The termites Reticuliterines virginicus and R. flavipes are sympatric in forests along the eastern United States from Florida to Maryland. These congeners construct subterranean nests, forage on surface and buried wood, and appear to have very similar ecological requirements. In the present study, I examined host-wood selection by these species in a coastal forest over two years. Logs inhabited by R. virginicus had significantly greater diameters than those used by R. flavipes. It is not known whether this pattern resulted from species-specific differences in preference for host size or competition for preferred logs. Host-wood temperature did not differ for …

Tick pheromones that regulate assembly, attraction/aggregation/attachment, and mating behavior have been described. Most of the compounds regulating these behaviors are purines, substituted phenols, or cholesteryl esters. Other pheromonal compounds include organic acids, hematin, or ecdysteroids. Novel devices have been developed that combine the specific compounds comprising these pheromones with an acaricide. When applied to tick-infested vegetation or directly to the body surfaces of livestock or companion animals, these devices are effective for tick control. This review summarizes the current state of knowledge of tick pheromones. In addition, this review also presents examples illustrating how devices using tick pheromones can offer …

A few years ago I published the results of a phylogenetic analysis of New World Exoristinae based on molecular data from two genes, 28S rDNA and Elongation Factor 1-alpha (Stireman 2002). In that study I employed parsimony, neighbor joining, and maximum likelihood inference methods to generate phylogenetic reconstructions, and explored a variety of weighting schemes and combinations of the sequence data (i.e. each gene separately and both together). The results of these analyses generally supported recent taxonomic hypotheses (e.g., Herting 1984; Wood 1987; O’Hara and Wood 2004). Injection of the hormone 20-hydroxyecdysone (20-E) into partially fed (virgin) female adults of the American dog tick, Dermacentor variabilis, while they are attached and feeding on the rabbit host, initiated the expression of the vitellogenin (Vg) gene, and Vg protein secretion and uptake by the ovary. The induction of egg production by 20-E in this bioassay was dose dependent in the range of 1-50 times the concentration normally found in a replete, vitellogenic female. Antimicrobial midgut proteins and peptides that result from blood digestion in feeding American dog ticks Dermacentor variabilis (Say) were identified. Midgut extracts from these ticks showed antimicrobial activity against Micrococcus luteus, regardless of whether they were challenged with peptidoglycan, blood meal components, rabbit blood, Bacillus subtilis, Escherischia coli or Borrelia burgdorferi. However, no peptide band co-migrating with defensin was found in midgut extracts from the challenged ticks. Partial purification of the midgut extracts using C18 Sep Paks and gel electrophoresis showed the presence of 4 distinct bands with rMW 4.1, 5.3, 5.7 and 8.0 kDa identified …

Chemical analysis (high-performance liquid chromatography) and bioassay demonstrated the presence of compounds that seem to be components of the Ixodes scapularis arrestment pheromone. Only two purines, guanine and xanthine, were found in acidified saline extracts made from cast skins after molting of fed nymphs, fed larvae, and fecal/excretory exudates deposited by unfed adults on substrates in their environment. The ratio of guanine to xanthine was 10.6:1 in an extract from the nymphal skins versus 0.95:1 in an extract from the larval skins. The factors that influence rates of soldier production in subterranean termites are poorly understood. In the present study, foraging groups of Coptotermes formosanus Shiraki were collected from baldcypress trees in Lake Charles, Lousiana, in November 1985, and February, April/May and August 1986. Termites were maintained in the laboratory for nine months, seven months, four/five months and zero months, respectively, and then examined for survivorship, termite biomass and soldier production after incubation for an additional five weeks at 30° C. Within collection periods, there was no association between survivorship and soldier production or between mean dry biomass and soldier production. However, …

