The Lipooligosaccharides Of Haemophilus Ducreyi Are Highly Sialylated, William Melaugh, A A. Campagnari, B W. Gibson Jan 1996

The Lipooligosaccharides Of Haemophilus Ducreyi Are Highly Sialylated, William Melaugh, A A. Campagnari, B W. Gibson

Chemistry Faculty Publications

The major lipooligosaccharides of the sexually transmitted pathogen Haemophilus ducreyi 35000 have been previously found to terminate in N-acetyllactosamine and sialyl-N-acetyllactosamine, Neu5Ac alpha 2-->3Gal beta 1-->4GlcNAc (W. Melaugh, N. J. Phillips, A. A. Campagnari, M. V. Tullius, and B. W. Gibson, Biochemistry 33: 13070-13078, 1994). In this study, mass spectrometry and composition analyses have shown that the lipooligosaccharides from three other H. ducreyi strains also contain N-acetyllactosamine and are highly sialylated (approximately 30 to 50%), although one African strain was found to contain neither of these structural features.

Membrane Topology Of The Colicin A Pore-Forming Domain Analyzed By Disulfide Bond Engineering, Denis Duche, Jacques Izard, Juan M. Gonzalez-Manas, Michael W. Parker, Marcel Crest, Daniel Baty Jan 1996

Membrane Topology Of The Colicin A Pore-Forming Domain Analyzed By Disulfide Bond Engineering, Denis Duche, Jacques Izard, Juan M. Gonzalez-Manas, Michael W. Parker, Marcel Crest, Daniel Baty

Department of Food Science and Technology: Faculty Publications

Four colicin A double-cysteine mutants possessing a disulfide bond in their pore-forming domain were constructed to study the translocation and the pore formation of colicin A. The disulfide bonds connected a-helices 1 and 2, 2 and 10, 3 and 9, or 3 and 10 of the poreforming domain. The disulfide bonds did not prevent the colicin A translocation through the Escherichia coli envelope. However, the mutated colicins were able to exert their in vivo channel activity only after reduction of their disulfide bonds. In vitro studies with brominated phospholipid vesicles and planar lipid bilayers revealed that the disulfide bond that …

Moraxella (Branhamella) Catarrhalis: A Molecular Epidemiology Study, Lyndell R. Gill May 1995

Moraxella (Branhamella) Catarrhalis: A Molecular Epidemiology Study, Lyndell R. Gill

Electronic Theses and Dissertations

Moraxella (Branhamella) catarrhalis is the third-most-frequently isolated microorganism associated with acute exacerbations of chronic bronchitis in patients during their stay at the Mountain Home VA Medical Center (MHVAMC). In order to develop a practical, epidemiologically-meaningful typing method for M. (B.) catarrhalis, we tested two methods based on analysis of chromosomal DNA for typeability, reproducibility, and ability to differentiate between unrelated strains (discriminatory power, D). M. (B.) catarrhalis isolants from MHVAMC from 7/1/87-6/30/88 were grown overnight in broth and embedded in agarose. DNA was isolated by standard methods. The DNA was subjected to: (1) restriction endonuclease digestion (with either Bgl II …

Pathogenicity Of Murine Cytomegalovirus Mutants, Victoria Jean Cavanaugh Apr 1995

Pathogenicity Of Murine Cytomegalovirus Mutants, Victoria Jean Cavanaugh

Theses and Dissertations in Biomedical Sciences

The purpose of this study was to identified nonessential murine cytomegalovirus (MCMV) genes involved in pathogenesis in vivo. Our approach to identifyjng these genes consisted of constructing MCMV mutants, and then analyzing these mutants in vitro and in vivo. Recombinant viruses (RV) expressing the β-glucuronidase marker gene were constructed by site-directed insertion and deletion mutagenesis of the MCMV Hind III-J and -I regions of the viral genome. Mutations were targeted to this region of the MCMV genome because the corresponding region of the human CMV genome is nonessential and is involved in down-regulating major histocompatibility complex (MHC) class I expression …

The Kinetic Characterization Of Mutant L289p Of Recombinant Hiv-1 Reverse Transcriptase, Tryn Thomas Stimart

