Black Bugbane & The Blues: Interactions Between Our Wildflower Of The Year And The Insect World, W. John Hayden Apr 2017

Black Bugbane & The Blues: Interactions Between Our Wildflower Of The Year And The Insect World, W. John Hayden

Biology Faculty Publications

No, this article has nothing to do with American Roots music. Black Bugbane is one of several common names for the 2017 VNPS Wildflower of the Year, Actaea racemosa. And Blues refers to a subfamily of lycaenid butterflies, commonly referred to as Blues or Azures. The interactions between Black Bugbane, a.k.a., Black Cohosh, Appalachian Azure butterflies (Celastrina neglectamajor), and ants was recently summarized by VNPS charter member and past president Nicky Staunton (2015). In brief, Black Bugbane is the sole food source for caterpillars of Appalachian Azure butterflies, a situation that, superficially, might seem like any other caterpillar and host …

Molecular And Morphological Data Reveal Three New Cryptic Species Of Chiasmocleis (Mehely 1904) (Anura, Microhylidae) Endemic To The Atlantic Forest, Brazil, Mauricio C. Forlani, João F. R. Tonini, Carlos A. G. Cruz, Hussam Zaher, Rafael O. De Sá Feb 2017

Biology Faculty Publications

Three new cryptic species of Chiasmocleis from the Atlantic Forest of Brazil are described. Two of these species occur in the northeastern states of Sergipe and Bahia, whereas the third species is found in the southeastern state of São Paulo. The new species can be distinguished from other congeneric species by the molecular data, as evidenced in the phylogeny, and by a combination of morphological characters including: size, foot webbing, dermal spines, and coloration patterns. Chiasmocleis species differ in osteological traits, therefore we also provide an osteological description of each new species and comparisons with data reported for other species …

The Progression Of Lyme Disease In The State Of Virginia, Margaret Michel Jan 2017

The Progression Of Lyme Disease In The State Of Virginia, Margaret Michel

Honors Theses

Lyme disease has spread in the United States from the northeast to more southern and western parts of the country. It has been shown that the tick which most often carries the Lyme disease pathogen (Borrelia burgdorferi) in North America, Ixodes scapularis, has also spread to these new areas of concern. In Virginia, the disease has progressed from the eastern shore to the western, more mountainous regions. Through obtaining historical museum specimens of mice from 11 locations in Virginia, we seek to determine whether the Lyme disease causing pathogen, B. burgdorferi, has always been present in …

Actaea Racemosa: The Slender Wands Of Flowering Common Black Cohosh Beckon Us To Explore Woodlands, W. John Hayden Jan 2017

Actaea Racemosa: The Slender Wands Of Flowering Common Black Cohosh Beckon Us To Explore Woodlands, W. John Hayden

Biology Faculty Publications

Common Black Cohosh is a perennial rhizomatous forest herb. Its horizontal rhizomes bear numerous adventitious roots on the underside and aerial stems of annual duration on the upper side, along with knobby scars left from aerial stems of previous years. Leaves are alternate, twice or thrice compound in ternate or pinnate patterns, and large—up to 1 m long. Individual leaflet size and shape vary with position in the large compound leaves, with position of a leaf on the stem, and from population to population. Most often leaflets are coarsely serrate, lobed to deeply incised, with a truncate to cuneate base …

Sepals And Petals And Stamens—Oh, My! Or, A Brief Discourse On Putative Homologies Of Perianth Elements Of Common Black Cohosh, W. John Hayden Jan 2017

Sepals And Petals And Stamens—Oh, My! Or, A Brief Discourse On Putative Homologies Of Perianth Elements Of Common Black Cohosh, W. John Hayden

Biology Faculty Publications

I encountered some contradictory information while preparing to write the 2017 Wildflower of the Year brochure: some sources describe flowers of Actaea racemosa, Common Black Cohosh, as having petals, while others say petals are absent. How can that be? How could there be such uncertainty about this common plant, one known to science since before the time of Linnaeus? After a little research, I decided to describe Black Cohosh flowers as having a series of organs interpretable either as staminodes (nonfunctional stamens) or as petals located between its sepals and stamens (Figure 1). Frankly, I waffled on the petal issue, …

The Hidden History Of Humming Frogs, Rafael O. De Sá Jan 2017

The Hidden History Of Humming Frogs, Rafael O. De Sá

Biology Faculty Publications

In the depths of Brazil’s Atlantic Forest, researchers are battling to identify species before they are lost for good. Dr Rafael O de Sá, Professor of Biology at the University of Richmond, Virginia, USA, specialises in a group known as the ‘narrow-mouthed frogs’ and has already found four species new to science.

