Phma, A Ph-Sensitive Gfp Reporter For Cell Engulfment In Drosophila Embryos, Tissues And Cells, Elane Fishilevich, James A. J. Fitzpatrick, Jonathan S. Minden Jan 2010

Phma, A Ph-Sensitive Gfp Reporter For Cell Engulfment In Drosophila Embryos, Tissues And Cells, Elane Fishilevich, James A. J. Fitzpatrick, Jonathan S. Minden

Department of Entomology: Faculty Publications

Engulfment of apoptotic cells by phagocytosis ensures the removal of unwanted and defective cells. We developed a genetically encoded marker for cell engulfment, pHMA, which consists of the pH-sensitive derivative of GFP, pHluorin, fused to the actin-binding domain of Moesin. In healthy cells of Drosophila embryos and cultured cells, pHMA resides at the cell cortex. In dying cells, pHMA loses its cortical localization and reports a modest decrease in pH. In embryos, the dying cells lose their apical contacts, then move basally and are ultimately engulfed by neighboring cells or macrophages. The cell corpse material is strongly acidified soon after …

Sugar In Moderation: Sugar Diets Affect Short-Term Parasitoid Behaviour, Danielle Lightle, Mario Ambrosino, Jana C. Lee

Entomology Papers from Other Sources

The biological control potential of parasitic wasps in the field is expected to increase with the provisioning of sugar sources, which increase longevity and replenish carbohydrate reserves. Apanteles aristoteliae is an important parasitoid of Argyrotaenia franciscana, the orange tortrix, an economic pest in fruit crops. In the present study, the effect of sugar diet on the physiological status of A. aristoteliae is investigated in the laboratory, as well as the effects of nutritional status on short-term olfactory orientation and parasitism behaviour, and the association between olfactory orientation and immediate parasitism activity. Levels of glycogen, fructose, total sugars, proportional weight …

Pollen Interception By Linyphiid Spiders In A Corn Agroecosystem: Implications For Dietary Diversification And Risk-Assessment, Julie A. Peterson, Susan Romero, James D. Harwood Jan 2010

Pollen Interception By Linyphiid Spiders In A Corn Agroecosystem: Implications For Dietary Diversification And Risk-Assessment, Julie A. Peterson, Susan Romero, James D. Harwood

West Central Research and Extension Center, North Platte

Dietary diversification, including consumption of plant tissues such as pollen, can enhance the fecundity of generalist predators, resulting in improved control of pest prey. Supplemental pollen feeding has been observed in many natural enemies, including sheet-web spiders (Araneae: Linyphiidae), which represent a major component of food webs in agroecosystems. Their horizontal, ground-based webs have the potential to intercept pollen grains during anthesis of crop plants, providing the opportunity for consumption of pollen to occur. In laboratory feeding trials, Frontinella communis and Tennesseellum formicum (Araneae: Linyphiidae) readily fed on pollen grains dusted on their webs, with 82 and 92% of spiders …

Diversity Of Interactions: A Metric For Studies Of Biodiversity, Lee A. Dyer, Thomas R. Walla, Harold F. Greeney, John O. Stireman Iii, Rebecca F. Hazen

Biological Sciences Faculty Publications

Multitrophic interactions play key roles in the origin and maintenance of species diversity, and the study of these interactions has contributed to important theoretical advances in ecology and evolutionary biology. Nevertheless, most biodiversity inventories focus on static species lists, and prominent theories of diversity still ignore trophic interactions. The lack of a simple interaction metric that is analogous to species richness is one reason why diversity of interactions is not examined as a response or predictor variable in diversity studies. Using plant-herbivore-enemy trophic chains as an example, we develop a simple metric of diversity in which richness, diversity indices (e.g., …

Modeling Evolution Of Diabrotica Virgifera Virgifera (Coleoptera: Chrysomelidae) To Transgenic Corn With Two Insecticidal Traits, David W. Onstad, Lance J. Meinke Jan 2010

Modeling Evolution Of Diabrotica Virgifera Virgifera (Coleoptera: Chrysomelidae) To Transgenic Corn With Two Insecticidal Traits, David W. Onstad, Lance J. Meinke

