A key idea in de novo modeling of a medium-resolution density image obtained from cryo-electron microscopy is to compute the optimal mapping between the secondary structure traces observed in the density image and those predicted on the protein sequence. When secondary structures are not determined precisely, either from the image or from the amino acid sequence of the protein, the computational problem becomes more complex. We present an efficient method that addresses the secondary structure placement problem in presence of multiple secondary structure predictions and computes the optimal mapping. Alternative splicing occurs throughout the human genome, leading to multiple proteins from a single gene. The resulting proteins can be nearly identical or vastly different in how they function. Alternative splicing within the perilipin family has been observed in perilipins 1 and 3, giving rise to proteins with varying functions. The alternansucrase (ALT, EC 2.4.1.140) catalyses transferring of glucose from sucrose to produce a polymer with α-1,6 and α- 1,3 linkages, so-called "alternan". The ALT-encoding gene from Leuconostoc citreum ABK-1 (WTalt) was successfully cloned and expressed in Escherichia coli BL21 (DE3). The two versions of truncated SH3-like domains, ∆3SHALT and ∆7SHALT, were successfully constructed. The WTALT, ∆3SHALT and ∆7SHALT possessed optimum temperature of 40 °C, while optimum pH of WTALT was at 5.0 but ∆3SHALT's and ∆7SHALT's were shifted to pH 4.0. Alpha-glucosidase [EC 3.2.1.20; α-D-glucoside glucohydrolase] is an exohydrolase which catalyzes non-reducing end of substrates to release D-glucose. In addition, alpha-glucosidase also displays a transferase activity, which bring about the formation of α-glucosylated compounds such as soluble starch and glycogen. Fisetin is a flavonoid which was widely exerted in pharmacological industries. However, fisetin has low water solubility and unstable in aqueous solution. So the uses of fisetin have been limited. The purpose of this study was to increase water solubility of fisetin by transglycosylation reaction using cyclodextrin glycosyltransferase (CGTase) from Paenibacillus sp. RB01. การวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมและอินเทอแรกโตมเพื่อค้นหาสารบ่งชี้สำหรับโรคออทิซึมสเปกตรัม, ฉัตรระวี พิชิตพันธ์พงศ์

ในปัจจุบัน ออทิซึมสเปกตรัม (Autism spectrum disorder, ASD) ยังคงเป็นโรคที่ยังคงไม่ทราบกลไกระดับโมเลกุล แน่ชัด ยีนและโปรตีนที่เคยมีการรายงานการแสดงออกผิดปกติไปในผู้ป่วยออทิซึมสเปกตรัมมีความสามารถในการทำซ้ำ (Reproducibility) ต่ำหรืออาจไม่มีเลย ซึ่งน่าจะมาจากความหลากหลายของอาการแสดงของผู้ป่วย จากการศึกษาล่าสุด แสดงให้เห็นว่าการจัดกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกสามารถช่วยในการระบุยีนและกลไกที่ผิดปกติไปในกลุ่มย่อยของผู้ป่วย อย่างไรก็ตาม การแบ่งกลุ่มผู้ป่วยเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกใน ออทิซึมสเปกตรัมยังไม่มีการใช้กันอย่างแพร่หลายในการค้นหาโปรตีน Biomarker ที่เกี่ยวข้องกับออทิซึมสเปกตรัม ในการศึกษาครั้งนี้ ผู้วิจัยทำการวิเคราะห์โดยการแบ่งกลุ่มผู้ป่วยจำนวน 85 รายออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R และทำการวิเคราะห์รูปแบบการแสดงออกของยีนในออทิซึมสเปกตรัมในแต่ละกลุ่มย่อยเปรียบเทียบกับกลุ่มควบคุมคนปกติที่อายุใกล้เคียงกันและเพศเดียวกันกับผู้ป่วย (Sex/age-matched control) ซึ่งสามารถระบุยีนที่มีการแสดงออกผิดปกติ (Differentially expressed genes, DEGs) ในออทิซึมสเปกตรัม นอกจากนี้ ผู้วิจัยได้ทำการวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมจากเซลล์ไลน์เม็ดเลือดขาว (Lymphoblastoid cell lines,LCLs) ด้วยเทคนิค Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) ร่วมกับ Nano-liquid chromatography-tandem mass spectrometry (Nano-LC-MS/MS) รายชื่อของโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญที่ได้จากการวิเคราะห์ถูกนำมาเปรียบเทียบกับ DEGs และผลการวิเคราะห์ Meta-analysis ของโปรตีนและโปรตีโอมในการศึกษาก่อนหน้า หน้าที่ทางพยาธิสภาพที่มีความเกี่ยวข้องกับโปรตีนถูกวิเคราะห์โดยโปรแกรม Ingenuity Pathway Analysis (IPA) และได้ทำการคัดเลือกโปรตีนที่น่าสนใจมาทำการตรวจสอบโดยใช้ Western blot analysis จากการศึกษาครั้งนี้พบว่าผลการวิเคราะห์การแบ่งกลุ่มผู้ป่วยออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R สามารถแบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อย ซึ่งพบว่าหนึ่งในกลุ่มนั้น เป็นกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง จากผลวิเคราะห์รูปแบบ ทรานสคริปโตมของเซลล์ไลน์เม็ดเลือดขาว พบว่ามีจุดโปรตีนจำนวน 82 จุดที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง และได้ทำการคัดเลือก 18 จุดโปรตีนจากทั้งหมดมาวิเคราะห์ชนิดของโปรตีนโดยเทคนิค Nano-LC-MS/MS ซึ่งพบว่ามีความเกี่ยวข้องกับการทำงานของระบบประสาทและกระบวนการอักเสบ จากการศึกษานี้พบว่าจากโปรตีนทั้งหมด การแสดงออกของ Isocitrate dehydrogenase (IDH2) และ Diazepam binding inhibitor (DBI) ลดลงในออทิซึมสเปกตรัม ซึ่งโปรตีนเหล่านี้ยังพบการแสดงออกผิดปกติในการวิเคราะห์ Meta-analysis ของทรานสคริปโตมและโปรตีโอมร่วมด้วย การแบ่งกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยทางคลินิกตามด้วยการวิเคราะห์ทรานสคริปโตมและโปรตีโอมแบบบูรณาการร่วมกัน ทำให้การศึกษานี้สามารถระบุโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มย่อยของออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษาอย่างรุนแรง ซึ่งโปรตีนเหล่านี้อาจทำหน้าที่เป็นตัวแทนในการช่วยระบุกลไกทางโมเลกุลของออทิซึมสเปกตรัมในอนาคต ตลอดจน อาจพัฒนาไปเป็น …

ออทิซึมสเปกตรัมเป็นความผิดปกติที่เกี่ยวกับการพัฒนาของระบบประสาทและสมองที่มีความชุก 1 ใน 68 คนของเด็กในสหรัฐอเมริกา โดยจะมีระดับความรุนแรงและอาการแสดงทางคลินิกแตกต่างกันออกไปในแต่ละบุคคล ออทิซึมสเปกตรัมจัดเป็นโรคที่ไม่ทราบสาเหตุการเกิดที่แน่ชัด แต่เป็นที่เชื่อกันว่าสาเหตุของการเกิดโรคนั้นอาศัยปัจจัยร่วมหลายปัจจัย ได้แก่ ทั้งปัจจัยทางพันธุกรรมและปัจจัยทางสิ่งแวดล้อม การวินิจฉัยออทิซึมสเปกตรัมในปัจจุบัน ใช้วิธีอิงจากการสังเกตพฤติกรรมผู้ป่วย