Mechanism Of Decarboxylation Of Glycine And Glycolate By Isolated Soybean Cells, David J. Oliver

Isolated soybean leaf mesophyll cells decarboxylated exogenously added [1-14C]glycolate and [1-14C]glycine in the dark. The rate of CO2 release from glycine was inhibited over 90% by isonicotinic acid hydrazide and about 80% by KCN, two inhibitors of the glycine to serine plus CO2 reaction. The release of CO2 from glycolate was inhibited by less than 50% under the same conditions. This indicates that about 50% of the CO2 released from glycolate occurred at a site other than the glycine to serine reaction. A number of' a-(N)-heterocyclic thiosemicarbazones have been synthesized and evaluated for potential antineoplastic activity since Brockman and colleagues2 reported the antineoplastic activity of' 2-f'ormylpyridine thiosemicarbazone (1). The primary objective of' this research project has been the synthesis and characterization of' the semicarbazone and the thiosemicarbazone derivatives of' the di-2-pyridyl ketone, (9-a) and (9-b), and the 1,8-diazaf'luorenone, (10-a) and (10-b), ring systems, as well as the evaluation of' these compounds for both in vitro and in vivo antineoplastic activity against Ehrlich ascites tumor cells in mice. Optical pumping of dye lasers deposits heat nonuniformly in the active medium. Because the refractive index of a liquid or gas is a function of temperature, gradients are created which refract the laser beam. This effect is sometimes serious enough to quench laser action. In this paper, we evaluate the effects caused by optical pumping with a coaxial flashlamp and suggest ultraviolet absorbers and wavelength shifters as materials for minimizing the problem

The role of ascorbic acid with regard to lipid peroxidation and aging has been examined. A thorough literature analysis indicates that free radical-induced lipid peroxidation is a plausible biochemical explanation for aging. Lipid peroxidation causes cellular damage due to altered enzyme activities, error-prone nucleic acid metabolism, and membrane dysfunction, as well as the accumulation of aging pigments in the lysomes. Ascorbic acid, a water soluble free radical quencher, was examined with regard to carbon tetra-chloride-induced lipid peroxidation and in vivo aging. A simple, quick, and sensitive radiometric assay for glutaminase has been established. This assay uses L-(U-14C)-glutamine as the substrate and measures the 14CO2 produced when the reaction is coupled to glutamate decarboxylase. An efficient purification procedure for cow brain glutaminase has also been developed; the yield is 12%. Steps include acetone extraction, ammonium sulfate fractionation, calcium phosphate gel treatment, and differential gel filtration on Sepharose 4B. The final preparation has a specific activity of 385 μmole/min/mg and shows a single protein band on polyacrylamide gel electrophoresis in sodium dodecyl sulfate; this band corresponds to a …

Photoelectric Properties And Detection Of The Aromatic Carcinogens Benzo[A]Pyrene And Dimethylbenzanthracene, William A. Houle, Hugh M. Brown, O. Hayes Griffith

The absolute photoelectron quantum yield spectra for benzo[ajpyrene and dimethylbenzanthracene are presented in the wavelength range 180-230 nm. These polycyclic aromatic carcinogens have photoelectron quantum yields of approximately 2 X 10-3 electrons per incident photon at 180 nm. The quantum yields fall off quickly and monotonically at wavelengths longer than 210 nm (5.9 eV). Threshold values for benzo[a]pyrene and dimethylbenzanthracene are 5.25 ± 0.06 eV and 5.27 ± 0.04 eV, respectively. The photoelectron quantum yields of benzo[a]pyrene and dimethylben-zanthracene are several orders of magnitude greater than typical components of biological membranes (amino acids, phospholipids, and polysaccharides). Analysis of historical nutrient data for Lake Mead indicates that the fertility of the reservoir has decreased which may be the cause for a corresponding decline in the largemouth bass population. However, it appears that fertility can be manipulated by altering the operation of the dam. The depletion of nutrients in the euphotic zone by phytoplankton and subsequent accumulation in the hypolimnion during summer and fall provide a natural nutrient gradient from which water of varying fertility can be drawn for discharge.
