The mosquito family Culicidae is divided into 2 subfamilies named the Culicinae and Anophelinae. Nix, the dominant male-determining factor, has only been found in the culicines Aedes aegypti and Aedes albopictus, 2 important arboviral vectors that belong to the subgenus Stegomyia. Here we performed sex-specific whole-genome sequencing and RNAseq of divergent mosquito species and explored additional male-inclusive datasets to investigate the distribution of Nix. Except for the Culex genus, Nix homologs were found in all species surveyed from the Culicinae subfamily, including 12 additional species from 3 highly divergent tribes comprising 4 genera, suggesting Nix originated at least 133 to …

Histamine is a biogenic amine that regulates multiple physiological functions in diverse organisms, specifically playing a central role in the mammalian immune response. The common bed bug, Cimex lectularius L. (Hemiptera: Cimicidae), excretes histamine in large amounts in its feces as a component of its aggregation pheromone, which contaminates homes. The potential health risks associated with the presence of indoor histamine are unclear, but to predict future exposure risks, it is critical that we understand if other arthropods excrete histamine in any discernible phylogenetic pattern. Histamine is a component of the bed bug aggregation pheromone. It was recently identified as an environmental contaminant in homes with active bed bug infestations, posing a potential health risk to humans via skin contact or inhalation. It remains unclear how histamine is distributed in homes and if histamine can become airborne. In the present study, histamine levels in household dust were quantified from multiple locations within bed bug infested and uninfested apartments. Laurel wilt disease (LWD) is a lethal vascular disease impacting lauraceous hosts caused by Harringtonia lauricola, the fungal symbiont of the redbay ambrosia beetle (Xyleborus glabratus Eichoff) (RAB) (Coleoptera: Curculionidae). LWD has caused tree mortality throughout the southeastern United States and is continuing to spread into new regions. Current management methods have not been successful in preventing spread, warranting investigation into innovative techniques including RNA interference (RNAi).

Cockroach baits are one of the most effective tools used for German cockroach (Blattella germanica L.) control. This is due in part to the number of cockroach baits on the market with various matrix compositions and active ingredients, aiding in the control of resistant (physiologically and behaviorally) cockroach populations through bait rotation. Death of individuals from the same species represents potential risks from disease, predation or competition for animals. Diverse responses associated with death have evolved ranging from simply avoiding or being attracted to the corpses in solitary animals to complicated undertaking behavioral repertoire in eusocial insects. A systematic review in chapter 1 suggested cannibalism is an ancestral and widespread death-related behavior in all non-human animals. Tick-borne diseases are an emerging threat to human and animal health. In Kentucky, tick-borne disease surveillance has identified rising incidences of spotted fever rickettsiosis, ehrlichiosis, and Lyme disease. Since these diseases occur through the bites of infected ticks, effective prevention efforts are reliant upon knowing where the risk of exposure to tick bites exists. Historical data on tick distribution in Kentucky is variable, with very little reported on a statewide scale, leaving vector control workers, public health personnel, physicians, veterinarians, and others to rely on outdated, intermittent, or out-of-state information. Small fruit production has been greatly challenged by the repeated invasion of non-native arthropod pests, impacting economic feasibility, and increasing the need for insecticides. Spotted wing drosophila (Drosophila suzukii (Matsumura)) is the most recent invasive pest of small fruits. It causes damage by ovipositing beneath the skin of ripening fruits. Its wide-spread distribution and development of resistance to insecticides has resulted in the need for improved alternative management strategies for small fruit growers.

Genetic strategies of insect pest control are receiving increased interest due to recent advances in genetic engineering. By introducing alleles that make males functionally sterile, genetically modified (GM) insects can be used in area-wide pest management programs similar to the Sterile Insect Technique (SIT). Genetic control strategies carry several potential benefits, including improved efficacy and specificity, lower operation costs, and reduced dependence on chemical insecticides, but their use is still controversial despite extensive technical documentation indicating their innocuity to humans and the environment. Urban ecosystems can support diverse communities of wild native bees. Because bloom times are conserved by geographic origin, incorporating some non-invasive non-native plants in urban landscapes can extend the flowering season and help support bees and other pollinators during periods when floral resources from native plants are limiting. A caveat, though, is the possibility that non-native plants might disproportionately host non-native, potentially invasive bee species. We tested that hypothesis by identifying all non-native bees among 11,275 total bees previously collected from 45 species of flowering woody landscape plants across 213 urban sites. BACKGROUND: Biopesticides and transgenic crops based on Bacillus thuringiensis (Bt) toxins are extensively used to control insect pests, but the rapid evolution of insect resistance seriously threatens their effectiveness. Bt resistance is often polygenic and complex. Mutations that confer resistance occur in midgut proteins that act as cell surface receptors for the toxin, and it is thought they facilitate its assembly as a membrane-damaging pore. However, the mechanistic details of the action of Bt toxins remain controversial.