Department of Entomology: Faculty Publications

Asimulation model of the population dynamics and genetics of western corn rootworm, Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), was created to evaluate the use of refuges in the management of resistance to transgenic insecticidal corn, Zea mays L., expressing one or two toxin traits. Hypothetical scenarios and a case study of a corn hybrid pyramided with existing toxins are simulated. In the hypothetical situations, results demonstrated that evolution is generally delayed by pyramids compared with deployment of a single-toxin corn hybrid. However, soil insecticide use in the refuge reduced this delay and quickened the evolution of resistance. Results were sensitive …

Categorizing The Resistance Of Soybean Genotypes To The Soybean Aphid (Hemiptera: Aphididae), L. M. Pierson, Tiffany M. Heng-Moss, Thomas E. Hunt, J. C. Reese

Department of Entomology: Faculty Publications

We evaluated selected soybean, Glycine max (L.) Merr., genotypes during their reproductive stages for resistance to the soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), under greenhouse conditions and documented the categories of aphid-resistant soybean. Two screening studies were performed to assess the level of resistance to the soybean aphid on six soybean genotypes during the reproductive stages of development. Significant differences in aphid damage ratings were detected among the soybean evaluated in the screening studies. Three genotypes (KS4202, K-1639-2, and K1621) were considered moderately resistant based on the assessed damage ratings. Two of these genotypes (K-1639-2 and KS4202), along with …

Degree-Day Requirements For Alfalfa Weevil (Coleoptera: Curculionidae) Development In Eastern Nebraska, A. R. Stilwell, Robert J. Wright, Thomas E. Hunt, Erin E. Blankenship

Department of Entomology: Faculty Publications

The alfalfa weevil, Hypera postica (Gyllenhal), is a serious, yet sporadic defoliator of alfalfa, Medicago sativa L., in Nebraska. A 2-yr study was conducted in 2005 and 2006 to test for variation in degree-day requirements by location in eastern Nebraska. Sampling took place along a latitudinal gradient in three regions of eastern Nebraska. Three fields were sampled in each region during the 2 yr of the study. Alfalfa weevil larval degree-day requirements were found to vary by latitude in eastern Nebraska. Alfalfa weevil larvae were discovered in southern regions after fewer developmental degree-days had accumulated than in fields in the …

Twenty-five species of African Blastobasinae (Lepidoptera: Coleophoridae) are reviewed; 12 species are redescribed, and 13 species are described as new. Rearing of Lepidoptera ancillary to sampling efforts targeted for fruit flies (Diptera: Tephritidae) and their parasitoids was conducted in and near forested areas in coastal, central highland, and western highland habitats in Kenya. Reared moths were associated with fruits of 64 plant species in 34 families. Two new species, Blastobasis millicentae and Neoblastobasis perisella, were discovered in mixed original type series of species described by Meyrick and also reared from fruit. Eight new species, Blastobasis acirfa, B. aynekiella, …

Impact Of Dietary Diversification On Invasive Slugs And Biological Control With Notes On Slug Species Of Kentucky, Anna K. Thomas Jan 2010

Impact Of Dietary Diversification On Invasive Slugs And Biological Control With Notes On Slug Species Of Kentucky, Anna K. Thomas

University of Kentucky Master's Theses

Increasing introductions of non-native terrestrial slugs (Mollusca: Gastropoda) are a concern to North American regulatory agencies as these generalists impact the yield and reduce the aesthetic value of crop plants. Understanding how the increase in diversification in North American cropping systems affects non-native gastropods and finding effective biological control options are imperative for pest management; however, little research has been done in this area. This study tested the hypothesis that dietary diversification affects the biological control capacity of a generalist predator and allows the slug pest Deroceras reticulatum (Müller) (Stylommatophora: Agriolimacidae) to more effectively fulfill its nutritional requirements. Results showed …

Environmental Correlates Of Abundances Of Mosquito Species And Stages In Discarded Vehicle Tires, Donald A. Yee, Jamie M. Kneitel, Steven A. Juliano Jan 2010

