Carrion Beetles Of The Blackwater Ecologic Preserve: Community Structure Seasonal Patterns And Habitat Use, Amy L. Simons

Carrion beetles (Silphidae) are important in the decomposition of carcasses in ecosystems. Two subfamilies, Nicrophorinae and Silphinae, differ in reproductive behaviors. The Nicrophinae, burying beetles, bury small carcasses to serve as food for the adults and their offspring. The Silphinae oviposit near larger carcasses. There is intense competition among all carrion beetles and other carrion feeders for carcasses, and beetle species have evolved seasonal activity patterns that minimize competition, such as when they are active and when they reproduce. Northern Silphidae communities are more diverse than southern communities, probably due to increased competition for carrion in the south.

Spiders and harvestmen are commonly captured by or reside upon the carnivorous purple pitcher plant, Sarracenia purpurea. Although spiders and harvestmen are often known to be prey of S. purpurea, other ecological interactions between these arthropods and the plant are poorly understood. Studies were undertaken at three pitcher plant populations, two in Virginia and one in North Carolina, to assess the ecological relationships between spiders and harvestmen and S. purpurea. Multiple plots containing pitcher plants (treatment) and plots lacking pitcher plants (control) were created at these locations. Trachymyrmex septentrionalis represents the only species of fungus-growing ant in the southeastern United States. T. septentrionalis cultivates a symbiotic fungus on which the ants feed. Worker ants collect plant and animal debris to feed the fungus. These ants are common in Florida, but increasingly scarce as they near the northern extent of their range in New Jersey. Colonies of T. septentrionalis occur in patches throughout Blackwater Ecological Preserve, a longleaf pine forest in southeastern Virginia. BioUD is an arthropod repellent that contains the active ingredient 2-undecanone originally derived from wild tomato plants. Repellency of BioUD was compared with five commercially available arthropod repellents against the ticks Amblyomma americanum (L.) and Dermacentor variabilis Say in two-choice bioassays on treated versus untreated cotton cheesecloth. Overall mean percentage repellency against both species was greatest for and did not differ significantly between BioUD (7.75% 2-undecanone) and products containing 98.1% DEET, 19.6% IR3535, and 30% oil of lemon eucalyptus. Products containing 5% and 15% Picaridin and 0.5% permethrin were also repellent compared with untreated controls but to a lesser degree …

1. Colonisation is a critical ecological process influencing both population and community level dynamics by connecting spatially discrete habitat patches. How communities respond to both natural and anthropogenic disturbances, furthermore, requires a basic understanding of how any environmental change modifies colonisation rates. For example, disturbance-induced shifts in the quantity of forest cover surrounding aquatic habitats have been associated with the distribution and abundance of numerous aquatic taxa. However, the mechanisms generating these broad and repeatable field patterns are unclear.

Background: Ticks are obligate blood feeders. The midgut is the first major region of the body where blood and microbes ingested with the blood meal come in contact with the tick's internal tissues. Little is known about protein expression in the digestive tract of ticks. In this study, for analysis of global gene expression during tick attachment and feeding, we generated and sequenced 1,679 random transcripts (ESTs) from cDNA libraries from the midguts of female ticks at varying stages of feeding.

Defensins are an important component of the innate immune system of ticks. These small peptides are produced by various genera of ticks, and expressed in various tissues. In this study we used RNA interference to silence the expression of the defensin varisin produced by the hemocytes of the American dog tick, Dermacentor variabilis. Ticks were injected with double stranded varisin RNA prior to being placed on a rabbit. After feeding, the ticks were removed, bled, and the hemolymph plasma and hemocytes separated. The aromatic polymer lignin protects plants from most forms of microbial attack. Despite the fact that a significant fraction of all lignocellulose degraded passes through arthropod guts, the fate of lignin in these systems is not known. Using tetramethylammonium hydroxide thermochemolysis, we show lignin degradation by two insect species, the Asian longhorned beetle (Anoplophora glabripennis) and the Pacific dampwood termite (Zootermopsis angusticollis). In both the beetle and termite, significant levels of propyl side-chain oxidation (depolymerization) and demethylation of ring methoxyl groups is detected; for the termite, ring hydroxylation is also observed. Antimicrobial peptides, including defensins, are components of the innate immune system in ticks that have been shown to provide protection against both gram-negative and gram-positive bacteria. Varisin, one of the defensins identified in Dermacentor variabilis, was shown to be produced primarily in hemocytes but transcript levels were also expressed in midguts and other tick cells. In this research, we studied the role of varisin in the immunity of ticks to the gram-negative cattle pathogen, Anaplasma marginale. That individuals attempt to minimize the ratio of mortality risk/growth rate (μ/g) when foraging within individual habitat patches is well established. Do species partition among spatially discrete communities embedded in complex landscapes in a similar manner? We investigated how 3 ovipositing species (2 Hyla treefrogs and a hydrophilid beetle, Tropisternus lateralis) responded to simultaneous gradients of nutrients and predation risk. Species partitioned our experimental metacommunity primarily by reducing oviposition with fish. Tropisternus positively responded to increased nutrients, but the effect decreased with increasing risk, as predicted by μ/g theory. Use of fish habitats by Tropisternus was unrelated to breeding …

