Turkish Journal of Biology
The architecture of testicular lobules, morphology of spermatozoa, and spermatophore in the mud crab, Scylla serrata, are described for the first time employing atomic force microscopy (AFM). A fast and simple squash preparation protocol was followed using crustacean physiological saline without distorting the natural structure. Observations provided new insight into the arrangement of lobules and morphology of testicular assembly. It consists of a close network of lobules of different sizes in a radial manner on radially branching basal tubules. Budding of new lobules is another distinguishable feature noted in the network of testicular organization. A thin membranous envelope of the …

Hepatopancreas Mitochondria Of Mytilus Galloprovincialis: Effect Of Zinc Ions On Mitochondrial Bioenergetics And Metabolism, Antonino De Lisi, Ermelinda Prato, Francesca Biandolino, Giulio Sarli, Donatella Negro, Gianluigi La Piana, Domenico Marzulli
Turkish Journal of Biology
Oxygen uptake, respiratory complexes, and metabolic activities have been studied in mitochondria isolated from the hepatopancreata of Mytilus galloprovincialis collected in the Mar Grande of Taranto (Ionian Sea, Italy). Although exposure to 5.0 µg Zn^{2+}/L resulted in a significant increase of states 3 and 4 respiration with glutamate + pyruvate as respiratory substrate, it was found that the exposure of mussels to different concentrations of Zn^{2+} (2.57.5 µg/L) neither inhibited mitochondrial respiration nor exerted any inhibitory effect on representative mitochondrial dehydrogenases. It rather stimulates these activities, producing an extra synthesis of adenosine triphosphate by hepatopancreas mitochondria and possibly increasing its …

Computational Design Of A Pentapeptide Inhibitor For Fibroblast Growth Factor Receptor 3b (Fgfr3b), Mehmet Ali̇ Öztürk
Turkish Journal of Biology
Fibroblast growth factor receptor (FGFR) is a cell membrane protein and a member of the tyrosine kinase family. It has extracellular domains that can be activated by ligand binding, followed by receptor dimerization. FGFR3 has 2 isoforms, 3b and 3c. The R248C mutation in FGFR3b leads to ligand independent receptor dimerization and results in different kinds of dermatological diseases such as seborrheic keratoses, acanthosis nigricans, and epidermal nevi. In order to prevent the increased cellular signaling caused by the R248C mutation, a pentapeptide ligand was designed that recognizes the mutation and binds to the receptor dimerization site. Molecular docking and …

Cytotoxic Activities Of Some Pseudomonas Aeruginosa Isolates: Possible Mechanisms And Approaches For Inhibition, Ghadir Saeed El-Housseiny, Mohammad Mabrouk Aboulwafa, Nadia Abdelhaleim Hassouna
Turkish Journal of Biology
The cytotoxicity of 6 highly cytotoxic Pseudomonas aeruginosa isolates was tested against Vero cells. Cytotoxicity was found to be cell-associated, high cytotoxicity was observed at a multiplicity of infection of 200:1, and this activity gradually decreased as the bacterial count decreased. This cytotoxicity caused rounding and detachment of the Vero monolayer as early as 1 h after infection. Moreover, viable metabolically active bacteria were necessary for Vero cell killing. These results imply that one or more of the type III secretion system effector proteins (ExoU, ExoS, ExoT, or ExoY) were responsible for the observed cytotoxic effects. This cytotoxicity was affected …

Concentration Of Sodium Dodecyl Sulfate Used In Occlusion Body Extraction Affects Spodoptera Littoralis Nucleopolyhedrovirus Biological Activity, Hi̇lal Susurluk, Umut Toprak, Mehmet Oktay Gürkan
Turkish Journal of Biology
Efficacy of a baculovirus as a biological control agent is evaluated according to biological parameters, median lethal dose (LD_{50}) of the virus, and median lethal time (LT_{50}) of infected larvae. Sodium dodecyl sulfate (SDS) is commonly used in concentrations of 0.1%-1% to extract infective units of the baculoviruses, occlusion bodies (OBs), from infected cadavers. Although several studies have shown disruption of OB structure by SDS, the effects of SDS concentrations on baculovirus biological parameters are not well known. Indeed, it is essential to detect the optimum SDS concentration for OB extraction to compare the activity of baculoviruses accurately. In our …

In this study, the genetic structures and relationships of native Turkish dog breeds were investigated using 20 polymorphic loci (17 microsatellites and 3 proteins). For this aim, a total of 141 blood samples were taken from Turkish shepherd dogs and Turkish Greyhounds located in several geographical regions of Turkey. Multilocus F_{ST} values indicated that around 1.92% of the total genetic variation could be explained by breed differences and the remaining 98.08% by differences among individuals. The gene flow between populations within each generation varied between 8.4 (Akbash-White Kars Shepherd dog pairs) and 62.3 (Black-Grey Kars Shepherd dog pairs). Four different …