Identification Of Rifampicin Resistance Mutations In Escherichia Coli, Including An Unusual Deletion Mutation, Eugene Y. Wu, Angela K. Hilliker Jan 2017

Identification Of Rifampicin Resistance Mutations In Escherichia Coli, Including An Unusual Deletion Mutation, Eugene Y. Wu, Angela K. Hilliker

Biology Faculty Publications

Rifampicin is an effective antibiotic against mycobacterial and other bacterial infections, but resistance readily emerges in laboratory and clinical settings. We screened Escherichia coli for rifampicin resistance and identified numerous mutations to the gene encoding the β chain of RNA polymerase (rpoB), including an unusual nine nucleotide deletion mutation. Structural modeling of the deletion mutant indicates locations of potential steric clashes with rifampicin. Sequence conservation in the region near the deletion mutation suggests a similar mutation may also confer resistance during the treatment of tuberculosis.

Photography-Based Taxonomy Is Inadequate, Unnecessary, And Potentially Harmful For Biological Sciences, Luis M. P. Ceríaco, Eliécer E. Gutiérrez, Alain Dubois, Rafael O. De Sá, Et Al. Nov 2016

Photography-Based Taxonomy Is Inadequate, Unnecessary, And Potentially Harmful For Biological Sciences, Luis M. P. Ceríaco, Eliécer E. Gutiérrez, Alain Dubois, Rafael O. De Sá, Et Al.

Biology Faculty Publications

The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & Nemésio 2007; Donegan 2008, 2009; Nemésio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per …

Jewels Of The Orchidaceae, W. John Hayden Apr 2016

Jewels Of The Orchidaceae, W. John Hayden

Biology Faculty Publications

To temperate-zone plant enthusiasts, the orchid family seems more than a little strange. On the one hand, native orchids grow wild without assistance from people, they are rooted in the soil, and they survive freezing cold winter temperatures. On the other hand, the tropical orchids that we encounter are ornamental plants, pampered by their human caregivers, cultured indoors in pots filled with fir bark or other media designed to mimic the plants’ natural epiphytic habit, and, as a group, these ornamental tropical orchids have essentially zero tolerance to frost. Of course, their flowers, fruits, and seeds define them all as …

Effects Of A Pyrrole-Based, Microtubule-Depolymerizing Compound On Raw 264.7 Macrophages, John A. Ciemnieckia, Clarke P. Lewis, John T. Gupton, Krista J. Stenger Feb 2016

Effects Of A Pyrrole-Based, Microtubule-Depolymerizing Compound On Raw 264.7 Macrophages, John A. Ciemnieckia, Clarke P. Lewis, John T. Gupton, Krista J. Stenger

Biology Faculty Publications

RAW 264.7 murine macrophages were exposed to the pyrrole-based compound 3,5-Dibromo-4-(3,4-dimethoxyphenyl)-1H-pyrrole-2-carboxylic acid ethyl ester (JG-03-14), which is a known microtubule depolymerizing agent with antitumor activity [1,2,3]. In this study exposure to JG-03-14 reduced the production of pro-inflammatory molecules by macrophages activated with lipopolysaccharide (LPS). Treatment with the pyrrole-based compound decreased the concentration of tumor necrosis factor-α (TNF-α) and nitric oxide (NO) released from the macrophages. Exposure to JG-03-14 also decreased TNF-α mRNA expression levels and the protein expression levels of inducible nitric oxide synthase (iNOS), the enzyme responsible for NO production in the activated macrophages. Furthermore, JG-03-14 treatment significantly changed …

High Genetic Diversity But Low Population Structure In The Frog Pseudopaludicola Falcipes (Hensel, 1867) (Amphibia, Anura) From The Pampas Of South America, José A. Langone, Arley Camargo, Rafael O. De Sá Feb 2016

High Genetic Diversity But Low Population Structure In The Frog Pseudopaludicola Falcipes (Hensel, 1867) (Amphibia, Anura) From The Pampas Of South America, José A. Langone, Arley Camargo, Rafael O. De Sá

Biology Faculty Publications

Relative to South America’s ecoregions, the temperate grasslands of the Pampas have been poorly studied from a phylogeographic perspective. Based on an intermediate biogeographic setting between subtropical forest (Atlantic Forest) and arid ecosystems (Chaco and Patagonia), Pampean species are expected to show unstable demographic histories due to the Quaternary climatic oscillations. Herein, we investigate the phylogenetic relatedness and phylogeographic history of Pseudopaludicola falcipes, a small and common frog that is widely distributed across the Pampean grasslands. First, we use molecular data to assess if P. falcipes represents a single or multiple, separately evolving cryptic lineages. Because P. falcipes is …

