Bex2 is well known for its role in the nervous system, and is associated with neurological disorders, but its role in the lung's physiology is still not reported. To elucidate the functional role of Bex2 in the lung, we generated a Bex2 knock-out (KO) mouse model using the CRISPR-Cas9 technology and performed transcriptomic analysis. A total of 652 genes were identified as differentially expressed between Bex2-/- and Bex2+/+ mice, out of which 500 were downregulated, while 152 were upregulated genes. Among these DEGs, Ucp1, Myh6, Coxa7a1, Myl3, Ryr2, RNaset2b, Npy, Enob1, Krt5, Myl2, Hba-a2, and Nrob2 are the most prominent …

In Silico Drug Repositioning Against Human Nrp1 To Block Sars-Cov-2 Host Entry, Şeref Gül Jan 2021

In Silico Drug Repositioning Against Human Nrp1 To Block Sars-Cov-2 Host Entry, Şeref Gül

Turkish Journal of Biology

Despite COVID-19 turned into a pandemic, no approved drug for the treatment or globally available vaccine is out yet. In such a global emergency, drug repurposing approach that bypasses a costly and long-time demanding drug discovery process is an effective way in search of finding drugs for the COVID-19 treatment. Recent studies showed that SARS-CoV-2 uses neuropilin-1 (NRP1) for host entry. Here we took advantage of structural information of the NRP1 in complex with C-terminal of spike (S) protein of SARSCoV- 2 to identify drugs that may inhibit NRP1 and S protein interaction. U.S. Food and Drug Administration (FDA) approved …

With the emergence of the new SARS-CoV-2 virus, drug repurposing studies have gained substantial importance. Combined with the efficacy of recent improvements in ligand- and target-based virtual screening approaches, virtual screening has become faster and more productive than ever. In the current study, an FDA library of approved drugs and compounds under clinical investigation were screened for their antiviral activity using the antiviral therapeutic activity binary QSAR model of the MetaCore/MetaDrug platform. Among 6733-compound collection, we found 370 compounds with a normalized therapeutic activity value greater than a cutoff of 0.75. Only these selected compounds were used for molecular docking …

Does Experimentally Quieting Traffic Noise Benefit People And Birds?, Mitchell J. Levenhagen, Christopher J.W. Mcclure, Jesse R. Barber Jan 2021

Does Experimentally Quieting Traffic Noise Benefit People And Birds?, Mitchell J. Levenhagen, Christopher J.W. Mcclure, Jesse R. Barber

Biology Faculty Publications and Presentations

Protected natural areas are not free from noise, especially noise generated by traffic within park boundaries. Natural soundscapes are important for maintaining community structure, providing positive visitor experiences, and increasing visitor support for management actions that reduce impacts on natural resources. To test experimental quieting as a strategy to increase both wildlife habitat quality and visitor experience, we enforced decreased speed limits and presented educational signage to reduce sound levels along a road system in an alternating, on–off block design within Grand Teton National Park, Wyoming, USA. We continuously recorded background sound levels while conducting bird space use assessments and …

Novel Role Of Endoplasmic Reticulum-Associated Degradation In The Regulation Of Ceruloplasmin, Stephen William Hippleheuser Jan 2021

Novel Role Of Endoplasmic Reticulum-Associated Degradation In The Regulation Of Ceruloplasmin, Stephen William Hippleheuser

Wayne State University Theses

Ceruloplasmin (Cp) is a secreted ferroxidase produced by the hepatocytes that assists in the transport of iron throughout the human body. In human aceruloplasminemia patients, due to Cp deficiency excess intracellular iron buildup leads to ailments like liver cirrhosis, neurodegeneration, and blindness. We recently found that the biogenesis of Cp in the endoplasmic reticulum (ER) is regulated by a principal ER quality-control process, ER-associated degradation (ERAD). ERAD clears misfolded ER proteins for cytosolic proteasomal degradation, with the Sel-1 suppressor of lin-12-like (Sel1L)-HMG-CoA reductase degradation 1 (Hrd1) protein complex representing the most conserved branch in mammals. Interestingly, we found that Sel1L-Hrd1 …

Bird Abundance And Diversity And The Impact Of Oyster Reef Restoration On The Bird Community In Mosquito Lagoon, Florida, Usa, Jessica Copertino

