Resolution Of Inflammation Rescues Axon Initial Segment Disruption, Nicholas M. George
Resolution Of Inflammation Rescues Axon Initial Segment Disruption, Nicholas M. George
Theses and Dissertations
Axonal domains are required for proper neuron function. These domains are unstable and degenerate concurrent with the inflammation in multiple sclerosis (MS) and the inflammatory disease models experimental autoimmune encephalomyelitis (EAE) and lipopolysaccharide (LPS) induced inflammation. Previous studies from our laboratory have shown that the axon initial segment (AIS) is maintained independently of the presence of myelin, but that AIS disruption is seen in MS as well as EAE and LPS-mediated inflammation. AIS loss can be interrupted in the early stage of EAE using the anti-inflammatory drug Didox. However, the potential for Didox directed repair of the AIS in later …
The Curiosity Of Con, Petrified Breath, And An Accident Known As Blue., Steven Randall
The Curiosity Of Con, Petrified Breath, And An Accident Known As Blue., Steven Randall
Theses and Dissertations
My thesis installation emerged from an interest in visualizing breath. The resulting work came to exist at the intersection between art, biology, and performance.
The unicorn tapestries were used as a generative point of departure to explore the preservation and transformation of images through time, by time, and with time. Reproductions of the six tapestries were each etched into paper and then submerged into solutions of Phenol Red dye, Ferric Ferrocyanide (also known as Prussian Blue), and various forms of sodium chloride. Exhaled breath was used to encrust these images of the tapestries into physical objects which gradually crystallized and …
Dietary Adaptations And Intra- And Interspecific Variation In Dental Occlusal Shape In Hominin And Non-Hominin Primates, Kelsey O'Neill
Dietary Adaptations And Intra- And Interspecific Variation In Dental Occlusal Shape In Hominin And Non-Hominin Primates, Kelsey O'Neill
Theses and Dissertations
Dental morphology and tooth shape have been used to recreate the
dietary adaptations for extinct species, and thus dental variation can provide
information on the relationship between fossil species and their
paleoenvironments. Variation in living species with known behaviors can provide
a baseline for interpreting morphology, and behavior, in the fossil record.
Tooth occlusal surface outlines in hominins and non-hominin primates, and other
mammals, have been used for assessments of taxonomic significance, with
variability often considered as being primarily phylogenetic. Few studies have
attempted to assess how diet might influence the pattern of variability in closely
related species. Here the …
Regulation Of Stress Response And Innate Immunity By DsRna-Binding Proteins Pact And Trbp, Lauren S. Vaughn
Regulation Of Stress Response And Innate Immunity By DsRna-Binding Proteins Pact And Trbp, Lauren S. Vaughn
Theses and Dissertations
An integral aspect of innate immune response to viral infections is the ability to detect non-self molecules to initiate antiviral signaling via pattern recognition receptors (PRRs). One subset of these receptors are cytoplasmic receptors that contain double stranded (dsRNA) binding domains, which allow them to identify non-self dsRNA produced during a viral infection and mount a protective cellular response. PKR is a dsRNA-activated eIF2α kinase that is a key regulator of cellular antiviral and stress response pathways. Activation of PKR’s catalytic activity requires binding to one of its activators, viral dsRNAs or the cellular protein PACT (PKR activator). Although PACT …
Establishing The Microcrustacean Daphnia As A Model System For Research On Aging, Charles Andrew Shumpert
Establishing The Microcrustacean Daphnia As A Model System For Research On Aging, Charles Andrew Shumpert
Theses and Dissertations
Aging is a ubiquitous process pertaining to all biological systems around the planet. Although much has been learned from studies so far on the molecular mechanisms that lead to aging, a complete understanding of a healthy life span and longevity still eludes us. In this dissertation, we will examine the use of a freshwater microcrustacean Daphnia as a model system for studies on the biology of aging. The Introduction chapter presents a review of the general molecular alterations associated with cellular and organismal aging, and discusses the core model organisms currently used to study the aging process. The introduction chapter …
The Effects Of Elevated Circulating Levels Of Lipocalin 2 On Liver Metastasis Of Colorectal Cancer, John Gerard Bonapart
The Effects Of Elevated Circulating Levels Of Lipocalin 2 On Liver Metastasis Of Colorectal Cancer, John Gerard Bonapart
Theses and Dissertations
In the United States, colorectal cancer (CRC) is the third most diagnosed cancer and the second leading cause of cancer deaths. In 2015, approximately 143,000 new cases will be diagnosed and 50,000 deaths are predicted. Liver metastasis is the major cause of death. To understand the molecular and cellular mechanisms of liver metastasis, we used an orthotopic mouse model of CRC liver metastasis that recapitulated all stages of tumor growth and metastasis to perform whole genome microarray analysis to identify genetic changes in the liver microenvironment in response to primary tumor growth in the cecum. We identified lipocalin 2 (Lcn2) …