วินิจฉัยโรคและความรุนแรงตามลักษณะอาการทางคลินิกเท่านั้น ยังไม่มีสารบ่งชี้ทางห้องปฏิบัติการทางการแพทย์ที่สามารถใช้ในการวินิจฉัย หรือติดตามความรุนแรงของโรคอย่างจำเพาะ โดยผลการศึกษาก่อนหน้านี้ แสดงให้เห็นว่าผู้ป่วยโรคออทิซึมสเปกตรัมมีความผิดปกติในระดับอณูชีวโมเลกุลมากมายทั้งในสมองและเลือด ซึ่งมีหลายประการที่สอดคล้องกับพยาธิสภาพระดับชีวโมเลกุลที่เกิดขึ้นในสมอง อย่างไรก็ตาม ยังไม่มียีนหรือโปรตีนชนิดใดที่สามารถใช้ในการอธิบายโรคออทิซึมสเปกตรัมได้ทั้งหมดในปัจจุบัน แสดงให้เห็นว่าอาจมีกลไกอื่น ๆ ที่สามารถทำให้เกิดพยาธิสภาพได้ ยิ่งไปกว่านั้น แม้โรคออทิซึมสเปกตรัมจะเป็นโรคที่มีอัตราการถ่ายทอดทางพันธุกรรมสูง แต่มีฝาแฝดร่วมไข่จำนวนไม่น้อยที่คนหนึ่งเป็นโรคแต่อีกคนไม่เป็น หรือมีระดับความรุนแรงของโรคไม่เท่ากัน เนื่องจากฝาแฝดร่วมไข่มีลำดับเบสของดีเอ็นเอที่เหมือนกันทุกประการเมื่อแรกเกิด แสดงให้เห็นว่า กลไกเหนือพันธุกรรมหรือเอพิเจเนติกส์อาจเป็นสาเหตุหรือมีความเกี่ยวข้องกับการเกิดโรคออทิซึมสเปกตรัม รีโทรทรานสโพซอนไลน์วัน (retrotransposon LINE-1) เป็นส่วนของจีโนมที่สามารถเคลื่อนที่ได้ และมีอยู่ถึงประมาณร้อยละ 20 และในจีโนมมนุษย์ อย่างไรก็ตามบทบาทของ LINE-1 ในผู้ที่เป็นออทิซึมสเปกตรัมยังคงไม่ทราบแน่ชัด แต่มีรายงานว่าพบการควบคุมที่ผิดปกติของ LINE-1 ในสมองของหนูที่ขาด MECP2 และมีพฤติกรรมคล้ายออทิซึมสเปกตรัม ดังนั้นคณะผู้วิจัยจึงทำการวิเคราะห์ระดับเมธิเลชันของ LINE-1 เพื่อตรวจสอบการเปลี่ยนแปลงระดับเมธิเลชันของ LINE-1 ในเซลล์ของผู้ที่เป็นออทิซึมสเปกตรัม และผลต่อการแสดงออกของยีน รวมไปถึงบทบาทหน้าที่ทางชีวภาพของยีนนั้น ๆ โดยทำการศึกษาในเซลล์ไลน์เม็ดเลือดขาว (lymphoblastoid cell lines; LCLs) ของผู้ที่เป็นออทิซึมสเปกตรัมและคนปกติ นอกจากนี้ยังได้ทำการวิเคราะห์ทางชีวสารสนเทศศาสตร์ร่วมด้วย โดยได้ทำการวิเคราะห์ร่วมระหว่างรายชื่อยีนที่มี LINE-1 แทรกตัวอยู่และมีการแสดงออกที่ผิดปกติในผู้ที่เป็นโรคออทิซึมสเปกตรัม และได้คัดเลือกรายชื่อยีนมาทำการศึกษาบาทหน้าที่ทางชีวภาพ และความสัมพันธ์ของเครือข่ายการควบคุมยีนของยีนเหล่านั้นด้วยโปรแกรม Ingenuity Pathway Analysis (IPA) ซึ่งพบว่ายีนเหล่านั้นอาจจะมีความสัมพันธ์กับโรคทางระบบประสาทและสมอง ในส่วนของการวิเคราะห์ระดับเมธิเลชันของ LINE-1 ในดีเอ็นเอซึ่งสกัดมาจาก LCLs ของผู้ที่เป็นโรคออทิซึมสเปกตรัมและคนปกติ และดำเนินการโดยใช้เทคนิคการวิเคราะห์ Combined Bisulfite Restriction Analysis (COBRA) ซึ่งพบว่าใน LCLs ของกลุ่มตัวอย่างที่เป็นออทิซึมสเปกตรัมมีระดับเมธิเลชันของ LINE-1 ที่เปลี่ยนแปลงไปเมื่อเทียบกับคนปกติ และเมื่อทำการวิเคราะห์ระดับเมธิเลชันของ LINE-1 ในแต่ละกลุ่มของ LCLs ที่เป็นออทิซึมสเปกตรัม ซึ่งถูกจัดแบ่งกลุ่มตามหลักเกณฑ์ของ ADI-R เนื่องจากออทิซึมสเปกตรัมเป็นโรคที่มีลักษณะอาการและความรุนแรงที่หลากหลายมาก คณะผู้วิจัยพบว่าแต่ละกลุ่มมีรูปแบบการเปลี่ยนแปลงของระดับเมธิเลชันของ LINE-1 ที่จำเพาะ และในบางกลุ่มไม่พบการเปลี่ยนแปลงของระดับเมธิเลชันของ LINE-1 นอกจากนี้ยังพบว่าการเมธิเลชันของ …

