Screen For Suppressors And Enhancers Of Excitotoxic Neurodegeneration, Anthony Omorodion Edokpolo
Screen For Suppressors And Enhancers Of Excitotoxic Neurodegeneration, Anthony Omorodion Edokpolo
Dissertations and Theses
Excitotoxicity is an important and frequently observed neurodegenerative process. Excitotoxicity mediates brain damage in a range of diseases and conditions including stroke, and is triggered by excessive stimulation of glutamatergic synapses. In spite of extensive studies, the molecular mechanisms involved in excitotoxicity following the over-activation of postsynaptic glutamate receptors are not well understood, and clinical trials based on our partial understanding of the process ended with disappointment. Genetic screens in simple animal models offer a powerful alternative approach, since screens are unbiased, analysis is facilitated by strong research tools, and cellular mechanisms are highly conserved through evolution. We produced a …
Experimental Studies Of Rctb- Initiator Of Vibrio Cholerae Chromosome Ii Replication: Genetic Engineering, Protein Purification, And Protein Crystals, Olha Ivashkiv
Dissertations and Theses
Vibrio cholerae, a member of the Vibrionaceae family, is notorious as the causative agent of acute gastrointestinal disease known as cholera. Genetic information in members of the Vibrionaceae family is distributed between two chromosomes that are distinctively maintained. In V. cholerae, the initiation of DNA replication in each chromosome involves distinct processes. While the initiation of replication of chromosome I is carried by DnaA, the initiation of chromosome II replication is controlled by RctB, a DNA-binding protein that is an exclusive attribute of all Vibrio species. RctB has no homologs among other members of bacteria proteins and can serve as …
Structure-Function Analysis Of Zapc, An Ftsz-Ring Stabilizer, In Escherichia Coli Cytokinesis, Lukasz Tchorzewski
Structure-Function Analysis Of Zapc, An Ftsz-Ring Stabilizer, In Escherichia Coli Cytokinesis, Lukasz Tchorzewski
Dissertations and Theses
In Escherichia coli, cell division is defined by the polymerization and constriction of a cytokinetic ring (Z ring) formed by FtsZ, a tubulin-like GTPase, at midcell. Division also involves the formation of a multi-protein complex at midcell known as the divisome. Several divisome proteins promote the assembly/disassembly processes of FtsZ, thereby exercising spatiotemporal control over division. Among FtsZ regulatory proteins are the FtsZ ringassociated proteins (Zap), which either directly or indirectly stabilize the Z-ring by increasing lateral interactions amongst FtsZ protofilaments in the Z-ring. ZapA-D are recruited during early cytokinesis and have overlapping functions in stabilizing FtsZ at midcell, but …
Methodological Advancements For Improving Performance And Generating Ensemble Ecological Niche Models, Robert Boria
Methodological Advancements For Improving Performance And Generating Ensemble Ecological Niche Models, Robert Boria
Dissertations and Theses
This study employs spatial filtering of occurrence data with the aim of reducing overfitting to sampling bias in ecological niche models (ENMs). Sampling bias in geographic space leads to localities that may also be biased in environmental space. If so, the model can overfit to those biases. As a preliminary test addressing this issue, we used Maxent, bioclimatic variables, and occurrence localities of a broadly distributed Malagasy tenrec, Microgale cowani (Family Tenrecidae: Subfamily Oryzorictinae). We modeled the abiotically suitable area of this species using three distinct datasets: unfiltered, spatially filtered, and rarefied unfiltered localities. To quantify overfitting and model performance, …

This research study investigated the effect of using the think aloud protocol while reading informational text on students' ability to learn from text in a secondary science classroom. The participants in this study were high school students (n=47) in three classes of a mixed-grade Integrated Biology, Chemistry, and Physics course. The study tracked student achievement during a four-week curriculum unit on the theory of evolution and evidence for biological evolution. All students received instruction on using the think aloud protocol, and all students practiced the think aloud protocol when reading short articles related to scientific evidence for evolution. The researcher …
Interspecific Variation In Leaf-Level Biogenic Emissions Of The Bambuseae, Andrea Natalie Melnychenko
Interspecific Variation In Leaf-Level Biogenic Emissions Of The Bambuseae, Andrea Natalie Melnychenko
Dissertations and Theses
Plants emit a diverse range of biogenic volatile organic compounds (BVOCs) into the atmosphere, of which isoprene is the most abundantly emitted. Isoprene significantly affects biological and atmospheric processes, but the range of isoprene and BVOCs present in bamboos has not been well characterized. In this thesis I explore the range of isoprene emission found in bamboos and relate it to plant morphological and physiological characteristics. In addition, I measure and relate the entire suite of BVOCs present in the bamboos to their fundamental isoprene emission rate.