The first full-length mRNA for vitellogenin (Vg) from ticks was sequenced. This also represents the first complete sequence of Vg from the Chelicerata and of a heme binding Vg. The Vg cDNA from the American dog tick, Dermacentor variabilis was 5744nt in length (GenBank Accession number AY885250), which coded for a protein of 1843aa with a calculated molecular weight of 208kD. This protein had an 18 aa signal sequence, a single RXXR cleavage signal that would generate two subunits (49.5 and 157K in molecular weight) and lipoprotein N-terminal and carboxy von Willebrand factor type D domains. The termites Reticuliterines virginicus and R. flavipes are sympatric in forests along the eastern United States from Florida to Maryland. These congeners construct subterranean nests, forage on surface and buried wood, and appear to have very similar ecological requirements. In the present study, I examined host-wood selection by these species in a coastal forest over two years. Logs inhabited by R. virginicus had significantly greater diameters than those used by R. flavipes. It is not known whether this pattern resulted from species-specific differences in preference for host size or competition for preferred logs. Host-wood temperature did not differ for …

Tick pheromones that regulate assembly, attraction/aggregation/attachment, and mating behavior have been described. Most of the compounds regulating these behaviors are purines, substituted phenols, or cholesteryl esters. Other pheromonal compounds include organic acids, hematin, or ecdysteroids. Novel devices have been developed that combine the specific compounds comprising these pheromones with an acaricide. When applied to tick-infested vegetation or directly to the body surfaces of livestock or companion animals, these devices are effective for tick control. This review summarizes the current state of knowledge of tick pheromones. In addition, this review also presents examples illustrating how devices using tick pheromones can offer …

Injection of the hormone 20-hydroxyecdysone (20-E) into partially fed (virgin) female adults of the American dog tick, Dermacentor variabilis, while they are attached and feeding on the rabbit host, initiated the expression of the vitellogenin (Vg) gene, and Vg protein secretion and uptake by the ovary. The induction of egg production by 20-E in this bioassay was dose dependent in the range of 1-50 times the concentration normally found in a replete, vitellogenic female. Antimicrobial midgut proteins and peptides that result from blood digestion in feeding American dog ticks Dermacentor variabilis (Say) were identified. Midgut extracts from these ticks showed antimicrobial activity against Micrococcus luteus, regardless of whether they were challenged with peptidoglycan, blood meal components, rabbit blood, Bacillus subtilis, Escherischia coli or Borrelia burgdorferi. However, no peptide band co-migrating with defensin was found in midgut extracts from the challenged ticks. Partial purification of the midgut extracts using C18 Sep Paks and gel electrophoresis showed the presence of 4 distinct bands with rMW 4.1, 5.3, 5.7 and 8.0 kDa identified …

Less than 1% of all Lepidoptera are aphytophagous; of these, a considerable proportion is found in the family Lycaenidae. The aphytophagous Lycaenidae are believed to have arisen from a mutualistic template involving ant attendance. With this association firmly in place, it is a relatively simple shift to exploitation, either of the ants themselves, through active carnivory on the brood/trophallactic feeding from adults, or by carnivory on ant-tended homopterans, with little to no interference by the ants. Chemical analysis (high-performance liquid chromatography) and bioassay demonstrated the presence of compounds that seem to be components of the Ixodes scapularis arrestment pheromone. Only two purines, guanine and xanthine, were found in acidified saline extracts made from cast skins after molting of fed nymphs, fed larvae, and fecal/excretory exudates deposited by unfed adults on substrates in their environment. The ratio of guanine to xanthine was 10.6:1 in an extract from the nymphal skins versus 0.95:1 in an extract from the larval skins. The factors that influence rates of soldier production in subterranean termites are poorly understood. In the present study, foraging groups of Coptotermes formosanus Shiraki were collected from baldcypress trees in Lake Charles, Lousiana, in November 1985, and February, April/May and August 1986. Termites were maintained in the laboratory for nine months, seven months, four/five months and zero months, respectively, and then examined for survivorship, termite biomass and soldier production after incubation for an additional five weeks at 30° C. Within collection periods, there was no association between survivorship and soldier production or between mean dry biomass and soldier production. However, …

Both soluble and cell-mediated components are involved in the innate immune response of arthropods. Injection of Borrelia burgdorferi, the Lyme disease agent, results in the secretion of defensin into the hemolymph of the ixodid tick, Dermacentor variabilis. The presence of the peptide is observed as early as 15 min post-challenge and remains present through 18 h post-challenge. As observed in insects and soft ticks, the transcript for defensin is detected as early as 1 h post-challenge in D. variabilis. RT-PCR resulted in an amplicon of 624 bp with a 225 bp region that translates to a 74 …