โรคออทิซึมสเปกตรัมเป็นกลุ่มโรคของความผิดปกติของพัฒนาการทางระบบประสาทที่มีสาเหตุมาจากหลายปัจจัย การศึกษาก่อนหน้าพบว่าในเลือดของผู้ป่วยออทิซึมสเปกตรัมจะรูปแบบการแสดงออกของยีนแตกต่างจากคนปกติ และยังพบว่ามีการเปลี่ยนแปลงของปฏิกิริยาเมธิเลชันบนดีเอ็นเอในเลือดและสมองของผู้ป่วยออทิซึมสเปกตรัมซึ่งปฏิกิริยาดีเอ็นเอเมธิเลชันเป็นกลไกที่ควบคุมการแสดงออกของยีน และยังเป็นกลไกที่ตอบสนองต่อสิ่งแวดล้อมที่เปลี่ยนแปลงไป ดีเอ็นเอเคลื่อนที่ Alu หรือ Alu elements เป็นดีเอ็นเอส่วนที่มีจำนวนซ้ำ ๆ ในจีโนม และสามารถเหนี่ยวนำการแสดงออกของยีนได้ผ่านกลไกเมธิเลชันของดีเอ็นเอ ซึ่งเป็นกลไกหนึ่งที่ควบคุมอัตราการเพิ่มจำนวนตนเองในจีโนม โดย Alu elements มีสัดส่วนการเกิดปฏิกิริยาเมธิเลชันสูงถึงร้อยละ 23 ของปฏิกิริยาเมธิเลชันที่เกิดขึ้นทั้งหมดบนจีโนมของมนุษย์ อย่างไรก็ตามยังไม่เคยมีผู้ทำการศึกษาว่ากลุ่มยีนที่แสดงออกผิดปกติในผู้ป่วยโรคออทิซึมสเปกตรัมนั้นมีความเกี่ยวข้องกับ Alu elements หรือไม่ ในงานวิจัยนี้ผู้ทำวิจัยได้ทำการค้นพบว่ากลุ่มยีนที่มี Alu elements แทรกตัวอยู่มีระดับแสดงออกของยีนที่ผิดปกติในเลือด หรือเซลล์ไลน์ที่พัฒนามาจากเลือดของผู้ป่วยออทิซึมสเปกตรัมผ่านการวิเคราะห์แบบ meta-analysis และโดยผู้ทำวิจัยพบว่ามียีนจำนวน 423 ยีนที่มีการแสดงออกผิดปกติซึ่งเมื่อนำมาวิเคราะห์หาบทบาทหน้าที่ทางชีวภาพของกลุ่มยีนเหล่านี้ด้วยเครื่องมือชีวสารสนเทศ พบว่ามีความเกี่ยวข้องกับกระบวนการพัฒนาการของระบบประสาท และมีความเกี่ยวกับโรคที่มีความผิดปกติคล้ายกับโรคออทิซึมสเปกตรัม และนอกจากนี้ผลการศึกษาในเซลล์โมเดลจากผู้ป่วยออทิซึมสเปกตรัมที่ใช้ในการศึกษาครั้งนี้ พบว่ามีการเปลี่ยนแปลงของระดับและรูปแบบการเกิดปฏิกิริยาเมธิเลชันของ Alu elements ในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่ผ่านการจัดจำแนกกลุ่มผู้ป่วยด้วยข้อมูลแบบสอบถามทางพฤติกรรมสำหรับการวินิจฉัยผู้ป่วยออทิซึมสเปกตรัม สอดคล้องกับผลการวิเคราะห์รูปแบบการแสดงออกของยีนในผู้ป่วยกลุ่มที่มีระดับเมธิเลชันผิดปกติจะมีรูปแบบการแสดงออกของยีนที่มี Alu elements แทรกตัวอยู่ผิดปกติด้วย ซึ่งข้อมูลการค้นพบใหม่นี้ได้แสดงให้เห็นความถึงสัมพันธ์ระหว่างระดับปฏิกิริยาเมธิเลชันของ Alu elements กับความผิดปกติของระดับการแสดงออกของยีนในผู้ป่วยออทิซึมสเปกตรัมเมื่อมีการจัดจำแนกผู้ป่วยตามลักษณะอาการพฤติกรรมทางคลินิก ซึ่งอาจใช้อธิบายความผิดปกติทางชีวโมเลกุลที่จำเพาะในผู้ป่วยแต่ละกลุ่มได้ และอาจพัฒนาเป็นตัวบ่งชี้ทางชีวภาพในเลือดของโรคออทิซึมสเปกตรัมในอนาคตได้

กระบวนการสร้างนิวไรท์ เป็นกระบวนการที่สำคัญในการเจริญพัฒนา การซ่อมแซมส่วนที่เสียหาย รวมถึงการสื่อสารระหว่างเซลล์ของเซลล์ประสาท ซึ่งหากกระบวนการการสร้างนิวไรท์เกิดความผิดปกติสามารถก่อให้เกิดโรคความผิดปกติของระบบประสาท เช่น โรคอัลไซเมอร์ โรคพาร์กินสัน และโรคฮันติงตัน เป็นต้น ในการศึกษาวิจัยนี้ศึกษาฤทธิ์การต้านสารอนุมูลอิสระ และการกระตุ้นการสร้างนิวไรท์จากสารสกัดเมล็ดของหมามุ่ยไทย และหมามุ่ยอินเดียในเซลล์ประสาทเพาะเลี้ยงชนิด Neuro2a การศึกษาคุณสมบัติในการต้านสารอนุมูลอิสระโดยวิธี DPPH และ ABTS assays นอกจากนั้นทำการศึกษาอัตราการรอดชีวิตของเซลล์ประสาทโดยวิธี MTT พร้อมกับศึกษาการแสดงออกของยีนที่เกี่ยวข้องกับการกระตุ้นการงอกของนิวไรท์ ประกอบด้วย Ten-4 และ GAP-43 โดยวิธี Real-time PCR ซึ่งจากการทดสอบพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียด้วยตัวทำละลายเอทานอลมีความสามารถในการต้านสารอนุมูลอิสระ และพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียไม่เป็นพิษต่อเซลล์ประสาท