Chulalongkorn University Theses and Dissertations (Chula ETD)
Electrochemical immunosensors for the simultaneous detection of antibiotics have been of interest and have been reported previously. However, those methods still require difficult labeling for the reporting of electrochemical signals. This research aimed to develop a novel label-free electrochemical immunosensor with an origami paper-based analytical device (oPAD) for the simultaneous detection of quinolone antibiotics. Norfloxacin (Nor) and Enrofloxacin (Enr) are among the quinolone antibiotics, and they are used as models to study antibiotic detection. Nor and Enr antibodies were immobilized separately onto the functionalized oPAD. Thionine and ferrocenecarboxylic acid were coated onto the redox species area to detect Nor and …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Urinary tract infections (UTIs) are commonly found in elderlies, infants, hospitalized patients, and most women. UTIs are caused by a variety of pathogens with uropathogenic Escherichia coli (UPEC) is mostly prevalence. Even though UPEC can be treated with several antibiotics, the rapid emergence of multidrug resistant (MDR)-UPEC renders the antibiotic treatment no longer successful. Therapy using phages, which are bacterial viruses, has been considered but it is limited by bacterial resistance that develops during evolution. Fortunately, the suppression of phage resistance can be improved when used several phages as a cocktail. Here, I aim to formulate an efficient phage cocktail …

Metabolic Engineering Of Saccharomyces Cerevisiae For 3-Hydroxypropionic Acid Production, Supattra Lertsriwong
Chulalongkorn University Theses and Dissertations (Chula ETD)
3-Hydroxypropionic acid (3-HP) is an important building block chemical that can be produced from renewable resources through the malonyl-CoA pathway. This route is the best candidate for 3-HP biosynthesis through the expression of the malonyl-CoA reductase (MCR). However, the problem of the imbalance between MCR-C and MCR-N proteins causes a low conversion rate of malonyl-CoA to 3-HP. Therefore, this study aimed to balance the bi-functional MCR enzyme by dissecting MCR into two fragments and enhancing the supply of intermediates to increase the production of 3-HP. It was found that the recombinant harboring dissected MCR produced 1.26 g/L of 3-HP, which …

Macrophage-Cancer Cell Interaction In Three Dimension Liver Cancer Model, Pornlapat Keawvilai
Macrophage-Cancer Cell Interaction In Three Dimension Liver Cancer Model, Pornlapat Keawvilai
Chulalongkorn University Theses and Dissertations (Chula ETD)
Hepatocellular carcinoma (HCC) is the one of the leading causes of cancer-related mortality. Liver cancer progression is significantly influenced by immune cells which infiltrate tumors and their interaction with liver cancer cells in the tumor microenvironment (TME). Circulating monocytes are recruited to tumor site by TME and differentiated into tumor-associated macrophages (TAMs) which are roughly divided into pro-inflammatory macrophages (M1) or pro-tumoral macrophages (M2). In order to study the interaction between macrophages and liver cancer cells. In in vitro, an effective liver cancer model which closely mimicks this crosstalk is necessary. This study aimed to fabricate a three-dimension (3D) liver …

Production Of Vanillate From Lignin Derivatives Using Genetically Modifiedescherichia Coli, Jedsadakorn Ninrat
Chulalongkorn University Theses and Dissertations (Chula ETD)
Vanillic acid is an important compound used in food, beverage, fragrance, and pharmaceutical industries. Vanillic acid can be produced by using either natural plant, native, and engineered microorganisms. However, the industrial scale production of vanillic acid compound still difficult due to low yielding production which can be improved by metabolic engineering of bacterial cells. Hence, the efficient biocatalyst, engineering, and biotechnology process can be a promising strategy to enhance vanillic acid production from lignin derivatives and strengthen the large-scale production for industrial approach. Among several lignin-derived compounds, ferulic acid is a widely used substrate for vanillic acid production. Recently, the …

Screening Of Clostridium Sp. For Biohydrogen Production From Synthetic Wastewater, Chonticha Srimawong
Screening Of Clostridium Sp. For Biohydrogen Production From Synthetic Wastewater, Chonticha Srimawong
Chulalongkorn University Theses and Dissertations (Chula ETD)
In this work, bacteria that can produce endospores were isolated from natural sources and industrial wastes, including mangrove forests, hot springs, and coconut factories. The bacterial isolates were identified as Clostridium spp. and Panibacillus spp. based on 16S RNA gene. When comparing the H2 produced by batch fermentation, two of the isolates with the best H2 production were selected for further study, namely CUEA01 and CUEA03. After genomics analysis, it was discovered that CUEA01 is a novel species named C. hydrogenum, and CUEA03 is C. felsineum. Both are capable of growing and producing H2 in alkaline conditions. The genomics data …
Screening Of Clostridium Sp. For Butanol Production From Sato (Thai Rice Wine) Factory Wastewater, Julalak Buranaprasopchai
Screening Of Clostridium Sp. For Butanol Production From Sato (Thai Rice Wine) Factory Wastewater, Julalak Buranaprasopchai
Chulalongkorn University Theses and Dissertations (Chula ETD)
The production of Sato, a Thai traditional beverage, yields Sato wastewater (STW), which contains residual rice constituents with high chemical and biological oxygen demands and so forms an environmental problem. In addition, STW often smells rotten from the formation of hydrogen sulfide. To solve this problem, the composition of STW was analyzed for its physiological and chemical characteristics to evaluate its potential as a substrate for ABE fermentation using Clostridium spp., which has never been reported for use in acetone-butanol-ethanol (ABE) fermentation before. Solvent-producing Clostridium isolates were obtained from environmental sources in Thailand, and that producing the highest butanol level …
