The trophic morphology of gape-limited predators constrains the shape and size of prey items they can ingest. Trophic morphology consists of any morphological feature that is involved in the handling and ingestion of food. Diet has a profound effect on the morphology of many gape-limited predators. Identifying how prey type and resource level affect the morphology of different populations is an essential step in understanding the mechanisms contributing to patterns of morphological diversity. Species interactions (Chapter 1) induce plasticity in morphology that can lead to increased fitness, morphological divergence, and eventually speciation.

Self-incompatibility is thought to have played a profound role in the evolution of the angiosperms. However, there is little evidence of self-incompatibility systems in early diverging lineages of flowering plants. Illicium parviflorum, one such early-divergent angiosperm, is an evergreen perennial species endemic to central Florida, particularly within the Ocala National Forest. Although locally abundant, I. parviflorum is currently listed as endangered at the state level due to being under constant threat of habitat disturbance and over-harvesting. Notably, this species had been described as self-incompatible due to its low seed-set. However, low seed set may also be a result of …

Systematics Of Leptopelis (Anura: Arthroleptidae) From The Itombwe Plateau, Eastern Democratic Republic Of The Congo, Francisco Portillo

Leptopelis, a genus of Central African treefrogs, includes 51 species that live in tropical forests and savannas. Currently, only two species of Leptopelis are known from the poorly explored Itombwe Plateau in eastern Democratic Republic of the Congo (DRC). Itombwe is renowned among conservationists for its rich and endemic amphibian fauna, including: Xenopus itombwensis, Chrysobatrachus cupreonitens, Laurentophryne parkeri, Hyperolius leleupi and at least three species of Arthroleptis. Evolutionary relationships of Itombwe Leptopelis were examined by sequencing two mitochondrial genes (16S: 557 bp [base pairs], cyt b: 620 bp) and one nuclear gene (RAG1: 761 bp). The gastrula stage represents the point in development at which the three primary germ layers diverge. At this point the gene regulatory networks that specify the germ layers are established and the genes that define the differentiated states of the tissues have begun to be activated. These networks have been well characterized in sea urchins, but not in other echinoderms. Embryos of the brittle star Ophiocoma wendtii share a number of developmental features with sea urchin embryos, including the ingression of mesenchyme cells that give rise to an embryonic skeleton. The vast diversity of insect appendages provides an excellent model for examining the cellular and molecular basis of phenotypic changes in nature. One of the hallmarks of evolution of insect appendages is the differential enlargement of hind (T3) legs compared to their fore (T1) and mid (T2) counterparts. While basic information regarding the genetic mechanisms that regulate the size of the T3 legs became available in the past several years, virtually nothing is known about cellular mechanisms involved. Botrylloides violaceus is a colonial ascidian with the ability to undergo sexual and asexual reproduction as well as regeneration. The canonical pathway starts with the extracellular protein Wnt and ends with β-catenin, a transcription factor, which also functions in cell adhesion. The Wnt signaling pathway is involved in embryogenesis and regeneration in a variety of other species. In our studies we attempt to isolate and sequence both a Wnt gene and from Botrylloides via degenerate primer design and PCR. Using bioinformatic methods we aligned sequences from other organisms, as the Botrylloides genome has not yet been sequenced. Using mouse, Ciona, …

Janus Kinase (JAK) activity specifies the cell fates of the follicular epithelium during Drosophila oogenesis by establishing a gradient of JAK activity with highest levels at the A/P poles. Unpaired (Upd), a ligand for the pathway, is expressed and secreted exclusively from the polar cells potentially establishing the JAK activity gradient. This project proposed that Upd acts as a morphogen to directly establish the JAK activity gradient, specifying the fates of the follicular epithelium. Two studies were conducted to investigate the physical and biological processes contributing to the dissolved oxygen (DO) availability in the Skidaway River Estuary (SRE), Savannah, GA during the summer of 2005. A temporal study looked at changes in DO concentrations, Chlorophyll a concentrations, bacterial abundance, water depth, and salinity, every hour, for 26 hours, over both a neap and a spring tide. A spatial study looked at changes in the above variables at 5 sites along the SRE while following the tide inland during a neap high and low tide, and a spring high and low tide. Distribution And Conservation Genetics Of The Cow Knob Salamander, Plethodon Punctatus Highton (Caudata: Plethodontidae), Matthew R. Graham

Being lungless, plethodontid salamanders respire through their skin and are especially sensitive to environmental disturbances. Habitat fragmentation, low abundance, extreme habitat requirements, and a narrow distribution of less than 70 miles in length, makes one such salamander, Plethodon punctatus, a species of concern (S1) in West Virginia. To better understand this sensitive species, day and night survey hikes were conducted through ideal habitat and coordinate data as well as tail tips (10 to 20 mm in length) were collected. DNA was extracted from the tail tips and polymerase chain reaction (PCR) was used to amplify mitochondrial 16S rRNA gene fragments. …

This investigation identified the morphological defects caused by different concentrations of chromium, copper, and arsenic (CCA) on the developing zebrafish embryo; in addition to defects caused by the three combined components (complete CCA). A change in the DNA of the developing embryos in response to the three components of CCA and complete CCA was also determined. Early Dania rerio embryos were cultured for five days with various concentrations of CCA components and complete CCA. The central Appalachian region of the United States receives some of the highest inputs of nitrogen (N) due to acidic deposition in the nation. It is believed that these high could levels contribute to a decline in forest soils within the next 50 to 70 yrs. This study examines factors that influence spatial variability in N-treated and untreated watersheds of the Fernow Experimental Forest, Parsons, West Virginia. Within each of the two watersheds [WS4 untreated control, > 100 yr.; WS3 N-treated, acidified, clear cut, ~ 31 yr.], two 0.04 ha plots, one high N and one low N, were selected for …

In West Virginia, as throughout its range, there is limited information about the life history of the cave salamander (Eurycea lucifuga). The purpose of this study was to describe the natural history of this species in West Virginia. The objectives of this study were to determine habitat selection of the cave salamander, to ascertain its environmental characteristics, and to study its reproductive and non-reproductive biology. Three study caves were located in Greenbrier County, West Virginia. All sites were examined at least once each month from May 1999 through April 2000. By implementing a mark-recapture study, I investigated the ecology of three species of sympatric plethodontid salamanders (Aneides aeneus, Desmognathus ochrophaeus, and Plethodon glutinosus) on rock outcrops in the Westvaco Wildlife and Ecosystem Research Forest, Randolph County, West Virginia. By examining activity patterns, vertical stratification, and habitat preference for A. aeneus, D. ochrophaeus, and P. glutinosus, I attempted to offer some insight into how these sympatric species avoid competition on small, isolated rock outcrops. The blacknose dace, Rhinichthys atratulus, is a cyprinid minnow common to shallow streams throughout North America. It has been shown to be sensitive to anthropogenic stress and a study of the genetic nature of R. atratulus populations may yield information about their habitats. In West Virginia, two subspecies of R. atratulus are present, R. a. atratulus and R. a. obtusus, which are separated by the Allegheny divide. In north central West Virginia, R. a. atratulus was observed in the Cheat River drainage, an area previously thought be within the range of R. a. obtusus. Fish of the R. a. atratulus …
