. 2018; McLaughlin & Mineau 1995; Perry et al. 2016), this research utilizes pitfall traps and yellow bowl traps in multiple locations throughout the two study areas to catch insects, which are then identified to their specific order. The functional basis of life history adaptation is a key topic of research in life history evolution. Studies of wing polymorphism in the cricket Gryllus firmus have played a prominent role in this field. However, prior in-depth investigations of morph specialization have primarily focused on a single hormone, juvenile hormone, and a single aspect of intermediary metabolism, the fatty-acid biosynthetic component of lipid metabolism. This category includes commercial and non-commercial applicators who use or supervise the use of restricted use pesticides in solid or gaseous formulations within enclosed tight spaces such as tents, vaults, tarped stacks, structures, vehicles or vessels for a wide variety of conditions or commodities, including raw agricultural products.

Species within the African stingless bee genus Hypotrigona are difficult to differentiate due to morphological similarities. Chemical profiles of whole head extracts from workers of three Hypotrigona species: H. gribodoi, H. araujoi and H. ruspolii were studied by gas chromatography–mass spectrometry. A total of 50 components belonging to six chemical classes: hydrocarbons, aldehydes, alcohols, terpenoids, steroids and fatty acids were identified. Edible insects are part of the diets of a significant proportion of rural populations in the tropics especially Africa and Asia, and their use as source of key nutrients for better nutrition is re-emerging. Indigenously, elemental methods are used to process the insects before they are consumed or sold in retail outlets. In recent years, better knowledge of processing, packaging and storage has become necessary because of commercialisation needs. It is largely accepted that larval experience influence host-plant preference of larvae. In general, larval experience with a host plant induces both an odour and feeding preference for that plant. In a previous study, exposure of three noctuid stem borer species, i.e. Sesamia nonagrioides, Busseola fusca and B. nairobica, to a vanillin-enriched medium induced an olfactory preference for the odours of this medium in the larvae of these species. The potato aphid, Macrosiphum euphorbiae, is an important agricultural pest that causes economic losses to potato and tomato production. To establish the transcriptome for this aphid, RNA-Seq libraries constructed from aphids maintained on tomato plants were used in Illumina sequencing generating 52.6 million 75±105 bp paired-end reads. The reads were assembled using Velvet/Oases software with SEED preprocessing resulting in 22,137 contigs with an N50 value of 2,003bp. After removal of contigs from tomato host origin, 20,254 contigs were annotated using BLASTx searches against the non-redundant protein database from the National Center for Biotechnology Information (NCBI) as well as IntereProScan. …
