Objective: To determine the impact of < 5% O2 tension on the in vitro development of human pre-implantation embryos. It was hypothesized that the optimal O2 tension would differ from the commonly utilized 5%, as shown by beneficial effects after the implementation of a more physiological O2 tension such as 2% for either the partial or full duration of in vitro culture.

Aim: To conduct a randomized controlled study to determine the effect of O2 tension during in vitro human embryo culture on the production of reactive oxygen species (ROS), apoptosis, gene expression and embryo development.

The purpose of this work was to study the relationships between age, measures of gait variability, and locomotor adaptability. Measures of gait variability are used to identify maladapted locomotor behavior, motor disease, and risk of falls. The first aim was to determine the relationships between age and measures of gait variability. Thirty-four participants (23-71 years old) walked on a treadmill for 6 minutes at their preferred speed. Variability of stride times and lengths was computed via linear measures (standard deviation and coefficient of variation) and nonlinear measures (sample entropy and detrended fluctuation analysis). Single-cell RNA sequencing of cells from cultured human blastocysts has enabled us to define the transcriptomic landscape of placental trophoblast (TB) that surrounds the epiblast and associated embryonic tissues during the enigmatic day 8 (D8) to D12 peri-implantation period before the villous placenta forms. We analyzed the transcriptomes of 3 early placental cell types, cytoTB (CTB), syncytioTB (STB), and migratoryTB (MTB), picked manually from cultured embryos dissociated with trypsin and were able to follow sublineages that emerged from proliferating CTB at the periphery of the conceptus. Previously, we demonstrated the ability of the normal mammary microenvironment (niche) to direct non-mammary cells including testicular and embryonic stem cells (ESCs) to adopt a mammary epithelial cell (MEC) fate. These studies relied upon the interaction of transplanted normal MECs with non-mammary cells within the mammary fat-pads of recipient mice that had their endogenous epithelium removed. Here, we tested whether acellular mammary extracellular matrix (mECM) preparations are sufficient to direct differentiation of testicular-derived cells and ESCs to form functional mammary epithelial trees in vivo. The hamster is a useful model of human reproductive biology because its oocytes are similar to those in humans in terms of size and structural stability. In the present study we evaluated fecundity rate, ovarian follicular numbers, ova production, mitochondrial number, structure and function, and cytoplasmic lamellae (CL) in young (2–4 months) and old (12–18 months) Syrian hamsters (Mesocricetus auratus). Young hamsters had higher fertilisation rates and larger litters than old hamsters (100 vs 50% and 9.3 +/- 0.6 vs 5.5 +/- 0.6, respectively). Squid rely on multiple sensory systems for predator detection. In this study we examine the role of two sensory systems, the lateral line analogue and vision, in successful predator evasion throughout ontogeny. Squid Doryteuthis pealeii and Lolliguncula brevis were recorded using high-speed videography in the presence of natural predators under light and dark conditions with their lateral line analogue intact or ablated via a pharmacological technique. Paralarval squid showed reduced escape responses when ablated; however, no differences were found between light and dark conditions in non-ablated paralarvae, as was previously shown in juveniles and adults, indicating that the lateral line …