Vulnerable Species In A Changing Climate: The Genomic Response Of Antarctic Notothenioid Fishes To Predicted Oceanic Conditions As A Model Of Physiological Plasticity And Adaptive Capability, Troy James Huth
Theses and Dissertations
In its fifth report in 2014 the IPCC reinforced the contribution of anthropogenic CO2 to global climate change predicting widespread and significant changes to the global climate over a relatively short time scale. The polar regions, including the Southern Ocean surrounding Antarctica, were identified as ecosystems that may experience the most rapid and severe changes. As the Southern Ocean is one of the coldest and most oceanographically stable regions on earth, the endemic fauna likely have no alternative habitats available for migration. Further compounding the challenge these species will face is the substantial degree of adaptation to the extreme cold …

Effects Of Shrub Encroachment And Removal On South Texas Coastal Prairie Flora, Parker Alex Watson
Theses and Dissertations
Watson, Parker Alex, Effects of shrub encroachment and removal on South Texas coastal prairie flora. Master of Science, December, 2015. 60 pages, 14 tables, 11 figures, 47 references, 5 appendices. Native-invasive honey mesquite (Prosopis glandulosa) and huisache (Acacia farnesiana) are aggressively encroaching onto South Texas coastal prairies, outcompeting Gulf cordgrass (Spartina spartinae) and associated prairie vegetation with potential legacy effects on the landscape. To measure conditions in the presence of shrubs, data loggers were installed for 16 mos in shrub understories and grass cover to monitor light, soil and air temperatures. Vegetation and canopy surveys were also conducted to evaluate …
Water Use Of Four Commonly Planted Landscape Tree Species In A Semi-Arid Suburban Environment, Michael Cameron Bunnell
Water Use Of Four Commonly Planted Landscape Tree Species In A Semi-Arid Suburban Environment, Michael Cameron Bunnell
Theses and Dissertations
Native plant communities and agricultural land are commonly converted to urban areas as cities across the Western United States continue to grow and expand. This expansion is typically accompanied by afforestation where a common goal among communities is to maximize shade tree composition. Planted forests in these regions are commonly composed of introduced tree species native to mesic environments and their ability to persist is dependent on consistent irrigation inputs. Many potential ecosystem services may be derived from planting trees in urban and suburban areas; however, there are also costs associated with extensive afforestation, and shade tree cover may have …
Virulence Of Photorhabdus Spp.: Examining The Roles Of Environment, Evolution, And Genetics In Insect Mortality, Dana Blackburn
Virulence Of Photorhabdus Spp.: Examining The Roles Of Environment, Evolution, And Genetics In Insect Mortality, Dana Blackburn
Theses and Dissertations
Entomopathogenic nematodes (EPNs) (genera Heterorhabditis and Steinernema) kill their invertebrate hosts with the aid of a mutualistic bacterium. The bacteria (Xenorhabdus spp. for steinernematids and Photorhabdus spp. for heterorhabditids) are primarily responsible for killing the host and providing the nematodes with nutrition and defense against secondary invaders. Photorhabdus is a Gram-negative bacterium in the Enterobacteriaceae family with high virulence towards their insect hosts. To achieve high mortality rates Photorhabdus produces a variety of virulence factors such as toxins, lipases, proteases, secretion systems, and fimbriae. EPNs are amenable to laboratory rearing and mass production for biocontrol applications against insects …
Validating Side Scan Sonar As A Fish Survey Tool, Michael A. Bollinger
Validating Side Scan Sonar As A Fish Survey Tool, Michael A. Bollinger
Theses and Dissertations
Hydroacoustic methods can be used to answer a variety of questions regarding fish populations and behavior. In this study, side scan sonar methodology was developed to quantify abundance and biomass and compared to established visual observation methods on SCUBA over artificial reef structures in the western Gulf of Mexico. Side scan sonar methods were equivalent to SCUBA surveys for measuring fish abundance over the same reef areas, however, abundances were significantly higher when the larger area sampled by side scan was utilized. Side scan sonar methods were also more time efficient than SCUBA, ROV and long line fishing methods (66.7%, …
Examination Of The Chagas Disease Vector, Triatoma Gerstackeri, In The Lower Rio Grande Valley, Alfredo Flores
Examination Of The Chagas Disease Vector, Triatoma Gerstackeri, In The Lower Rio Grande Valley, Alfredo Flores
Theses and Dissertations
Chagas disease is caused by the protozoan parasite Trypanosoma cruzi, which is carried by an insect vector of the family Reduviidae, and of the subfamily Triatomina. The species Triatoma gerstaeckeri is identified as the primary vector of T. cruzi within the Rio Grande Valley. Species of Triatoma gerstaeckeri were trapped in La Sal del Rey National Refuge in 2012 and 2013.
