This report provides details for the construction of a hot-bead thermistor current meter which is capable of measuring water velocities on a millimeter spatial scale and for the construction of a compact and accurate calibration system. Hot-bead thermistor current meters can be built with response times of 200 ms capable of measuring velocities between 0.1 and 80 cm s-1. The construction of a sturdy probe for application in lotic systems such as high gradient Hawaiian streams was achieved by the use of heavy-duty acrylic tubing, small stainless steel gas-chromatography tubing, and flexible Tygon spaghetti tubing. Biochemical Systematics Of Notothenioid Fishes From Antarctica, Mara A. Mcdonald, Michael H. Smith, Michael W. Smith, James M. Novak, Paul E. Johns, Arthur L. Devries

Genetic variation at 30 protein-coding loci was examined in seven forms of notothenioid fishes from Antarctica. Multilocus heterozygosity varied from 0.018 to 0.078 across taxa. An analysis of the allozyme data revealed the probable existence of an unrecognized cryptic species within Trematomu5 bemacchii. Pagothenia borchgrevinki is as closely related to some species of Trematomus as are some species of Trematomus to each other. Speciation among the species of Trematomus and Pagothenia appears to have taken place primarily after the separation of Antarctica from Australia.

After mating, hypotrichous ciliated protozoa transform a set of their micronuclear chromosomes into thousands of short, linear DNA molecules that form the macronuclear genome. To examine micronuclear genome organization in the hypotrich Euplotes crassus, we have analyzed two cloned segments of micronuclear DNA as well as the macronuclear DNA molecules that are derived from them. E. crassus was found to display a number of features characteristic of other hypotrich genomes, including (i) clustering and close spacing of the precursors of macronuclear DNA molecules, (ii) the frequent occurrence of internal eliminated sequences within macronuclear precursors, (iii) overlapping macronuclear precursors, (iv) lack …

In its report to the Ministier for Fisheries in November 1986, the Southern Western Australian Shark Working Group recommended the establishment of an Industry/Government Mangement Advisory Committee to provide managememt advice on the fishery to the Minister for Fisheries. The Advisory Committee held its first meeting in Albany on 23 February 1989. After being briefed on the state of shark stocks, the Committee's deliberations fell into three broad categories. The total catch of about 12.1 million kilograms for the 1987/88 season, based on processor returns, was an increase of 44% on the 1986/87 season. This very high catch, the third highest on record, resulted from an above average perulus settlement in 1983/84 and a "flow-on" of fast growing animals stemming from the expected high recruitment in 1988/990 caught as "reds" in 1987/88. Following the sexual cycle, hypotrichous ciliated protozoans fragment a set of their micronuclear chromosomes to generate the thousands of short, linear DNA molecules present in the transcriptionally active macronucleus. We have used a hybrid selection procedure to examine macronuclear DNA molecules for subtelomeric length heterogeneity to determine whether chromosome fragmentation occurs at unique or multiple sites. The results suggest that multiple, but closely spaced, chromosome fragmentation sites are used by Oxytricha nova. In contrast, Euplotes crassus uses unique chromosome fragmentation sites in a reproducible manner to generate the ends of macronuclear DNA molecules. Additional studies compared DNA sequences in …

The Yellow-shafted Flicker (Colaptes auratus auratus) and Red-shafted Flicker (C. a. cafer) form a stable, narrow hybrid zone on the western Great Plains of North America. Allozyme data were obtained from 31 structural gene loci for 33 samples representing 246 Northern Flickers from throughout the Great Plains. Flickers were approximately equivalent to other birds in terms of proportion of polymorphic loci (P = 0.207) and average heterozygosity (H = 0.056). There was no concordant variation between plumage characters and allelic frequencies.

Seasonal filtration rates of a pond population of the freshwater pisidiid clam, Musculium partumeium (Say), were assessed by measuring the clearance of 2.02 µ.m latex beads from suspension and expressed in terms of FR = a(AFDW}b where FR = filtration rate (ml H20 · hr- 1 ) , AFDW = mg ash-free dry weight of a whole clam and "a" and "b" are constants. The a-values ( = FR of a 1-mg AFDW clam} vary seasonally with the highest rates corresponding to periods of maximum growth and reproduction in the spring and fall. Treatment of Schneider's line 3 Drosophila cells with the steroid hormone ecdysterone rapidly stimulated the synthesis and accumulation of the polypeptide previously designated p7 [Berger, E. M., Ireland, R. C. & Wyss, C. (1980) Somatic Cell Genet. 6, 119-129]. In this report, p7 is identified as the 23,000-dalton heat shock polypeptide (hsp23). In addition to hsp23, the synthesis of the low molecular weight heat shock polypeptides hsp22, hsp26, and hsp27 was also stimulated by ecdysterone, although to different extents. Thirty-five cryptic variant lines were used to examine the mechanisms involved in genetic modulation of alcohol metabolism in Drosophila. Late third-instar larval, preemergence pupal, and adult stages cultured at 18 and 28 C were examined. Spectrophotometric analyses for native alcohol dehydrogenase (ADH) activity and residual ADH activity after treatment with guanidine hydrochloride and heat were performed. Differential response of cryptic variants to treatment with the denaturants during development suggested that this variation may have an adaptive significance.

Natural populations of the North American stream limpet, F. rivularis, were studied in upstate New York [USA], in a set of localities whose waters have a 15-fold range of dissolved Ca (4.6-67.6 mg/liter) and also range from oligotrophy to eutrophy. Shell component analyses (CaCO3, total organic C and total N) are reported as component mass-fractions (mg/g or .mu.g/g dry weight) and as values for a standard limpet shell of 35 mm aperture length (AL). Seasonal metabolic rates were examined for the sphaeriid clam, Musculium partumeium (Say) from a permanent and a temporary pond. For the fall-born generation from the permanent pond and for the single generation from the temporary pond, metabolic rates peak during periods of greatest growth and reproduction. Metabolic rates were consistently higher for the permanent pond population. These rates for Musculium partumeium extend the available information on the family Sphaeriidae to the genus Musculium.

Clarke (1973) states that Pisidium casertanum (Poli) is perhaps the most common species of Pisidium and is truly cosmopolitan, occurring throughout Eurasia, Africa, Australia, and South, Central and North America. Populations of these clams are reported in habitats ranging from ephemeral ponds to benthic zones of deep lakes. The ampullariid Pomacea urceus uses its ctenidium and lung in respiration. The snails reach the surface with their siphon and ventilate their lung by withdrawing their head-foot in a pumping action. The lung gas also serves to change the buoyancy of the snails. After a ventilation a snail does not necessarily return to the same buoyancy level. An Analysis Of Gene-Enzyme Variability In Natural Populations Of Drosophila Melanogaster And D. Simulans, Stephen J. O'Brien, Ross J. Macintyre

Nine populations of D. melanogaster and two populations of D. simulans were analyzed for polymorphism in 10 gene-enzyme systems by the technique of gel electrophoresis. In the eight natural populations of D. melanogaster, an average of 54% of the enzymes were polymorphic, and the average heterozygosity was 22.7%. An experimental population of D. melanogaster, which has been maintained in a laboratory cage for 20 years, showed levels of polymorphism equivalent to those of natural populations. The D. simulans populations had much less variability. The possible factors involved in maintaining these polymorphisms are discussed.