Environmental Correlates Of Abundances Of Mosquito Species And Stages In Discarded Vehicle Tires, Donald A. Yee, Jamie M. Kneitel, Steven A. Juliano

Faculty Publications – Biological Sciences

Discarded vehicle tires are a common habitat for container mosquito larvae, although the environmental factors that may control their presence or abundance within a tire are largely unknown. We sample discarded vehicle tires in six sites located within four counties of central Illinois during the spring and summer of 2006 to determine associations between a suite of environmental factors and community composition of container mosquitoes. Our goal was to find patterns of association between environmental factors and abundances of early and late instars. We hypothesized that environmental factors correlated with early instars would be indicative of oviposition cues, whereas environmental …

Quantifying Tarnished Plant Bug, Lygus Lineolaris (Palisot De Beauvois), Resistance To Acephate In Louisiana, Josh T. Copes Jan 2010

Quantifying Tarnished Plant Bug, Lygus Lineolaris (Palisot De Beauvois), Resistance To Acephate In Louisiana, Josh T. Copes

LSU Master's Theses

The tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), is one of the most yield- limiting insect pests attacking cotton in the Mid-Southern region of the U.S. This pest is almost exclusively managed with chemical control strategies. The organophosphate insecticide, acephate, has been one of the most important insecticides recommended to control tarnished plant bugs. In recent years, reports of unsatisfactory acephate performance have become common and actual field rates have been increased to improve control. The objective of this study was to survey acephate susceptibility in Louisiana populations of tarnished plant bug using laboratory bioassays and evaluate acephate efficacy …

Association Of Esterases In Resistance To Naled And Resmethrin In The Southern House Mosquito, Culex Quinquefasciatus, Jennifer R. Gordon Jan 2010

Association Of Esterases In Resistance To Naled And Resmethrin In The Southern House Mosquito, Culex Quinquefasciatus, Jennifer R. Gordon

LSU Master's Theses

The vector-competency and its affinity for humans make C. quinquefasciatus an important target of mosquito abatement programs. Whenever such control programs are implemented, protocols to monitor frequencies and mechanisms of resistance to the insecticides used are necessary to optimize the efficacy of the management strategy and to slow development of resistance to the insecticides used. In the current study, susceptibilities to the two adulticides used by EBRMARC (naled and resmethrin) were monitored using field-collected mosquitoes and a topical and contact bioassay, respectively. My hypothesis was that esterase-mediated enhanced metabolism conferred resistance to both insecticides in populations of C. quinquefasciatus from …

Toxicity Interaction Of Fipronil And Imidacloprid Against Coptotermes Formasanus, Pan Luo

LSU Master's Theses

The Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae), is considered one of the most destructive structural pests in the world, especially in warm and humid areas. Insecticide application is an effective strategy in termite control. In recent years, non-repellent insecticides have become popular for their high efficacy due to delayed toxicity and horizontal transfer. Fipronil (registered name Termidor®) and imidacloprid (registered name Premise®) have been applied to the perimeter of millions of houses in the United States. Fipronil and imidacloprid have different modes of action which may produce a synergistic effect when combined. There have been no studies on …

Symptoms, Distribution And Abundance Of The Stem-Boring Caterpillar, Blastobasis Repartella (Dietz), In Switchgrass, Jarrad R. Prasifka, Jeffrey D. Bradshaw, Arvid A. Boe, Dokyoung Lee, David Adamski, Michael E. Gray

Agronomy, Horticulture and Plant Science Faculty Publications

A potential pest of switchgrass, Panicum virgatum L., was first detected in South Dakota in 2004, where death of partially emerged leaves was noted in a small proportion of tillers. Similar “dead heart” symptoms were observed in switchgrass in Illinois during 2008 and adults of a stem-boring caterpillar were collected and identified as Blastobasis repartella (Dietz). In 2009, a survey of the central United States was used to estimate the distribution and abundance of this insect. In eight northern states, B. repartella was consistently found in both cultivated plots and natural stands of switchgrass. In four southern states, B. repartella …