This study investigated the role of milk fat globule-epidermal growth factor-factor 8 (MFGE8) in TGF-β-induced epithelial– mesenchymal transition (EMT) of endometrial epithelial cells. These were in vitro studies using human endometrial epithelial cells and mouse blastocysts. We investigated the ability of TGF-β to induce EMT in endometrial epithelial cells (HEC-1A) by assessment of cytological phenotype (by light and atomic force microscopy), changes in expression of the markers of cell adhesion/differentiation E- and N-cadherin, and of the transcription factor Snail (by immunofluorescence and immunoblotting), and competence to support embryo attachment in a mouse blastocyst outgrowth assay. Mutant huntingtin protein (mhtt)– the protein responsible for cellular dysfunction in Huntington’s disease (HD) –is a product of an expanded trinucleotide repeat (TNR) cytosine-adenine-guanine (CAG) sequence in exon 1 of the huntingtin (HTT) gene. The pathology of HD has been extensively researched; however, the mechanism by which the disease-causing TNR expansions occur in somatic cells remains elusive. Costal cartilage is a type of hyaline cartilage that forms rod-like structures that connect the ribs to the sternum. The most common chest wall deformities, pectus excavatum and pectus carinatum involved efective costal cartilage resulting in sternal displacement. Costal cartilage is not widely studied leaving little insight into possible factors involved in the pathogenesis of these pectus deformities. This study focused on the presence and distribution of two important regulators of collagen fibrillogenesis and organization, biglycan and decorin. Drosophila melanogaster is a dominant model organism for studying the function of animal genes in initial stages of embryogenesis. Usually, images containing Drosophila gene expression patterns are captured at different developmental stages to study the interconnection of animal genes. To achieve most biologically meaningful results, gene expression images from a similar stage should be compared. Currently, biologists manually classify embryos in images into different stages, which is time intensive and infeasible for current massively produced gene expression images. Sea turtles reside in different acoustic environments with each life history stage and may have different hearing capacity throughout ontogeny. For this study, two independent yet complementary techniques for hearing assessment, i.e. behavioral and electrophysiological audiometry, were employed to (1) measure hearing in post-hatchling and juvenile loggerhead sea turtles Caretta caretta (19-62 cm straight carapace length) to determine whether these migratory turtles exhibit an ontogenetic shift in underwater auditory detection and (2) evaluate whether hearing frequency range and threshold sensitivity are consistent in behavioral and electrophysiological tests. Cephalopods have visual and mechanoreception systems that may be employed to sense and respond to an approaching predator. While vision presumably plays the dominant role, the importance of the lateral line analogue for predator evasion has not been examined in cephalopods. To test the respective roles of vision and the lateral line analogue, brief squid, Lolliguncula brevis, were observed in the presence of summer flounder, Paralichthys dentatus, under light and dark conditions with their lateral line analogue intact and ablated. We have previously shown that non-mammary and tumorigenic cells can respond to the signals of the mammary niche and alter their cell fate to that of mammary epithelial progenitor cells. Here we tested the hypothesis that paracrine signals from mammary epithelial cells expressing progesterone receptor (PR) are dispensable for redirection of testicular cells, and that re-directed wild-type testicular-derived mammary cells can rescue lobulogenesis of PR-null mammary epithelium by paracrine signaling during pregnancy. Vemurafenib is a selective B-RafV600E inhibitor in melanoma targeted therapy which also inhibits the wild type B- and C-Raf. Experiments were conducted to redirect mouse Embryonic Stem (ES) cells from a tumorigenic phenotype to a normal mammary epithelial phenotype in vivo. Mixing LacZ-labeled ES cells with normal mouse mammary epithelial cells at ratios of 1:5 and 1:50 in phosphate buffered saline and immediately inoculating them into epithelium-divested mammary fat pads of immune-compromised mice accomplished this. Our results indicate that tumorigenesis occurs only when normal mammary ductal growth is not achieved in the inoculated fat pads. When normal mammary gland growth occurs, we find ES cells (LacZ+) progeny interspersed with normal mammary cell progeny in the mammary epithelial structures. We …

Chromosome organization is regarded as an important factor in regulating gene expression. In addition to the histone code, it is a part of the epigenetic mechanisms participating in fertilization and early embryonic development. Recent studies have demonstrated that chromosomes of human spermatozoa have well-defined spatial organization that includes preferred intranuclear positioning. The hypothesis of this dissertation is that sperm chromosome architecture participates in controlled transformation and activation of the paternal genome following fertilization.

Understanding the extent to which changes in muscle form and function underlie ontogenetic changes in locomotory behaviors and performance is important in understanding the evolution of musculoskeletal systems and also the ecology of different life stages. We explored ontogenetic changes in the structure, myosin heavy chain (MHC) expression and contractile properties of the circular muscles that provide power for jet locomotion in the long-finned squid Doryteuthis pealeii. The dynamics of pulsed jetting in squids throughout ontogeny is not well understood, especially with regard to the development of vortex rings, which are common features of mechanically generated jet pulses (also known as starting jets). Studies of mechanically generated starting jets have revealed a limiting principle for vortex ring formation characterized in terms of a 'formation number' (F), which delineates the transition between the formation of isolated vortex rings and vortex rings that have 'pinched off' from the generating jet. To understand the role of lipids during early embryogenesis, major lipid classes together with individual fatty acid and sterol composition were determined in embryos from multiple developmental stages of the euphausiids Euphausia pacifica and Thysanoessa spinifera. Average lipid content in embryos of E. pacifica and T. spiniferafrom the earliest stage (multicell) were 4.45 and 3.69 µg embryo-1,respectively. During development, the lipid content decreased at similar rates in the embryos of both species. In contrast to many crustacean eggs, phospholipids were the dominant lipid class in all embryonic stages, with decreasing concentrations seen during development. Individual …