Data indicates a seasonal dispersal pattern of activity for T. gerstaeckeri. Optimal time of capture was between 1-2 hours after sunset. Different methods were utilized to determine the best combination of lures. Results indicate that the UV-light alone is …
Decomposition And Nitrogen Dynamics Of Turtle Grass (Thalassia Testudinum) In A Subtropical Estuarine System, Monica Delgado
Decomposition And Nitrogen Dynamics Of Turtle Grass (Thalassia Testudinum) In A Subtropical Estuarine System, Monica Delgado
Theses and Dissertations
Seagrass beds are pivotal in the functioning of coastal ecosystems in terms of productivity, organic matter turnover and nutrient cycling. Aiming to document decay and nitrogen (N) dynamics of turtle grass (Thalassia testudinum) in a subtropical estuarine system, decomposition patterns of leaves and rhizomes were characterized and compared. Nitrogen usage during decomposition of tissues, and of live tissues and epiphytes growing on live leaves, was also quantified and compared. Stable isotope ratios allowed tracing N within the seagrass bed, following N incorporation into seagrass tissues from the surrounding media (water, sediment). Leaves had a higher N content and decomposed at …
Activation Of Microglia In The Rotenone Lymnaea Stagnalis Model Of Parkinson's Disease, Xiomara Galvan
Activation Of Microglia In The Rotenone Lymnaea Stagnalis Model Of Parkinson's Disease, Xiomara Galvan
Theses and Dissertations
Parkinson’s disease (PD) is the most common irreversible neurodegenerative movement disorder affecting approximately ten million people worldwide. In the central nervous system (CNS), PD is known by the selective loss of Dopamine (DA) cells of the substantia nigra pars compacta. Although the disease has been very widely studied, the underlying mechanisms are not yet understood. Several hypotheses have arisen as result of injury of the mitochondria, some experiments include microglia activation, microtubule damage, and reactive oxidative species, which all result in the death of DA cells. The theory of microglia activation upon inflammation was explored in this study. Using the …

Diversification And Speciation Across Sundaland And The Philippines: The Effects Of 30 Million Years Of Eustatic Flux, Luke J. Welton
Theses and Dissertations
I highlight two squamate lineages endemic to Southeast Asia, the Asian water monitors (Varanus salvator Complex) and the Angle-headed lizards (Agamidae: Gonocephalus), and elucidate their systematic affinities and historical biogeography. My results represent novel phylogenetic inferences, with biogeographic histories and diversification events corresponding to major climatic fluctuations over the past 30 million years. Additionally, I solidify the taxonomy and systematics of the Varanus salvator Complex, and enumerate more than a dozen Gonocephalus lineages as candidate species in need of taxonomic scrutiny. Lastly, I investigate contemporary and historical patterns of dispersal throughout Sundaland, and between Sundaland and the Philippines.