จากนั้นทำการศึกษาฤทธิ์ และกลไกของสารสกัดจากสมุนไพรต่อการกระตุ้นการงอกของนิวไรท์ โดยพบว่าสารสกัดจากเมล็ดหมามุ่ยไทยด้วยตัวทำละลายเอทานอล สามารถกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 และ GAP-43 ในขณะที่สารสกัดจากเมล็ดของหมามุ่ยอินเดียด้วยตัวทำละลายเฮกเซนกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 อีกทั้งศึกษาผลการยับยั้งการแสดงออกของยีน Ten-4 ต่อการกระตุ้นการสร้างนิวไรท์ของสารสกัดสมุนไพร ซึ่งพบว่ามีการสร้างนิวไรท์ และความยาวของนิวไรท์ลดลง จากการศึกษาสามารถบ่งชี้ได้ว่าสารสกัดจากเมล็ดหมามุ่ยไทย และหมามุ่ยอินเดียสามารถปกป้องเซลล์ประสาทจากสารอนุมูลอิสระได้ และสามารถกระตุ้นการงอกของนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน GAP-43 และ Ten-4 ซึ่งเป็นอีกทางเลือกหนึ่งในการพัฒนายาการกระตุ้นการเจริญพัฒนาของเซลล์ประสาทสำหรับป้องกัน หรือรักษาโรคความผิดปกติของระบบประสาท

โรคออทิซึมสเปกตรัม (Autism Spectrum Disorder, ASD) คือ โรคทางพัฒนาการระบบประสาทที่พบในเพศชายมากกว่าเพศหญิงโดยไม่ทราบสาเหตุ สาเหตุการเกิดโรคออทิซึมสเปกตรัมยังไม่เป็นที่ทราบแน่ชัดแต่การได้รับบิสฟีนอลเอขณะตั้งท้องมีความสัมพันธ์ต่อการเกิดโรคออทิซึมสเปกตรัมในลูกหนู อย่างไรก็ตามยังไม่มีการศึกษาแน่ชัดว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกของยีนที่เกี่ยวข้องกับโรคออทิซึมสเปกตรัมหรือไม่ โดยเฉพาะเมื่อวิเคราะห์แบบแยกเพศ ในการศึกษาครั้งนี้ ทางคณะผู้วิจัยจึงทำการศึกษาผลของการได้รับบิสฟีนอลเอในขณะตั้งท้องต่อการแสดงออกของยีนในสมองและความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมรวมไปถึงหน้าที่ของเซลล์ระบบประสาท ในการทำนายว่าการได้รับบิสฟีนอลเอสามารถเปลี่ยนแปลงการแสดงออกของยีนและมีความสัมพันธ์กับโรคออทิซึมสเปกตรัมหรือไม่ ทางผู้วิจัยได้ทำการวิเคราะห์แบบ meta-analysis โดยใช้ข้อมูลการแสดงออกของยีนที่ผิดปกติจากการได้รับบิสฟีนอลเอที่ได้ตีพิมพ์ไปแล้วและใช้การวิเคราะห์แบบ hypergeometric distribution หาความสัมพันธ์ระหว่างกลุ่มยีนดังกล่าวกับรายชื่อยีนจากฐานข้อมูลโรคออทิซึมสเปกตรัมต่าง ๆ สำหรับการศึกษาถึงผลของการได้รับบิสฟีนอลเอขณะตั้งท้องต่อการแสดงออกของยีนที่มีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมนั้น คณะผู้วิจัยได้นำสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลขณะตั้งท้องมาวิเคราะห์ทางทรานสคริปโตมิกส์ด้วยเทคนิค RNA-seq และใช้โปรแกรม Ingenuity Pathway Analysis วิเคราะห์อินเตอร์แอกโตมและบทบาทหน้าที่ทางชีวภาพของกลุ่มยีนที่มีการแสดงออกผิดปกติ ยีนที่มีความสัมพันธ์กับโรคออทิซึมสเปกตรัมถูกคัดเลือกเพื่อยืนยันด้วยเทคนิค quantitative RT-PCR นอกจากนี้เซลล์ระบบประสาทที่แยกได้จากลูกหนูถูกนำมาเพาะเลี้ยงเพื่อศึกษาความยาวของสายใยประสาท การศึกษาพบว่า จากการวิเคราะห์ทาง meta-analysis ยีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความสัมพันธ์อย่างมีนัยสำคัญกับหน้าที่และโรคออทิซึมสเปกตรัม การวิเคราะห์ RNA-seq ในสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลเอขณะตั้งท้องพบว่ายีนที่มีการแสดงออกผิดปกติมีความสัมพันธ์กับโรคออทิซึมสเปกตรัม เป็นที่น่าสนใจอย่างยิ่งว่ากลุ่มยีนที่เคยมีการรายงานว่ามีความผิดปกติในโรคออทิซึมสเปกตรัม ได้แก่ Auts2 Foxp2 และ Smarcc2 แสดงออกผิดปกติอย่างจำเพาะต่อเพศ การวิเคราะห์เชิงอินเตอร์แอกโตมพบว่ายีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความเกี่ยวข้องอย่างมีนัยสำคัญกับบทบาทหน้าที่ทางระบบประสาทที่เคยมีรายงานในผู้ป่วยออทิซึมสเปกตรัม การวิเคราะห์ความยาวของสายใยประสาทพบว่าการได้รับบิสฟีนอลเอส่งเสริมความยาวของสายใยประสาทและการแตกแขนงของสายใยประสาทแต่ลดขนาดของตัวเซลล์อย่างมีนัยสำคัญทางสถิติ จากการศึกษาครั้งนี้พบว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกที่ผิดปกติของยีนในสมองส่วนฮิปโปแคมปัสและกลุ่มยีนดังกล่าวมีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมและแสดงออกจำเพาะในแต่ละเพศประกอบกับการได้รับบิสฟีนอลเอขณะตั้งท้องส่งเสริมความยาวของสายใยประสาท การศึกษานี้แสดงให้เห็นว่าการได้รับบิสฟีนอลเอขณะตั้งครรภ์อาจเป็นสาเหตุหรือเพิ่มความเสี่ยงต่อโรคออทิซึมสเปกตรัมและอาจจะเป็นหลักฐานที่อธิบายถึงความชุกของโรคนี้ในเพศชายที่มากกว่าเพศหญิงซึ่งควรศึกษาเพิ่มเติมในอนาคต

SMYD3 plays a key role in cancer cell viability, adhesion, migration and invasion. SMYD3 promotes formation of inducible regulatory T cells and is involved in reducing autoimmunity. However, the nearly “closed” substrate-binding site and poor in vitro H3K4 methyltransferase activity have obscured further understanding of this oncogenically related protein. Here we reveal that SMYD3 can adopt an “open” conformation using molecular dynamics simulation and small-angle X-ray scattering. This ligand-binding-capable open state is related to the crystal structure-like closed state by a striking clamshell-like inter-lobe dynamics. Gap junctions are intercellular channels that permit the free passage of ions, small metabolites, and signaling molecules between neighboring cells. In the diseased human heart, altered ventricular gap junction organization and connexin expression (i.e., remodeling) are key contributors to rhythm disturbances and contractile dysfunction. Connexin43 (Cx43) is the dominant gap junction protein isoform in the ventricle which is under tight regulation by serine/tyrosine phosphorylation. Phosphorylation and dephosphorylation regulate many aspects of Cx43 function including trafficking, assembly and disassembly, electrical and metabolic coupling at the plaque, as well as to modulate the interaction with other proteins.

Despite multiple DNA repair pathways, DNA lesions can escape repair and compromise normal chromosomal replication, leading to genome instability. Cells utilize specialized low-fidelity Translesion Synthesis (TLS) DNA polymerases to bypass lesions and rescue arrested replication forks. TLS is a highly conserved two-step process that involves insertion of a nucleotide opposite a lesion and extension of the resulting aberrant primer terminus. Peatlands contain one-third of soil carbon (C), mostly buried in deep, saturated anoxic zones (catotelm). The response of catotelm C to climate forcing is uncertain, because