Interspecific variation in isoprene emission documented in a comprehensive survey of bamboos. Two …
First Bacterial Endosymbionts Found In The Phylum Ascomycota, Eileen Elizabeth Fitzpatrick
First Bacterial Endosymbionts Found In The Phylum Ascomycota, Eileen Elizabeth Fitzpatrick
Dissertations and Theses
Organisms belonging to the Kingdom Fungi are known to occupy a wide variety of ecological niches and are found globally in virtually all environments. Two members of the smallest of the fungal phylum, the Zygomycota, have also been found to harbor intercellular bacteria initially described as being from or closely related to organisms from the Genus Burkholderia. In this study two microaerophilic members of the species Verticilium from the phyla Ascomycota were characterized. Both appear to carry two bacterial endosymbionts. This is the first evidence of bacterial endosymbionts found within a member of the Ascomycota. Through the use of fluorescent …
Microbial Biomineralization Of Iron, Wen Fang
Microbial Biomineralization Of Iron, Wen Fang
Dissertations and Theses
Iron is a common cation in biomineral sand; it is present for example in magnetite produced by magnetotactic bacteria and in iron sulfides produced by sulfate reducing microorganisms. The work presented in this thesis focused on two types of microorganisms capable of forming iron biominerals. In the first project I have studied the effect of O2 on the respiratory physiology and the formation of magnetosomes by Magnetospirillum magneticum AMB-1. In the second project I have studied the relationship between olivine and the activity of dissimilatory sulfate reducing (DSR) microorganisms. For the first project, I grew cells of AMB-1 in cultures …
Carcass Feeding For Captive Vultures: Testing Assumptions About Zoos And Effects On Birds And Visitors, Hannah Gaengler
Carcass Feeding For Captive Vultures: Testing Assumptions About Zoos And Effects On Birds And Visitors, Hannah Gaengler
Dissertations and Theses
Carcass feeding is a potentially controversial feeding method for zoo animals. The common assumption is that many North American zoos refrain from feeding large carcasses to their carnivorous animals because zoo visitors might not approve of this feeding method. However, since there are several species of carnivores in zoos that feed from large carcasses in nature, this food type also has the potential to be beneficial to their welfare. In intelligent and usually inquisitive scavengers like vultures, a lack of activity and behavioral opportunities could be a welfare problem in captivity; providing them with a more complex food item might …
Effects Of Reactive Oxygen Species On Life History Traits Of Caenorhabditis Elegans, Samson William Smith
Effects Of Reactive Oxygen Species On Life History Traits Of Caenorhabditis Elegans, Samson William Smith
Dissertations and Theses
Evolutionary life history theory predicts that tradeoffs among fitness-related phenotypes will occur as a result of resource limitations and/or physiological constraints. Such tradeoffs are defined as the cost(s) incurred on one component of fitness (e.g., reproduction) by the increased expression of another fitness-related trait (e.g., longevity). Only recently have researchers begun to investigate the mechanistic bases of life history tradeoffs. A recent proposal is that reactive oxygen species (ROS) have a central role in shaping life history traits and tradeoffs. Research on disparate animal taxa has highlighted strong correlations between oxidative stress resistance and fitness-related life history traits, for example. …
The Role Of Nucleotide Excision Repair In Restoring Replication Following Uv-Induced Damage In Escherichia Coli, Kelley Nicole Newton
The Role Of Nucleotide Excision Repair In Restoring Replication Following Uv-Induced Damage In Escherichia Coli, Kelley Nicole Newton
Dissertations and Theses