Chemistry & Biochemistry Theses & Dissertations

The Human Immunodeficiency Virus, type 1 (HIV-1) is the causative agent of the Acquired immunodeficiency Syndrome (AIDS). Currently, AIDS is the leading cause of death amongst young people in the United States. The large increase of HIV-1 infections over the past decade has spawned considerable research in fighting the spread of this disease. One area of intense HIV-1 research has focused on the enzyme Reverse Transcriptase (RT) which is an essential enzyme involved in the replication and life cycle of HIV-1. HIV-1 RT is composed of p66 and p51 subunits and is only active as a heterodimer in vivo. …

Structural Characterization Of A Novel Inhibitor Of Hiv Reverse Transcriptase (Hiv Rt), Greggory Jon Woitte Apr 1995

Structural Characterization Of A Novel Inhibitor Of Hiv Reverse Transcriptase (Hiv Rt), Greggory Jon Woitte

Chemistry & Biochemistry Theses & Dissertations

Human immunodeficiency virus (HIV) infections have become a leading cause of death among young people in the United States today. As the number of HIV infections increases, so too does the cost of treatment. Together, these numbers have prompted an increase in the development of pharmaceutical interventions. HIV reverse transcriptase (HIV RT) has become a suitable target for drug therapy because it is the sole enzyme responsible for HIV replication.

Fucoidan, a sulfated polysaccharide isolated from the brown algae Fucus vesiculosus, has been shown to block a variety of cell adhesion related events including metastasis. In addition, fucoidan has also …

Translational Regulation Of The C-Jun Proto-Oncogene, Anil Sehgal Apr 1994

Translational Regulation Of The C-Jun Proto-Oncogene, Anil Sehgal

Theses and Dissertations in Biomedical Sciences

The v-jun oncogene was originally isolated from the ASV17 virus in 1987. Ever since its isolation, extensive work has been done to understand the role of the v-jun oncogene in cell transformation. The c-Jun protein is a transcription factor which binds to the DNA target TGACTCA. The c-Jun protein binds to DNA in the form of dimers. It can form homodimers with itself and heterodimers with Jun family (JunB and JunD), Fos family (FosB, Fra1 and Fra2), or with CREB family members through the leucine zipper motif. Because the c-jun proto-oncogene plays an important role in cell transformation, extensive work …

Use Of Pyocin To Select A Haemophilus Ducreyi Variant Defective In Lipooligosaccharide Biosynthesis, A A. Campagnari, R Karalus, M A. Apicella, William Melaugh, A J. Lesse, B W. Gibson

Chemistry Faculty Publications

Haemophilus ducreyi, a cause of genital ulcer disease in developing countries, appears to facilitate the heterosexual transmission of the human immunodeficiency virus in Africa. Despite an increase in studies of this gram-negative human pathogen, little is known about the pathogenesis of chancroid. Our studies have shown that the lipooligosaccharides (LOS) of H. ducreyi may play an important role in ulcer formation. Monoclonal antibody and mass spectrometric analyses identified a terminal trisaccharide present on H. ducreyi LOS that is immunochemically similar to human paragloboside. This epitope is present on the LOS of Neisseria gonorrhoeae, and it may be the site of …

A Characterization Of Extractable, Hydroxylated Fatty Acid Bearing Components In Legionella Pneumophila, Jonathan R. Lane Dec 1993

A Characterization Of Extractable, Hydroxylated Fatty Acid Bearing Components In Legionella Pneumophila, Jonathan R. Lane

Electronic Theses and Dissertations

Extraction of the lipids of Legionella pneumophila yields phases unlike those produced from other Gram-negative bacteria. A viscous interface forms between the aqueous (wash) and organic phases. More than half of the hydroxylated fatty acids were found distributed between the aqueous phase and the interfacial material, fractions in which such constituents have not been reported in other Gram-negative species. It was further observed that after the material from the aqueous/interfacial phase was dissolved in methanol or chloroform/methanol (2:1 (V/V)), the addition of acetone would create a white, flocculent precipitate. Analyses showed that the supernatant contained fatty acids that were nonhydroxylated …

