In vivo analysis of equine feed digestibility has been the gold standard since the late 1800's, although it can be time consuming, costly, and labor intensive. In vitro digestibility analysis may be more economical and beneficial to both feed manufacturers and consumers. The objectives of this study were to: (1) determine an optimal cryoprotectant of mice embryos; and (2) determine whether morula or blastocyst stage is ideal for vitrification in both medias. One experiment was performed using two different medias for vitrification with open pulled straws (OPS) with morulae and blastocysts. In the first protocol, we called V, embryos were exposed to 10% ethylene glycol (EG) for 5 minutes, then 40% ethylene glycol and 0.6 M galactose for about 30 seconds, loaded into an OPS, and plunged into liquid nitrogen. This senior project report discusses the design, construction, and evaluation of a portable artificial insemination chute for the use of the Cal Poly Beef Department. The design was based on the suggested modifications from operators of several other chutes currently in use.
The main areas considered for the design were; Ergonomics, cattle safety, factors effecting maximum breeding conception rates, and Portability.
The breeding chute proved superior over traditional chutes in a variety of ways. Improvement occurred in the areas of consideration as follows, user operation was simplified, animal stress was decreased, and maximum breeding efficiency was obtained.

Leydig cells produce testosterone in response to luteinizing hormone (LH) via the cyclic adenosine monophosphate (cAMP)/protein kinase A pathway. Additionally, these cells are responsible for producing insulin-like peptide 3 (INSL3), a peptide hormone that is essential for testicular descent. The insl3 promoter in Leydig cells can be activated by cAMP through the transcription factor Nur77, which has also been shown to regulate the promoters of the steroidogenic enzymes, cyp17 and 3b-hsd. While the mechanism of LH action on testosterone production is well characterized, the effect of LH on insl3 abundance has yet to be shown directly. The MA-10 …

Cytochrome P450 (CYP) enzymes activate or eliminate medications and other xenobiotics, and Caffeine has been used as an indicator of activity for some CYPs. The natural genetic variation in the human population for caffeine metabolizing CYPs does not yet have an established animal model for comparison, but the natural variation in inbred mouse strains presents an attractive starting point. The project gave four different strains of mice a 40mg/kg dose of caffeine and sampled blood and liver tissue after 30 minutes. Samples were analyzed using High Performance Liquid Chromatography (HPLC), reverse transcription PCR (rtPCR), and quantitative PCR (qPCR). The DBA/2J …

This rangeland condition assessment was conducted as part of the Cal Poly Rangeland Condition Assessment Project. Five pastures comprising 31.7 acres at the Cal Poly sheep unit were assessed to collect baseline ecological information. Methods used were Line-point Intercept, Belt Transect, Residual Dry Matter, and the rangeland health indicators recommended by Pyke et al., 2002. Additionally, a preliminary species list was collected. The rangeland health assessment method found all pastures to have no to slight departure from the local reference area when evaluated for soil and site stability, hydrologic function, and biotic integrity.

This study was designed to compare the benefits and costs of two different synchronized artificial insemination programs on two groups of beef first calf heifers. A financial budget was developed that would include cost of synchronized insemination per animal and the added value of a bred heifer compared to an open one.
The two synchronized programs were called controlled internal drug release (CIDR) and Ovsynch. They used a specific timeline and drugs to complete the synchronization process. The facility will be an upgrade to the facility currently used at the ranch in Pine Valley, California. The design parameters for the facility were narrowed down to the following: 1. Must reduce stress to animals during handling. 2. Must be cost effective to save labor and maintenance. 3. Must show improvement from the current set- up. The mammary alveolar cell-T (MAC-T) cell line is able to uniformly differentiate and secrete casein proteins in response to dexamethasone, insulin and prolactin and is extensively used to study bovine mammary epithelial cell function. Growth hormone (GH) has been shown to increase milk protein synthesis both in vivo and in mammary cell models, and induce cytoskeletal rearrangement in 3T3 fibroblast cell line and a Chinese hamster ovary (CHO) cell line. Few studies have focused on identifying the mechanisms involved in differentiated MAC-T cells’ response to GH. We tested the hypothesis that MAC-T cells would respond directly to GH and that …

I will be assisting in the placement of a product in the field and evaluating response. It is the first step in evaluating if this product is an appropriate and marketable product.