Computational Studies Of Paradifluorobenzene Cations And Hydrogen Cyanide Molecule, John C. Rowe Iv Jan 2016

Computational Studies Of Paradifluorobenzene Cations And Hydrogen Cyanide Molecule, John C. Rowe Iv

Honors Theses

Polycyclic aromatic hydrocarbons (PAHs) are hypothesized to comprise a significant portion of interstellar carbon identified from the Diffuse Interstellar Bands (DIBs). Gas phase cation-molecule reactions between compounds that previously existed as weakly bound species provides the best explanation for the emergence of PAHs in the interstellar medium (ISM). In this work, we use computational methods to characterize one of these weakly-bound systems. We discovered that intramolecular charge distribution (calculated using the Natural Population Analysis) dictate the complexes formed between a paradifluorobenzene cation and either one or two HCN molecules. Additionally, the relative stabilities of the complexes and the binding energies …

Impact Of Vector Range Expansion On Pathogen Transmission Dynamics Of Lyme Disease In Southwestern Virginia, Bishan Bhattarai Jan 2016

Impact Of Vector Range Expansion On Pathogen Transmission Dynamics Of Lyme Disease In Southwestern Virginia, Bishan Bhattarai

Honors Theses

Blacklegged ticks, Ixodes scapularis is the primary vector of Lyme disease in eastern United States. Borrelia Burgdorfei, the etiological agent of Lyme disease is transferred by ticks of Ixodes species. In recent years, its congener, Ixodes affinis has been expanding its range northwards from its southern population. We were interested in studying how the introduction of this new vector affected the interaction between the pathogen genotype and the host. We hypothesized that differential host use by I. affinis and I. scapularis would partly explain observed differences in B. burgdorferi infection prevalence and genotypic structure in southeastern Virginia. The result …

Systematics Of The Neotropical Genus Leptodactylus Fitzinger, 1826 (Anura: Leptodactylidae): Phylogeny, The Relevance Of Non-Molecular Evidence, And Species Accounts, Rafael O. De Sá, Taran Grant, Arley Camargo, W. R. Heyer, María Laura Ponssa, Edward Stanley Sep 2014

Systematics Of The Neotropical Genus Leptodactylus Fitzinger, 1826 (Anura: Leptodactylidae): Phylogeny, The Relevance Of Non-Molecular Evidence, And Species Accounts, Rafael O. De Sá, Taran Grant, Arley Camargo, W. R. Heyer, María Laura Ponssa, Edward Stanley

Biology Faculty Publications

A phylogeny of the species-rich clade of the Neotropical frog genus Leptodactylus sensu stricto is presented on the basis of a total evidence analysis of molecular (mitochondrial and nuclear markers) and non-molecular (adult and larval morphological and behavioral characters) sampled from > 80% of the 75 currently recognized species. Our results support the monophyly of Leptodactylus sensu stricto, with Hydrolaetare placed as its sister group. The reciprocal monophyly of Hydrolaetare and Leptodactylus sensu stricto does not require that we consider Hydrolaetare as either a subgenus or synonym of Leptodactylus sensu lato. We recognize Leptodactylus sensu stricto, Hydrolaetare, Adenomera, and Lithodytes …

A New Species Of Chiasmocleis (Microhylidae, Gastrophryninae) From The Atlantic Forest Of Espírito Santo State, Brazil, João Filipe Tonini, Maruicio Forlani, Rafael O. De Sá Jul 2014

A New Species Of Chiasmocleis (Microhylidae, Gastrophryninae) From The Atlantic Forest Of Espírito Santo State, Brazil, João Filipe Tonini, Maruicio Forlani, Rafael O. De Sá

Biology Faculty Publications

Among Neotropical microhylids, the genus Chiasmocleis is exceptionally diverse. Most species ofChiasmocleis were described in recent years based on external morphology, but recent studies using molecular data did not support the monophyly of the species groups clustered based on feet webbing. Furthermore, a phylogeographic study of C. lacrimae estimated high genetic divergence and low gene flow among populations across small geographic ranges. Increasing the molecular and geographic sampling, and incorporating morphological data, we identified new cryptic species. Herein, we used novel genetic and morphological data to describe a new species of Chiasmocleis.