Following low levels of UV exposure, Escherichia coli cells deficient in nucleotide excision repair recover and synthesize DNA at near wild type levels, an observation that formed the basis of the post replication recombination repair model. In this study, we characterized the DNA synthesis that occurs following UV-irradiation in the absence of nucleotide excision repair and show that although this synthesis resumes at near wild type levels, it is coincident with a high degree of cell death. We confirm that the replication occurring under these conditions involves extensive levels of strand exchange. However, cells undergoing this form of replication accumulate …
Externally-Expressed Fluorescence Across Sexes, Life Stages, And Species Of Spiders, Erin Brandt
Externally-Expressed Fluorescence Across Sexes, Life Stages, And Species Of Spiders, Erin Brandt
Dissertations and Theses
Although all spiders possess fluorophores in their hemolymph, the expression of external fluorescence is much more restricted. The purpose of this study was to evaluate differences in externally-expressed fluorescence between sexes, life stages, and species of spiders. To approach this question, we developed novel instrumentation to capture fluorescence with photographs of our specimens. We paired these fluorescence measurements with spectrometer measurements to attempt to determine the role that fluorescence plays in the overall coloration in spiders. The study was divided into four sections. First, we examined how fluorescence varies in sexes and life stages in Misumena vatia, an ambush predator …

Use Of Two-Dimensional Agarose-Gel Analysis To Characterize Processing Of Uv-Irradiated Plasmids And The Composition Of The Replisome Following Uv-Induced Arrest, Harout Arthur Jeiranian
Dissertations and Theses
In this thesis, I address two fundamental questions related to our understanding of how DNA damage is processed and repaired during replication. Using Two-dimensional (2-D) agarose gel analysis, I first examine whether DNA damage on plasmids introduced by transformation is processed in a manner similar to that observed on endogenously replicating plasmids and the chromosome. The original intent for using this approach was to develop a technique that could examine how different DNA adducts would be repaired in various sequence contexts. However, I found that distinct differences exist between the processing of DNA damage on transforming plasmids and the chromosome. …
Causes And Consequences Of Mitochondrial Variation In Caenorhabditid Nematodes, Kiley Ann Hicks
Causes And Consequences Of Mitochondrial Variation In Caenorhabditid Nematodes, Kiley Ann Hicks
Dissertations and Theses
Mitochondria are dynamic organelles that harbor their own stream-lined genome and generate much of the ATP necessary to sustain eukaryotic life via an electron transport chain (ETC). Because of the central role for mitochondria in organismal physiology, mitochondrial genetic and phenotypic variation can alter organismal fitness and affect population genetic and evolutionary outcomes. Despite the far-reaching relevance of mitochondria to evolutionary processes and human health, we lack a basic understanding of the causes and consequences of mitochondrial genetic and phenotypic variation. In this thesis, I quantified mitochondrial reactive oxygen species (ROS), membrane potential (δΨM), and mitochondrial morphological traits within Caenorhabditis …

Feasibility Study Of Trans-Cranial Direct Current Stimulation In Presence Of Brain Tumor, Abhilash Nair
Feasibility Study Of Trans-Cranial Direct Current Stimulation In Presence Of Brain Tumor, Abhilash Nair
Dissertations and Theses
Trans-cranial Direct Current Stimulation has been shown to modulate cortical neuronal activity. Weak constant current is applied to the scalp using electrodes, leading to sub-threshold changes in neuronal membrane potential. Before actual clinical stimulation is performed, such systems are validated by simulating the stimulation and the resulting current flow patterns using finite element solvers. However, the reliability of such models depends upon the accuracy with which the underlying anatomy has been modeled [1]. In earlier studies it has been shown that the current flow patterns within the brain are altered due to the presence of lesioned brain tissues[2]. Here we …

