Malnutritional stress during gestation is a well-established driver of metabolic disfunction in offspring. Extended exposure to malnutrition requires metabolic plasticity as the animal shifts toward a catabolic state. In this paper we demonstrate the influence of malnutrition throughout gestation on uterine artery hemodynamics and the metabolism of the dam and neonate. We hypothesized that gestational malnutrition reduces blood flow of the maternal uterine artery and regulates the metabolic profile of the dam and offspring. Further, the combination of these factors consequently influences the concentration of metabolites in the cerebrospinal fluid of the neonate at birth. To test our hypotheses, pregnant …

Fibroblast growth factor 21 (FGF21) has been identified in multiple mammalian species as a molecular marker of energy metabolism while also providing negative feedback to the gonads. However, the role of FGF21 in regulating the energetic and reproductive physiology of beef heifers and cows has yet to be characterized. Herein, we investigated the temporal concentrations of FGF21 in female beef cattle from the prepubertal period to early lactation. Circulating concentrations of FGF21, non-esterified fatty acids, plasma urea nitrogen, glucose, and progesterone were assessed. Prolific use of supplementation strategies, including the utilization of urea, are practiced in beef cattle production systems. Unfortunately, the influence of urea supplementation on metabolics, adipose tissue mobilization, and mammary secretions is limited in beef cows. Therefore, the objectives of this experiment were to assess the influence of urea supplementation on metabolic profiles, morphometrics, and mammary secretions. Pregnant, multiparous beef cows were fed individually and assigned to treatment (n = 4/treatment) as Control or Urea Supplementation. Microarray analysis using Affymetrix Bovine GeneChip 1.0 ST Array to determine RNA expression analysis was performed on somatic granulosa cells from two different groups of cows classified based on androstenedione concentration within the follicular fluid (Control vs High A4) of estrogen-active dominant follicles. The normalized linear microarray data was deposited to the NCBI GEO repository (GSE97017 - RNA Expression Data from Bovine Ovarian Granulosa Cells from High or Low Androgen-Content Follicles). Bison are an icon of the American West and an ecologically, commercially, and culturally important species. Despite numbering in the hundreds of thousands today, conservation concerns remain for the species, including the impact on genetic diversity of a severe bottleneck around the turn of the 20th century and genetic introgression from domestic cattle. Pooling individual samples prior to DNA extraction can mitigate the cost of DNA extraction and genotyping; however, these methods need to accurately generate equal representation of individuals within pools. The objective of this study was to determine accuracy of pool construction of blood samples based on white blood cell counts compared to two common DNA quantification methods. Yes-associated protein 1 (YAP1) is a major component of the Hippo signaling pathway. Although the exact extracellular signals that control the Hippo pathway are currently unknown, increasing evidence supports a critical role for the Hippo pathway in embryonic development, regulation of organ size, and carcinogenesis. Granulosa cells (GCs) within the ovarian follicle proliferate and produce steroids and growth factors, which facilitate the growth of follicle and maturation of the oocyte.We hypothesize that YAP1 plays a role in proliferation and estrogen secretion of GCs. After ovulation, somatic cells of the ovarian follicle (theca and granulosa cells) become the small and large luteal cells of the corpus luteum. Aside from known cell type-specific receptors and steroidogenic enzymes, little is known about the differences in the gene expression profiles of these four cell types. Analysis of the RNA present in each bovine cell type using Affymetrix microarrays yielded new cell-specific genetic markers, functional insight into the behavior of each cell type via Gene Ontology Annotations and Ingenuity Pathway Analysis, and evidence of small and large luteal cell lineages using Principle Component Analysis. A study investigated the use of Anti-Mullerian hormone (AMH) and/or follicle counts as a predictor of subsequent superovulatory response and embryo production in 79 beef cows. Before initiation of superovulation, ultrasonography was used to scan the ovaries of each donor cow to record the number of 3 to 5 mm follicles present, and a blood sample was collected for measure of serum AMH. At the time of embryo collection, the ovaries of donor cows were palpated to estimate the number of corpora lutea (CL) present on each ovary. Improving Production Efficiency Of Beef Cow-Calf Operations, Amanda Jo Davis

Three studies were conducted to improve and incorporate reproductive management practices into beef cow-calf production. First study evaluated serial use of Estrotect estrous detection patches as a simple, cost-effective reproductive management tool to identify cyclic animals before breeding, distinguish between females conceiving to AI versus natural service, and determine seasonal pregnancy rate after bull removal. Also determined, was effectiveness of altered timing of GnRH treatment (1 d ± CIDR removal) in a modified 14-d CIDR-Select Synch protocol. When evaluated over a 4-wk period, estrous detection patches correctly (P < 0.01) identified 79% of cyclic and 86% of non-cyclic heifers. Patches were 96 and 98% accurate in identifying heifers and cows pregnant by AI, respectively, and were 76 and 87% accurate in identifying pregnant heifers and cows at the end of the breeding season (P < 0.01). Treatment with GnRH at CIDR removal reduced labor costs and animal handling without compromising estrous response (both ~63.0%) and AI pregnancy rates (~76 and 77%; P > 0.1). Forty-eight Gelbvieh x Angus steers (265 ¡À 40 kg) were utilized to determine the relationships among molecular breeding values (MBV), steer performance, and carcass traits. Body weight (BW), hip height (HH), hip width (HW), exit velocity (EV; rate at which steers exited the squeeze chute and traversed 1.8 m) and body ultrasound measurements of steers were recorded at d 0, 93 and 154 of grazing mixed stockpiled endophyte-infected and -free tall fescue.