Investigation Of The Structural Heterogeneity Of Lipooligosaccharides From Pathogenic Haemophilus And Neisseria Species And Of R-Type Lipopolysaccharides From Salmonella Typhimurium By Electrospray Mass Spectrometry, B W. Gibson, William Melaugh, Nancy J. Phillips, M A. Apicella, A A. Campagnari, J M. Griffiss Jan 1993

Griffiss

Chemistry Faculty Publications

Heterogeneity in the lipooligosaccharides (LOS) of pathogenic Haemophilus and Neisseria species is evident from the multiplicity of components observed with electrophoretic analyses. Knowledge of the precise structures that make up these diverse LOS molecules is clearly the key to reaching an understanding of pathogenic processes such as phase variation and molecular mimicry. Except for a few cases, little is known about the specific structural features of LOS that underlie phase variation and molecular mimicry, partly because of the inherent difficulties in the structural elucidation of these complex glycolipids. In the lipopolysaccharides (LPS) from Salmonella typhimurium and Escherichia coli, rough, or …

Kinetic Characterization Of A Recombinant C-Terminal Mutant Of Reverse Transcriptase From The Human Immunodeficiency Virus, Thomas S. Heard Jul 1992

Kinetic Characterization Of A Recombinant C-Terminal Mutant Of Reverse Transcriptase From The Human Immunodeficiency Virus, Thomas S. Heard

Chemistry & Biochemistry Theses & Dissertations

The human immunodeficiency virus (HIV) reverse transcriptase (RT) (EC 2.7.7.49) is the central replication enzyme for HIV. In general, the kinetic mechanism for this and all other polymerases involves the ordered binding of two substrates: a primer-template (PT) followed by a deoxyribonucleoside triphosphate (dNTP). Previous investigations prompted this research when it was discovered that the substrate dNTP, in absence of PT, could protect a recombinant c-terminal mutant HIV-1 RT from inhibition by pyridoxal-5'-monophosphate (PLP), an active-site dNTP inhibitor. In contrast, the non-mutant recombinant HIV-1 RT required both substrates for protection from PLP inhibition. This investigation sought to determine if this …

Aerotaxis In Halobacterium Halobium, Jemima Clara Lindbeck Jun 1991

Aerotaxis In Halobacterium Halobium, Jemima Clara Lindbeck

Loma Linda University Electronic Theses, Dissertations & Projects

Aerotaxis, previously studied in eubacteria was investigated in the archaebacterium Halobacterium halobium. A quantitative temporal assay for aerotaxis of H. halobium was developed using computer assisted motion analysis. Aerotaxis was most pronounced in early log phase cultures. Peak aerotaxis was found to coincide with peak respiration consistent with a model in which respiration is required for aerotaxis. In a spatial assay a mutant strain lacking the proton motive force (pmf)-generating pigments, bacteriorhodopsin (bR) and halorhodopsin (hR), exhibited enhanced aerotaxis bands relative to the bR+hR+ parent. The enhanced aerotaxis in the mutant cells was not due solely to …

The Study Of Natural Product(S) From Opsanus Tau (Linnaeus), The Oyster Toadfish, Miki Taira

Chemistry & Biochemistry Theses & Dissertations

Opsanus tau (Linnaeus), the Oyster toadfish, commonly inhabits the Chesapeake Bay. The Oyster toadfish has been categorized as a venomous and/or ichthyotoxic fish; however, chemical studies to substantiate these claims have not been forthcoming.

The oyster toadfish has dorsal and opercular spines which are believed to secrete a venom(s). Based on our findings, the oyster toadfish does not secrete venom(s) from these spines.

The oyster toadfish secretes large amounts of a mucus when it is irritated. 3-Octanone was believed to be the major ichthyotoxic component of the mucus from the oyster toadfish. Based on our investigation, 3-octanone is present only …

Immunochemical Characterization Of The Antigenic And Allergenic Components Of The Plant Pollens Of Paspalum Notatum. Callistemon Citrinis And Melaleuca Leucadendron, Saeed S. Hosseiny Jan 1991

Immunochemical Characterization Of The Antigenic And Allergenic Components Of The Plant Pollens Of Paspalum Notatum. Callistemon Citrinis And Melaleuca Leucadendron, Saeed S. Hosseiny