Both soluble and cell-mediated components are involved in the innate immune response of arthropods. Injection of Borrelia burgdorferi, the Lyme disease agent, results in the secretion of defensin into the hemolymph of the ixodid tick, Dermacentor variabilis. The presence of the peptide is observed as early as 15 min post-challenge and remains present through 18 h post-challenge. As observed in insects and soft ticks, the transcript for defensin is detected as early as 1 h post-challenge in D. variabilis. This study examined the ability of ticks to maintain multiple species of spotted fever group rickettsiae via transovarial transmission. Using a capillary feeding method, previously established Rickettsia montana- and Rickettsia rhipicephali-infected cohorts of Dermacentor variabilis (Say) were exposed to R. rhipicephali and R. montana, respectively, in two reciprocal challenge experiments. Eggs collected from individual females, for two successive generations, of each cohort were assessed for rickettsial infection by polymerase chain reaction for each challenge experiment. In addition to a soluble response, many invertebrates control bacterial infections by means of phagocytosis or melanotic encapsulation. In some insects, Escherichia coli growth is reported to be inhibited by aggregation/encapsulation. Soluble and phagocytic responses to bacterial challenge have been reported in ticks, but evidence of an aggregation/encapsulation response was reported only for inanimate (araldite) implants. Ticks were challenged by direct inoculation of bacteria into the hemocoel cavity. By plating, no viable E. coli were detected 6 h postinoculation. Hemolymph from partially fed virgin Dermacentor variabilis females was collected following Borrelia burgdorferi challenge and assayed for antimicrobial activity against Bacillus subtilis and B. burgdorferi. A small inducible cationic peptide was identified by SDS-PAGE in the hemolymph of these ticks as early as 1h post challenge. Following purification by a three-step procedure involving sequential SepPak elution, reversed phase high performance liquid chromatography (RP-HPLC) and gel electrophoresis, the yield of the active peptide was approximately 0.1% of the total protein in the hemolymph plasma. In this study we examined the efficiency of an in vitro feeding technique using glass microcapillaries as a method of establishing rickettsiae-infected lines of ticks. To quantify the volume ingested by ticks during microcapillary feeding, the incorporation of radiolabeled amino acids in tick gut and hemolymph was calculated. Fifteen of 18 ticks consumed between 0.06 μl and 6.77μl. However, ingestion of fluid was not correlated to weight gain during capillary feeding. Response Of The Tick Dermacentor Variabilis (Acari: Ixodidae) To Hemocoelic Inoculation Of Borrelia Burgdorferi (Spirochetales), Robert Johns, Daniel E. Sonenshine, Wayne L. Hynes

When Borrelia burgdorferi B31 low passage strain spirochetes were directly injected into the hemocoel of Dermacentor variabilis(Say) females, the bacteria were cleared from the hemocoel within < 24 h. Viable spirochetes were not found in hemolymph, salivary gland, or ovary tissues by subculturing or by IFA. The hemocyte population increased ≈6 times within the first 6 h after inoculation compared with the uninoculated controls. In contrast, the soluble total hemolymph protein content decreased inversely with the increase in hemocytes. Borreliacidal activity was demonstrated with cell-free hemolymph from D. variabilis. In vitro antimicrobial assays using hemolymph from borrelia-challenged and nonchallenged ticks resulted in 72% spirochete reductions compared with only 11.5%, respectively, within 1 h. Additional evidence of induced antimicrobial hemolymph protein activity was demonstrated by SDS-PAGE, which revealed upregulation of a lysozyme-like peptide (≈ 15 kDa) (22% increase) and the induction of a ≈ 5.8 kDa peptide in the B. burgdorferi-challenged ticks. 1. Termite nitrogenase activity was highest in autumn and spring (≈ 3 µg N2 fixed termite fresh mass (g)–1 day–1) and lowest in winter and summer (≈ 0·8 µg N2 fixed termite fresh mass (g)–1 day–1).

2. The nitrogenase activity of worker termites was significantly higher than all other castes (1·58 ± 0·27 µg N2 fixed termite fresh mass (g)–1 day–1).

3. Worker termites constituted the largest proportion of all the castes throughout the study period (≈ 90%).

The Lincoln index (Peterson method) is frequently used to estimate animal population size in mark-release-recapture studies. We tested the accuracy of this method to estimate termite colony size using logs infested with termites that were maintained in the laboratory. Termites were fed paper towels dyed either with 0.05% or 0.1% (w/w) of the dye marker Nile blue and released into their host logs in the laboratory. Following recapture a week later, estimates of termite population size for termites dyed with 0.05% Nile blue, and were approximately 3 times greater for termites dyed with 0.1% Nile blue. Termites of the genus Reticulitermes Holmgren and the wood roaches Cryptocercus punctulatus Scutter were observed feeding on wood stained green by the ascomycete fungus, Chlorociboria aeruginascens aeruginascens (Leotiaceae), in a Virginia forest. However, in laboratory choice trials, both termites and wood roaches preferred unstained wood to stained wood. In no-choice feeding trials, Reticulitermes fed stained birch ate significantly less wood and weighed significantly less than termites fed unstained birch. These results indicate that natural host utilization patterns cannot be relied upon to indicate feeding preferences.

The effects of block size and species on preference were examined for the subterranean termite Reticulitermes sp. (Rhinotermitidae) in choice and no-choice tests. Termites significantly preferred Douglas fir over Ponderosa pine and red oak, irrespective of block size. The less-preferred woods were eaten in large amounts in no-choice tests, and all three woods appeared to be favorable for the termites and their gut protozoans. In single species tests, termites ate significantly more from larger blocks of Douglas fir, but ate similar amounts from large and small blocks of Ponderosa Pine and red oak.
