The potato aphid, Macrosiphum euphorbiae, is an important agricultural pest that causes economic losses to potato and tomato production. To establish the transcriptome for this aphid, RNA-Seq libraries constructed from aphids maintained on tomato plants were used in Illumina sequencing generating 52.6 million 75±105 bp paired-end reads. The reads were assembled using Velvet/Oases software with SEED preprocessing resulting in 22,137 contigs with an N50 value of 2,003bp. After removal of contigs from tomato host origin, 20,254 contigs were annotated using BLASTx searches against the non-redundant protein database from the National Center for Biotechnology Information (NCBI) as well as IntereProScan. …

Emerald ash borer (EAB), Agrilus planipennis (Coleoptera: Buprestidae), is an aggressive invader from Asia that has killed millions of trees in North America. Recently EAB has been documented developing in a novel host, white fringetree, Chionanthus virginicus. I evaluated larval performance in two common ash species and white fringetree by infesting excised bolts with emerald ash borer eggs. In addition I evaluated several plant characteristics to determine which most influence larval development. I also conducted choice and no choice assays using the classical biological control agent, Tetrastichus planipennisi (Hymenoptera: Eulophidae), to assess its ability to locate larval EAB in …

Changes to traditional taxonomic methods to incorporate new technologies and methods have already improved the quality of species hypotheses, but more work can be done to improve the speed of new species documentation. The mitochondrial COI DNA barcode has been successfully used to identify species with high accuracy since the early 2000s, and has been used in conjunction with morphological examinations and other DNA markers to discover and delimit new species. This thesis explores the application of DNA barcodes as the primary data for delimitation and diagnosis of new species of ichneumonoids.

Throughout the 2016 and 2017 growing season, field research experiments were replicated across South Dakota. Many times seed treatments are used prophylactic, which is neither good for the producers or the environment. Producers will be able to reduce production costs, if they only use a seed treatment when necessary. The purpose of the first experiment was to determine the effects of seed treatments in combination with planting date and seeding rate on soybean yield. On June 11, 2018, Amanda Grady, Entomologist, Arizona Zone, Forest Health Protection, performed a site visit to evaluate aspen health on the Nature Conservancy lands at Heart Prairie Preserve (HPP) near Flagstaff. Specifically, the evaluation was to determine whether or not the aspen within exclosures on Fern Mountain have symptoms or signs of Oystershell scale (OSS), an emergent forest pest contributing to aspen decline in northern Arizona.

Pesticides, poor nutrition, parasites and diseases work synergistically to contribute to the decline of the honey bee. Heritable sub-lethal behavior/immune effects may also contribute to the decline. Maternal stress is a common source of heritable immune/behavior deficits in many species. A stressed honey bee queen has the potential to pass such deficits on to worker bees. Using a repeated measures design, this study will determine whether the health of worker bee is reduced by a cold stress on the queen by analyzing egg hatch rate and protein content, emergence rate, and adult aggression and immune function for offspring laid before …

Many insects form close relationships with microbial symbionts. Insect symbionts can provide novel phenotypes to their hosts, including influencing dietary breadth. In the polyphagous cowpea aphid, Aphis craccivora, the facultative symbiont Arsenophonus improves aphid performance on one host plant (locust), but decreases performance on other plants. The goal of my thesis was to investigate the mechanism by which Arsenophonus facilitates use of locust. First, I assembled an Aphis craccivora-Arsenophonus-Buchnera reference transcriptome to conduct RNAseq analysis, comparing gene expression in aphids feeding on locust and fava, with and without Arsenophonus infection. Cucurbits (i.e. squash, melons, pumpkins, gourds) are high value crops of global importance. Insect pests in these systems are often controlled by chemical insecticides, which are not always effective and can be damaging to the environment. Many integrated pest management (IPM) techniques have been developed for the control of pests in these systems, with a goal of improving system stability and reducing chemical inputs. The overarching goal of my research was to investigate the impact of select IPM techniques on arthropod populations and yield in organic and conventional cucurbit systems.

RNA interference (RNAi) is a useful reverse genetics tool for investigation of gene function as well as for practical applications in many fields including medicine and agriculture. Due to the variability in RNAi efficiency, RNAi-based methods are currently being developed for controlling only coleopteran insects which are known to be amenable to RNAi. The first chapter of my thesis includes findings from research to investigate what are the factors that make coleopteran insects relatively more efficient in RNAi. Public awareness of declining pollinator populations has increased interest in creating “bee–friendly” urban landscapes. I quantified bee visitation and assemblages of 72 species of flowering woody plants common in urban landscapes. I found strong plant species effects and variation in seasonal activity of particular bee taxa but no overall differences in bee visitation or genus diversity between native versus nonnative species or trees versus shrubs. Analysis of pollen from a subset of these plants revealed small but statistically significant differences in total and essential amino acids between native and nonnative species and trees and shrubs, although each group had species …

Tibraca limbativentris Stål (Hemiptera: Pentatomidae) is a major pest causing economic losses to rice cultivation in Brazil. The objectives of this work were to assess different sources of silicon in the induction of plant resistance, to examine the interaction of rice varieties with silicon-based resistance, and to determine the effects of varieties and silicon on stink bug biology. The interaction of rice cultivars BRS Esmeralda, IRGA 429 (widely cultivated in Brazil), and Canela de Ferro (a resistant cultivar), and 2 sources of silicon (K silicate, and Ca + Mg silicate) were studied. The relationship between biodiversity and ecosystem functioning is important in maintaining agroecosystem sustainability. Plant-microbe symbioses, such as exists between the grass tall fescue (Schedonorus arundinaceum) and the asexual fungal endophyte Epichloë coenophiala, can be utilized to enhance agroecosystem functions, such as herbivore resistance. “Novel” E. coenophiala strains that vary in the production of mammal- and insect-toxic compounds have been identified, inserted into tall fescue cultivars, and are planted in pastures globally. Novel fungal endophyte-tall fescue associations may have divergent ecosystem function effects. The objective of this study was to estimate the economic impact of the sugarcane aphid (SCA) outbreak in the Rio Grande Valley (RGV), Texas. Local producers were surveyed to gather detailed information about crop production and pest control practices. Collected data were used to estimate the reduction in profit associated with the SCA infestation, as well as the monetary value of the prevented loss attributed to control efforts. Sorghum industry losses were then used to assess the overall economic impact of the SCA outbreak in the RGV economy.

The soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is an invasive species from Asia that has been the major economic insect pest of soybeans, Glycine max (L.) Merrill, since 2000. While use of soybeans expressing antibiosis and antixenosis is a well-studied strategy to manage this pest, aphid-tolerant soybeans remain underexplored. This study examined the relationship between cumulative aphid-days (CAD) and yield loss in the tolerant soybean KS4202 during two growing seasons to determine the economic injury levels (EILs) for soybean aphids on KS4202. While at NOAA my research was directed by the inquiry of whether or not HFO-1234yf, a new compound being introduced into coolant system and replacing greenhouse gases HFC, is harmful to organic life. Using the computer program Matlab I was able to develop figures that represented the concentration of HFO-1234yf (HFO) in 12 different tower locations throughout the country. Comparing and contrasting concentrations of this HFO compound at each site led me to the question the possible harmful side effects this compound could bring to organic life due to the fact that concentration levels are increasing, with one factor being …
