Kremen Is Beyond A Subsidiary Co-Receptor Of Wnt Signaling: An In Silico Validation, Hemn Mohammadpour, Saeed Khalili, Zahra Sadat Hashemi

Turkish Journal of Biology

Kremen-1 is a co-receptor of Wnt signaling, interacting with the Dkk-3 protein, which is an antagonist of the Wnt/ß catenin pathway. In the present study we attempted to shed some light on the possible orientations of Kremen/Dkk-3 interactions, explaining the mechanisms of Kremen and Dkk-3 functions. Employing state-of-the-art software, a Kremen model was built and subsequently refined. The quality of the final model was evaluated using RMSD calculations. Ultimately, we used the Kremen model for docking analysis between Kremen and Dkk-3 molecules. A model built by the Robetta server showed the best quality scores. Near native coordination of the final …

Turkish Journal of Biology

The light phase of photosynthesis is considered a joint operation of 2 functional pigment-protein complexes: a light harvesting antenna, absorbing sunlight in a wide spectral range, and a reaction center, utilizing the energy of absorbed light quanta in photochemical charge separation reactions. These complexes allow converting solar energy to the energy of specific biomolecules with high quantum efficiency. However, before being transferred to reaction centers, the solar energy is stored in the lowest excited state of pigment molecules of the light harvesting antenna that partially convert light quantum energy into heat. These energy losses bring a significant reduction of energy …

Turkish Journal of Biology

Our earlier studies found that in vitro estradiol modulates mitochondrial Ca2+ transport in discrete brain regions. The present study examined the role of estradiol receptors (ERs) in estradiol-induced inhibition of Ca^{2+} efflux from synaptosomal mitochondria isolated from rat caudate nuclei and brain stems. Radioactively labeled CaCl_2 (0.6?0.75 µCi ^45CaCl_{2}) was used for Ca^{2+} transport monitoring. The results revealed that in the presence of ER antagonist 7\alpha,17ß-[9[(4,4,5,5,5-pentafluoropentyl)sulfinyl]nonyl]estra-1,3,5(10)-triene-3,17-diol (ICI 182,780) (1 µmol/L), the inhibitory effect of estradiol on mitochondrial Ca^{2+} efflux was more than 60% decreased, suggesting the involvement of ER in this mode of estradiol neuromodulatory action. When particular contributions of …

Turkish Journal of Biology

This letter to the editor highlights the important aspects of cancer stem cells and provides a deeper insight into the origin and characterization of these cells as well as the mechanisms contributing to drug resistance and relapse. The two currently available models (CSC and the clonal evolution model) need not be mutually exclusive to explain the origin of CSCs. The identification of CSCs (based on surface markers, drug efflux, enzyme activity, and signal transduction molecules and proteins involved in EMT processes) can help in discriminating these cells from other cell types. Moreover, CSCs prefer a hypoxic environment. This microenvironmental cue …

The affinity of ß-lactamase inhibitory protein (BLIP) for TEM-1 ß-lactamase has raised hopes in the challenge of protein-based inhibitor discovery for ß-lactamase-mediated antibiotic resistance. Currently, the effect of the formation of the ß-lactamase:BLIP complex in vivo in ß-lactam resistant bacteria is an open question. The scarcity of information to the extent to which BLIP can impair ß-lactamase activity inside cells has urged us to assess the in vivo efficacy of BLIP as a potent ß-lactamase inhibitor. To this end, ß-lactamase and BLIP were coexpressed in Escherichia coli. Simultaneous expression of ß-lactamase and BLIP and the formation of the TEM-1 ß-lactamase:BLIP …