Distribution Of Eastern Hemlock, Tsuga Canadensis, In Eastern Kentucky And The Susceptibility To Invasion By The Hemlock Woolly Adelgid, Adelges Tsugae, Joshua Taylor Clark

University of Kentucky Master's Theses

The hemlock woolly adelgid, an invasive non-native insect, is threatening eastern hemlock in Kentucky. This study examined three techniques to map the distribution of eastern hemlock using decision trees, remote sensing, and species distribution modeling. Accuracy assessments showed that eastern hemlock was best modeled using a decision tree without incorporating satellite radiance. Using the distribution from the optimal model, risk maps for susceptibility to hemlock woolly adelgid infestation were created using two species distribution models. Environmental variables related to dispersal were used to build the models and their contributions to the models assessed. The models showed similar spatial distributions of …

Creating An Interactive And Dichotomous Key To The World Subfamilies Of Braconidae, Kacie Jo Johansen Jan 2010

Creating An Interactive And Dichotomous Key To The World Subfamilies Of Braconidae, Kacie Jo Johansen

University of Kentucky Master's Theses

Members of Braconidae (Hymenoptera: Ichneumonoidea) are mostly parasitoids of other holometabolous insects. It is a large family with a little over 18,000 described species and many more to be described. Subfamily classification in this group has been unstable and resolution of phylogenetic history at the subfamily level has been problematic. Since 1993, no new keys to the subfamilies of Braconidae have been produced. Many taxonomic and phylogenetic changes have taken place since then, including a recent phylogeny proposing 47 subfamilies and representing the most robust and resolved analysis to date. A pressing need for new keys to the world subfamilies …

Niche Specialization And Conservation Biology Of Cicindela Nevadica Lincolniana, Tierney R. Brosius

Department of Entomology: Dissertations, Theses, and Student Research

As with many organisms across the globe, Cicindela nevadica lincolniana is threatened with extinction. Understanding ecological factors that contribute to extinction vulnerability and what methods aid in the recovery of those species is essential in developing successful conservation programs. Here we examine behavioral mechanisms for niche partitioning along with improving techniques for captive rearing protocol and increasing public awareness about the conservation of this local insect. Ovipositional selectivity was examined for Cicindela nevadica lincolniana, Cicindela circumpicta, Cicindela togata, Cicindela punctulata, and Cicindela fulgida. Models reflect that these species of co-occurring tiger beetles select different ranges of salinity in which to …

Biotic And Abiotic Influences On Within-Plant Distribution Of Soybean Aphid (Hemiptera: Aphididae: Aphis Glycines), Tierney R. Brosius, Leon G. Higley, Thomas E. Hunt

Department of Entomology: Faculty Publications

The within-plant distribution of soybean aphid, Aphis glycines Matsumura has been attributed to various causes, yet it is unclear which factor or combination of factors most influence aphid within-plant distribution. Understanding soybean aphid distribution within the soybean canopy is important both in developing sampling programs and in evaluating injury. Because natural enemies have the potential to alter aphid dispersion, we examined how within-plant distribution of soybean aphid was influenced by different natural enemies. Field cages of different mesh diameters were used to exclude different sizes of natural enemies from aphid infested plots. Plots were surveyed and both natural enemy density …

The Prospects For Sequencing The Western Corn Rootworm Genome, Nicholas Miller, S. Richards, T. W. Sappington Jan 2010

The Prospects For Sequencing The Western Corn Rootworm Genome, Nicholas Miller, S. Richards, T. W. Sappington

Department of Entomology: Faculty Publications

Historically, obtaining the complete sequence of eukaryotic genomes has been an expensive and complex task. For this reason, efforts to sequence insect genomes have largely been confined to model organisms, species that are important to human health and representative species from a few insect orders. This situation is set to change as a number of ‘next generation’ sequencing technologies are making largescale DNA sequencing both affordable and accessible. Sequencing the genome of the western corn rootworm, Diabrotica virgifera virgifera, is likely to become a realistic proposition within the next 2 years. In the meantime, there is an active community …