The Neotropical Genus Austrolebias: An Emerging Model Of Annual Killifishes, Nibia Berois, María J. Arezo, Rafael O. De Sá Jun 2014

The Neotropical Genus Austrolebias: An Emerging Model Of Annual Killifishes, Nibia Berois, María J. Arezo, Rafael O. De Sá

Biology Faculty Publications

Annual fishes are found in both Africa and South America occupying ephemeral ponds that dried seasonally. Neotropical annual fishes are members of the family Rivulidae that consist of both annual and non-annual fishes. Annual species are characterized by a prolonged embryonic development and a relatively short adult life.

Males and females show striking sexual dimorphisms, complex courtship, and mating behaviors. The prolonged embryonic stage has several traits including embryos that are resistant to desiccation and undergo up to three reversible developmental arrests until hatching. These unique developmental adaptations are closely related to the annual fish life cycle and are the …

Sex Ratio Bias And Extinction Risk In An Isolated Population Of Tuatara (Sphenodon Punctatus), Kristine L. Grayson, Nicola J. Mitchell, Joanne M. Monks, Susan N. Keall, Joanna N. Wilson, Nicola J. Nelson Apr 2014

Sex Ratio Bias And Extinction Risk In An Isolated Population Of Tuatara (Sphenodon Punctatus), Kristine L. Grayson, Nicola J. Mitchell, Joanne M. Monks, Susan N. Keall, Joanna N. Wilson, Nicola J. Nelson

Biology Faculty Publications

Understanding the mechanisms underlying population declines is critical for preventing the extinction of endangered populations. Positive feedbacks can hasten the process of collapse and create an ‘extinction vortex,’ particularly in small, isolated populations. We provide a case study of a male-biased sex ratio creating the conditions for extinction in a natural population of tuatara (Sphenodon punctatus) on North Brother Island in the Cook Strait of New Zealand. We combine data from long term mark-recapture surveys, updated model estimates of hatchling sex ratio, and population viability modeling to measure the impacts of sex ratio skew. Results from the mark-recapture …

Evolutionary Relationships Of The Critically Endangered Frog Ericabatrachus Baleensis Largen, 1991 With Notes On Incorporating Previously Unsampled Taxa Into Large-Scale Phylogenetic Analyses, Karen Siu-Ting, David J. Gower, Davide Pisani, Roman Kassahun, Fikirte Gebresenbet, Michele Menegon, Abebe A. Mengistu, Samy A. Saber, Rafael O. De Sá, Mark Wilkinson, Simon P. Loader

Biology Faculty Publications

Background: The phylogenetic relationships of many taxa remain poorly known because of a lack of appropriate data and/or analyses. Despite substantial recent advances, amphibian phylogeny remains poorly resolved in many instances. The phylogenetic relationships of the Ethiopian endemic monotypic genus Ericabatrachus has been addressed thus far only with phenotypic data and remains contentious.

Results: We obtained fresh samples of the now rare and Critically Endangered Ericabatrachus baleensis and generated DNA sequences for two mitochondrial and four nuclear genes. Analyses of these new data using de novo and constrained-tree phylogenetic reconstructions strongly support a close relationship between Ericabatrachus and …

Host-Specialization Of Bartonella In Flea Vectors Collected From Black-Tailed Prairie Dogs, Mark D. Massaro Jan 2014

Host-Specialization Of Bartonella In Flea Vectors Collected From Black-Tailed Prairie Dogs, Mark D. Massaro

Honors Theses

At least 22 species of Bartonella bacteria have been named and many are associated with one of a list of potential mammalian reservoirs and arthropod vectors. One example of such a system is the parasitism of black-tailed prairie dogs (Cynomys ludovicianus) of the Western U.S. by Oropyslla hirsuta and Pulex simulans fleas. These vectors are thought to maintain B. washoensis infection in these mammals, but little is known about their role in the specialization of this strain to this reservoir. We found that the more specialist O. hirsuta fleas were much more likely to be infected with B. washoensis than …

First Report Of Satellite Males During Breeding In Leptodactylus Latrans (Amphibia, Anura), Gabriel Laufer, Noelia Gobel, José M. Mautone, María Galán, Rafael O. De Sá

Biology Faculty Publications

Individual males can adopt alternative mating tactics. The occurrence of satellite males is a common behaviour across anuran taxa (e.g., Lithobates clamitans, Wells, 1977; Anaxyrus cognatus, Krupa, 1989; Dendropsophus ebraccatus, Miyamoto and Cane, 1980; Rhinella crucifer, Forester and Lynken, 1986). Satellite males take peripheral positions to calling males, and adopt alternate mating tactics in an attempt to intercept females that are attracted to calling males (Wells, 2007) to increase their own mating success. Satellite males could have an inexpensive form of mate-locating, avoiding predators, and saving energy (Arak, 1983). Furthermore, this strategy could play an important role in the genetic …