prior experiments have focused on surface warming. We show that deep peat heating of a 2 m-thick peat column results in an exponential increase in CH4 emissions. However, this response is due solely to surface processes and not degradation of catotelm peat. Incubations show that only the top 20–30 cm of peat from experimental plots have higher CH4 production rates at elevated temperatures. A new class of N-linked protein glycosylation – arginine rhamnosylation – has recently been discovered as a critical modification for the function of bacterial elongation factor P (EF-P). Herein, we describe the synthesis of suitably protected α- and β-rhamnosylated arginine amino acid “cassettes” that can be directly installed into rhamnosylated peptides. Preparation of a proteolytic fragment of Pseudomonas aeruginosa EF-P bearing both α- and β-rhamnosylated arginine enabled the unequivocal determination of the native glycosidic linkage to be α through 2D NMR and nano-UHPLC-tandem mass spectrometry studies.

Plasticization of PVC was carried out by covalently linking phthalate derivatives via copper-free (thermal) or copper catalyzed azide-alkyne cycloadditions. Di(2-ethylhexyl) phthalate derivatives (DEHP-ether and DEHP-ester) were synthesized and appended to PVC at two different densities. The glass transition temperatures of the modified PVC decreased with increasing content of plasticizer. PVC-DEHP-ether gave lower glass transition temperatures than PVC-DEHP-ester, reflecting the enhanced flexibility of the ether versus ester linker.

Four peptides were synthesized using solid phase peptide synthesis. The two target peptides synthesized were based off peptide 18-4 and its negative analog. Each were synthesized once manually, and once using an automatic peptide synthesizer. Peptide 18-4 has a high affinity for breast cancer cells, allowing it to be used to detect circulating tumor cells (CTCs) in blood (Kaur et. al, 2015). Manual solid phase peptide synthesis is performed by anchoring the first amino acid to Wang resin and coupling each Fmoc protected amino acid individually until the target sequence is achieved. Twenty years ago, a major curriculum revision at a large, comprehensive university in the Western United States led to the implementation of an integrated lecture/laboratory (studio) experience for our engineering students taking general chemistry. Based on these twenty years of experience, construction of four purpose-built studio classrooms to house the majority of the remaining general chemistry courses was completed in 2013. A detailed study of the effects of the entire ecology of the studio experience on student success was initiated at that time. Data from content knowledge pre- and post-tests, learning attitudes surveys, and student course evaluations show positive effects …