Retrospective Theses and Dissertations

Antigenic as well as allergenic cross-reactivity was observed among the pollen extracts of Paspalum notatum, bahia (BA); Callistemon citrinis, bottlebrush (BO); and Melaleuca leucadendron, melaleuca (ME). lmmunoblot analysis of SOS-PAGE separated proteins revealed multiple antigenic components in the extracts of all three allergens which cross-reacted with rabbit antisera made to each pollen extract (anti-BA, anti-BO and anti-ME). A unique antigenic component of 15 kD molecular size reacting only to rabbit anti-BA antisera was detected in the bahia extract. Allergenic cross-reactivity among these pollens was strongly suggested by results obtained with skin testing of allergic individuals. A clinical study of 55 …

Blood lead levels of healthy Karachi population were estimated. Mean levels for males, females, soldiers and school children were 34.4, 31.8, 29.9 and 38.2 micrograms/dl respectively. About 93% cases of either sex had elevated lead levels, of whom 30% males and 10% females had levels above the safety limits (40 micrograms/dl). Soldiers living in relatively pollution free area though had levels lower than the rest of the population but 91% had levels over 25 micrograms/dl and only two had acceptable levels. Ninety-two percent children showed levels above 25 micrograms/dl with a large number having levels over 40 micrograms/dl. A very …

Maternal Immunomodulation Of Neonatal Alloantigen Response, Leh Chang Jun 1990

Maternal Immunomodulation Of Neonatal Alloantigen Response, Leh Chang

Loma Linda University Electronic Theses, Dissertations & Projects

Remarkable success has been achieved in the transplantation of allogeneic cardiac grafts into newborn infants at Loma Linda University Medical Center. The superior graft survival rate documented in these patients has not correlated with the degree of immunosuppression rendered, or the selection of genetically matched donors. However, the clinical success has correlated with the age of the recipient at the time of receiving a transplant. Patients receiving an allograft within the first few weeks of life are unique in that they seem to accept the alloantigens of their cardiac graft while responding aggressively to antigens in their environment. These observations …

Characterization Of The Aquatic Environment In Lake Mead Near The Proposed Spring Canyon Pumped-Storage Project, And Assessment Of Potential Aquatic Impacts, Charles R. Liston, Stephen J. Grabowski, Bureau Of Reclamation Jun 1988

Characterization Of The Aquatic Environment In Lake Mead Near The Proposed Spring Canyon Pumped-Storage Project, And Assessment Of Potential Aquatic Impacts, Charles R. Liston, Stephen J. Grabowski, Bureau Of Reclamation

Publications (WR)

A pumped storage system consists of an upper reservoir and lower reservoir separated by an elevation difference. During low demand energy periods such as nights and weekends water is pumped from the lower to the upper reservoir using available energy from conventional steam electric power plants. During high energy demand periods, such as mornings and afternoons of weekdays, upper reservoir water is allowed to drop back down through the same system of water conduits and turbines, generating electricity to conveniently meet abrupt electrical energy requirements. The same water turbines thus act both as pumps and as conventional hydroelectric turbines. Because …

Isolation And Characterization Of A Novel Motility Mutant In Salmonella Typhimurium, David Matthew Chacko Sep 1985

Isolation And Characterization Of A Novel Motility Mutant In Salmonella Typhimurium, David Matthew Chacko

Loma Linda University Electronic Theses, Dissertations & Projects

A new mutant, BT40, isolated from mutagenized cultures of S. typhimurium was deficient in chemotaxis (Che- phenotype) in tryptone semisoft agar, but had random motility with a tumbling frequency (37%) similar to that of the parental strain, ST23 (39%). In spatial assays accumulation of BT40 cells in capillary tubes containing attractants was decreased by 33%-59% compared to ST23, but BT40 had a normal response to attractants in temporal assays. The methylation level of the chemotactic proteins was decreased in BT40. At room temperature, the mean swimming speed was 11 μm/s for BT40 and 20 μm/s for ST23; more BT40 …

Response Of A Nitrifying Biofilm To Copper, Denise Moldroski Mott Jan 1985

Response Of A Nitrifying Biofilm To Copper, Denise Moldroski Mott

Masters Theses

In pure cultures of Nitrosomonas, copper is known to cause a stimulation of nitrification at low levels (≤ 0.03 mg Cu+2/1) and an inhibition affect at higher levels (≥ 0.05 mg Cu+2/1). Nitrifying activated sludges are much more tolerant of copper showing only 75% inhibition at 150mg Cu+2/1. The purpose of this paper was to determine the copper sensitivity of a nitrifying biofilm grown in a continuous flow trickling filter system.