The Genus Odontophrynus (Anura: Odontophrynidae): A Larval Perspective, Filipe Augusto C. Do Nascimento, Tamí Mott, José A. Langone, Christine A. Davis, Rafael O. De Sá

Biology Faculty Publications

The genus Odontophrynus consists of 11 species of medium-sized frogs distributed across south and east South America. This study examines and describes the chondrocrania and oral cavities of 0. americanus, 0. maisuma, 0. carvalhoi, and 0. cu/tripes, and reviews current knowledge about the larval external morphology of the genus. Twenty-one tadpoles were cleared and double-stained for chondrocranium description and five tadpoles were dissected for analysis in a scanning electron microscope. The presence of a tectum parientale may be considered here as a putative synapomorphy of the genus. The 0. americanus and 0. cu/tripes species groups were partially differentiated by the …

Species Boundaries And Biogeography Of East African Torrent Frogs Of The Genus Petropedetes (Amphibia: Anura: Petropeditidae), Simon P. Loader, F. Sara Ceccarelli, Mark Wilkinson, Michele Menegon, Jean Mariaux, Rafael O. De Sá, Et Al. May 2013

Species Boundaries And Biogeography Of East African Torrent Frogs Of The Genus Petropedetes (Amphibia: Anura: Petropeditidae), Simon P. Loader, F. Sara Ceccarelli, Mark Wilkinson, Michele Menegon, Jean Mariaux, Rafael O. De Sá, Et Al.

Biology Faculty Publications

Based on morphology there are currently three described East African species of torrent frogs belonging to the tropical African genus Petropedetes (P. dutoiti, P. martiennseni and P. yakusini). Taxonomic uncertainties are evident within this group: a putative new species reported from the Nguru Mountains and the species P. yakusini have a fragmented distribution across the Southern Tanzanian highlands. We conducted a molecular systematic study, sampling populations of East African petropedetids occurring in the mountains of Tanzania (P. martiennseni and P. yakusini), to investigate species boundaries and biogeography. Data provide evidence for the recognition of …

The Identity Of Rana Margaritifera Laurenti, 1768 (Anura, Bufonidae), Esteban O. Lavilla, Ulisses Caramaschi, José A. Langone, José P. Pombal Jr., Rafael O. De Sá

Biology Faculty Publications

Rana margaritifera was described by Laurenti in 1768 and currently is associated to the genus Rhinella, under the combination Rhinella margaritifera. Currently, the R. margaritifera species group consists of 16 recognized species. Further-more, many additional species have been suggested to exist in this group which highlights the ambiguity surrounding the identity of Rhinella margaritifera and impend further description of the species in this group. After an exhaustive bibliographic review, we concluded that the recent designation of a lectotype for R. margaritifera is invalid according with Art. 73, ICZN, 1999. Herein, we designate and provide the description of a …

De Sá, Miguel Trefaut Rodrigues

Biology Faculty Publications

Despite major progress in deciphering the amphibian tree of life by molecular phylogenetics, we identified two questions remaining to be answered regarding relationships within Hyloidea, the clade of South American origin that comprises most extant anuran diversity. A few genera like Rupirana and Crossodactylodes have enigmatic phylogenetic positions, and relationships among major lineages within some families like Leptodactylidae remain ambiguous. To resolve these specific questions we used two approaches (1) a complete matrix approach representing >6.6 kb, including most major Hyloidea lineages (61 terminals) combining different methods of phylogenetic reconstruction and measures of node support; and (2) a supermatrix approach …

Potent Mutagens Have Positive And Negative Effects On Viral Fitness Of Reovirus In Vitro, Jennie Altman Apr 2013

Potent Mutagens Have Positive And Negative Effects On Viral Fitness Of Reovirus In Vitro, Jennie Altman

Honors Theses

Due to the inherently error-prone nature of RNA replication, mutations to genomes of RNA viruses occur frequently and accumulate. We hypothesized that RNA versions of nucleoside analogues that increase mutation rates in DNA could cause increases in the mutation rate of a model RNA virus, Reovirus, and decrease the fitness of the virus in vitro due to the accumulation of deleterious mutations. After conducting multiple passages of Reovirus in mouse L929 cells in the presence of these potential RNA mutagens in four separate trials, there were not only virus samples with the expected decreased infectivity, but surprisingly, samples with marked …
