Characterizing The Maternally Inherited Endosymbionts Of Solitary Bees, Abiya Saeed
Theses and Dissertations--Entomology
Solitary bees are important pollinators of crops, with species in the family Megachilidae (mason bees) being used for orchard pollination. Commercial movement of these bees also moves their microbiota, including bacterial endosymbionts capable of reproductive manipulation. To test for presence of these bacteria, I screened commercially available species of US orchard pollinators and locally captured solitary bees from Kentucky. I also set up mason bee boxes in five apple orchards to examine recruitment of local pollinators. I conducted 454-pyrosequencing to determine bacterial diversity within four species followed by diagnostic PCR of 30 collected species (184 individuals) to determine infection frequency …

Reticulitermes flavipes is one of the most common termite species in the world, and has been an intriguing research model due to its ecological and biological and economic significance. The fundamental biological question addressed by this study is to elucidate the role of miRNAs in termite development and how miRNA can influence labor division. miRNAs are short non-coding RNAs that have an important role in gene regulation at post-transcriptional level, and can potentially be involved in the regulation of caste polyphenism. The Role Of The Bacterial Endosymbiont, Arsenophonus, In The Soybean Aphid, Aphis Glycines, Jason A. Wulff

Bacterial endosymbionts can have profound impacts on their host’s ecology. Notably, endosymbionts can protect their hosts against natural enemies and influence host plant interactions. The endosymbiont Candidatus Arsenophonus infects a wide taxonomic range of arthropod hosts, and is suspected of an uncharacterized mutualistic role in hemipterous insects. In the soybean aphid, Aphis glycines, an introduced pest of soybeans in the United States, Arsenophonus is the sole facultative endosymbiont. The focus of this dissertation is to characterize the role of Arsenophonus in the aphid, with an overall emphasis on its impact on aphid management strategies.

Changing Litter Resources Associated With Hemlock Woolly Adelgid Invasion Affect Benthic Communities In Headwater Streams, Christopher J. Strohm

Hemlock woolly adelgid is an invasive herbivore causing extensive mortality of eastern hemlock, an important foundation species that provides stable conditions influencing biological communities. Hemlock is often found in riparian areas and following its decline, broadleaved species, including birch, beech, and rhododendron, will replace it. These plants differ from hemlock in patterns of canopy cover and leaf properties, which influence conditions and resources within streams.
My goal was to evaluate potential impacts of adelgid-induced alterations to riparian canopies and litter on benthic communities and litter breakdown in streams. I characterized benthic invertebrate communities, litter colonization and litter breakdown in streams …

Assessing And Mitigating Lawn Insecticide Hazards To Bees And Other Beneficial Invertebrates, Jonathan Lane Larson

Turfgrass settings, including lawns, golf courses, and sports fields, support many beneficial invertebrates that provide important ecosystem services. These non-target organisms and their associated predation, decomposition, and pollination services can be disrupted by the use of certain insecticides. I compared the ecotoxicity of representatives from three major turf insecticide groups, the neonicotinoids, premix formulations, and the anthranilic diamides, in lab and field realistic settings in order to inform industry initiatives towards environmental sustainability.
In lab and field bioassays clothianidin, a neonicotinoid, and a premix clothianidin/pyrethroid spray were acutely toxic to beneficial insects. Populations of predators, springtails, and earthworms, as well …

Populations of Cimex lectularius, the bed bug, have resurged around the world posing significant challenges for pest management professionals and causing physical, economic, and emotional strife. Pyrethroid resistance has been found in the vast majority of populations making pest management more difficult. The objectives of my dissertation research were to document the evolution of resistance to pyrethroid and neonicotinoid combination products (called combination products here) and to a neonicotinoid in the laboratory, to record potential fitness costs to resistance to the combination products, and to compare the efficacy of nine insecticides on six populations. Influence Of Edible Fruit Coatings On Drosophila Suzukii (Matsumura) (Diptera: Drosophilidae) Oviposition And Development, Katharine A. Swoboda-Bhattarai, Hannah Burrack