This study examined the ability of ticks to maintain multiple species of spotted fever group rickettsiae via transovarial transmission. Using a capillary feeding method, previously established Rickettsia montana- and Rickettsia rhipicephali-infected cohorts of Dermacentor variabilis (Say) were exposed to R. rhipicephali and R. montana, respectively, in two reciprocal challenge experiments. Eggs collected from individual females, for two successive generations, of each cohort were assessed for rickettsial infection by polymerase chain reaction for each challenge experiment. In addition to a soluble response, many invertebrates control bacterial infections by means of phagocytosis or melanotic encapsulation. In some insects, Escherichia coli growth is reported to be inhibited by aggregation/encapsulation. Soluble and phagocytic responses to bacterial challenge have been reported in ticks, but evidence of an aggregation/encapsulation response was reported only for inanimate (araldite) implants. Hemolymph from partially fed virgin Dermacentor variabilis females was collected following Borrelia burgdorferi challenge and assayed for antimicrobial activity against Bacillus subtilis and B. burgdorferi. A small inducible cationic peptide was identified by SDS-PAGE in the hemolymph of these ticks as early as 1h post challenge. Following purification by a three-step procedure involving sequential SepPak elution, reversed phase high performance liquid chromatography (RP-HPLC) and gel electrophoresis, the yield of the active peptide was approximately 0.1% of the total protein in the hemolymph plasma. In this study we examined the efficiency of an in vitro feeding technique using glass microcapillaries as a method of establishing rickettsiae-infected lines of ticks. To quantify the volume ingested by ticks during microcapillary feeding, the incorporation of radiolabeled amino acids in tick gut and hemolymph was calculated. Fifteen of 18 ticks consumed between 0.06 μl and 6.77μl. However, ingestion of fluid was not correlated to weight gain during capillary feeding. While much research has been done on periodical cicadas, apparently no quantitative studies have been carried out to determine the vegetational structure of periodical cicada brood emergence sites or oviposition sites. We determined large tree and small tree densities and dominance in five forest stands which experienced high densities of Brood XIX cicada emergence in 1998. Paired observations were made at one of these sites to determine whether there was a preference for egg deposition in woodland edges versus the forest interior. When Borrelia burgdorferi B31 low passage strain spirochetes were directly injected into the hemocoel of Dermacentor variabilis(Say) females, the bacteria were cleared from the hemocoel within < 24 h. Viable spirochetes were not found in hemolymph, salivary gland, or ovary tissues by subculturing or by IFA. The hemocyte population increased ≈6 times within the first 6 h after inoculation compared with the uninoculated controls. In contrast, the soluble total hemolymph protein content decreased inversely with the increase in hemocytes. Borreliacidal activity was demonstrated with cell-free hemolymph from D. variabilis. In vitro antimicrobial assays using hemolymph from borrelia-challenged and nonchallenged ticks resulted in 72% spirochete reductions compared with only 11.5%, respectively, within 1 h. Additional evidence of induced antimicrobial hemolymph protein activity was demonstrated by SDS-PAGE, which revealed upregulation of a lysozyme-like peptide (≈ 15 kDa) (22% increase) and the induction of a ≈ 5.8 kDa peptide in the B. burgdorferi-challenged ticks. 1. Termite nitrogenase activity was highest in autumn and spring (≈ 3 µg N2 fixed termite fresh mass (g)–1 day–1) and lowest in winter and summer (≈ 0·8 µg N2 fixed termite fresh mass (g)–1 day–1).
2. The nitrogenase activity of worker termites was significantly higher than all other castes (1·58 ± 0·27 µg N2 fixed termite fresh mass (g)–1 day–1).

The Lincoln index (Peterson method) is frequently used to estimate animal population size in mark-release-recapture studies. We tested the accuracy of this method to estimate termite colony size using logs infested with termites that were maintained in the laboratory. Termites were fed paper towels dyed either with 0.05% or 0.1% (w/w) of the dye marker Nile blue and released into their host logs in the laboratory. Following recapture a week later, estimates of termite population size for termites dyed with 0.05% Nile blue, and were approximately 3 times greater for termites dyed with 0.1% Nile blue. Termites host symbiotic bacteria that fix atmospheric nitrogen, which makes termites important in the biogeochemical cycling of nitrogen in ecosystems. The objective of this study was to estimate annual nitrogen contributions by termites by examining seasonal rates of nitrogen fixation, seasonal abundance of different termite castes and their nitrogenase activity, the effects of dietary nitrogen on nitrogenase activity, and low oxygen and high carbon dioxide atmospheres on termite nitrogenase activity, and the accuracy of mark-recapture studies to estimate termite population size.