Anatomical Analysis Of The Feedback Projections From Extrastriate Cortex To Area 18 In Ferret Visual Cortex, Moody Saint Louis
Anatomical Analysis Of The Feedback Projections From Extrastriate Cortex To Area 18 In Ferret Visual Cortex, Moody Saint Louis
Dissertations and Theses
The purpose of this thesis is to characterize the cortical inputs to area 18 of ferret visual cerebral cortex. Contrary to feedforward connections, feedback connections are presumed to have a modulatory influence on the responses of lower order neurons providing information already processed. Input from feedback connections can supposedly elicit changes in the response to stimuli within the receptive field and may be involved in the role of discriminating objects relative to the background. The aim of our set of experiments was to fully analyze and compare the anatomical characteristics of feedback connections to area 18 from extrastriate areas as …

Morphological And Neuroanatomical Changes Associated With The Induction Of Neurodegeneration And Life Span Reduction In Drosophila Melanogaster By Glia Targeted Expression Of Cdh1/Rap/Fzr, An Activator Of The Anaphase Promoting Complex, Israel Nnah
Dissertations and Theses
"The retina aberrant in pattern (rap) gene encodes the Fizzy-related protein (Fzr), an activator of the E3 ubiquitin ligase complex: the Anaphase Promoting Complex/Cyclosome (APC/C). APC/C facilitates the cell cycle stage-specific degradation of cyclins and other mitotic regulators, resulting in the inactivation of Cdk (Cyclin Dependent Kinase) complexes, thus promoting completion of mitosis. Previous studies from our laboratory have shown that Rap/Fzr (the Drosophila homolog of the mammalian Cdh1) regulates glia differentiation and promotes neuron formation. Recent studies have shown that the glia specific over expression of Rap/Fzr causes progressive neurodegeneration and life span reduction preceded by temperature sensitive paralysis, …
Population Structure Of Island-Associated Pantropical Spotted Dolphins (Stenella Attenuata) In Hawaiian Waters, Sarah Shelby Courbis
Population Structure Of Island-Associated Pantropical Spotted Dolphins (Stenella Attenuata) In Hawaiian Waters, Sarah Shelby Courbis
Dissertations and Theses
Understanding gene flow, diversity, and dispersal patterns is important for predicting effects of natural events and anthropogenic activities on dolphin populations. With the very recent exceptions of false killer whales (Pseudorca crassidens), spinner dolphins (Stenella longirostris), and common bottlenose dolphins (Tursiops truncatus), Hawaiian odontocete species are managed as single stocks within the U.S. Hawaiian Exclusive Economic Zone. These exceptions are a result of recent studies that have indicated that some species have populations that show fidelity to individual islands or groups of islands, resulting in genetic differentiation, often with management implications. The first part …
Molecular Genetic Analysis Of Creld1 In Patients With Heterotaxy Disorder, Samaneh Zhian
Molecular Genetic Analysis Of Creld1 In Patients With Heterotaxy Disorder, Samaneh Zhian
Dissertations and Theses
Heterotaxy refers to the abnormal arrangement of internal organs in relation to each other. Model organism studies have shown that functions of more than eighty genes are required for normal asymmetric left-right organ development. CRELD1 has been shown to be necessary for proper heart development and mutations in CRELD1 are known to increase risk of cardiac atrioventricular septal defects (AVSD). AVSD is the most common form of heart defect associated with heterotaxy, and we have previously shown that some individuals heterotaxy-related AVSD have mutations in CRELD1.
Therefore, we propose to examine the CRELD1 gene in a large sample of …
