Although North America contains the largest diversity of freshwater mussels in the world, only ~24% of species in the United States and Canada have stable populations. This group provides unique hurdles for surveying, and current unionid survey protocols include neither longitudinal studies nor randomization of sites. This project conducts a metadata analysis of freshwater mussels in the Greenup Pool of the Ohio River (GPOR) to answer three questions: 1. Studies of threats that bats face during hibernation have increased in response to white-nose syndrome (WNS), a fungal disease that has ravaged North American bat populations. However, impacts of WNS on bat ecology during spring emergence, when bats are potentially recovering from infection and allocating resources for reproduction, is relatively understudied. As more bat species become listed at the federal and state level, the need to understand the factors associated with spring emergence is critical for improving conservation guidelines and habitat management practices. The goal of this study is to examine the effect of urbanization and latitude on bone morphology, specifically limb length and bone density among gray squirrels endemic to the eastern United States. This study’s hypotheses are as follows: gray squirrels occupying lower latitudes will have larger body sizes and longer limbs relative to body size than those at higher latitudes following Bergmann’s and Allen’s rules. Further, squirrels in urban habitats will have greater bone density than those in rural habitats. Results show moderate correlation between body mass and respective proxies and latitude following Bergmann’s rule. Weak correlations were found between …

Eastern diamondback rattlesnakes (Crotalus adamanteus; EDB) are an imperiled species native to the Southeastern United States longleaf pine (Pinus palustris) ecosystem. Several markrecapture methods have been used range-wide in hopes of increasing knowledge on their population numbers. I explored the efficacy of two mark-recapture methods (scale cauterization and dorsal head photography) using the Marine Corps Recruit Depot, Parris Island, SC EDB population in this multi-part study. The first method I evaluated was ventral scale branding, which has been widely implemented without knowledge of brand permanence. Determining the survey effort required to reliably detect population change can be challenging for cryptic, elusive species. The secretive nature of amphibians makes it difficult to monitor population status and gather information about their natural history, including habitat use, which is essential for amphibian monitoring programs. In wildlife management, translocation is defined as the purposeful movement of an individual(s) from one place to another with a wildlife management goal. Translocation has become a common tool for mitigating negative human-wildlife interactions. We conducted a translocation study on timber rattlesnakes (Crotalus horridus) in response to numerous human-rattlesnake encounters with the goal of developing a translocation protocol that would allow natural resources personnel to mitigate negative encounters between the public and rattlesnakes and reduce impacts to local timber rattlesnake populations. Translocations have the potential to aim conservation efforts as well as to reduce mortality caused by human activities. Eastern diamondback rattlesnakes (Crotalus adamanteus, EDB) have a limited ability to adapt to habitat loss and fragmentation due to the species’ slow life history and minimal dispersal ability. Because they are a venomous species, they are viewed as nuisance animals and are often killed on sight. We translocated a cohort of EDBs to investigate the potential of using translocations as a conservation and mitigation tool for this species. The decline of amphibians has been of international concern for more than two decades, and the global spread of introduced fauna is a major factor in this decline. Conservation management decisions to implement control of introduced fauna are often based on diet studies. One of the most common metrics to report in diet studies is Frequency of Occurrence (FO), but this can be difficult to interpret, as it does not include a temporal perspective. Coyotes (Canis latrans) are now found throughout North and Central America, but before European colonization were restricted to west of the Mississippi. Migration occurred in two major paths to the East; north over the Great Lakes (through Canada) and south below the Great Lakes. The location of these routes is significant because those migrating north interbred with the wolves that reside there. These hybrid animals are larger and behaviorally different from their western counterparts. It is possible to differentiate these hybrids morphologically and genetically. As anthropogenic environmental stressors such as urbanization continue to increase, it is necessary to understand their impact on amphibian ecology. Due largely to their biphasic life history amphibians are negatively impacted by the destruction of lotic habitats i.e., wetlands for the creation of roads and urban centers. I examined the effects of three human-made road rut pools on the reproductive success and species richness of amphibians in southwest West Virginia from May to October 2012. Internal implantation of radio-transmitters is the preferred attachment technique for snakes, but the high costs and invasive nature of the surgery make a functional alternative desirable. External radio-transmitters are cost-effective alternatives to surgical implantation. Rattlesnake rattles are unique morphological features that can serve as an attachment site for external radio-transmitters. Using thread and epoxy, I attached transmitters to the rattles of eastern diamondback rattlesnakes (Crotalus adamanteus; EDB). I calculated average monitoring duration using radio telemetry data collected from 49 adult EDBs telemetered from 2014 to 2017 in coastal South Carolina. The Wood Turtle (Glyptemys insculpta) is listed as vulnerable in West Virginia and only occurs in the Eastern Panhandle. The study population is unique in West Virginia because it occurs at a much higher mean elevation than other Wood Turtle populations in the state. The primary objective of this study was to determine if, and to what extent, altitude affected this population’s ecology. Turtles were located with opportunistic surveys and captured by hand. The Cumberland Plateau Salamander, Plethodon kentucki, is a member of the Plethodon glutinosus complex comprising 16 sibling species, which are best differentiated by range. Few studies have been conducted to gain information on the natural history of P. kentucki. To alleviate this, two sites at Beech Fork State Park in Wayne County, West Virginia were used to study the salamander’s general life history with emphasis on reproduction, phenology, and population size. The Queensnake (Regina septemvittata) is a small secretive water snake found throughout the eastern United States. Once common, their numbers have declined to the extent that they are now threatened throughout most of their range, largely the result of pollutant-based reduction in prey species. These snakes are assumed to eat molted crayfish exclusively. For some common crayfish species, molting happens only twice a summer during a two- week period. Calls of male anurans during breeding seasons are species-specific identification tools. However, males cease calling after any nearby disturbance, including those of researchers. I proposed a variation on current methods that attempts to reduce this lag in calling after researcher-created disturbance by utilizing the propensity for competition in male frogs. I surveyed 14 breeding sites in Cabell and Wayne counties during the 2010 and 2011 breeding seasons. First, I used traditional visual encounter surveys (VESs). Large river studies for freshwater mussel populations and habitat in the Monongahela River within Pennsylvania have been almost non-existent over the past century. Aquatic diversity and water quality have been impaired in the Monongahela River since the Industrial Revolution and early impoundments were constructed to control the river. To date, there have been no thorough mussel population studies conducted on the Pennsylvanian Monongahela River proper since A.E. Ortmann in 1919. Amphibian declines have been well documented, specifically in the last few decades. The Hellbender, Cryptobranchus alleganiensis, one of North America’s largest salamander species, has suffered dramatic declines throughout much of its range, with estimated declines of up to 77 percent recently documented in some populations. With diurnal and nocturnal searches and mark-recapture techniques, I collected data on the status of Eastern Hellbender populations in Eastern West Virginia. Since 1971, documented Rough Greensnake (Opheodrys aestivus) occurrences in West Virginia have declined from approximately 100 to less than 20 occurrences in the last 10 years. In contrast to the decline of Rough Greensnakes in WV, Smooth Greensnake (O. vernalis) populations appear to be stable over the same period of time. Historic sites from the West Virginia Biological Survey were cross referenced with habitat descriptions in published literature to establish research locations. The Eastern Hellbender, Cryptobranchus alleganiensis alleganiensis, is a species of concern in West Virginia and is in need of management. An important component of conservation efforts will involve identifying suitable habitat for protection. The goal of this research was to locate populations and examine hellbender habitat preferences to help managers identify habitat for protection. Populations were located using rock turning surveys from May through November, 2006. Hellbenders may be sensitive to water chemistry, so the dissolved oxygen, pH, turbidity, specific conductivity, and water temperature were measured. Substrate composition may influence populations, so substrate was characterized with Wolman pebble counts. …