A continuous flow trickling filter system was built using limestone chips for the biofilm substrate. The biofilm was established from secondary effluent. …

A Comparison Of The Acidic Extracellular Polysaccharides From Rhizobium Trifolii Mutants Affected In Trifolium Repens Root Hair Curling, Brian Andrew Hanley Jan 1985

A Comparison Of The Acidic Extracellular Polysaccharides From Rhizobium Trifolii Mutants Affected In Trifolium Repens Root Hair Curling, Brian Andrew Hanley

Masters Theses

The compositions and sugar linkages of the acidic extracellular polysaccharide (EPS) from several Rhizobium trifolii mutants were compared. These mutants were derived from a wild type Rhizobium trifolii, ANU843, and are unable to cause normal root hair curling, Hac-, or nodulate clover, Nod-. Three of the mutants contain a Tn5 insertion in a 5.3 kb BglII fragment of the symbiotic plasmid (pSym). A fourth mutant is missing the pSym, which includes much of the 5.3 kb BglII fragment, both of these deletions result in Hac-Nod- phenotypes. When the 5.3 kb BglII fragment is cloned (pRT150) and reinserted into the …

Non-Parental-Type Recombinants In Crosses Between Different Restriction-Modification Systems In Salmonella, Mary Ball May 1978

Non-Parental-Type Recombinants In Crosses Between Different Restriction-Modification Systems In Salmonella, Mary Ball

Loma Linda University Electronic Theses, Dissertations & Projects

Many new restriction-modification (R-M) system which are closely linked to serB and allelic to the hsdSB system of S. typimurium occur in different Salmonella serotypes. In PI transduction studies with these systems, recombinants were isolated which fell into one of three classes with reference to their specific restriction phenotype. Recombinants that restricted like the donor, recombinants that restricted like the recipient and recombinants that had lost the ability to restrict. This latter group having lost the recipient SB restriction without gaining the specific donor restriction and here called "zero recombinants,” were of special interest and were investigated.

Zero recombinants …

An Immunochemical Study Of The Combining Sites Of The Second Lectin Isolated From Bandeiraea Simplicifolia (Bs Ii), Charles Wood, E. A. Kabat, S. Ebisu, Irwin J. Goldstein Jan 1978

An Immunochemical Study Of The Combining Sites Of The Second Lectin Isolated From Bandeiraea Simplicifolia (Bs Ii), Charles Wood, E. A. Kabat, S. Ebisu, Irwin J. Goldstein

Nebraska Center for Virology: Faculty Publications

Two lectins with different binding specificities have been isolated from extracts of seeds of Bandeiraea simplicifolia. The first, Bandeiraea lectin I [11] was specific for terminal non-reducing αDGalactosyl residues. It reacted with B substances from human ovarian cysts and with several galactomannans to form precipitin lines in agar gels. Polysaccharides with terminal αDGalactosyl residues, such as larch galactan, did not react. The lectin agglutinated B erythrocytes strongly but also reacted to a lower titre with A1 and very weakly with A2 erythrocytes [15, 28] indicating that terminal non-reducing αDGalNAc [24] can …

A Diurnal Zooplankton Migration Study In Lake Mead, Robert D. Staker Jan 1974

A Diurnal Zooplankton Migration Study In Lake Mead, Robert D. Staker

Publications (WR)

The diurnal vertical movement of zooplankton was first recorded in freshwater lakes by Weismann (1877) in Lake Constance (Bodensee), although Cuvier was credited with observing the migration of Daphniae in 1817 (Gushing, 1955). The migration is best observed in deep oligotrophic lakes and migrations of 50 m per day are described (Worthington, 1931). In addition, two species of chaetognaths are reported to migrate 400 m a day in marine waters off of Lisbon (Waterman and Berry, 1967) while Birge (1895) found little evidence of the phenomenon at all in Lake Mendota, Wisconsin. Most zooplankton that migrate rise at night and …