Electronic Theses and Dissertations, 2020-2023

Birds are often used as indicators for biodiversity and ecosystem health. While birds have been monitored in other parts of the Indian River Lagoon (IRL), there has been little research on birds in Mosquito Lagoon (ML, the northernmost portion of the IRL). This thesis first examines the abundance and diversity of birds in ML by using two years of photographic observations to assess seasonal variations and the use of various habitat features by specific bird taxa. Abundance and species richness were highest in winter, while evenness and Simpson's diversity were highest in summer. Moreover, natural and artificial habitat features were …

Non-Lethal Predictors Of Organ Level Metal Contaminants In Central Florida Red-Shouldered Hawks, Jennifer Bouchenot Jan 2021

Non-Lethal Predictors Of Organ Level Metal Contaminants In Central Florida Red-Shouldered Hawks, Jennifer Bouchenot

Electronic Theses and Dissertations, 2020-2023

Elemental contaminants aggregate in the environment due to human activity and can induce detrimental physiological effects within organisms. Birds exhibit exceptional susceptibility to many pollutants and are popular biomonitoring agents throughout the world. Birds of prey are of special interest due to their substantial biomagnification potential. Lethal sampling of internal organs yields accurate contaminant measurements, but recent sampling trends are endeavoring towards non-destructive sampling such as feathers. This methodological shift stems from researchers' preference for less-regulated samples, enhanced animal welfare, and broadening the possible collaborative audience with simpler protocols. Unfortunately, investigations of feathers have shown that their accuracy is capricious …

Maternal Cannabis Use Is Associated With Suppression Of Immune Gene Networks In Placenta And Increased Anxiety Phenotypes In Offspring, Gregory Rompala, Yoko Nomura, Yasmin L. Hurd Jan 2021

Maternal Cannabis Use Is Associated With Suppression Of Immune Gene Networks In Placenta And Increased Anxiety Phenotypes In Offspring, Gregory Rompala, Yoko Nomura, Yasmin L. Hurd

Publications and Research

While cannabis is among the most used recreational drugs during pregnancy, the impact of maternal cannabis use (mCB) on fetal and child development remains unclear. Here, we assessed the effects of mCB on psychosocial and physiological measures in young children along with the potential relevance of the in-utero environment reflected in the placental transcriptome. Children (~3-6 years) were assessed for hair hormone levels, neurobehavioral traits on the behavioral assessment system for children (BASC-2) survey, and heart rate variability (HRV) at rest and during auditory startle. For a subset of children with behavioral assessments, placental specimens collected at birth were processed …

More Than A Myth: The Benefits Of Incorporating Holistic Healing Methods From Native American And Mexican Cultures To Approach Opioid Use Disorder (Oud) Treatment, Juliana Favela Jan 2021

More Than A Myth: The Benefits Of Incorporating Holistic Healing Methods From Native American And Mexican Cultures To Approach Opioid Use Disorder (Oud) Treatment, Juliana Favela

CMC Senior Theses

Opioid Use Disorder (OUD) is a substance use disorder that characterizes a dependence on and an addiction to opioids. Opioids attack the prefrontal cortex, the dopaminergic reward system, and the hypothalamic-pituitary-adrenal (HPA) axis which is responsible for the body’s stress response. Opioids constantly shift one’s “set point” to the point where normal actions that cause joy and excitement no longer cause those feelings, resulting in craving and dependence. The opioid epidemic is currently ravaging the United States and has disproportionately affected Native American populations specifically in the rural areas (Tipps, et.al, 2018). The current methods for addressing OUD include medicine …

Assessing The Role Of Disturbance In Estuarine Fish Community Dynamics, Dakota Lewis Jan 2021

Assessing The Role Of Disturbance In Estuarine Fish Community Dynamics, Dakota Lewis

Electronic Theses and Dissertations, 2020-2023

Ecological disturbances alter biotic communities and ecosystems. In many coastal zones disturbances are increasing, including algal blooms, storms, hypoxic events, and fish kills. These disturbances are often related, for example blooms releasing toxins or depleting dissolved oxygen, ultimately killing fish. Depending on the intensity, duration, and geographic extent of a disturbance, the fish community can take days to years to recover from disturbances. To explore the relationships among environmental disturbances, sport fish, and forage fish communities, this thesis examined two Florida estuaries with differing disturbance regimes. Using an ensemble modelling approach combining generalized linear models (GLM), Bayesian modelling, and Bayesian …

Polycystic Kidney Disease: An Examination And Review Of Disease Type, Presentation, Treatment, And Prognosis, Zoey Mann