Being lungless, plethodontid salamanders respire through their skin and are especially sensitive to environmental disturbances. Habitat fragmentation, low abundance, extreme habitat requirements, and a narrow distribution of less than 70 miles in length, makes one such salamander, Plethodon punctatus, a species of concern (S1) in West Virginia. To better understand this sensitive species, day and night survey hikes were conducted through ideal habitat and coordinate data as well as tail tips (10 to 20 mm in length) were collected. DNA was extracted from the tail tips and polymerase chain reaction (PCR) was used to amplify mitochondrial 16S rRNA gene fragments. …

Chapter 1 is a literature review of Pseudacris feriarum, largely based on published P. triseriata Complex species accounts. Chapter 2 presents the natural history of P. feriarum in West Virginia and compares some parameters with other Pseudacris species. Chapter 3 looks at the phenology of a wetland in eastern West Virginia. Chapter 4 examines the current range of P. feriarum and compares it with the historical range in the state. Minnow traps were placed along three drift fences with additional traps placed throughout the study area. From February 5 to March 27, 2005, 85 captured adults were identified to species, marked by elastomer injection for mark-recapture analysis, and measured for morphometrics. Microbial surveying using antibiotic resistant bacteria, fecal coliforms, and virulence genes is an approach not previously tested on the Great Kanawha River. Research objectives were to test antibiotic resistant and fecal coliform bacteria as bioindicators of water quality, and develop a multiplex- polymerase chain reaction (mPCR) system for identification of stx1, stx2, and eaeA DNA sequences in isolated fecal coliforms (FC). Fecal indicator populations and antibiotic resistant populations were demonstrated to be independent.