Drosophila suzukii (Matsumura) is a highly invasive vinegar fly recently detected in the United States that severely threatens the viability of soft skinned fruit production. Insecticides mitigate some of this damage, but alternative methods to manage D. suzukii infestation are needed. We tested three edible coatings to determine if they could prevent or reduce oviposition by D. suzukii females or affect immature survivorship and development in two important host crops, blueberry and raspberry. None of the coatings prevented oviposition, but some reduced the number of eggs laid. Two carnauba wax-based coatings, PrimaFresh 45 and Raynox, dramatically reduced survivorship of immature …

Background: Transgenic corn hybrids that express toxins from Bacillus thuringiensis (Bt) have suppressed European corn borer populations and reduced the pest status of this insect throughout much of the US corn belt. A major assumption of the high-dose/refuge strategy proposed for insect resistance management and Bt corn is that the frequency of resistance alleles is low so that resistant pests surviving exposure to Bt corn will be rare. Results: The frequency of resistance to the Cry1F Bt toxin was estimated using two different screening tools and compared with annual susceptibility monitoring based on diagnostic bioassays and LC50 …

The adverse effect that insecticides have on spider populations is of major concern since these predators are a component in the biological control of rice insect pest populations. This study was designed to determine the effect of the pyrethroid insecticide Lambda-cyhalothrin on spider populations in rice fields. Treatments consisted of plots treated once with the insecticide and untreated plots (control). Spiders were sampled four times per crop year over a two-year period in each of three sites in the state of Rio Grande dol Sul, Brazil. Treatments were replicated four times. Management Of Potato Leafhopper And Maple Spider Mite On Nursery Grown Maples, Julia Prado Beltran

Potato leafhopper Empoasca fabae (Harris) (Homoptera: Cicadellidae) and maple spider mite Oligonychus aceris (Shimer) (Acarina: Tetranychidae) are important pests of maple trees. Investigations determined how insecticide use and fertilization impacted the abundance of E. fabae and O. aceris on `Red Sunset' red maple and `Autumn Blaze' Freeman maples. Bifenthrin applications directed against leafhoppers reduced damage to both cultivars, but increased O. aceris on Autumn Blaze. Using a threshold of one leafhopper per branch to trigger pesticide applications protected Red Sunset maples from leafhopper injury. It also prevented outbreaks of O. aceris on Autumn Blaze because few trees reached this density. …

Frequency And Distribution Of Extended Diapause In Nebraska Populations Of Diabrotica Barberi (Coleoptera: Chrysomelidae), Ryan W. Geisert, Lance J. Meinke

The frequency of extended diapause in populations of the northern corn rootworm, Diabrotica barberi Smith & Lawrence (Coleoptera: Chrysomelidae), was measured in eastern Nebraska. Adult collections were made during late summer in 2008 and 2009 from eight sites each year (seven sites were consistent over years). Eggs were obtained from 12 to 20 females per site and were held on moist soil under appropriate temperature profiles to facilitate egg survival, diapause development, and diapause termination. Percentage egg hatch was recorded after the first and second year for the 2008 and 2009 collections. Additional extended diapause expression was estimated for the …

Characterization Of Resistance To The Cry1f Toxin From Bacillus Thuringiensis In Resistant Fall Armyworm, Spodoptera Frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) From Puerto Rico, Ana Maria Velez

Transgenic corn expressing Cry1F protein from Bacillus thuringiensis Berliner has been registered for Spodoptera frugiperda control since 2003. Unexpected damage to Cry1F corn was reported in 2006 in Puerto Rico and Cry1F resistance in S. frugiperda was documented. The inheritance of Cry1F resistance was characterized in a S. frugiperda resistant strain from Puerto Rico which displayed >387-fold resistance to Cry1F. Concentration-response bioassays of reciprocal crosses of resistant and susceptible parental populations indicated that resistance is recessive and autosomal. Bioassays of the backcross of the F1 generation crossed with the resistant parental strain suggest that a single locus is responsible …