Biology and Microbiology Graduate Students Plan B Research Projects

Polycystic Kidney Disease (PKD) is a ciliopathy that primarily presents as renal cysts. Inherited as either a dominant (ADPKD) or recessive (ARPKD) mutation, PKD is one of the most commonly inherited kidney diseases. ADPKD is caused by the inheritance of a mutation in either PKD1 or PKD2, which code for the polycystin-1 and -2 proteins, respectively. The less severe form of PKD, ADPKD is typically adult-onset, with the possibility of extremely late-stage presentation. In addition to renal cysts, hepatic and pancreatic cysts are common, as well as other non-cystic symptoms including headache and hypertension. ARPKD is caused by the inheritance …

Guillain-Barre Syndrome: A Review Of Current Diagnostic Tools, Treatments, And Research, Kaitlyn Hespen

Biology and Microbiology Graduate Students Plan B Research Projects

Guillain-Barre Syndrome is a rare post-infection inflammatory disorder. It is caused by the cross-reactivity of antibodies produced from a recent infection which then attack the gangliosides of the peripheral nervous system. Guillain-Barre syndrome includes multiple subtypes which are categorized based on both disease course and symptoms, which include length of progression phase, pain, and cranial nerve involvement. This paper will serve as a review of diagnostic tools and treatment options used for Guillain-Barre syndrome patients. It will also discuss recent research on these topics as well as studies on viral outbreaks which may be linked to Guillain-Barre syndrome. These will …

Current And Future Perspectives In The Treatment Of Fracture Nonunions, Zebadiah Nelson Jan 2021

Current And Future Perspectives In The Treatment Of Fracture Nonunions, Zebadiah Nelson

Biology and Microbiology Graduate Students Plan B Research Projects

Fracture nonunions comprise one of the serious clinical complications in orthopedics. Fracture nonunions result when the time to union is greater than 6 to 9 months. It is estimated that approximately 5% to 10% of all fractures progress to fracture nonunion due to a host of complicating patient variables and risk factors. New scoring systems have emerged which help in directing the treatment strategies. Fracture nonunion management should be directed toward maintaining the diamond concept which introduces osteogenic cells lines, an osteoconductive scaffold for those cells to grow, as well as the necessary growth factors, and a stable mechanical microenvironment. …

The rise in multimorbidity among various patient populations has made fragmentation of care an increasingly common issue due to the need for specialized care of each chronic condition and complexity of treatment plans. Implementation of care coordination intervention models has been identified as an essential component in combating this fragmented nature of our current health care system. This is done by providing an organized model of care and improving communication between all parties involved in care and treatment plans. This literature review will begin with a brief discussion on the importance of care coordination and components needed for implementation and …

The Relationship Between Land Use Change And Disease Prevalence, Allison Bjornstad Jan 2021

The Relationship Between Land Use Change And Disease Prevalence, Allison Bjornstad

Biology and Microbiology Graduate Students Plan B Research Projects

Zoonotic disease outbreaks are becoming more novel and are increasing in frequency. This increase can be partially attributed to land use changes including deforestation, urbanization, and cultivation. Land use change alters disease prevalence by causing an increase in contact rates between disease hosts and humans. Land use changes also alter species compositions in the area to contain more competent disease hosts. Each pathogen can react to these changes in different ways. The specific reactions that each pathogen has to land use changes should be further researched so proper mitigation steps can be taken. Once the effects of land use change …

A Repeated Triple Lysine Motif Anchors Complexes Containing Bone Sialoprotein And The Type Xi Collagen A1 Chain Involved In Bone Mineralization, Jeff P. Gorski, Nichole T. Franz, Daniel Pernoud, Andrew Keightley, David R. Eyre, Julia Thom Oxford

Biology Faculty Publications and Presentations

While details remain unclear, initiation of woven bone mineralization is believed to be mediated by collagen and potentially nucleated by bone sialoprotein (BSP). Interestingly, our recent publication showed that BSP and type XI collagen form complexes in mineralizing osteoblastic cultures. To learn more, we examined the protein composition of extracellular sites of de novo hydroxyapatite deposition which were enriched in BSP and Col11a1 containing an alternatively spliced “6b” exonal sequence. An alternate splice variant “6a” sequence was not similarly co-localized. BSP and Col11a1 co-purify upon ion-exchange chromatography or immunoprecipitation. Binding of the Col11a1 “6b” exonal sequence to bone sialoprotein was …

Monitoring Tropical Forest Succession At Landscape Scales Despite Uncertainty In Landsat Time Series, T. Trevor Caughlin, Cristina Barber, Gregory P. Asner, Nancy F. Glenn, Stephanie A. Bohlaman, Chris H. Wilson