Comparing Reproductive Capacity Of Nearshore And Offshore Red Snapper, Lutjanus Campechanus, On Artificial Reefs In The Western Gulf Of Mexico, Ricky J. Alexander
Theses and Dissertations
Red snapper collected quarterly from four artificial reefs in south Texas were examined to compare total length, total weight, age, and the proportion of male to female red snapper, calculate gonadosomatic index values, batch fecundity, annual spawning frequency, and annual fecundity, and estimate size and age at maturity. Spawning occurred from April to September, with most active spawning observed in June (58%). Despite larger red snapper producing more eggs, and fish being larger offshore, most active spawning (68%) and egg production (74%) was observed nearshore. Female red snapper reached 50% maturity (L50) between 350-550 mm, and L75 by 600 mm. …
The Effects Of Modafinil And Atomoxetine On Dopamine Signaling In The Striatal Subregions Of The Rat, Martin Bobak
The Effects Of Modafinil And Atomoxetine On Dopamine Signaling In The Striatal Subregions Of The Rat, Martin Bobak
Theses and Dissertations
In an effort to combat diseases and disorders that impede our health, comfort and well-being, an abundance of prescription drugs have emerged in the past 60 years. Many prescription drugs have remarkable efficacy for treating the primary symptoms of these diseases and disorders; however, some drugs carry negative side effects that impose their own adverse symptoms, albeit, often to a lesser degree than the primary symptoms. Thus, one of the main objectives of the pharmaceutical industry is to innovate and develop novel therapeutics, which remediate the primary symptoms of disease and lack undesirable negative side effects. However, in order to …
Repeated Trait Evolution Driven By Divergent Natural Selection At Early And Late Stages Of Speciation, Spencer J. Ingley
Repeated Trait Evolution Driven By Divergent Natural Selection At Early And Late Stages Of Speciation, Spencer J. Ingley
Theses and Dissertations
Speciation – the process by which new species arise – is of fundamental importance in the biological sciences. The means by which new species arise, and the relationship among living species, has been a topic that has captivated both lay and scientific observers for centuries. In recent years, the study of speciation has enjoyed increased attention, resulting in significant advances in our understanding of how species form. Although our understanding of the processes that contribute to speciation has increased dramatically in recent years, our knowledge of how reproductive barriers accumulate as speciation proceeds is still limited. Thus, studies that evaluate …

Life History, Oviposition Characteristics, And Management Of The Ozark Baltimore Checkerspot (Euphydryas Phaeton Ozarkae), Stephen Robertson
Theses and Dissertations
The Ozark Baltimore Checkerspot (Euphydryas phaeton ozarkae Masters) is an understudied nymphalid butterfly endemic to the Ozarks that specializes on False Foxglove (Aureolaria sp.) as the larval food plant. Few studies have focused on this animal and much of what is known has been derived from studies of the eastern subspecies (Euhpydryas phaeton phaeton Drury). Genetic evidence shows only slight variation between subspecies, but critical ecological differences are found between the two taxa. Here I present an in-depth life history with respect to general ecology and offer evidence towards species identity. Oviposition behavior (search and egg-site selection) is a key …

Bacterially-Based Immune Challenges And Trauma Elicit Terminal Investment In Male Aedes Aegypti, Molly K. Schumacher
Bacterially-Based Immune Challenges And Trauma Elicit Terminal Investment In Male Aedes Aegypti, Molly K. Schumacher
Theses and Dissertations
Investment in life history traits such as immune function and reproduction is constrained by finite available resources. A cost-of-immunity trade-off may occur in response to infection when resources are diverted away from reproductive effort and into an immune response. Alternatively, an infected individual may enhance reproductive effort to maximize terminal reproductive success in response to the survival threat inherent to infection (terminal investment). We measured male Aedes aegypti reproductive behavior following inoculations with: living bacteria; killed bacteria as an immune elicitor; and a sham control. Mating competitiveness relative to naïve males was also determined through a binary mate choice experiment …
Delimiting Species And Varieties Of Cycladenia Humilis (Apocynaceae), Holly Kathryn Brabazon
Delimiting Species And Varieties Of Cycladenia Humilis (Apocynaceae), Holly Kathryn Brabazon
Theses and Dissertations
Taxonomic delimitation of rare species is vital for accurate assessments of diversity and for their conservation. Cycladenia humilis, the sole species of Cycladenia, is an enigmatic perennial widely dispersed across the western United States. Within this species there are three currently recognized varieties: C. humilis var. humilis in Northern California, C. humilis var. venusta in Southern California, and C. humilis var. jonesii in Utah and Northern Arizona. Some populations occur geographically in areas between the typical distribution of each variety and the presently accepted taxonomy inadequately addresses these populations. Using five nDNA regions, we seek to clarify relationships between current …

Theses and Dissertations