This investigation identified the morphological defects caused by different concentrations of chromium, copper, and arsenic (CCA) on the developing zebrafish embryo; in addition to defects caused by the three combined components (complete CCA). A change in the DNA of the developing embryos in response to the three components of CCA and complete CCA was also determined. Early Dania rerio embryos were cultured for five days with various concentrations of CCA components and complete CCA. To determine if Antarctic krill employ body shrinkage as one of its overwintering mechanisms in the field, Euphausia superba and Euphausia crystallorophias were collected during fall and winter in and around Marguerite Bay through US Southern Ocean GLOBEC field programs during fall and winter 2001 and 2002. The relationships between the body length and weight of both krill species were exponentially correlated with no significant differences between the two species (p>0.05). The blue crab (Callinectes sapidus) plays an important economic and ecological role in estuaries and coastal habitats from the Gulf of Mexico to the east coast of North America, but demographic assessments are limited by length-based methods. We applied an alternative aging method using biochemical measures of metabolic byproducts (lipofuscins) sequestered in the neural tissue of eyestalks to examine population age structure. From Chesapeake Bay, subsamples of animals collected from the 1998-99 (n-769) and 1999-2000 (n=367) winter dredge surveys were collected and lipofuscin was measured. Leukemia inhibitory factor (LIF) enhances in vitro murine preimplantation development in a time- and dose-dependent fashion. Knockout experiments have demonstrated that endometrial LIF is essential for in vivo murine implantation. We assessed the impact of LIF and an anti-LIF polyclonal antibody (pab) on in vivo development and developed a novel and successful nonsurgical method of embryo transfer for this species, a transcervical blastocyst transfer technique. To better understand growth and age-pigment (lipofuscin) accumulation rates of the blue crab Callinectes sapidus under natural conditions, juveniles (33 to 94 mm carapace width) were reared in outdoor ponds for over 1 yr. Growth rates, measured by carapace width, during summer and fall exceeded all those reported in the literature; the initial carapace width of 59 ± 14 mm (mean ± SD) increased to 164 ± 15 mm within a 3 mo period. The blue crab Callinectes sapidus is an economically and ecologically important species in many temperate estuaries, yet stock assessments have been limited to length-based methods for demographic analyses. We evaluated the potential of age pigments (lipofuscins) sequestered in neural tissue of eye-stalks and brains to estimate the age of blue crabs collected from Chesapeake Bay and Chincoteague Bay. The rate of lipofuscin accumulation was determined using crabs of known age reared in the laboratory. Age pigments were extracted from neural tissues (eye-stalk or brain), quantified, and normalized to protein content to allow comparisons across tissue types and crab sizes. Field-collected …

Leukemia inhibitory factor (LIF) is a pleiotropic cytokine which demonstrates perplexing physiological effects. It has been demonstrated that LIF is essential for implantation in mice. Little is known relating to the manner by which LIF effects pre-implantation and post-implantation development. The objectives of this project were to determine the effects LIF on pre-implantation development, to determine the effects that it may have on implantation rates, successful pregnancy rates, and resorption rates, and to determine the effects that LIF has on the skeletal development of mice. We reared the harpacticoid copepod Nitocra spinipes on diets of bacteria, a diatom, or a macroalga, evaluating survivorship and growth in short-term (≤ 1 generation) experiments. Lipid content of the copepods and their diets was measured and used as an index of nutrition. Although growth, survivorship and lipid content of N. spinipes were significantly greater when fed the diatom, which had the highest lipid content of the 3 diets, the copepod was able to develop from egg to adult when fed a lipid- poor bacterial diet. The incorporation of lipids into the copepod Acartia tonsa and its eggs was measured when it was fed either a bacterivorous ciliate (Pleuronema sp.) or a diatom (Thalassiosira weissflogii). Egg production was 10-fold higher on the diatom diet, whereas hatch success of eggs was the same for algal and ciliate diets. Adult copepods fed diatoms contained more total fatty acid and sterols than copepods fed the ciliate diet, and individual lipids reflected the dietary source. Eggs from diatom-fed copepods had fewer fatty acids but more sterols than eggs from copepods on a ciliate diet. Ciliate-fed copepods and …