Biology Faculty Publications and Presentations

Forecasting rates of forest succession at landscape scales will aid global efforts to restore tree cover to millions of hectares of degraded land. While optical satellite remote sensing can detect regional land cover change, quantifying forest structural change is challenging. We developed a state-space modeling framework that applies Landsat satellite data to estimate variability in rates of natural regeneration between sites in a tropical landscape. Our models work by disentangling measurement error in Landsat-derived spectral reflectance from process error related to successional variability. We applied our modeling framework to rank rates of forest succession between 10 naturally regenerating sites in Southwestern …

What Is The Underlying Cause Of Infantile Colic?, Tamar Itzkowitz Jan 2021

What Is The Underlying Cause Of Infantile Colic?, Tamar Itzkowitz

The Science Journal of the Lander College of Arts and Sciences

Infantile colic (IC) is an important area of current research due to the extreme distress it causes parents and their infants. It is vital that a cause is isolated so that treatment can be found because IC is a risk factor for child abuse. In this paper, two major theories were posed to elucidate the underlying cause of IC—the gastrointestinal model and the neurological model. The gastrointestinal model suggests that IC stems from issues such as an immature gut. The neurological model suggests that infantile migraines are the causative agents of IC. Both theories supply correlational evidence but are subject …

The Riddle Of The Fetal Allograft, Rachel Tepper Jan 2021

The Riddle Of The Fetal Allograft, Rachel Tepper

The Science Journal of the Lander College of Arts and Sciences

The immunological paradox of nurturing a fetus with paternal antigens poses some perplexing questions. Peter Medawar, an immunologist, asked at a lecture, “How does the pregnant mother contrive to nourish within itself, for many weeks or months, a fetus that is an antigenically foreign body?” Researchers have since then struggled to answer this question. The research on this topic has led to a few general hypotheses that try to explain this phenomenon. The downregulation of T cells toward paternal alloantigens is an accepted hypothesis. Another hypothesis discusses the significance of the decidua and its ability to impair dendritic cells, which …

Do People With Hashimoto’S Disease Need A Thyroidectomy?, Miriam Minkowitz Jan 2021

Do People With Hashimoto’S Disease Need A Thyroidectomy?, Miriam Minkowitz

The Science Journal of the Lander College of Arts and Sciences

Hashimoto’s Thyroiditis (HT) is one of the most common autoimmune diseases (Hiromatsu et al. 2013 p.13). It primarily affects the thyroid gland. The thyroid gland influences growth and regulates the body’s metabolism by manipulating hormonal levels. Hashimoto’s thyroiditis affects thyroid function through mechanisms that cause the hormone levels to become dysregulated. The standard therapy for Hashimoto’s thyroiditis is hormone replacement. This approach helps most patients by regulating their thyroid hormones, though there are some individuals who fail treatment. Untreatable patients are plagued with weight gain, sleep disturbances, and other symptoms. Certain factors in thyroid disease may cause various secondary complications …

Response Of Fish And Invertebrate Larvae To Backreef Sounds At Varying Distances: Implications For Habitat Restoration, Emily R. Anderson, Jack Butler, Mark J. Butler Jan 2021

Response Of Fish And Invertebrate Larvae To Backreef Sounds At Varying Distances: Implications For Habitat Restoration, Emily R. Anderson, Jack Butler, Mark J. Butler

Biological Sciences Faculty Publications

Underwater sound is used by many marine larvae to orient to coastal habitats including backreef, sponge-dominated hardbottom habitat in the Florida Keys (FL, United States)—a particularly “noisy” coastal habitat. However, the distance over which acoustic cues are attractive to settlement-stage larvae is generally unknown. We examined this phenomenon in a region of the Florida Keys where mass sponge die-offs have diminished both underwater soundscapes and larval settlement. The absence of pronounced hardbottom-associated sound over such a large area allowed us to experimentally test in situ the response of fish and invertebrate larvae to broadcasted sounds at different distances from their …

Mitchell Iii, Daniel E. Sonenshine, Charles S. Apperson, Richard Michael Roe

Biological Sciences Faculty Publications

The transovarial transmission of tick-borne bacterial pathogens is an important mechanism for their maintenance in natural populations and transmission, causing disease in humans and animals. The mechanism for this transmission and the possible role of tick hormones facilitating this process have never been studied. Injections of physiological levels of the tick hormone, 20-hydroxyecdysone (20E), into part-fed (virgin) adult females of the American dog tick, Dermacentor variabilis, attached to the host caused a reduction in density of Rickettsia montanensis in the carcass and an increase in the ovaries compared to buffer-injected controls. This injection initiates yolk protein synthesis and uptake …