Dictyostelium discoideum is a primitive eukaryote that has recognized useful in studying various aspects of lysosomal enzymes as well as development since it can differentiate from unicellular amoebae into a multicellular. In this study, we investigated the localization, ecological and molecular function of two lysosomal enzymes, hexosaminidase (NagA) and,,-glucosidase (GluA) proteins, by overexpressing the genes encoding them in D. discoideum. Separate cell-lines overexpressing a GFP-tagged versions of the full length DdNagA gene (GFP-NagA), mutant version of the NagA gene (GFP-NagA (N214S)), a GFP-tagged version of the full length DdGluA gene (GFP-GluA) and a mutant version of first 83 amino acids DdGluA …
Factors Constraining The Reproductive Output Of Baptisia Alba Macrophylla, Ashley Morgan Hembrough
Factors Constraining The Reproductive Output Of Baptisia Alba Macrophylla, Ashley Morgan Hembrough
Theses and Dissertations
Baptisia alba macrophylla, a native, herbaceous perennial legume, produces inflorescences with a large number of flowers, yet matures relatively few fruits. We hypothesized that the number of seeds matured by B. alba macrophylla is maximized based on resource availability at each stage of development, but this optimal number is further compromised by extrinsic factors including pollen limitation and pre-dispersal seed predation by the weevils Apion rostrum and Tychius sordidus. To test this hypothesis, we conducted a field experiment at two sites: (i) the John English Prairie, located in Hudson, IL, and also (ii) the Sugar Grove Nature Center and Funks …

ANALYSIS OF PLANT LIGNIN FORMATION AND ENGINEERING FOR BIOFUELS PRODUCTION
Deborah L. Petrik
149 Pages August 2015
Grasses such as switchgrass and Miscanthus are attractive feedstock candidates for second generation biofuels because they are perennials that can grow on less fertile or "marginal" lands not used for food crops, require little fertilizer and modest pest and disease management, and are not a human food source. Releasing the energy potential contained in plant-based cellulosic material requires either conversion to a liquid biofuel such as ethanol or burning. Thus, much of the current research focuses on not only understanding grass development as …
Bioinformatics For The Comparative Genomic Analysis Of The Cotton (Gossypium) Polyploid Complex, Justin Thomas Page
Bioinformatics For The Comparative Genomic Analysis Of The Cotton (Gossypium) Polyploid Complex, Justin Thomas Page
Theses and Dissertations
Understanding the composition, evolution, and function of the cotton (Gossypium) genome is complicated by the joint presence of two genomes in its nucleus (AT and DT genomes). Specifically, read-mapping (a fundamental part of next-generation sequence analysis) cannot adequately differentiate reads as belonging to one genome or the other. These two genomes were derived from progenitor A-genome and D-genome diploids involved in ancestral allopolyploidization. To better understand the allopolyploid genome, we developed PolyCat to categorize reads according to their genome of origin based on homoeo-SNPs that differentiate the two genomes. We re-sequenced the genomes of extant diploid relatives of tetraploid cotton …
Environmental And Adaptive Buffers That Mediate The Response Of Subalpine Ecosystems To Environmental Change, Lafe G. Conner
Environmental And Adaptive Buffers That Mediate The Response Of Subalpine Ecosystems To Environmental Change, Lafe G. Conner
Theses and Dissertations
This document reports the results of 4 studies of subalpine ecosystem ecology, describing ways that spatial heterogeneity in soils and plant communities mediate ecosystem responses to environmental change. Ecosystem responses to environmental change are also mediated by regional climate patterns and interannual variability in weather. In the first chapter we report the results of an experiment to test for the mediating effects of associational resistance in a forest community that experienced wide-spread beetle kill. We found that Engelmann spruce were more likely to survive a beetle outbreak when growing in low densities (host dilution) and not through other types of …
Switching Tactics: Phenotypic Plasticity In The Alternative Mate-Finding Tactics Of Burying Beetles, Theresa Mulrey
Switching Tactics: Phenotypic Plasticity In The Alternative Mate-Finding Tactics Of Burying Beetles, Theresa Mulrey
Theses and Dissertations
Male Nicrophorus burying beetles utilize two alternative mate-finding tactics, representing an ideal model system to test the influence of environmental cues on the expression of alternative tactics. The "searching" tactic involves flying in search of a carcass on which to mate. This tactic can result in high levels of paternity, but is risky in that carcasses are rare and competition for carcasses is fierce. The "signaling" tactic, which involves emitting pheromone in the absence of a carcass to attract females, is energetically less costly, but a signalling male must mate with multiple females to achieve the same reproductive returns as …
Investigations Into The Genomes Of Two Ascomyetes, Jay Wilbur Pyle
Investigations Into The Genomes Of Two Ascomyetes, Jay Wilbur Pyle
Theses and Dissertations
Fusarium verticillioides is a filamentous ascomycete that is both a plant endophyte and pathogen, causing disease during any life stage of the plant. When F. verticillioides grows in maize the fungus can synthesis a number of mycotoxins including the fumonisins, which have been linked with human esophageal cancer and neural tube associated birth defects. In an attempt to control fumonisin production our lab is searching the genome of F. verticillioides for a selfish genetic element known as Spore killer. The plan is simple; we envision creating a bio-control strain capable of harnessing the genetic transmission-distorting properties of Spore killer to …