The benefits of biopesticides and transgenic crops based on the insecticidal Cry-toxins from Bacillus thuringiensis (Bt) are considerably threatened by insect resistance evolution, thus, deciphering the molecular mechanisms underlying insect resistance to Bt products is of great significance to their sustainable utilization. Previously, we have demonstrated that the down-regulation of PxmALP in a strain of Plutella xylostella (L.) highly resistant to the Bt Cry1Ac toxin was due to a hormone-activated MAPK signaling pathway and contributed to the resistance phenotype. Adult Cycloneda munda were collected from Fayette County, Kentucky (38.12°N, 84.54°W). Eggs were collected daily from 4 to 6 females and placed in L:D 16:8, 14:10, 12:12,10:14, at 22°C ± 1.0°C. F1 offspring were individually reared in glass vials at each photoperiod on pea aphids and Ephestia kuehniella eggs to examine the effects of photoperiod on development (Data file 1: C-munda-Dev-Times-at-LD.jmp). Massive economic damage by spotted-wing drosophila (SWD), Drosophila suzukii, results from their unique egg laying behavior. Gravid SWD flies employ a complex suite of behaviors to find fruit with the optimum firmness and chemistry. We tested for SWD attraction to various fruits and found overripe raspberries to be most attractive to gravid flies which was unexpected since they oviposit in ripe fruit. We next tested the same choice assay in a T-maze and found that the results were comparable. We also found discrete odor profiles among the ripening stages. Agricultural intensification and expansion have reduced biodiversity in agroecosystems, jeopardizing the ecosystem services that nature provides to humans in these landscapes including wildlife-mediated pest control. Among these purveyors of ecosystem services are birds, who can provide services to agroecosystems by consuming arthropod pests. Yet some bird species also act as pests by consuming crops. Herein, I use molecular diet analysis through high-throughput sequencing on DNA extracted from bird fecal samples to examine birds’ diet and classify bird species as consumers of major strawberry pests (Lygus spp.). The midgut structure and function have been extensively studied in adult Ae. aegypti, because adult females need to blood feed to reproduce, and the alimentary canal is the first barrier that encounters pathogens ingested through the blood meal. Dendroctonus bark beetles are among the most economically and ecologically significant forest pests in North America and play a critical role in the overall health of conifer forest ecosystems. Dendroctonus bark beetles influence ecosystem benefits and biodiversity and drive forest succession, and adversely affect timber production, forest management, and recreation. As temperatures surge and climatic fluctuations become more extreme, catastrophic bark beetle outbreaks are increasing in frequency, escalating pressures on highly vulnerable conifer forests already compromised by heat and drought. Insects living in temperate and polar environments have developed numerous adaptations to increase survival at low temperatures. A majority of insects are freeze-intolerant and die from internal ice formation, but some are freeze-tolerant and can survive ice formation in extracellular spaces. Both categories of insects prepare for winter with a combination of seasonal and rapid acclimation responses, which differ both in time course and in underlying mechanisms. Detailed social and phenological data collected from nesting aggregations exist for relatively few sweat bee species because nesting aggregations are rarely found in large numbers, even when local populations are highly abundant. This limits researchers’ abilities to assess the social status of many species, which in turn, limits our ability to trace the sequence of evolutionary steps between alternative social states. To address this problem, we demonstrate the utility of rehydrated, pinned specimens from pan trap and netting collections for generating inferences about the phenology and social status of a well-studied sweat bee species, Lasioglossum (Dialictus) laevissimum. …

Codling moth (CM) (Cydia pomonella L.), a devastating pest, creates a serious issue for apple production and marketing in apple-producing countries. Therefore, effective nondestructive early detection of external and internal defects in CM-infested apples could remarkably prevent postharvest losses and improve the quality of the final product. In this study, near-infrared (NIR) hyperspectral reflectance imaging in the wavelength range of 900–1700 nm was applied to detect CM infestation at the pixel level for three organic apple cultivars, namely Gala, Fuji and Granny Smith. The field release of Wolbachia trans-infected male mosquitoes, as well as the use of toxic sugar baits, is a novel and promising candidate technique for integrated mosquito management programs. However, the methods of action of the two techniques may not be complementary, because the Wolbachia method releases mosquitoes into the environment expecting a wild population reduction in subsequent generations while the toxic baits are intended to reduce the wild population by killing mosquitoes. Bed bugs (Cimex lectularius) have proliferated globally and have become one of the most challenging pests to control indoors. They are nocturnal and use multiple sensory cues to detect and orient towards their human hosts. After feeding, usually on a sleeping human, they return to a shelter on or around the sleeping surface, but not directly on the host. We hypothesized that although human skin odors attract hungry bed bugs, human skin compounds may also prevent arrestment on hosts.
