Evaluation Of The Virulence Potential Of Avian Pathogenic Escherichia Coli Isolated From Broiler Breeders With Colibacillosis In Mississippi, Jiddu Joseph

Theses and Dissertations

Avian pathogenic Escherichia coli (APEC) is a bacterium that is responsible for colibacillosis in birds. However, information about broiler breeder APEC isolates is limited, but the data is critical due to the transfer of this bacteria down the production pyramid to progenies resulting in high mortality. Therefore, we evaluated the phenotypic virulence characteristics of 28 isolates using embryo lethality and day-old chick challenge assays. Also, the in vitro adhesion and invasion potential of selected nine isolates were identified. Results showed more than 1/3rd of the isolates were highly virulent and the virulence increased as the number of virulence-associated genes …

Theses and Dissertations

Campylobacteriosis is a leading foodborne illness worldwide, primarily caused by Campylobacter jejuni (C. jejuni) which is associated with poultry consumption. The emergence of antibiotic resistance has emphasized the need for alternative strategies to control C. jejuni colonization in poultry. To assess the prevalence of C. jejuni in poultry, 270 cloacal swab samples were collected from broilers raised under No-Antibiotics Ever system. Among these samples, 16.3% were identified as C. jejuni positive. Notably, these isolates exhibited a diverse range of virulence factors and antimicrobial resistance genes, with 61.36% of isolates showing hyper-motile and 20.45% demonstrating multidrug resistance. Following isolation, whole genome …

Effects Of Sampling Methodologies On Mycoplasma Gallisepticum Tissue Populations In Commercial Layer Pullets, Sethulakshmi Kattupparayil Sasidharan May 2023

Effects Of Sampling Methodologies On Mycoplasma Gallisepticum Tissue Populations In Commercial Layer Pullets, Sethulakshmi Kattupparayil Sasidharan

Theses and Dissertations

Mycoplasma gallisepticum (MG) can cause chronic respiratory disease in chickens and infectious sinusitis in turkeys. Impacts of MG infections can include increased morbidity and mortality, decreased egg production, hatchability and feed efficiency. Biosecurity and bio-surveillance remain the primary means of deterrence as the current means of control are not wholly effective. Further applications towards the characterization of MG-related disease control requires proper understanding of the characteristics of MG infections with accurate and efficient quantification of in vivo MG populations. To this end, study was conducted to determine the in vivo MG populations in infected pullets and to determine impact of …

Molecular And Phenotypic Characterization Of Escherichia Coli Isolated From Broiler Chicken Flocks In Mississippi, Priyanka Devkota

Theses and Dissertations

Avian pathogenic Escherichia coli (APEC) is the causative agent of colibacillosis. APEC causes significant economic losses to the poultry industry. In this study, 66 E. coli isolates were collected from broiler flocks across Mississippi and categorized as clinical and non-clinical isolates. Genotypic and phenotypic characterization of virulence factors and antimicrobial resistance profiles of these E. coli were examined. The data disclosed a higher prevalence of virulence genes in clinical isolates than in non-clinical isolates. High differences on the genotypic and phenotypic antimicrobial resistance among clinical and non-clinical isolates were observed. Whole genome sequences of avian E. coli elaborated a diverse …

Development Of A Novel Loop-Mediated Isothermal Amplification (Lamp) Assay For The Rapid Detection Of Clostridium Perfringens, Deepa Chaudhary Dec 2022

Development Of A Novel Loop-Mediated Isothermal Amplification (Lamp) Assay For The Rapid Detection Of Clostridium Perfringens, Deepa Chaudhary

Theses and Dissertations

Clostridium perfringens (C. perfringens) causes diseases such as necrotic enteritis in poultry. Current methods for early detection of C. perfringens, including culture, and molecular techniques (PCR and qPCR), have logistical limitations in detecting pathogens on-site because they require specialized equipment and long sample preparation time. This study describes developing a highly sensitive on-site LAMP assay combined with a simple DNA extraction method to detect C. perfringens. Results showed that the LAMP assay was highly specific and sensitive with the optimal reaction conditions: 63°C for 30 mins with primer concentration of 1.2 mM FIP/BIP and 0.2 mM F3/B3. The HotSHOT method …

The Effect Of Sprinkler Cooling On Water Conservation, House Environment, And Broiler Performance, Jonathan W. Moon May 2022

The Effect Of Sprinkler Cooling On Water Conservation, House Environment, And Broiler Performance, Jonathan W. Moon

Theses and Dissertations

Maintaining performance and mitigating heat stress of the modern broiler is a challenging task during hot weather conditions. Increased demand of high quality reasonably priced animal protein and predictions of future limited water availability make sustainability and water conservation a worthwhile goal for the poultry industry. The most used form for cooling broilers after wind speed is evaporative cool cell (CC) systems. Though highly effective at cooling the incoming air, they substantially increase the relative humidity inside the house, which hinders the bird’s ability to cool itself through evaporative respiration. Cool cells also utilize mass amounts of water. The objective …
